Abstract
PURPOSE: To determine the activity of the titled substance, by adding a solution containing hydroxybenzoylcholine, o-hydroxybenzoic acid hydroxylase and NADH to the specimen, and determining the light absorbance of the consumed NADH by colorimetry.
CONSTITUTION: The hydroxybenzoylchloline of formula is used as the synthetic substrate, and a solution containing hydroxybenzoylcholine, o-hydroxybenzoic acid hydroxylase and NADH is added to a liquid containing cholinesterase. The o-hydroxybenzoic acid produced from the hydroxybenzoylcholine by the action of the cholinesterase is converted to catechol by the action of the o-hydroxybenzoic acid hydroxylase in the presence of NADH. The amount of the consumed NADH is determined as the decrease of the light absorbance of NADH by colorimetry, and the cholinesterase activity is calculated from the value.
