Abstract
PURPOSE: To obtain a β-(R)-aminoglutaric monoester, having an antimicrobial activity, and capable of being a synthetic intermediate for thienamycin, by hydrolyzing a dialkyl β-aminoglutarate with an ester hydrolytic enzyme.
CONSTITUTION: A dialkyl β-aminoglutarate of formulaIis treated with an esterase derived from a hog liver in a phosphoric acid buffer solution at a pH of 8.0 and 25°C for 1.5hr, and the resultant product is then extracted under neutral conditions and purified to give a monoalkyl β-(R)-aminoglutarate of formula II. The methyl and the ethyl esters exhibit an antimicrobial activity against staphylococci and is easily converted into a synthetic intermediate for thienamycin. Furthermore, the resultant monoalkyl ester can be converted into a β-(S)-aminoglutaric monoester by the ester interchange.
