Description

【発明の詳細な説明】 (産業上の利用分野) 本発明は、クロマトグラフに関し、より詳細には選択性
を改良し長寿命とした液体クロマトグラフ・カラムの新
規な充填サポートに関する。
(従来の技術) 血液、血清あるいは面禁中における疎水性交物の存在を
確認するとともに、これの濃度を測定することがしばし
ば必要である。このためには、液体クロマトグラフが有
効的な手段であり、これはいわゆる逆状相(疎水性)充
屓月を使用する。そのような充填材は試料から疎水性薬
物を吸着するが、試料に含まれる池の疎水性物質から薬
物を連続的に分別するのに妨げとなり易いタンパク質物
質も吸着ヅ−る。従って、予備的な試料の前処理操作を
実施する必要がある。−例として、人の血清の場合、試
料の前処理は現在幾つかの方法で行なわれている。
最も普通の方法では、タンパク質が沈澱され、上澄み液
は水と混合しない打(幾溶剤で油出され、有量溶剤は蒸
発により抽出物から除去され、そして被分析1カ(an
alyte )の残留物は高圧液体クロマトグラフ(H
PLC)による分析面に移動状相に戻される(  re
constrtutod> 。この方法は非常に時間が
かかり、費用的に不能率である。
現在採用されている第2の方法は、小さな使い后てカラ
ムにおいて、シリカに結合したオクタデシルシラン(O
DS)の逆状相充填材に被分析物を吸着させるものであ
る。この技法は自動化できるが、タンパク質が充填材に
残るため、カラムは1つの試料だけしか使用できず、結
果的にこの技法も多数の試料では費用的に不能率ひある
。
第3の方法では、シリカに結合されたオクタデシルシラ
ンの逆状相充填材が予備カラムに導入される。この予備
カラムは分析カラムから分離されているが、切換弁機構
により分析カラムに接続可能でおる。血清の試料は直接
予備カラムに注入され、この子偏カラムではタンパク質
が変性されるとともに蓄積され、除タンパクされた被分
析溶液は分別のために分析カラムに通過される。(Jぼ
こ3回の注入操作後、予[1iIiカラムはタンパク質
のTf5留物を除去するために逆洗しむけれに[ならな
い。この間欠的な逆i・λは多数の試431の場合、l
Li I::I的に不能率である。史には、タンパク質
はオクタデシルシラン・バッキングから完全に除去り゛
ることができないため、このバッキングは結果的には劣
化する。
(問題点を解決するための手段) 本発明は、充填材料の内面の疎水性と外面の親水性と被
分析物の試料との各々箕なる相互作用に基づき、被分析
物の試料を識別−りる)1タ体クロマトグラフ充填材料
を設(プるという点で新しい概念である。
内面逆状相(Is−RP)液体クロマトグラフ充填材料
を称呼しlこ本発明の新規な充填材料は、多孔サポート
であり、(1)多孔サポートの内面は疎水性分配状相を
結合し、(2)多孔サポートの外面は(例えば)人の血
漿に見られるタンパク質物質に対し非吸着性となる。よ
うに現水性となし、(3)多孔サポートの孔径はタンパ
ク質物質が疎水性分配状相を帯びる内面に立ち入らない
ようにサイズを制限しである。
本発明のl5−RP充J?7リボー1〜は、タンパク質
含有の生物学的風質(例えば人の血清または血漿)中の
疎水性低分子(例えば交換)を分離し、6逆状相高性能
クロマトグラフにより定量化す、る□前に従来必要とさ
れていた試料の前凱理操作の必要性を排除するように設
計されている。本発明の1S−RP充填材料は、市販の
孔径調整済み粒状シリカ(平均孔径は凡そ100オング
ストローム付近)から簡便に製造され、そのl5−RP
充填材料は親水性を帯びるとともにグリセロイルプロピ
によりタンパク質に対し非吸着性となし、それとともに
孔径を約80オングストローム以下、好すしくは40オ
ングストローム以下に縮小されている。
疎水性ポリペプチドの分配部分く例えば、アミノ酸フェ
ニルアラニン、トリプトファン、チロシン、ロイシン、
インロイシン、バリン、リジンおよび/またはアルギニ
ンを含む)は酵素分解を受は易い化学結合を含み、当業
者にとって周知の多種多様の反応を用いることにより、
水酸官能基を介してナボートの充@面にある幾分mのグ
リセロイルプロピル基に共有結合される。この誘導は多
孔微粒子へ浸透できる分子で実施されるため、微粒子の
内外面共疎水性分配状相を保有することになる。微粒子
の外面にある疎水性分子がタンパク質に対し吸着性を帯
びるようになるため、充填材はキモトリプシンやカルボ
キシペブチダーピなどのような酵素で処理される。これ
らの酵素は、微粒子の外面悲りから分子分配状相の疎水
性部分を分解するが、内面からは分解しない。これは多
孔微粒子の孔径が小さくてそのような大きな酵素が多孔
微粒子に浸入できないことによる。醇累氾理後に外面の
グリセロイルプロピルMに残留する化学的な官能性は、
最初のグリセロイルア1コビル結合物と同じくタンパク
質に対して非吸着性である。
その生成物がl5−RP充填サポートである。
本発明のl S −RP充填(Aを1皆造1する際の基
礎として、シリカやガラスなどを含む多数の出発物質が
利用できる。このような物質は以下に例示する表の通り

