Abstract
PURPOSE: To obtain high-purity urokinase free from pyrogenous substances, by a combination of gel filtration with a crosslinked dextran and affinity chromatography with urokinase adsorbent.
CONSTITUTION: Crude urokinase (U) obtained from human urines is dissolved in a phosphoric acid buffer and passed through a crosslinked dextran gel column. The effluent of pyrogenous substances is discharged from the column befor U. The column is then connected to a urokinase adsorbent column, wherein a group of the formula (R is amidino or guanidino group; n is an integer 5W12) is linked to a water-insoluble carrier. The pure U flows out of the column and is adsorbed to the adsorbent column.
