Abstract
본 발명은 피토스핑고신 유도체를 포함하는 기능성 화장료 조성물에 관한 것으로, 더욱 상세하게는 UV-필터링 물질과 더불어 피토스핑고신 유도체인 피토스핑고신-1-포스페이트(PhS-1-P)를 활성 성분으로 함유함으로써, 자외선에 의한 피부 노화를 방지하고 피부의 산화적 스트레스를 억제하며 세포 활성화 효능을 구현할 수 있는, 기능성 화장료 조성물, 피부 보호용 조성물, 피부 노화 방지용 조성물 또는 피부 질환 예방용 조성물에 관한 것이다.
Description
본 발명은 피토스핑고신 유도체를 포함하는 기능성 화장료 조성물에 관한 것으로, 더욱 상세하게는 UV-필터링 물질과 더불어 피토스핑고신 유도체인 피토스핑고신-1-포스페이트(PhS-1-P)를 활성 성분으로 함유함으로써, 자외선에 의한 피부 노화를 방지하고 피부의 산화적 스트레스를 억제하며 세포 활성화 효능을 구현할 수 있는, 기능성 화장료 조성물, 피부 보호용 조성물, 피부 노화 방지용 조성물 또는 피부 질환 예방용 조성물에 관한 것이다.
저파장(100~400nm)에 존재하는 자외선은 광-기인성 피부반응의 주된 원인이 되고 있다. 구체적으로 단파장 자외선(290~320nm)인 UV-B는 표피층까지만 도달하고 이에 과다 노출시 화상에 해당하는 홍반이 일어난다. 장파장 자외선(320~400nm)인 UV-A는 파장이 상대적으로 길어 유리창을 잘 통과하며 피부에서는 진피층까지 도달하여 피부 흑화나 광노화를 일으킨다.
자외선에 노출시 초래되는 여러 가지 피부 손상을 방지하고, 피부를 보호하기 위해 다양한 종류의 화장품들이 사용되고 있다. 특히 피부에 손상을 주는 자외선에 대한 효과적인 보호를 위해 UV-A와 UV-B를 동시에 차단할 수 있는 화장품들이 선호되고 있다. 현재까지 안전성이 확보된 자외선 흡수제로는 벤조페놀 유도체, p-아미노안식향산 유도체, p-메톡시계피산 유도체 및 살리실산 유도체 등을 들 수 있으며, 이들 유기물에 흡수된 자외선 에너지는 열 에너지로 변환된 후 방출되는바 피부와의 접촉을 최소화할 수 있다.
피부 노화의 유력한 메커니즘은 자유라디칼 생성에 따른 신체의 산화 또는 '산화적 스트레스'라고 알려져 있다. 자유라디칼은 전자를 잃게 되는 거의 모든 분자로부터 만들어지며, 이것은 세포에서 자유에너지가 안정적인 산소와 결합할 때 세포호흡과정 중 미토콘드리아에서 생성된다. 세포는 산소를 ROS(reactive oxygen species) 혹은 과산화수소(H2O2)로 전환하는 효소를 가지고 있다. 특히 ROS는 세포를 둘러싼 유체 막에서 불포화 지방을 산화시키기 때문에 이로 인해 유체 막은 세포를 적절하게 보호하지 못하게 된다. 또한 몇몇 종류의 산화된 지방이 세포로 유입되면 독성화학물질을 발생시키게 되고 그로 인해 아포토시스(apoptosis)를 유발하게 된다. 또한 ROS는 피부의 지지 조직을 분해하는 단백질 효소인 콜라게나아제(collagenase)를 증가시켜 피부의 견고함을 떨어지게 한다.
이에, 최근 다양한 원료와 UV 흡수제를 적용하여 피부의 항노화 효능을 도모하려는 기능성 화장품에 대한 연구가 다수 이루어지고 있으나, 아직까지도 만족할만한 자외선 차단 효과 및 피부 노화 방지 효과를 구현한 제제는 개발되지 못하고 있다.
본 발명은 상기와 같은 종래기술의 문제점을 해결하고자 한 것으로, 자외선을 효과적으로 차단함과 더불어 산화적 스트레스를 억제하여 우수한 피부 노화 방지를 구현할 수 있는 새로운 기능성 화장료 조성물을 제공함을 기술적 과제로 한다.
상기한 기술적 과제를 달성하고자, 본 발명은 피토스핑고신-1-포스페이트(PhS-1-P)를 활성 성분으로 포함하는 기능성 화장료 조성물을 제공한다.
구체적으로, 본 발명은 피토스핑고신-1-포스페이트(PhS-1-P); 및 UV-필터링 물질;을 포함하는 것을 특징으로 하는 기능성 화장료 조성물을 제공한다.