グリセロイルプロピル結合シリカは、内面逆法相充填材
の製造において、使用に適するジオール法相の一例であ
る。原則的には、充分な数の以下に示す有機水酸官能基
を含むものであれば、どのような多孔親水性基質(ma
[r!X)も使用でさる。

法でサポートまたは他の部分に結合される。別のり“ボ

多種多様の疎水性状相がうまく使用でさ、原則的にはポ
リペプチドに上記の疎水性のJ、Lを含むアミノ酸のい
かなる組合Vも含まれ、このポリペプチドは太さすぎて
内面に浸入できない酵素によって分解することができる
。予想される組合ぼにも殆ど制限がない。有用なポリペ
プチドには、ジペプチド、トリペプチド、さらにはく調
整が困難になるものの)テトラペプチド、およびペンタ
ペプチドを含む。実例のポリペプチドはグリシン・フェ
ニルアラニン・フェニルアラニン、グリシン・フェニル
アラニン・イソロイシン、グリシン・イソロイシン・イ
ソロイシン、グリシン・フェニルアラニン・トリプトフ
ァンなどを含む。ざらに有用なものとしては、N−ター
−ブトオキシカルボニルフェニルアラニン、N−カルポ
ベンズノtキシフTニルアラニン、アセデルフェニルア
ラニンおよびベンズオキシフェニルアラニンがあり、こ
れらはポリペプチドではないが、ポリペプチド同様に空
間的条件によって支配される。後者は好ましくはスペー
サとしてのテトラメヂレンジアミンを介してグリt?[
」イルプロピル基に結合される。
秤々の酵素が疎水性状相の分解に使用でさ゛る。
ペプチドが末端カルボン酸を介して支持基質(SUpp
orting  majrix)に付着されると、キモ
トリブシンはアミノ酸フェニルアラニン l−1ノブ1
〜フアン、J夕よびブーロジンに使用でき、一方トリブ
シンはリジンとアルギニンに使用できる。もしペプチド
が末端のアミンを介して支持風質に1」霜されると、ペ
プシンは隣接するアミノ酸フェニルアラニン、トップト
ノ1ン、ヂロシン、またはロイシンを介してペプチド仝
体を分解するのに使用でさる。
あるいは、好ましくはカルボキシペプチダーゼ△は、末
端カルボキシルペブヂド連gJを具1希するアミノ酸の
全てを連続的に(即ら順次)分解するのに使用’C&る
。たIどしリジンおよびアルギニンを除く、これらはカ
ルボキシペプチダーゼBにより分解される。
l5−RPサポートは液体クロマトグラフ・カラム内に
充填される。タンパク質と小さい疎水性被分析分子の両
方を含む試料が動的な液状非変性移動法相としてl5−
RPクロマトグラフ・カラムに流入さけると、タンパク
質はカラムの空隙部分で溶離する。これはタンパク質の
全体サイズが多孔l5−RP充填材の孔より人き(、ま
たl5−RPサポートの外面かタンパク1′1を吸着し
ないからである。小さな内因性の親水性分子(よl5−
RP多孔充填月に浸透できるが、内部疎水性法相で分配
されず、これも保持されない。しかしながら、l5−R
P多孔充填材に浸透できる小さな疎水性被分析分子は内
面逆法相で分配され、タンパク質と親水性の小さな分子
基質部分からの分離にQf+ l、、た時間で保持され
る。装置は、1ステツプで小さな疎水性被分析分子を複
錐なタンパク貿合有混合物から動的に分別することが可
能である。小さな疎水性被分析物は、それからさらに、
切換弁の手段により通常のオクタデシルシラン分析カラ
ムに尋人されて分離することができる。もしTS−RP
 15填材が高性能微粒子(≦10μm)でできている
ならば、タンパク質の分離と分析的な被分析物の分解を
同じl5−RPカラムで同時に行なうことができる。
