- 【特許請求の範囲】 1.媒体中で他の物質から生物学的目標物質を単離する方法であって、 a.生物学的目標物質を含有する媒体を用意し、生物学的目標物質と可逆的 に結合する事の出来るシリカ磁気粒子を用意し、 b.シリカ磁気粒子と媒体とを組合せて、シリカ磁気粒子と生物学的目標物 質との複合体を形成し、 c.外部磁場を適用して媒体から複合体を除去し、及び、 d.生物学的目標物質を溶出させる事によって、複合体から生物学的目標物 質を分離して、単離された生物学的目標物質を得る、 事を特徴とする方法。 2.該方法によって単離される生物学的目標物質が、核酸から成る、請求項1に 記載の生物学的目標物質を単離する方法。 3.工程(b)で用意されるシリカ磁気粒子が、粒子1mg当たり、生物学的目 標物質の少なくとも2μgと可逆的に結合する事が出来る、請求項1に記載の生 物学的目標物質を単離する方法。 4.工程(b)で用意されるシリカ磁気粒子が、シリカ質酸化物被覆磁気粒子で ある、請求項3に記載の生物学的目標物質を単離する方法。 5.複合体中の生物学的目標物質の少なくとも60%が、工程(d)で粒子から 溶出される、請求項1に記載の生物学的目標物質を単離する方法。 6.工程(d)において、複合体から溶出される生物学目標物質が、50ppm 以下の遷移金属混入物を含む、請求項1に記載の生物学的目標物質を単離する方 法。 7.媒体中で他の物質から生物学的目標物質を単離する方法であって、 a)生物学的目標物質を含有する媒体を用意する工程、 b)粒子1mg当たり、生物学的目標物質の少なくとも2μgと可逆的に結 合する能力を持つシリカ磁気粒子を用意する工程、 c)媒体とシリカ磁気粒子とから成る混合物を形成する工程、 d)混合物中で、生物学的目標物質をシリカ磁気粒子に接着させる工程、 e)外部磁場を適用して、混合物から、シリカ磁気粒子と共に、粒子に接着 した生物学的目標物質を除去する工程、及び、 f)溶出溶液に粒子を暴露する事によって、シリカ磁気粒子から、少なくと も60%の生物学的目標物質を溶出させる工程、 を含む事を特徴とする方法。 8.該方法により単離される生物学的目標物質が、核酸物質から成る、請求項7 に記載の生物学的目標物質を単離する方法。 9.該方法により単離される核酸生物学的目標物質が、プラスミドDNA物質か ら成る、請求項8に記載の生物学的目標物質を単離する方法。 10.単離される核酸生物学的目標物質が、DNAフラグメント物質から成る、請 求項8に記載の生物学的目標物質を単離する方法。 11.該方法の工程(b)で用意されるシリカ磁気粒子が、シリカ質酸化物被覆磁 気粒子である、請求項7に記載の生物学的目標物質を単離する方法。 12.工程(c)で形成される混合物が、媒体、シリカ磁気粒子及びカオトロピッ ク塩を含み、カオトロピック塩濃度が、工程(d)において、生物学的目標物質 をシリカ磁気粒子に接着させるのに十分に高い、請求項7に記載の生物学的目標 物質を単離する方法。 13.工程(c)で形成される混合物中のカオトロピック塩が、塩酸グアニジン又 はチオシアン酸グアニジンから成る、グアニジニウムカオトロピック塩から成る 、請求項12に記載の生物学的目標物質を単離する方法。 14.工程(c)で形成される混合物中のカオトロピック塩の濃度が、少なくとも 2モルである、請求項12に記載の生物学的目標物質を単離する方法。 15.生物学的目標物質が、混合物をインキュベートする事によって、工程(d) においてシリカ磁気粒子に接着する、請求項7に記載の生物学的目標物質を単離 する方法。 16.