또한, 상기 조성물은 O/W형, W/O형, O/W/O형 또는 W/O/W형의 에멀젼 또는 마이크로에멀젼 타입으로 제형화된 것임을 특징으로 하는 기능성 화장료 조성물을 제공한다.
또한, 피토스핑고신-1-포스페이트(PhS-1-P); 및 UV-필터링 물질; 외에 피부 연화제; 점증제; 계면활성제; 보존제; 피부컨디셔닝제; 유화제; 보습제; 중화제; 및 물;을 더 포함하는 것을 특징으로 하는 기능성 화장료 조성물을 제공한다.
구체적으로, 피토스핑고신-1-포스페이트(PhS-1-P); 옥틸 메톡시신나메이트; 호모살레이트; 코코-카프릴레이트/카프레이트; 아이소스테아릴 네오펜타노에이트; PEG-40-스테아레이트; 세틸알코올; 메틸파라벤 또는 프로필파라벤; EDTA-이나트륨 염; 다이에탄올아민 세틸포스페이트; 아크릴레이트/C10-C30 알킬아크릴레이트-크로스폴리머; 1,2-프로판다이올; 수산화칼륨; 및 탈이온수;를 포함하는 것을 특징으로 하는 기능성 화장료 조성물을 제공한다.
또한, 피토스핑고신-1-포스페이트(PhS-1-P) 0.001~10 중량%; UV-필터링 물질 0.1~30 중량%; 피부 연화제 1~20 중량%; 점증제 0.1~10 중량%; 계면활성제 0.1~10 중량%; 보존제 0.1~10 중량%; 피부컨디셔닝제 0.1~10 중량%; 유화제 0.01~20 중량%; 보습제 0.5~8 중량%; 중화제 0.01~8 중량%; 및 물 30~70 중량%;를 포함하는 것을 특징으로 하는 기능성 화장료 조성물을 제공한다.
또한, 본 발명의 다른 측면으로, 피토스핑고신-1-포스페이트(PhS-1-P); 및 UV-필터링 물질;을 포함하는 피부 보호용 조성물, 피부 노화 방지용 조성물 및 피부 질환 예방용 조성물을 제공한다.
본 발명의 조성물은 우수한 자외선 차단능, 피부의 산화적 스트레스 억제능 및 세포 활성화 효능을 지녀 광 노출에 의해 야기되는 피부의 노화를 방지하고 피부를 효과적으로 보호할 수 있다.
본 발명의 조성물은 피토스핑고신-1-포스페이트(PhS-1-P)를 자외선에 대한 피부 보호 및 노화 방지를 위한 활성 성분으로 포함한다.
즉, 본 발명은 피토스핑고신 유도체인 피토스핑고신-1-포스페이트(PhS-1-P)를 광보호를 위한 유효량으로 포함하는 조성물로서, 전형적으로 자외선 방사, 특히 태양광 방사에 민감한 대상, 예컨대 피부를 효과적으로 보호할 수 있다.
본 발명의 조성물은 적합한 지지체 내지 기제로 제형화되어 다양한 광 흡수제, 예컨대 외피용약 및/또는 제제, 특히 피부 보호 및 일광 차단을 위한 생활 화장품으로 매우 적합하게 사용될 수 있다.
본 발명의 활성 성분인 상기 피토스핑고신-1-포스페이트(PhS-1-P)는 혈소판에서 분비되는 스핑고신-1-포스페이트(S1P)의 구조 유사체로서, ROS에 의한 산화적 스트레스를 억제하여 효과적으로 항노화 특성을 나타내게 한다.
상기 피토스핑고신-1-포스페이트(PhS-1-P)는 본 발명의 조성물 총 중량을 기준으로 0.001~10 중량%(더욱 상세하게는, 0.005~1 중량%) 포함되는 것이 적절하다.
구체적으로, 본 발명의 기능성 화장료 조성물은 피토스핑고신-1-포스페이트(PhS-1-P); 및 UV-필터링 물질;을 동시에 포함하는 것이다.
상기 UV-필터링 물질은 자외선을 차단(흡수)하는 성분으로서, 특히 UV-A 차단제, UV-B 차단제 및 이들의 혼합물 중에서 선택된 차단제 원료를 사용할 수 있다.
구체적으로, 상기 UV-필터링 물질은 친수성 또는 친유성을 지닌 UV-A 또는 UV-B 차단제이거나 이들의 혼합물일 수 있으며, 예를 들어 실온에서 액상인 호모멘틸산살리실레이트(INCI: 호모살레이트); 2-에틸헥실-2-시아노-3,3-디페닐아크릴레이트(INCI: 옥토크릴렌); 2-에틸헥실-2-하이드록시벤조에이트(2-에틸헥실살리실레이트, 옥틸살리실레이트, INCI: 옥틸 살리실레이트); 및 신남산의 에스테르, 바람직하게는 4-메톡시신남산(2-에틸헥실)에스테르(2-에틸헥실-4-메톡시신나메이트, INCI: 옥틸 메톡시신나메이트) 및 4-메톡시신남산이소펜틸에스테르(이소펜틸-4-메톡시신나메이트, INCI: 이소아밀 p-메톡시신나메이트); 중에서 선택된 1종 이상을 UV-필터링 물질로 사용할 수 있다.