従って、l5−RPサポート材を液体クロマトグラフ・
カラムに充填したとき、このサポート材は2つの機能を
果すことができる。第1に、l5−RP予備カラム(サ
ポートの粒径が凡そ30〜70tIJ)は、タンパク質
含有基質(即ち、血清と血漿)から疎水性の小さい分子
を抽出することにより1ステツプの動的な試料前処理技
法として使用できる。第2に、l5−RP分析カラム(
サポートの粒径が≦10〃)は、基質中のタンパク質か
ら疎水性の小さい分子を直接分離するのに使用でき、そ
れと共に小さい分子の被分析物を互いに分離することも
可能である。新規なナポート材のこれら2つの薇能性に
より、タンパク質含有の被分析混合物(例えば、血清や
血漿ンを、試料の前処理なしで、液体クロマトグラフ系
に直接注入することが可能である。小さい疎水性被分析
分子からのタンパク質高分子部分の分離は、サイズによ
る完全な除外と、1s−RPサポート充填材のもつタン
パク質の非吸着性の結果とし−(’ J5 @、その際
タンパク質をカラム外側空隙部分の中に分離し、疎水i
り内面での吸着により小ざい被分析分子を分離し、この
疎水性内面に小さい被分析分子が保持され、それによっ
てタンパク質部分から分N[される。
切換弁機構を介して典型的なオクタデシルシラン逆法相
HPtyC分析カラムに接続した小さい予備カラムにl
5−RP充填材を入れ、疎水性薬物を含む人の血漿の試
料を直接l5−RP予備カラムに注入すると、タンパク
買部分は完全に排除されて予備カラムの空隙部分に現わ
れ、それに対し疎水性薬物は内部充填領域への浸透およ
び疎水性コーティングへの吸着によってl5−RPカラ
ムに保持される。タンパク質部分は廃物として除去され
、動的に抽出された薬物部分は、問題の貼物被分析物を
さらに分離するために(切換弁を介して)HPLC分析
カラムに差し向けられる。
充填材を予備カラムに使用したときの利点は、(1)試
料の面処理操作がなくなること(即ち、タンパク質の沈
澱がない、仙111がないなど)、(2)l−IPLC
分析カラムを内因tr[(endogenous )タ
ンパク質による汚染から保護すること、J5よび安全性
(航速充填材料は、被分析物としてノエニトインを抽出
する能力上の性能低下を示すことなく、はぼ100個の
人の血漿試r1に使用された)である。
l5−RP充填材が小さい微粒子サポート(即ち、O径
が≦10u)で作られ、カラムにかなりの長さく15〜
25cm)充填寸れば、血uU試料を直接71人した際
、タンパク質の蓄積によるカラム劣化を招くことなく、
タンパク質の除去と被分析物の高訃能な分析的分離の両
方を同時に行なうことが可能となる。
I S −RP 払では、*1青タンパク質がカラム充
填材に保持されないため、充填されたカラム(予備カラ
ムや分析カラム)は多種多様の試別について反復して沈
用できる。クロマトグラフ系の一部としてl5−RP充
填材を含ませることにより、血清や血漿の試料の直接注
入をr4′う完全自動化が可能となる。この技法は完全
に新規且つ独特の方法で血清や血漿試料の前処理の問題
を解決する。
本発明の↑要な商業的適用は、製薬業界で行なわれてい
る血清および血漿中のトI P L C祭物検査にある
であろう。−例として、−製薬会社では、年に約30.
000の血清試料についてHP L C分析を行なって
いるそうである。そのような操r1三を改良することの
重要性が明らかである。。
以下の操作実施例で本発明を説明する。実施例は説明に
役立つのみで、これに限定されるものではない。

