Claims (1)
- 【特許請求の範囲】 1.CD3と、変異されたFcレセプター結合領域を含むヒトFc領域とに結合する抗 原結合領域を含むモノクローナル抗体であって、抗体OKT3に対して減少したT細 胞活性化特性を有し、235位でロイシンからアラニンへの変異を含む、モノクロ ーナル抗体。 2.前記抗体が、OKT3と呼ばれるネズミ抗体の抗原結合領域を含む、請求項1に 記載のモノクローナル抗体。 3.前記抗体が、234位で第2の変異をさらに含む、請求項1に記載のモノクロ ーナル抗体。 4.234位でフェニルアラニンからロイシンへの変異を含む、請求項3に記載の モノクローナル抗体。 5.234位でフェニルアラニンからアラニンへの変異を含む、請求項3に記載の モノクローナル抗体。 6.前記抗原結合領域が、CD4またはCD8にさらに結合する、請求項1に記載のモ ノクローナル抗体。 7.CD3と、変異されたFcレセプター結合領域を含むヒトFc領 域とに結合する抗原結合領域を含むモノクローナル抗体であって、抗体OKT3に対 して減少したT細胞活性化特性を有し、234位でフェニルアラニンからアラニン への変異を含む、モノクローナル抗体。 8.前記抗体が、OKT3と呼ばれるネズミ抗体の抗原結合領域を含む、請求項7に 記載のモノクローナル抗体。 9.前記抗体が、235位で第2の変異をさらに含む、請求項7に記載のモノクロ ーナル抗体。 10.235位でロイシンからグルタミン酸への変異を含む、請求項9に記載のモ ノクローナル抗体。 11.前記抗原結合領域が、CD4またはCD8にさらに結合する、請求項7に記載の モノクローナル抗体。 12.CD3と、変異されたFcレセプター結合領域を含むヒトFc領域とに結合する 抗原結合領域を含むモノクローナル抗体であって、抗体OKT3に対して減少したT 細胞活性化特性を有し、234位での第1の変異と235位での第2の変異とを含む、 モノクローナル抗体。 13.前記抗体が、OKT3と呼ばれるネズミ抗体の抗原結合領 域を含む、請求項12に記載のモノクローナル抗体。 14.前記ネズミ抗原結合領域が、CD4またはCD8にさらに結合する、請求項12 に記載のモノクローナル抗体。 15.前記ヒトFc領域が、IgG1またはIgG4 Fc部分である、請求項1、7、また は12に記載のモノクローナル抗体。 16.前記ヒトFc領域が、IgG1である、請求項15に記載のモノクローナル抗体 。 17.請求項1、7、または12に記載のモノクローナル抗体および生理学的に 受容可能なキャリヤを含む薬学的組成物。 18.請求項1、7、または12に記載のモノクローナル抗体の、免疫応答を引 き金とする移植器官組織の拒絶反応の抑制用薬剤の製造のための使用であって、 該薬剤が、生理学的に受容可能なキャリヤ中に含まれて、移植前、移植中、また は移植後のいずれかに、器官移植患者に投与される、使用。 19.請求項1、7、または12に記載の抗体および生理学的に受容可能なキャ リヤを含む請求項17に記載の薬学的組成物の、免疫応答を引き金とする移植器 官組織の拒絶反応の抑制用薬剤の製造のための使用であって、該薬剤が、移植前 、 移植中、または移植後のいずれかに、器官移植患者に投与され、そして該抗体が 、CD3と呼ばれる第1のT細胞表面タンパク質および同時に第2のT細胞表面タ ンパク質に結合することによって、免疫応答をモジュレートする、使用。 20.前記第2のT細胞表面タンパク質が、CD4およびCD8からなる群から選択さ れる、請求項19に記載の使用。 21.免疫応答を引き金とする移植器官組織の拒絶反応を抑制する方法であって 、移植前、移植中、または移植後のいずれかに、生理学的に受容可能なキャリヤ 中の請求項1に記載のモノクローナル抗体を器官移植患者に投与する工程を含む 、方法。 22.免疫応答を引き金とする移植器官組織の拒絶反応を抑制する方法であって 、移植前、移植中、または移植後のいずれかに、請求項1に記載の抗体および生 理学的に受容可能なキャリヤを含む請求項17に記載の薬学的組成物を器官移植 患者に投与する工程を含み、そして該抗体が、CD3と呼ばれる第1のT細胞表面 タンパク質および同時に第2のT細胞表面タンパク質に結合することによって、 免疫応答をモジュレートする、方法。 23.前記第2のT細胞表面タンパク質が、CD4、およびCD8 から選択される、請求項22に記載の方法。