生物学的目標物質が、混合物を室温で少なくとも30秒間インキュベートす る事によって、工程(d)においてシリカ磁気粒子に接着する、請求項15に記 載の生物学的目標物質を単離する方法。 17.シリカ磁気粒子を媒体から除去した後であって、該粒子から生物学的目標物 質を溶出する前に、シリカ磁気粒子を洗浄する工程を更に含む、請求項7に記 載の生物学的目標物質を単離する方法。 18.洗浄工程が、アルコールと塩とから成る洗浄溶液を使用して行われる、請求 項17に記載の生物学的目標物質を単離する方法。 19.洗浄工程が、少なくとも30容量%のアルコールと緩衝液とから成る洗浄溶 液を使用して行われる、請求項18に記載の生物学的目標物質を単離する方法。 20.生物学的目標物質が、水又は低イオン強度の溶出溶液を使用して、工程(f )で、シリカ磁気粒子から溶出される、請求項7に記載の生物学的目標物質を単 離する方法。 21.工程(f)で、シリカ磁気粒子から溶出される生物学的目標物質が、高分子 又は金属混入物を実質的に含まない、請求項7に記載の生物学的目標物質を単離 する方法。 22.媒体中で、他の物質からプラスミドDNA物質を単離する方法であって、 a)プラスミドDNAを含有する媒体を用意する工程、 b)粒子1mg当たり、生物学的目標物質の少なくとも2μgと可逆的に結 合する能力を持つシリカ質酸化物被覆磁気粒子を用意する工程、 c)媒体、シリカ質酸化物被覆磁気粒子及びカオトロピック塩を含む混合物 を形成する工程であって、混合物中のカオトロピック塩濃度が、プラスミドDN Aを粒子に接着させるのに十分に高い工程、 d)少なくとも或程度の生物学的目標物質が、シリカ質酸化物被覆磁気粒子 に接着するまで、混合物をおよそ室温でインキュベートする工程、 e)外部磁場を使用して、混合物からシリカ質酸化物被覆磁気粒子を除去す る工程、及び、 f)粒子を溶出溶液に暴露する事によって、シリカ質酸化物被覆磁気粒子に 接着したプラスミドDNAの少なくとも60%を溶出させる工程、 を含む事を特徴とする方法。 23.工程(c)で形成された混合物中のカオトロピック塩が、塩酸グアニジン又 はチオシアン酸グアニジンから成るグアニジニウムカオトロピック塩である、請 求項22に記載のプラスミドDNA物質を単離する方法。 24.工程(c)で形成された混合物中のカオトロピック塩の濃度が、約0.1M 〜7Mである、請求項22に記載のプラスミドDNA物質を単離する方法。 25.シリカ質酸化物被覆磁気粒子を媒体から除去した後であって、該粒子からプ ラスミドDNA物質を溶出する前に、シリカ質酸化物被覆磁気粒子を洗浄する工 程を更に含む、請求項22に記載のプラスミドDNA物質を単離する方法。 26.洗浄工程が、アルコールと塩とを含む洗浄溶液を使用して行われる、請求項 25に記載のプラスミドDNA物質を単離する方法。 27.洗浄工程が、少なくとも30容量%のアルコールと緩衝液とを含む洗浄溶液 を使用して行われる、請求項25に記載のプラスミドDNA物質を単離する方法 。 28.工程(f)でシリカ質酸化物被覆磁気粒子から溶出されるプラスミドDNA が、更なる処理又は分析を妨害する様な高分子又は金属混入物を実質的に含まな い、請求項22に記載のプラスミドDNA物質を単離する方法。 29.媒体から生物学的目標物質を単離する為のキットであって、第一容器の水溶 液中に懸濁されたシリカ質酸化物被覆磁気粒子のアリコットを含む、粒子が、粒 子1mg当たり、生物学的目標物質の少なくとも2μgと可逆的に結合する能力 を有するキット。 30.第二容器にカオトロピック塩を、及び第三容器に洗浄溶液を更に含む、請求 項29に記載のキット。