疎水性分配状相 2種類の充填材を作った。これらの充填材における疎水
性分配状相は、端末グリシン部分(gly )と1個ま
たは2個のし一フェニルアラニン(L−phe)部分を
含むそれぞれジペプチドどトリペプチドである。周知の
カルボキシルジイミダゾール(CD I )結合法を用
いた場合、ペプチドは既にCP G / 40  G 
1ycophascサボー1−に存するグリセロイルプ
ロピル官能基を介して該CP G / 40 G 1y
cophascサポートに付着された。反応の概略を添
附図面の第1図に示す。

seの外面にあるポリペプチド部分は、カルボキシペプ
チダーゼへ(アミノ酸連鎖のカルボキシル末ψ本基を加
水分解するエキソベブチターU〉で分解して除去した。
カルボキシペプチダーピ八は、他のアミノ酸に比べ、芳
香族アミノ酸の分解にJ5いて最も活性であり、従来よ
りアミノ酸のシーケンシング(5equenc i n
o >においてソリッドサポ−ト(solidsupp
ort )からアミノ酸を除去1ノることが実IすnT

的とした過去の研究では、タンパク質の浸透を許容する
ために、500オングストローム程麿の大きな孔径のソ
リッドサポートが使用されていた。しかし、ここに述べ
た適用例でit、ペプチド結合CρG / 40  G
 Iycopl+aseサボー1−の孔芹tユ、約40
オングストロームである。そのため、人さ゛なカルボキ
シペプチダーゼA(4よば34,000ダルトン)は充
填材の最外部分に留められ、微粒子のタト面からのみポ
リペプチドを分解する。
M索分解手順 0.01 NII IJ ンaFatiN液(D)−1
7,8>2mlにおいて、約1gのペプチド結合CPG
、/40  G+ycophaseナポートを1111
0のカルボキシペプチダーゼ八と化合させ、分解が本質
的に完了する37℃でほぼ12時間時々振盪しながら培
養させた。
それから、0.01 Mオルトリン酸塩緩衝液5Qml
、次いで1.0M塩化ナトリウム水溶液5Qmlでl5
−RPサポートから酵素を洗い出した。次に、サポート
は、分解したフェニルアラニンを除去するために、順次
501のH2O,50m1の蒸溜メタノール、および5
0m1のH2Oで洗浄した。

定酵素処理の前と後においてCPG/40  Gl)1
cophascに結合したポリペプチドの屯を求めるた
めに、フェニルアラニン部分を溶液中に解離させるよう
に充填材を酸の加水分vf?、(6N  l−I CL
、24時間、100℃)に晒した。その後、254nn
lの光学検知(optical dejec目00)目
通法相HPLCによるW11フェニルアラニン部分を定
房化により、CP G / 40  G 1.ycop
hase lナボートに結合したOを求めた。典型的な
実例どして、単一合成をした充填材は、酵素処理の前に
はジペプチドを1グラム当たりQ、  17mmol、
トリペプチドを1グラム当たり0 、04 mmol含
み、また酵素処即後にはそれぞれ0.10ml1lol
および0.02mm01含んでいた(第1表)。
反復合成 もし内面における疎水性状相の被覆率が、酵素分解後に
おいて、被分析物の充分な保持にとって必要な充分な疎
水性を表面にイ・」与するのに不充分であったら、同じ
材料について合成と酵素分解を繰り返した。
モ5ルl5−RPオミの性l評1「 <a )試験被分析物 モデルl5−RP材料の性能を評価するのに使用した疎
水性被分析分子はフェニトイン(5,5−ジフェニルヒ
ダントイン)であった。
(b )試験高分子基質 タンパク質基質は純化した標準釣人の血清アルブミンと
人の面素を含んでいた。
(c )液体クロマトグラフ系 モデルl5−RP材料は、添附した第2図に示す通り、
普通のオクタデシルシラン分析カラムと共に予備カラム
抽出モードで試験した。1s−RP材料は、内径3 m
mx長さ100IIIIllのガラス製予備カラム(A
)の中に充頃した。前記予備カラム(A)は、6ポート
・スライダー注入弁(Vl)と25cmの分析用オクタ
デシルシランカラム(C)に接続した6ボート切換弁(
V2)との間に配設した。分析用オクタデシルシランカ
ラムの短い5am部分は、分析用カラムへの移送に際し
、l5−RP予備カラムから溶離フェニトインを捕捉づ
るために弁v2と共に使用した。
(〔1)試験方法 フェニトイン被分析物をa有覆るタンパクr′1試・ 
料基質は、試料に溶は合う溶離剤液S1のポンプ(Pl
)連転(0,1M71ルトリン酸塩10.21VtNa

