Claims (1)
- 【特許請求の範囲】 1.カソード溜め(16又は18)と、アノード溜め(16又は18)と、 電源(24)とを含み、前記溜め(16、18)の各々が体表とイオン伝導関係 に配置されるために適し、前記溜め(16、18)の1つが投与すべき作用剤を 含み、電源(24)が前記溜めと前記体表とに通して所定レベルの電流を供給す るためにアノード溜め及びカソード溜め(16、18)を横切って電圧を印加す ることができる、体表を通して作用剤を投与するための電気駆動式デバイス(1 0)であって、電気駆動式作用剤投与中に、 (a)前記アノード溜めを約4より大きいpHに維持し;及び/又は (b)前記カソード溜めを約4未満のpHに維持する 前記デバイス。 2.アノード溜めpHを約4〜10の範囲内に維持する請求項1記載のデバ イス。 3.前記アノード溜めを緩衝化する請求項1又は2に記載のデバイス。 4.前記緩衝剤がアニオン緩衝剤である請求項3記載のデバイス。 5.前記アニオン緩衝剤がヒスチジン、リシン、アルギニン、アスパラギン 酸、グルタミン酸、システイン、チロシン及びこれらの組合せから成る群から選 択される請求項4記載のデバイス。 6.前記アノード溜めを、 (a)アスパラギン酸、グルタミン酸、クエン酸、コハク酸、リン酸、酢酸、 EDTA、乳酸、ホウ酸、炭酸、一塩基性リン酸ナトリウム、安息香酸、酒石酸 、マレイン酸、フマル酸、硫酸、ギ酸、リンゴ酸、マロン酸、グルタル酸及びア ジピン酸から成る群から選択される緩衝剤; (b)トロメタミン、トリエタノールアミン、イミダゾール、アンモニア、エ タノールアミン、ジエタノールアミン、ヒスチジン、リシン及びアルギニンから 成る群から選択される緩衝剤; (c)塩化カルシウム2水和物、トリエタノールアミン塩酸塩、塩化マグネシ ウム7水和物、ジエタノールアミン塩酸塩、塩化アンモニウム、エタノールアミ ン塩酸塩、塩化カリウム及び塩化ナトリウムから成る群から選択される緩衝剤; (d)2−(N−モルホリノ)エタンスルホン酸、1,4−ピペラジン−ビス −(エタンスルホン酸)、N−2−アセトアミド イミノ二酢酸、N−2(2− アセトアミド)−2−アミノエタンスルホン酸、N,N−ビス(2−ヒドロキシ エチル)−2−アミノエタンスルホン酸、2−(N−モルホリノ)プロパンスル ホン酸、N−トリス(ヒドロキシメチル)メチル−2−アミノエタンスルホン酸 、N−2−ヒドロキシエチルピペラジン−N’−2−エタンスルホン酸、4−( 2−ヒドロキシエチル)−1−ピペラジンプロパンスルホン酸、N−トリス(ヒ ドロキシメチル)メチル−2−アミノプロパンスルホン酸、2−シクロヘキシル アミノ−1−エタンスルホン酸、3−シクロヘキシルアミノ−1−プロパンスル ホン酸及びこれらの混合物から成る群から選択される両性イオン緩衝剤;又は (e)ポリアクリル酸、ポリメタクリル酸、ポリ(スチレン−無水マレイン酸 )、メタクリル酸/ジビニルベンゼンコポリマー、ポリ(2−アクリルアミド− 2−メチルプロパンスルホネート)、アクリル酸と長鎖アクリレートエステルと のコポリマー、及びポリ(メチルビニルエーテル−マレイン酸)から成る群から 選択されるポリマー緩衝剤 によって緩衝化する請求項3記載のデバイス。 7.カソード溜めpHを約2〜4の範囲内に維持する請求項1記載のデバイ ス。 8.カソード溜めを緩衝化する請求項1又は7に記載のデバイス。 9.前記カソード溜めをカチオン緩衝剤によって緩衝化する請求項7記載の デバイス。 10.前記カチオン緩衝剤がヒスチジン、リシン、アルギニン、アスパラギ ン酸、グルタミン酸、システイン、チロシン及びこれらの組合せから成る群から 選択される請求項9記載のデバイス。 11.前記カソード溜めを、 (a)アスパラギン酸、グルタミン酸、クエン酸、コハク酸、リン酸、酢酸、 EDTA、乳酸、安息香酸、酒石酸、マレイン酸、フマル酸、硫酸、ギ酸、リン ゴ酸、マロン酸、グルタル酸及びアジピン酸から成る群から選択される緩衝剤; 又は (b)ビニルピロリドン/第4級化ジメチルアミノエチルメタクリレートコポ リマー、ビニルカプロラクタム/ビニルピロリドン/ジメチルアミノエチルメタ クリレートターポリマー、ポリビニルピロリドン、及びメタクリレート/ジビニ ルベンゼンコポリマーから成る群から選択されるポリマー緩衝剤 によって緩衝化する請求項9記載のデバイス。 12.