Claims (1)
- 【特許請求の範囲】 1.適度なストリンジェント条件下で、図2(配列表の配列番号1)に示されて いるヌクレオチド配列に対応する、又は相補的なヌクレオチド配列を有する核酸 に対し、ハイブリダイズできる精製された核酸。 2.適度なストリンジェント条件下で、機能的に活性なグリコシルトランスフェ ラーゼをコードする図2(配列表の配列番号1)に示されているヌクレオチド配 列の一部に対応する、又は相補的なヌクレオチド配列を有する核酸に対し、ハイ ブリダイズできる、請求項1記載の核酸。 3.機能的に活性なグリコシルトランスフェラーゼをコードする請求項2記載の 核酸。 4.機能的に活性なグリコシルトランスフェラーゼをコードする図2(配列表の 配列番号1)に示されているヌクレオチド配列の一部に対応する、又は相補的な ヌクレオチド配列を有する請求項1記載の核酸。 5.機能的に活性なグリコシルトランスフェラーゼをコードする請求項4記載の 核酸。 6.図2(配列表の配列番号の配列番号1)に示されているヌクレオチド配列に 対応する、又は相補的なヌクレオチド配列を有する請求項1記載の核酸。 7.前記機能的に活性なグリコシルトランスフェラーゼが、下記a)〜c)からなる 群から選ばれた反応を触媒する請求項3記載の核酸: a) Galβ1 →4 をGlcNAc又はGlc に付加する; b) GalNAc 又はGlcNAcβ1 →3 をGal に付加する;及び c) Galα1 →4 をGal に付加する。 8.配列表の配列番号3のアミノ酸配列を有するグリコシルトランスフェラーゼ をコードする請求項3記載の核酸。 9.配列表の配列番号8のアミノ酸配列を有するグリコシルトランスフェラーゼ をコードする請求項3記載の核酸。 10.配列表の配列番号4のアミノ酸配列を有するグリコシルトランスフェラーゼ をコードする請求項3記載の核酸。 11.配列表の配列番号5のアミノ酸配列を有するグリコシルトランスフェラーゼ をコードする請求項3記載の核酸。 12.配列表の配列番号6のアミノ酸配列を有するグリコシルトランスフェラーゼ をコードする請求項3記載の核酸。 13.発現制御配列と機能的に結合している請求項3記載の核酸を含む発現ベクタ ー。 14.請求項13記載の発現ベクターで形質転換された組換え宿主細胞。 15.下記工程を含むグリコシルトランスフェラーゼの製造方法: a) グリコシルトランスフェラーゼの発現を許容する条件下で請求項14記載の 組換え宿主細胞を培養する工程;及び b) 発現されたグリコシルトランスフェラーゼを回収する工程。 16.配列表の配列番号3のアミノ酸配列、又はその機能的に活性なフラグメント を有するグリコシルトランスフェラーゼ。 17.配列表の配列番号8のアミノ酸配列、又はその機能的に活性なフラグメント を有するグリコシルトランスフェラーゼ。 18.配列表の配列番号4のアミノ酸配列、又はその機能的に活性なフラグメント を有するグリコシルトランスフェラーゼ。 19.配列表の配列番号5のアミノ酸配列、又はその機能的に活性なフラグメント を有するグリコシルトランスフェラーゼ。 20.配列表の配列番号6のアミノ酸配列、又はその機能的に活性なフラグメント を有するグリコシルトランスフェラーゼ。 21.固相支持体に結合したグリコシルトランスフェラーゼを含む組成物であって 、該グリコシルトランスフェラーゼが下記a)〜e)からなる群から選ばれたもので ある組成物: a) 配列表の配列番号3のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼ; b) 配列表の配列番号8のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼ; c) 配列表の配列番号4のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼ; d) 配列表の配列番号5のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼ;及び e) 配列表の配列番号6のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼ。 22.GalNAc又はGlcNAcβ1 →3 をGal に付加する方法であって、請求項16記載の グリコシルトランスフェラーゼの存在下でGal 残基を含む受容体成分に、活性化 されたGalNAc又はGlcNAcを含む反応混合物を接触させることを含む方法。 23.Gal β1 →4 をGlcNAc又はGlc に付加する方法であって、請求項17記載のグ リコシルトランスフェラーゼの存在下でGlcNAc又はGlc 残基を含む受容体成分に 、活性化されたGal を含む反応混合物を接触させることを含む方法。 24.Gal α1→4 をGal に付加する方法であって、請求項18記載のグリコシルト ランスフェラーゼの存在下で、Gal残基を含む受容体成分に、活性化されたGalを 含む反応混合物を接触させることを含む方法。 