)を介してl5−RPP備カラムに導入した。溶離剤S
1は、l5−RPP備カラム、弁(V2)(点線で示し
た位置にある)、および低圧抽出系を溝成する検出Z(
Di)を通過した。それと同時に、溶離剤S2 (20
%アセトニトリル/33%メタノール/47%0゜12
Mオルトリンll!2塩緩衝液、pト1〜6.O)を、
ポンプ(P2)により弁(V2>(点線[)γ置)を介
して1 、0ml/minの流量でオクタデシルシラン
カラムB、Cを経て高圧分析分離系を溝成する検出器D
2に送出した。
試料を導入すると、タンパク質はl5−RP予予力カラ
ムA、)の空隙で澄離し、弁V2(点線位置)を介して
検出器D1に至る。タンパク質の溶離後、弁V2は切換
り(実線位置>、rs−Rpカラムに保持されていた疎
水iil波分1斤1勿はカラムBに移し換えられる。全
ての被分析物かカラムBに捕捉されると、切換弁V2は
元の状rぶ(点線位置)に復帰され、被分析物はオクタ
デシルシランカラムでの分析分離および検出器D2によ
る定ぢ1化のために右段性移動法相($2)でカラムC
に溶離される。
8K M1及 2つのモデルl5−RP充填祠の性能評師、共に酵素処
理した(Is−RP)場合と共に酵素α理しない(ES
−TR−RP)場合のジペプチド(G40−oph 、
 )およびトリペプチド(G 40gph 2)のデー
タを第1表に示す。