(a)可逆的に酸化及び還元されることができる物質から構成される 第2電極であって、アノード溜めと、アノード溜めpHを約4より大きく維持す るように第2電極を操作する手段とに連通する第2電極;又は (b)可逆的に酸化及び還元されることができる物質から構成される第2電極 であって、カソード溜めと、カソード溜めpHを約4未満に維持するように第2 電極を操作する手段とに連通する第2電極 を含む請求項1記載のデバイス。 13.カソード溜め内のカリウム含量を感知するためのカリウムセンサーを 含む請求項1記載のデバイス。 14.カリウムセンサーが体表からカソード溜め中への所定レベルのカリウ ム流出を感知するときに、デバイスの操作を変えるための手段を含む請求項13 記載のデバイス。 15.(a)カソード溜めが投与すべき作用剤を含み、アノード溜めが約4 を越えるpHに維持される;又は (b)アノード溜めが投与すべき作用剤を含み、カソード溜めが約4未満のp Hに維持される 請求項1記載のデバイス。 16.電気駆動式作用剤投与中の体表の電気抵抗の低下、電気駆動式作用剤 投与によって生ずる体表の刺激の軽減、電気駆動式作用剤投与によって生ずる体 表の紅斑の軽減、電気駆動式作用剤投与によって生ずる体表の損傷の軽減、及び これらの組合せから成る群から選択される効果を有する請求項1記載のデバイス 。 17.体表を通して電気駆動によって作用剤を投与する方法であって、電気 駆動式作用剤投与デバイス(10)のアノード溜め(16又は18)及びカソー ド溜め(16又は18)(溜め(16、18)の少なくとも1つは投与すべき作 用剤を含む)を前記体表とイオン伝導関係に配置し、作用剤を電気駆動によって 体表を通して投与するためにアノード溜め及びカソード溜め(16、18)を横 切って電圧を印加し、電気駆動式作用剤投与中に、 (a)前記アノード溜めを約4より大きいpHに維持し;及び/又は (b)前記カソード溜めを約4未満のpHに維持する 前記方法。 18.アノード溜めpHを約4〜10の範囲内に維持する請求項17記載の 方法。 19.前記アノード溜めを緩衝化することをさらに含む請求項17又は18 に記載の方法。 20.前記アノード溜めをアニオン緩衝剤によって緩衝化する請求項19記 載の方法。 21.前記アニオン緩衝剤がヒスチジン、リシン、アルギニン、アスパラギ ン酸、グルタミン酸、システイン、チロシン及びこれらの組合せから成る群から 選択される請求項20記載の方法。 22.前記アノード溜めを、 (a)アスパラギン酸、グルタミン酸、クエン酸、コハク酸、リン酸、酢酸、 EDTA、乳酸、ホウ酸、炭酸、一塩基性リン酸ナトリウム、安息香酸、酒石酸 、マレイン酸、フマル酸、硫酸、ギ酸、リンゴ酸、マロン酸、グルタル酸及びア ジピン酸から成る群から選択される緩衝剤; (b)トロメタミン、トリエタノールアミン、イミダゾール、アンモニア、エ タノールアミン、ジエタノールアミン、ヒスチジン、リシン及びアルギニンから 成る群から選択される緩衝剤; (c)塩化カルシウム2水和物、トリエタノールアミン塩酸塩、塩化マグネシ ウム7水和物、ジエタノールアミン塩酸塩、塩化アンモニウム、エタノールアミ ン塩酸塩、塩化カリウム及び塩化ナトリウムから成る群から選択される緩衝剤; (d)2−(N−モルホリノ)エタンスルホン酸、1,4−ピペラジンービス −(エタンスルホン酸)、N−2−アセトアミド イミノ二酢酸、N−2(2− アセトアミド)−2−アミノエタンスルホン酸、N,N−ビス(2−ヒドロキシ エチル)−2−アミノエタンスルホン酸、2−(N−モルホリノ)プロパンスル ホン酸、N−トリス(ヒドロキシメチル)メチル−2−アミノエタンスルホン酸 、N−2−ヒドロキシエチルピペラジン−N’−2−エタンスルホン酸、4−( 2−ヒドロキシエチル)−1−ピペラジンプロパンスルホン酸、N−トリス(ヒ ドロキシメチル)メチル−2−アミノプロパンスルホン酸、2−シクロヘキシル アミノ−1−エタンスルホン酸、3−シクロヘキシルアミノ−1−プロパンスル ホン酸及びこれらの混合物から成る群から選択される両性イオン緩衝剤;又は (e)ポリアクリル酸、ポリメタクリル酸、ポリ(スチレン−無水マレイン酸 )、メタクリル酸/ジビニルベンゼンコポリマー、ポリ(2−アクリルアミド− 2−メチルプロパンスルホネート)、アクリル酸と長鎖アクリレートエステルと のコポリマー、及びポリ(メチルビニルエーテル−マレイン酸)から成る群から 選択されるポリマー緩衝剤 によって緩衝化する請求項19記載の方法。 23.カソード溜めpHを約2〜4の範囲内に維持する請求項17記載の方 法。 24.前記カソード溜めを緩衝化することをさらに含む請求項17又は23 に記載の方法。 25.前記カソード溜めをカチオン緩衝剤によって緩衝化する請求項24記 載の方法。 