Claims (1)
- 【特許請求の範囲】 1.治療用細胞の有効量を哺乳類に投与することを含み、治療用細胞が少なく とも二つの膜結合性蛋白様キメラ受容体を発現しており、 該受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合する ことが可能な細胞外部分、および(b)受容体結合標的細胞または受容体結合標的 感染体を破壊するよう該治療用細胞へシグナルを送ることが可能な細胞内または 膜通過部分を含み、かつ 該受容体のもう一つが、(a)標的細胞または標的感染体を特異的に認識し結合 することが可能な細胞外部分、および(b)CD28に由来する細胞内部分を含む、 哺乳類において細胞性免疫反応を指示する方法。 2.標的細胞が、感染体、腫瘍もしくは癌細胞、または自己免疫産生細胞に感 染した宿主細胞である、請求項1記載の方法。 3.細胞性反応がMHC-非依存的である、請求項1記載の方法。 4.細胞内または膜通過部分が、T細胞受容体蛋白質、B細胞受容体蛋白質、も しくはFc受容体蛋白質、またはその機能的誘導体のシグナル伝達部分である、請 求項1記載の方法。 5.標的感染体または標的細胞に対する細胞外部分の結合後に、膜通過部分が 治療用細胞の細胞溶解性シグナル伝達蛋白質と重合し、その結果該受容体結合細 胞または結合体の破壊が起こる、請求項1記載の方法。 6.細胞内または膜通過ドメインが、T細胞受容体、ζ、η、CD3デルタ、もし くはT3ガンマ蛋白質;Fc受容体γ蛋白質;またはB細胞受容体mb1もしくはB29蛋 白質に由来する、請求項4記載の方法。 7.キメラ受容体が以下のいずれかを含む、請求項6記載の方法: (a) 配列番号:6のアミノ酸421〜532もしくはその機能的細胞障害性シグナル 伝達誘導体; (b) 配列番号:6のアミノ酸(a)423〜455;(b)438〜455;(c)461〜494;もし くは(d)494〜528; (c) 配列番号:6のアミノ酸400〜420; (d) 配列番号:4のアミノ酸421〜575もしくはその機能的細胞障害性シグナル 伝達誘導体; (e) 配列番号:4のアミノ酸(a)423〜455;(b)438〜455;(c)461〜494;もし くは(d)494〜528; (f) 配列番号:4のアミノ酸400〜420; (g) 配列番号:5のアミノ酸421〜462もしくはその機能的細胞障害性シグナル 伝達誘導体; (h) 配列番号:5のアミノ酸402〜419; (i) 図15Aを包括するアミノ酸チロシン282〜チロシン298; (j) 図16のアミノ酸132〜171(配列番号:24); (k) 図17のアミノ酸140〜182(配列番号:25); (l) 図18のアミノ酸162〜220(配列番号:26);または (m) 図19のアミノ酸183〜228(配列番号:27)。 8.Fc受容体蛋白質が、ヒトFcγRIII、ヒトFcRIIγA、またはヒトFcRIIγCで ある、請求項4記載の方法。 9.治療用細胞が、 (a) Tリンパ球; (b) 細胞障害性Tリンパ球; (c) ナチュラルキラー細胞; (d) 好中球; (e) 顆粒球; (f) マクロファージ; (g) 肥満細胞; (h) HeLa細胞;および (i) 胎児幹細胞(ES) からなる群より選択される、請求項4記載の方法。 10.標的感染体が免疫不全ウイルスである、請求項4記載の方法。 11.細胞外部分に、CD4のHIVエンベロープ結合部分またはそのHIVエンベロー プ結合性誘導体が含まれる、請求項4記載の方法。 12.CD4のHIV-エンベロープ結合部分に、配列番号:1のヌクレオチド1〜369 に コードされるペプチドが含まれる、請求項4記載の方法。 13.治療用細胞が受容体結合標的細胞または標的感染体を細胞溶解によって破 壊する、請求項4記載の方法。 14.受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)受容体結合標的細胞または受容体結合標 的感染体を破壊するよう該細胞へシグナルを送ることが可能な、T細胞受容体、 B細胞受容体、またはFc受容体に由来する細胞内または膜通過部分を含み、かつ 受容体のもう一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)CD28に由来する細胞内部分を含む、 少なくとも二つの蛋白様膜結合性キメラ受容体を発現する細胞。 15.標的細胞が、感染体に感染した宿主細胞、腫瘍もしくは癌細胞、または自 己免疫産生細胞である、請求項14記載の細胞。 16.結合がMHC-非依存的である、請求項14記載の細胞。 17.細胞内または膜通過ドメインが、T細胞受容体、ζ、η、CD3デルタ、もし くはT3ガンマ蛋白質;Fc受容体γ蛋白質;またはB細胞受容体mb1もしくはB29蛋 白質のいずれかに由来する、請求項14記載の細胞。 18.