人の血清タンパク質を吸着なしで系に通過さぼるl5−
RPP填材の能力は、(i )サポート面におけるペプ
チド部分に対する残留クリセロイルプロピル基の相対比
、  (ii)血清タンパク質に対するペプチドの準融
和性、(iii)酵素分解により外面から除去されるペ
プチドの有限F?q、  (iv)充填材の小ざな公称
孔径(≦40オンゲス[・ローム)を含む特性および処
理の特異な組合ゼによって支配される。CP G / 
’40  G 1ycophase I、:、おけるグ
リレロイルプロピル結合法相の初明表面被mElが33
9 μmol /+11の場合、G 40−oph 。
はペプチド法相の総被覆笛(内外)として166μmo
l 7g  (49%)を生じ、これの42%は酵素分
解により除去された。G40−IJI)112は42μ
mol /a  (,12%)の総ペプチド被覆邑を産
出し、これの61%は酵素分解によって除去された(第
1表)。疎水性ペプチドが外面に残ると、タンパク質の
吸着性が大きくなるとともに充屓月からのタンパク質の
回収が低くなることが観察される。血清タンパク質のカ
ラl\に残る比率が最小であっても最高性能とならない
が、これはタンパク質の蓄積につれ被分析物を保持する
カラムの8退が次第に1員失づるためである。疎水性ペ
プチドを除去するためにlff?索で処理しなかったサ
ボーl−からは、水性移動法相での溶離による標準釣人
の血tXiアルブミン(+−I S A )の回収率は
約80乃至95%の範囲であった。酵素で処理したl5
−RPザボートからのHS Aの回収率は全ての場合、
100%±2%であった。
UF−aA導CPG/40  Glycophasc 
(即ち、ポリペプチドを添加しない)からの人の血漿お
よびフェニトインの溶離を第3図に示す。
トリペプチドで誘導したCPG、/40GIycoDh
aseからの血漿およびフェニトインの溶離を、酵素迅
理(ES−Is−RP)により外面から疎水性法相を除
去しない場合につぎ、第4図に示す。
本発明のモデルl5−RPカラムにおける人の血漿およ
びフェニトインの溶離を第5図に承り。
外面にペプチド法相を伴う充填材に残る極端なテーリン
グ(tailin(] )  (第4図)は、疎水性被
分析物の溶離をひどく不鮮明にする。従って、被分析物
は未処理ノコラムではタンパク質から分l1i1するこ
とができなかった。
l5−RPfJラムからのタンパク質の溶離はそのよう
なテーリングを示さず(第5図)、従って枝分<l’r
物の分離が可能である。
ペプチドカラムからのフェニトイン被分析物の溶離を第
4.5図に示す。完全にペプチドで被覆されたサポート
の8但ファクタ(k′)はl5−RPカラムより大きく
(第1表)、またトリペプチド(G40−(It)11
2 > (1)容量フy’/夕ハ、予想通りジペプチド
(G40−(]I)h l >より大きかった。1s−
RPカラムにおけるフェニトインの保持は、カラムの切
換えで血漿タンパク質からフェニトインを分離するのに
十分であった。
l5−RPカラムの効率は、非酵素処理材料の効率より
潰れていることが判った(第1表)。これは外面からペ
プチド部分を除去して孔の入口領域への出入口が広くな
ったことによると思われる。
Is  RP  G40 0phzにより人の血漿から
フェニトインを定量的に抽出することは、rS−RPカ
ラムからの溶離後にオクタデシルシランカラムB(第2
図)にフェニトインを捕捉する事により実証された。続
いて、オクタデシルシランカラムC(第2図)に捕捉さ
れた被分析物をシャンティングづるが、これで254 
nmの一定波長を検出してピーク高さを測定することに
よりフェニトインの)−IPLc定■化(第6図)が可
能となる。
この方法(より、発生した補正曲線は、メタノール中の
フェニトインの濃度範囲が5μmし乃至25μ(J/m
Lであり、第7図に示す。
人の血漿中のフェニトインについて同じ濃度範囲に亘る
直線性が第8図に示される。
l5−RP法による抽出で人の血漿からのフェニトイン
の回収は、フェニトインをメタノール中に溶解させ同じ
l5−RP法で抽出した場合と比べると、タンパク質基
貿に付加されたフェニトインの約97乃至100%の範
囲にあった。〕にI〜インタンパク貿が90%以上人の
血清アルブミンに結合することが知られているため、こ
れはタンパク質結合のフェニトインがl5−RPカラム
への導入で解放されることを意味する。限外濾過の研究
によれば、この解放は水性移動法相における成分の置換
によらないこと、及び高率のフェニトインが本当にl5
−RPカラムへの注入に先立らタンパク貿基質に結合さ
れたタンパク質であったことが判る。最初は、l5−R
Pカラムはタンパク質風質から遊離非結合フェニトイン
のみを抽出するのであろうと予想されていた。しかしな
がら、モデルl5−RPカラムは全濃度の非分析物が抽
出されることを示している。rS−RPカラムがどうし
て人の血清アルブミンからのフェニトインの解放を亢進
するかという正確な理由は明らかではない。しかしなが
ら、これはモデルl5−RPカラムの特異な特性である
ことが明白である。
I 5−It P予備カラム抽出法により人の血漿試料
から全フェニトインg!瓜を定Ih化する相対的な精度
は平均で、存在する被分析物の♀の2,1%であること
が判っており、それに対し反復測定の精度(即ら「変動
係数」)は、平均で2,5%であることが判った(第2
表に示す〉。
Is  RP  G40 0Dh2モデルカラムは、約
100個の人の血漿試料につきフェニトインの分析に使
用され、カラム性能の劣化または変化の兆候は何も観察
されなかった。
本発明のl5−RP充唄サポートは、液体クロマトグラ
フ材料において新しい概念を捉供する。
疎水性ペプチド結合状相を伴う小さい疎水性被分析物分
子の分配作用は、直径を≦80オングストローム、好ま
しくは≦40オンゲス1−ロームに調整した孔をもつ剛
性親水性サポートの内面に制限され、これにより大きな
高分子タンパク貿を含む基質から小さな疎水性被分析物
を動的に1ステツプで分離することが可能となる。l5
−RP充充填車ボー1〜設計、及びサポートの外面から
疎水性分配状相を除去するのに酵素を利用するという基
本的な概念は、液体クロマトグラフ充填材の中で特異で
ある。