26.前記カチオン緩衝剤がヒスチジン、リシン、アルギニン、アスパラギ ン酸、グルタミン酸、システイン、チロシン及びこれらの組合せから成る群から 選択される請求項25記載の方法。 27.前記カソード溜めを、 (a)ビニルピロリドン/第4級化ジメチルアミノエチルメタクリレートコポ リマー、ビニルカプロラクタム/ビニルピロリドン/ジメチルアミノエチルメタ クリレートターポリマー、ポリビニルピロリドン、及びメタクリレート/ジビニ ルベンゼンコポリマーから成る群から選択されるポリマー緩衝剤;又は (b)アスパラギン酸、グルタミン酸、クエン酸、コハク酸、リン酸、酢酸、 EDTA、乳酸、安息香酸、酒石酸、マレイン酸、フマル酸、硫酸、ギ酸、リン ゴ酸、マロン酸、グルタル酸及びアジピン酸から成る群から選択される緩衝剤に よって緩衝化する請求項17又は24記載の方法。 28.アノード溜め又はカソード溜めと連通する第2電極を、第2電極と連 通する前記溜めのpHを維持するために有効な形式で第2電極を可逆的に酸化及 び/又は還元することによって、操作することを含む請求項17記載の方法。 29.カソード溜めのカリウム含量を感知することを含む請求項17記載の 方法。 30.カソード溜め内のカリウム含量の所定レベルを感知したときに、電気 駆動式作用剤投与を変えることを含む請求項29記載の方法。 31.(a)前記カソード溜めが投与すべき作用剤を含み、この作用剤をア ノード溜めから体表を通して電気駆動によって投与することをさらに包含する; 又は (b)前記アノード溜めが投与すべき作用剤を含み、この作用剤をカソード溜 めから体表を通して電気駆動によって投与することをさらに包含する 請求項17記載の方法。 32.電気駆動式作用剤投与中の体表の電気抵抗の低下、電気駆動式作用剤 投与によって生ずる体表の刺激の軽減、電気駆動式作用剤投与によって生ずる体 表の紅斑の軽減、電気駆動式作用剤投与によって生ずる体表の損傷の軽減、及び これらの組合せから成る群から選択される効果を有する請求項17記載の方法。 33.アノード電極アセンブリ(12,16又は14,18)と、カソード 電極アセンブリ(12,16又は14,18)と、前記電極アセンブリと電気的 に接続した電源(24)と、前記電極アセンブリの少なくとも1つにおける作用 剤溜め(16,18)とを有する電気駆動式投与デバイス(10)において、前 記カソード溜め中のカリウムセンサーと、カリウムセンサーが(i)カソード電 極アセンブリ中の所定カリウム含量と(ii)カソード電極アセンブリ中のカリウ ム含量の所定変化速度とから成る群から選択されるパラメータを感知するときに デバイスの操作を変化させる手段とを含む改良デバイス。 34.投与デバイスの操作を変える前記手段がデバイスによって供給される 電流レベルを低下させる手段を含む請求項33記載のデバイス。 35.低下手段が供給電流を実質的に零にまで低下させる請求項34記載の デバイス。 36.カソード電極アセンブリ中のカリウム含量が前記所定値に達するとき にシグナルを発する手段を含む請求項33記載のデバイス。 37.カリウム含量の変化速度が前記所定値に達したときにシグナルを発す る手段を含む請求項33記載のデバイス。 38.カリウムセンサーがカリウム感知電極を含む請求項33記載のデバイ ス。 39.アノード電極アセンブリと、カソード電極アセンブリと、これらの電 極アセンブリに電気的に結合した電源と、電極アセンブリの少なくとも1つにお ける作用剤溜めとを有する、皮膚部位を通して電気駆動によって作用剤を投与す る電気駆動式投与デバイスの操作方法において、 電気駆動式作用剤投与中の皮膚部位からのカリウム流出の監視と; カリウム流出が所定値に達したときの投与デバイスの操作の変更と を含む改良方法。 40.投与デバイスの操作の前記変更がデバイスによる電流供給レベルの低 下を含む請求項39記載の方法。 41.供給電流を実質的に零にまで低下させる請求項39記載の方法。 42.投与デバイスの操作の前記変更がシグナル発生を含む請求項39記載 の方法。 43.投与デバイスの操作の前記変更が皮膚部位からの前記投与デバイスの 除去と、このデバイスの別の皮膚部位への再装着とを含む請求項39記載の方法 。 44.投与デバイスの操作の前記変更が皮膚部位からの前記投与デバイスの 除去と、その後の別の皮膚部位からの電気駆動による作用剤投与とを含む請求項 39記載の方法。 45.カソード電極アセンブリ中のカリウム含量の監視を含む請求項39記 載の方法。 46.カリウム含量をセンサーによって監視する請求項45記載の方法。