キメラ受容体が以下のいずれかを含む、請求項17記載の細胞: (a) 配列番号:6のアミノ酸421〜532もしくはその機能的細胞障害性シグナル 伝達誘導体; (b) 配列番号:6のアミノ酸(a)423〜455;(b)438〜455;(c)461〜494;もし くは(d)494〜528; (c) 配列番号:6のアミノ酸400〜420; (d) 配列番号:4のアミノ酸421〜575もしくはその機能的細胞障害性シグナル 伝達誘導体; (e) 配列番号:4のアミノ酸(a)423〜455;(b)438〜455;(c)461〜494;もし くは(d)494〜528; (f) 配列番号:4のアミノ酸400〜420; (g) 配列番号:5のアミノ酸421〜462もしくはその機能的細胞障害性シグナル 伝達誘導体; (h) 配列番号:5のアミノ酸402〜419; (i) 図15Aを包括するアミノ酸チロシン282〜チロシン298; (j) 図16のアミノ酸132〜171(配列番号:24); (k) 図17のアミノ酸140〜182(配列番号:25); (l) 図18のアミノ酸162〜220(配列番号:26);または (m) 図19のアミノ酸183〜228(配列番号:27) 19.Fc受容体蛋白質が、ヒトFcγRIII、ヒトFcRIIγA、またはヒトFcRIIγCで ある、請求項14記載の細胞。 20.細胞外部分に、受容体のリガンド結合部分、リガンドの受容体結合部分、 抗体の抗原結合部分、またはその機能的誘導体が含まれる、請求項14記載の細胞 。 21.標的感染体が免疫不全ウイルスである、または標的細胞が免疫不全ウイル スに感染した宿主細胞である、請求項20記載の細胞。 22.細胞外部分に、CD4のHIVエンベロープ結合部分またはその機能的誘導体が 含まれる、請求項21記載の細胞。 23.CD4のHIV-エンベロープ結合部分に、配列番号:1のヌクレオチド1〜369 にコードされるペプチドが含まれる、請求項22記載の細胞。 24.細胞が、受容体結合標的細胞または標的感染体を細胞溶解によって破壊す る、請求項14記載の細胞。 25.受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)T細胞受容体CD3、ゼータ、もしくはイー タポリペプチド、B細胞受容体、またはFc受容体に由来する細胞内または膜通過 部分を含み、かつ 受容体のもう一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)CD28に由来する細胞内部分を含む、 少なくとも二つの蛋白様膜結合性キメラ受容体を発現する細胞。 26.キメラ受容体が以下のいずれかを含む、請求項14または25記載の細胞 (a) CD16、CD7、もしくはCD5の細胞外部分; (b) CD5、もしくはCD7の膜通過部分;または (c) CD5細胞内部分。 27.受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)受容体結合標的細胞または受容体結合標 的感染体を破壊するよう該細胞へシグナルを送ることが可能な、T細胞受容体、B 細胞受容体、またはFc受容体蛋白質に由来する細胞内または膜通過部分を含み、 かつ 受容体のもう一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)CD28に由来する細胞内部分を含む、 一対の蛋白様膜結合性キメラ受容体。 28.受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)T細胞受容体CD3、ゼータ、もしくはイー タポリペプチド、B細胞受容体、またはFc受容体に由来する細胞内または膜通過 部分を含み、かつ 受容体のもう一つが、(b)CD28に由来する細胞内部分を含む、 一対の蛋白様膜結合性キメラ受容体。 29.受容体の一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)受容体結合標的細胞または受容体結合標 的感染体を破壊するよう該細胞へシグナルを送ることが可能な、T細胞受容体、B 細胞受容体、またはFc受容体蛋白質に由来する細胞内または膜通過部分を含み、 かつ 受容体のもう一つが、(a)標的細胞または標的感染体を特異的に認識し結合す ることが可能な細胞外部分、および(b)CD28に由来する細胞内部分を含む、 各々が蛋白様膜結合性キメラ受容体をコードする、一対のDNA分子。 30.受容体の一つが、(a)標的細胞または標的感染対を特異的に認識し結合す ることが可能な細胞外部分、および(b)T細胞受容体CD3、ゼータ、もしくはイー タポリペプチド、B細胞受容体、またはFc受容体に由来する細胞内または膜通過 部分を含み、かつ 受容体のもう一つが、(a)標的細胞または標的感染対を特異的に認識し結合す る細胞外部分、および(b)CD28に由来する細胞内部分を含む、 各々が蛋白様膜結合性キメラ受容体をコードする、一対のDNA分子。