より外面から疎水性法相を除去しない場合にJ3いて示

Claims (19)

    【特許請求の範囲】
  1. (1)充填材料へのタンパク質物質の浸入を阻止するの
    に十分に小さい孔径をもつ多孔サポートと、該多孔サポ
    ートの内面にのみ結合した疎水性分配状相(parti
    tioning phase)と、該多孔サポートの外
    面に結合した親水性分配状相とからなり、被分析混合物
    中のタンパク質物質が該充填材料により拒絶されるもの
    の、該被分析混合物の疎水性成分が該充填材料に吸着さ
    れ保持されることを特徴とする液体クロマトグラフ・カ
    ラムの内面逆状相充填材料。
  2. (2)前記多孔サポートがシリカである特許請求の範囲
    第1項記載の充填材料。
  3. (3)前記多孔サポートがガラスである特許請求の範囲
    第1項記載の充填材料。
  4. (4)孔径が約80オングストロームに等しいか、また
    はそれ以下である特許請求の範囲第1項記載の充填材料
    。
  5. (5)孔径が約40μm以下である特許請求の範囲第1
    項記載の充填材料。
  6. (6)粒径が約10μm以下である特許請求の範囲第1
    項記載の充填材料。
  7. (7)前記親水性分配状相がシリカに結合したグリセロ
    イルプロピル基からなる特許請求の範囲第1項記載の充
    填材料。
  8. (8)前記疎水性分配状相がグリセロイルプロピル基を
    介してシリカに結合したポリペプチド基からなる特許請
    求の範囲第1頃記載の充填材料。
  9. (9)前記ポリペプチド基がグリシン−L−フェニルア
    ラニンである特許請求の範囲第8項記載の充填材料。
  10. (10)前記ポリペプチド基がグリシン−L−フェニル
    アラニン−L−フェニルアラニンである特許請求の範囲
    第8項記載の充填材料。
  11. (11)多孔サポートの内外面に親水性分配状相を結合
    すること、それにより該多孔サポートの孔径が充填材料
    へのタンパク質物質の浸入を阻止するのに十分小さいサ
    イズに縮小されることと、該多孔サポートの内外面にあ
    る該親水性分配状相に疎水性分配状相を結合することと
    、該多孔サポートの外面から該疎水性分配状相を酵素で
    除去することとからなることを特徴とする液体クロマト
    グラフ・カラムの内面逆状相充填材料を製造する方法。
  12. (12)前記親水性分配状相が結合有機水酸基からなる
    特許請求の範囲第11項記載の方法。
  13. (13)前記親水性分配状相がシリカに結合したグリセ
    ロイルプロピル基からなる特許請求の範囲第11項記載
    の方法。
  14. (14)前記疎水性分配状相がグリセロイルプロピル基
    を介してシリカに結合したポリペプチド基からなる特許
    請求の範囲第11項記載の方法。
  15. (15)前記ポリペプチド基がグリシン−L−フェニル
    アラニンである特許請求の範囲第14項記載の方法。
  16. (16)前記ポリペプチド基がグリシン−L−フェニル
    アラニン−L−フェニルアラニンである特許請求の範囲
    第14項記載の方法。
  17. (17)前記疎水性分配状相がカルボキシペプチダーゼ
    Aでの処理により前記多孔サポートの外面から除去され
    る特許請求の範囲第11項記載の方法。
  18. (18)前記疎水性分配状相がキモトリプシンでの処理
    により前記多孔サポートの外面から除去される特許請求
    の範囲第11項記載の方法。
  19. (19)充填材料へのタンパク質物質の侵入を阻止する
    のに十分小さいサイズの孔径を有し、該充填材料の内外
    面に親水性分配状相を結合した多孔サポートで出発し、
    該多孔サポートの内外面にある該親水性分配状相に疎水
    性分配状相を結合することと、該多孔サポートの外面か
    ら該疎水性分配状相を酵素で除去することとからなるこ
    とを特徴とする液体クロマトグラフ・カラムの内面逆状
    相充填材料を製造する方法。
