Description
Claims (1)
- 【特許請求の範囲】 1. 特性エネルギーの光子により励起可能な分子を含む目的物の3つ又はそ れ以上の光子の励起技術による顕微鏡検査法であって、 nが上記目的物により吸収されるべき光子の数に等しく、3より大きい若しく はそれに等しい場合に、上記目的物に、上記特性エネルギーの約1/nのエネル ギーの光子を有するレーザ光の強いサブピコ秒のパルスのビームを照射し、 少なくとも3つの入射光子の同時吸収により分子励起を生じさせるべく、十分 に高い照射強度をもたらすように、上記目的物の範囲内に照射光を結像させるス テップを有する方法。 2. 上記目的物が、かご閉じ込めの生物学的に活性な分子を含み、上記照射 強度が、n個の入射光子の同時吸収により、かご閉じ込めされた生物学的に活性 な化合物を解放するのに十分であり、各エネルギーが、上記特性エネルギーの約 1/nに等しい請求の範囲第1項の方法。 3. 上記目的物が、蛍光性の分子を含み、上記照射強度が、n個の入射光子 7の同時吸収により、上記目的物の蛍光を生じさせるのに十分であり、各エネル ギーが、上記特性エネルギーの約1/nに等しい請求の範囲第1項の方法。 4. 上記照射光を結像させるステップが、上記n個の入射光子の同時吸収に より分子励起を生じさせるべく、上記焦点体積においてのみ十分に高い照射強度 をもたらすように、上記目的物の範囲内における小さな焦点体積へ照射光を結像 させることを有している請求の範囲第3項の方法。 5. 上記目的物を通じて、上記焦点体積を走査すべく、上記ビームを走査し 、 上記目的物により生じさせられた蛍光を検出するステップを更に含む請求の範 囲第4項の方法。 6. 上記目的物が組織であり、また、上記分子励起が、内在的な発色団,外 部から与えられた発色団、若しくは上記組織内の内在的な蛍光体による少なくと も3つの照射光子の同時吸収により発生する請求の範囲第1項の方法。 7. 蛍光光子を生じさせるための特性エネルギーの光子による励起に応答す る蛍光成分を含む目的物のレーザ走査蛍光顕微鏡検査用の装置であって、 上記目的物を受合うためのステージ手段と、 nが3よりも大きい、あるいはそれに等しい場合に、上記特性エネルギーの1 /nのエネルギーで構成されるサブピコ秒のコヒーレント光パルスの少なくとも 1つの光源と、 上記目的物に少なくとも3つの光子を吸収させ、上記蛍光光子を生じさせるべ く、上記目的物上にコヒーレント光パルスを結像させる手段と、 上記蛍光光子を検出する検出手段と、 上記蛍光光子を検出手段に対して指向させる手段とを備えた装置。 8. 上記目的物を通じて、上記光パルスを走査する走査手段を更に備えた請 求の範囲第7項の装置。 9. 上記検出手段が、共焦点光電子増倍管を有する請求の範囲第7項の装置 。 10. 上記検出手段が、フーリエ平面検出器を有する請求の範囲第7項の装 置。 11. 上記検出手段が、画像形成検出器を有する請求の範囲第7項の装置。