25.GalNAc又はGlcNAcβ1 →3 をGal に付加する方法であって、請求項19記載の グリコシルトランスフェラーゼの存在下で、Gal 残基を含む受容体成分に、活性 化されたGalNAc又はGlcNAcを含む反応混合物を接触させることを含む方法。 26.Gal β1 →4 をGlcNAc又はGlc に付加する方法であって、請求項20記載のグ リコシルトランスフェラーゼの存在下で、GlcNAc又はGlc 残基を含む受容体成分 に、活性化されたGal を含む反応混合物を接触させることを含む方法。 27.下記工程を連続的に実施することを含む、構造Gal α1 → 4Gal β1 →4Glc を有するオリゴ糖類を製造する方法: a) 配列表の配列番号6のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Glc 残基を含む受容体成 分に、活性されたGal を含む反応混合物を接触させる工程;及び b) 配列表の配列番号4のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Gal β1 →4Glc残基を含 む受容体成分に、活性化されたGal を含む反応混合物を接触させる工程。 28.構造Gal β1 →4 Glc を有するオリゴ糖類を製造する方法であって、請求項 20記載のグリコシルトランスフェラーゼの存在下で、Glc 残基を含む受容体成 分に、活性化されたGal を含む反応混合物を接触させることを含む方法。 29.構造GlcNAcβ1 →3Galβ1 →4Glcを有するオリゴ糖類を製造する方法であっ て、請求項16記載のグリコシルトランスフェラーゼの存在下で、Gal β1 →4Glc 残基を含む受容体成分に、活性化されたGalNAcを含む反応混合物を接触させるこ とを含む方法。 30.構造Gal β1 →4GlcNAc β1 →3Galβ1 →4Glcを有するオリゴ糖類を製造す る方法であって、請求項17記載のグリコシルトランスフェラーゼの存在下で、Gl cNAcβ1 →3Galβ1 →4Glc残基を含む受容体成分に、活性化されたGal を含む反 応混合物を接触させることを含む方法。 31.構造GalNAcβ1 →3Galβ1 →4GlcNAc β1 →3Galβ1 →4Glcを有するオリゴ 糖類を製造する方法であって、請求項19記載のグリコシルトランスフェラーゼの 存在下で、Gal β1 →4GlcNAc β1 →3Galβ1 →4Glc残基を含む受容体成分に、 活性化されたGalNAcを含む反応混合物を接触させることを含む方法。 32.下記工程を連続的に実施することを含む、構造GalNAcβ1 →3Galβ1 →4Glc NAc β1 →3Galβ1 →4Glcを有するオリゴ糖類を製造する方法: a) 配列表の配列番号6のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Glc 残基を含む受容体成 分に、活性化されたGal を含む反応混合物を接触させる工程; b) 配列表の配列番号3のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Gal β1 →4Glc残基を含 む受容体成分に、活性化されたGlcNAcを含む反応混合物を接触させる工程; c) 配列表の配列番号8のアミノ酸配列を有するグリコシルトランスフェラー ゼの存在下で、GlcNAcβ1 →3Galβ1 →4Glc残基を含む受容体成分に、活性化さ れたGal を含む反応混合物を接触させる工程;及び d) 配列表の配列番号5のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Gal β1 →4GlcNAc β1 →3Galβ1 →4Glc残基を含む受容体成分に、活性化されたGalNAcを含む反応混合 物を接触させる工程。 33.下記工程を連続的に実施することを含む、構造Gal β1 →4GlcNAc β1 →3G alβ1 →4Glcを有するオリゴ糖類を製造する方法: a) 配列表の配列番号6のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Glc 残基を含む受容体成 分に、活性化されたGal を含む反応混合物を接触させる工程; b) 配列表の配列番号3のアミノ酸配列、又はその機能的に活性なフラグメン トを有するグリコシルトランスフェラーゼの存在下で、Gal β1 →4Glc残基を含 む受容体部分に、活性化されたGlcNAcを含む反応混合物を接触させる工程;及び c) 配列表の配列番号8のアミノ酸配列を有するグリコシルトランスフェラー ゼの存在下で、GlcNAcβ1 →3Galβ1 →4Glc残基を含む受容体成分に、活性化さ れたGal を含む反応混合物を接触させる工程。
