Abstract

PURPOSE: To purify the titled compound having peripheral blood vessel dilating activity and effective for the remedy of cardio-vascular diseases, easily, in high purity, at a large amount, by using an affinity adsorbent composed of an insoluble carrier, a spacer, and a specific ligand.
CONSTITUTION: An inactive Nα-tosyl-L-arginine-agarose or N-acetylphenylalanine- L-arginine-agarose is prepared by chemically bonding Nα-tosyl-L-arginine or N- acetylphenyl-alanine-L-arginine to agarose which is an insoluble carrier. The affinity chromatography is carried out using the resultant compound as the adsorbent, whereby kallikrein is specifically adsorbed and the other proteins and the similar enzymes are left unadsorbed. By this procedure, kallikrein is separated from the other proteins, and purified in high purity. The reaction is carried out at 0W30°C for 5W25hr.
