Description
Claims (1)
- 【特許請求の範囲】 1.液体サンプル中の1以上の被検体の濃度を決定するための方法であって、 (a)固体支持体上に1以上の捕獲剤を固定化させるステップであって、各々 の捕獲剤が、供された結合剤に特異的に結合することができることを特徴とする ステップと、 (b)1以上の結合剤に前記液体サンプルを接触させるステップであって、各 々の結合剤が、結合部位のフラクションが被検体により占有されるに至るような 供された前記被検体に特異的な結合部位と、対応する捕獲剤に結合するのに適合 した尾部と、を有することを特徴とするステップと、 (c)前記ステップ(b)と同時又は連続的のいずれかにおいて、前記結合剤 が該結合剤各々の捕獲剤に結合するに至るように、前記固定化された捕獲剤に前 記液体サンプルを接触させるステップと、 (d)被検体により占有された供された結合剤の前記結合部位の前記フラクシ ョンを表す値を決定することにより、前記液体サンプル中の被検体の濃度を決定 するステップと、 を含むことを特徴とする方法。 2.支持体上に1以上の結合剤を固定化する方法であって、各々の結合剤が、 供された被検体に特異的な結合部位と、捕獲剤に結合するのに適合した尾部と、 を有し、 (a)支持体上に1以上の捕獲剤を固定化させるステップであって、各々の捕 獲剤が、供された結合剤の前記尾部に結合することができることを特徴とするス テップと、 (b)前記結合剤が該結合剤の尾部を通して該結合剤各々の捕獲 剤に特異的に結合するに至るように、前記支持体上に固定化された前記捕獲剤を 有する前記支持体に前記結合剤を接触させるステップと、 を含むことを特徴とする方法。 3.前記方法が、前記支持体上に固定化された前記捕獲剤に前記結合剤をさら す前に、該結合剤に前記尾部を付着させるステップを更に含むことを特徴とする 請求項2に記載の方法。 4.前記捕獲剤が、前記対応する結合剤の尾部における相補的配列にハイブリ ッド形成することができる配列を有するオリゴヌクレオチドであることを特徴と する請求項1〜3のいずれか一に記載の方法。 5.前記オリゴヌクレオチドが8〜30塩基長であることを特徴とする請求項4 に記載の方法。 5.前記結合剤が、被検体に特異的な結合部位を有する抗体であることを特徴 とする先の請求項のいずれか一に記載の方法。 6.前記結合剤が特異的である前記被検体の周辺濃度が大きく乱されないよう に、各々の結合剤の少量が用いられることを特徴とする先の請求項のいずれか一 に記載の方法。 7.前記結合剤の少量が0.1 V/Kモル未満であり、ここでVは、前記サンプ ルの容量であり、Kは、前記被検体が前記結合剤に結合することについての有効 アフィニティー定数であることを特徴とする請求項6に記載の方法。 8.各々の捕獲剤が、実質的に全ての供された結合剤に結合するために、過剰 に用いられることを特徴とする先の請求項のいずれか一に記載の方法。 9.前記捕獲剤が、分離された位置で支持体上に固定化されることを特徴とす る先の請求項のいずれか一に記載の方法。 10.前記分離された位置がミクロスポット(microspots)であることを特徴と する請求項9に記載の方法。 11.供された被検体の濃度の一連の測定が同時に行われ得るように、供された 捕獲剤が、複数の位置において前記支持体上に固定化されることを特徴とする先 の請求項のいずれか一に記載の方法。 12.前記被検体により占有される結合部位のフラクションを表す値が、競合及 び/又は非競合法において発現剤を用いて決定され、前記発現剤がマーカーで標 識されることを特徴とする先の請求項のいずれか一に記載の方法。 13.前記マーカーが蛍光又は化学発光マーカーであることを特徴とする請求項 12に記載の方法。 14.請求項1〜13のいずれか一に記載の方法において、液体サンプル中の1以 上の被検体の濃度を決定するためのキットであって、該キットが、 (a)固体支持体に付着され、供された結合剤に特異的に結合することができ る捕獲剤を複数の位置に有する前記固体支持体と、 (b)1以上の結合剤であって、各々の結合剤が、被検体に特異的な結合部位 と、1以上の捕獲剤に結合するのに適合した尾部と、 を有することを特徴とする結合剤と、 (c)占有された結合剤結合部位もしくは結合剤に結合した被検体又は未占有 の結合剤結合部位に結合することができるマーカーを有する1以上の発現剤と、 を含むことを特徴とするキット。 14.液体サンプル中の1以上の被検体の濃度を決定するためのアッセイ用のキ ットであって、該キットが、 (a)1以上の尾部であって、各々の尾部が結合剤への付着のためであること を特徴とする尾部と、 (b)固体支持体に付着され、尾部に特異的に結合することができる1以上の 捕獲剤を複数の位置に有する前記固体支持体と、 を含み、前記アッセイの使用者が前記尾部を前記結合剤に付着させることにより 、請求項1〜13のいずれか一に記載の方法において、前記捕獲剤が付着された前 記固体支持体と共に用いられ得る結合剤を供することを特徴とするキット。
