Abstract

NEW MATERIAL:L-lysine α-oxidase which is an L-amino acid oxidase capable of producing α-keto-ε-aminocaproic acid, NH3 and H2O2, having low Km value to L-lysine, high substrate specificity, and a molecular weight of about 112,000 measured by sephadex G-200 filtration.
USE: Determination of lysine in foods and drinks; diagnosis of lysinosis, etc., biochemical reagent, etc.
PROCESS: An electrophoretically pure L-lysine α-oxidase is prepared by (1) culturing microorganisms belonging to Trichoderma genus and capable of producing L-lysine α-oxidase, (2) extracting the cultured cells with water, and (3) purifying the resulting crude enzyme solution by filtering or dialyzing in the presence of oxygen, thereby effecting the oxidative deamination of the product.
