Claims (1)
- 【特許請求の範囲】 1.a)試料を、ホスファチジルセリンに対する高い親和性を有する検出可能な 試薬と接触させ、 b)ホスファチジルセリンに対する高い親和性を有する検出可能な試薬と反 応した細胞、好ましくは無損傷の細胞を定性的及び/又は場合により定量的に検 出し、 該検出段階は c)非−枯死細胞から枯死細胞を単離する ための段階の前又は後に行われ、その単離は枯死細胞が段階a)においてホスフ ァチジルセリンに対する高い親和性を有する検出可能な試薬を備えており、該試 薬がそれ自体既知の方法で選択可能でもあるという事実の故に行うことができる ことを含む試料中の枯死細胞を検出及び/又は場合により定量及び/又は場合に より単離するための方法。 2.ホスファチジルセリンに対する高い親和性を有する試薬がアネキシンとして 分類されるポリペプチド又はタンパク質である請求の範囲第1項に記載の方法。 3.フローサイトメトリーなどのそれ自体既知の方法におけるプロピジウムヨー ダイドの利用によるなどの、それ自体既知の方法でライシスを受けた細胞を検出 するためのマーカーを用いることにより、細胞をライシスを受けた細胞と無損傷 の細胞に区別することができる請求の範囲第1又は2項に記載の方法。 4.ホスファチジルセリンに対する高い親和性を有する試薬がアネキシンV又は ホスファチジルセリンに対する高い親和性を示すそれらの誘導 体である請求の範囲第1〜3項のいずれかに記載の方法。 5.ホスファチジルセリンに対する高い親和性を有する試薬を、試薬がホスファ チジルセリンに対する親和性を示すためにカチオン、好ましくは2価のカチオン の存在下で試料と接触させる請求の範囲第1〜4項のいずれかに記載の方法。 6.2価のカチオンがCd2+、Zn2+、Mn2+、Co2+及び好ましくはCa2+か ら成る群より選ばれる請求の範囲第5項に記載の方法。 7.ホスファチジルセリンに対する高い親和性を有する試薬を、試薬がホスファ チジルセリンに対する高い親和性を示すためにCa2+及びZn2+の両方の存在下 で試料と接触させる請求の範囲第5又は6項に記載の方法。 8.カチオンがTb3+である請求の範囲第5項に記載の方法。 9.ホスファチジルセリンに対して特異的な試薬に蛍光マーカー、放射性マーカ ー、酵素、金属、色素、検出可能な免疫グロブリン又はタンパク質部分などの検 出可能なマーカーが与えられている請求の範囲第1〜8項のいずれかに記載の方 法。 10.放射性マーカーが125I、131I、111In、32P、35S及び99Teから成 る群より選ばれる請求の範囲第9項に記載の方法。 11.蛍光マーカーがフルオレセイン、フィコエリスリン及びローダミンから成 る群より選ばれる請求の範囲第9項に記載の方法。 12.酵素がアルコールデヒドロゲナーゼ、パーオキシダーゼ及びアルカリ性ホ スファターゼから成る群より選ばれる請求の範囲第9項に記載の方法。 13.タンパク質部分がフェリチン、ビオチン、アビジン及びストレプ タビジン、ならびにそれらの誘導体から成る群より選ばれる請求の範囲第9項に 記載の方法。 14.色素がエバンスブルー及びクーマシーブリリアントブルーから成る群より 選ばれる請求の範囲第9項に記載の方法。 15.枯死細胞がFACSなどのフローサイトメーターの利用により単離される 請求の範囲第1〜14項のいずれかに記載の方法。 16.ミクロタイタープレートに接着性細胞を設け、枯死を多数の細胞試料にお いて測定する請求の範囲第1〜14項のいずれかに記載の方法。 17.アネキシン又はアネキシンの共役形態に高い親和性を示す固相の利用によ り非−枯死細胞から枯死細胞を分離する請求の範囲第1〜14項のいずれかに記 載の方法。 18.調べられるべき化合物及び/又は処置の存在に供された試料を用いて請求 の範囲第1〜17項のいずれかに記載の方法を行い、結果を、標準試料を用いて 、及び/又は調べるべき化合物及び/又は処置の存在の前に採取された試料を用 いて同じ条件下で請求の範囲第1〜17項に記載の方法を行って得られる結果と 比較することを含む、患者及び/又は試料中の枯死の量に対する化合物又は特定 の処置の効果を決定するための方法。 19.a)検出可能な又は検出可能にされることができる、ホスファチジルセリ ンに対する高い親和性を有する試薬、 b)無損傷の細胞からライシスを受けた細胞を区別し、好ましくは分離す るための検出可能な及び好ましくは選択可能な、例えばプロピジウムヨーダイド のようなマーカー、ならびに場合により c)カチオン、及び d)適した媒体 を含む請求の範囲第1〜18項のいずれかに記載の方法を行うために適したキッ ト。 20.例えば同原又は異種幹細胞あるいは骨髄細胞の移植のために、不適切な細 胞から移植に適した細胞を分離するための請求の範囲第1〜16項のいずれかに 記載の方法ならびに/又は請求の範囲第18項に記載のキットの利用。
