Claims (1)
- 【特許請求の範囲】 1.下記の分子式を有する段階的オリゴヌクレオチド合成に用いるための保護さ れたモノマー。 上記式中、R’は、2’オルトエステル保護基、H、エーテル、アルキルエー テル、エステル、ハロゲン、保護されたアミン及び保護されたヒドロキシル部か らなる第1のグループから選択されたものであり;R”は、任意のリガンドを含 むものであってよく;R1,R2及びR3は、少なくとも一つのアルコキシもしく はシロキシ置換基であり;かつBASEは核酸塩基である。 2.前記R’がホスホルアミダイト部である、請求項1に記載のモノマー。 3.前記ホスホルアミダイト部が、下記の式を有する、請求項2に記載のモノマ ー。 式中、R4は、任意の有機リガンドである。 4.前記R1,R2及びR3が、 からなる第2のグループから選択されたものである、請求項1に記載のモノマー 。 5.前記R1,R2及びR3が、 からなる第2のグループから選択されたものである、請求項1に記載のモノマー 。 6.前記R1,R2及びR3のそれぞれが、アルコキシもしくはシロキシ部分から なる第2のグループから選択されたものである、請求項1に記載のモノマー。 7.前記R1が、前記オルトエステル保護基である、請求項1に記載のモノマー 。 8.前記オルトエステル保護基が、非環式オルトエステルである、請求項7に記 載のモノマー。 9.前記R’が、下記の式の置換基を有するものである、請求項8に記載のモノ マー。 式中、R4、R5及びR6はオルトエステル保護基置換基である。 10.前記Rが、 からなる第3のグループから選択されたものである、請求項9に記載のモノマー 。 11.前記BASEが、アデニン、グアニン、シトシン、チミン、ウラシル、N −保護されたアデニン、N−保護されたグアニン及びN−保護されたシトシン、 並びにこれらの機能的に同属な物質からなる群から選択されている、請求項1に 記載のモノマー。 12.段階的ポリマー化反応において用いられる形式の保護されたモノマーにお いて、前記モノマーが、前記反応を容易とする複数の保護基を有し、前記保護基 のうちの1つが、シリル基であり、前記保護基の他の1つのがオルトエステルで あることを特徴とする保護されたモノマー。 13.第1のシリル保護基を有する少なくとも1つの置換されたモノマーを含む 第1のモノマー鎖を調製するステップと、 前記第1のシリル保護基を除去することにより前記1つの置換されたモノマー を脱保護して脱保護された鎖を得る脱保護ステップと、 特定の部位に第2のシリル保護基を有する第2の置換されたモノマーを与える 供給ステップと、 前記第2の置換されたモノマーを、前記脱保護された鎖と反応させて長いモノ マー鎖を得る反応ステップとを備える、部位特異的段階的ポリマー化方法。 14.前記脱保護ステップが、ヨウ素イオンを添加して前記第1のシリル保護基 を除去するステップを含む、請求項13に記載の方法。 15.前記第1のモノマー鎖が第1のホスホルアミダイト置換基を含み、前記反 応ステップが、前記第1の鎖を長くするために、前記脱保護されたモノマーと前 記第1のホスホルアミダイト置換基との反応により、前記第1のホスホルアミダ イト置換基を亜リン酸トリエステル結合に変換する変換ステップを含む、請求項 13に記載の方法。 16.前記第1の置換されたモノマー及び前記第2の置換されたモノマーが、そ れぞれ、アデニン、グアニン、シトシン、チミン、ウラシル及びこれらと機能的 に同属な物質からなるグループから独立に選択された相当の塩基にそれぞれ結合 されている、請求項15に記載の方法。 17.前記第2の置換された糖が、第2のホスホルアミダイト置換基であり、そ れによって、前記第1のモノマー鎖の延長後に第1のモノマー鎖において前記第 1の置換されたモノマーをもたらし、かつ前記供給ステップ、脱保護ステップ、 反応ステップ及び変換ステップを繰り返すことを含む、請求項15に記載の方法 。 18.前記繰り返しステップが、前記亜リン酸トリエステル結合を最初に酸化す ることなく行われる、請求項13に記載の方法。 19.第1の核酸塩基を有する少なくとも1つのホスホルアミダイトヌクレオシ ドを含み、かつ第1のシリル保護基部を有する第1の糖を含む第1の鎖を調製す るステップと、 前記第1のシリル保護基部の除去により前記1つのホスホルアミダイトヌクレ オシドを脱保護して脱保護された鎖を得る脱保護ステップと、 第2の核酸塩基部分を有する構造と、第2のシリル保護基部分を有する第2の 糖とを含む保護されたホスホルアミダイトヌクレオシドを供給する供給ステップ と、 前記保護された鎖を、前記1つの保護されたホスホルアミダイトヌクレオシド と反応させて、前記第2の核酸塩基部分を有する第2の鎖を形成する反応ステッ プとを備える、オリゴヌクレオチド配列の合成方法。 20.前記供給ステップが、下記の式を有する前記脱保護されたヌクレオシドを 含む、請求項19に記載の方法。 式中、R’は、2’オルトエステル保護基、H、エーテル、アルキルエーテル 、エステル、ハロゲン、保護されたアミン及び保護されたヒドロキシル部からな る第1のグループから選択されており、R”は任意のリガンドであってよく、R1 ,R2及びR3が、少なくとも1つのアルコキシ置換基もしくはシロキシ置換基 であり、BASEが核酸塩基である。 21.前記R1,R2及びR3のうち少なくとも1つが、 からなる群から選択されたものである、請求項20に記載の方法。 22.前記R1,R2及びR3のうち少なくとも1つが、 からなる群から選択されたものである、請求項20に記載の方法。 23.前記R’が、前記2”保護基であり、かつオルトエステルを含む、請求項 20に記載の方法。 24.前記R’が、 で示される構造を含み、式中、Rは、約20個までの炭素原子を有する2’保護 基置換基である、請求項23に記載の方法。 25.前記Rが、 からなる保護基グループから選択したものである、請求項24に記載の方法。 26.請求項19に記載の方法であって、さらに、前記鎖におけるリン(IIII)結 合を安定化するために、酸化を行うことなく、前記脱保護、供給及び反応ステッ プを複数サイクル繰り返す方法。 27.前記複数のサイクルが5回を超える、請求項26に記載の方法。 28.前記反応ステップが、シリルエーテルを分裂させてかつ前記保護基を除去 するために、ヨウ素を添加することにより前記保護されたヌクレオシドを脱保護 するステップを備える、請求項19に記載の方法。 29.前記調製ステップが、前記1つのヌクレオチドを不溶性非ガラス支持体に 結合するステップを含む、請求項19に記載の方法。 30.前記非ガラス支持体が、ポリスチレンビーズである、請求項29に記載の 方法。