상기 UV-필터링 물질은 조성물 총 중량을 기준으로 0.1~30 중량%(상세하게는 0.5~20 중량%, 더욱 상세하게는 1~10 중량%) 포함되는 것이 적절하다.
본 발명에 따른 활성 성분인 피토스핑고신 유도체를 함유하는 조성물은 다양한 형태의 화장품 및 피부 질환 예방/개선용 제품으로 제형화될 수 있다.
예를 들어, 로션, 점증화된 로션, 젤, 크림, 밀크, 연고, 분말 또는 고체 튜브 막대의 형태로 제공될 수 있고, 선택적으로 에어로졸의 형태로 포장될 수 있으며, 무스(mousse), 폼(foam) 또는 스프레이(spray)의 형태로 제공될 수도 있다. 또한 본 발명에 따른 조성물이 UV 조사에 대해 인체 표피를 보호하기 위한 또는 일광 방지/일광 차단을 위한 조성물로 사용될 경우, 용매 또는 지방 물질 중의 현탁액 또는 분산액 형태로 제공될 수 있고, 선택적으로 크림 또는 밀크와 같은 에멀젼 또는 마이크로에멀젼, 수포성 분산액의 형태가 될 수 있으며, 연고, 젤, 고체튜브 막대 또는 에어로졸 무스의 형태로 제공될 수 있다.
구체적으로, 본 발명의 조성물은 용액, 워터-인-오일(W/O) 타입의 에멀젼, 오일-인-워터(O/W) 타입의 에멀젼, 예컨대 워터-인-오일-인-워터(W/O/W) 타입 또는 오일-인-워터-인-오일(O/W/O) 타입과 같은 다중 에멀젼, 하이드로분산액 또는 리포분산액, 겔, 고체 스틱, 피부 투과성 치료 시스템 또는 에어로졸의 형태로 제품화될 수 있다.
이때, 본 발명의 조성물은 소정의 유화제(유화안정제)를 사용하여 O/W형, W/O형, O/W/O형 또는 W/O/W형의 에멀젼/마이크로에멀젼 타입으로 제형화될 수 있다.
상기 유화제로는 1종 이상의 합성 중합체, 예컨대 아크릴레이트/C10-C30 알킬아크릴레이트-크로스폴리머를 0.01~20 중량%(더욱 상세하게는, 0.1~20 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
본 발명의 조성물은 상기 피토스핑고신-1-포스페이트(PhS-1-P) 및 UV-필터링 물질 외에, 화장품 내지 일광 차단/일광 방지 제품에서 통상적으로 허용될 수 있는 보조제, 첨가제 및 부형제, 예컨대 피부 연화제, 점증제, 계면활성제, 보존제, 피부컨디셔닝제, 유화제, 보습제, 중화제, 물, 지방 물질/오일, 미용적으로 허용되는 유기 용매, 실리콘, 유연제, 추가의 일광 차단제, 소포제, 방향제, 충진제, 격리제, 음이온성, 양이온성, 비이온성 또는 양쪽성 중합체 또는 이들의 혼합물, 산성화제 또는 염기화제, 염료, 착색제, 안료 또는 나노안료 등을 더 포함할 수 있다. 특히 자외선 방사를 물리적으로 차단함으로써 추가의 광보호 효과를 제공하기에 적합한 보조제 및 첨가제, 예컨대 자외선 산란제를 추가로 포함할 수 있다.
상기 피부 연화제는 피부의 수화 또는 재수화를 유지하기 위한 습윤제로서, 피부에 보호막(protective coating)을 제공하여 피부로부터 물이 증발되는 것을 막는 역할을 한다. 피부 연화제는 추가적으로 피부 표면에 유연 또는 진정 효과를 제공하며, 이의 국소적인 사용은 일반적으로 안전한 것으로 간주된다.
상기 피부 연화제로는 코코-카프릴레이트/카프레이트를 1~20 중량%(상세하게는 2~15 중량%, 더욱 상세하게는 4~12 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
본 발명의 조성물의 수성 상(phase)은 1종 이상의 점증제를 포함할 수 있다. 상기 점증제는 제형에 적합한 점조도를 제공하는 역할을 한다.
상기 점증제로는 아이소스테아릴 네오펜타노에이트를 0.1~10 중량%(더욱 상세하게는, 1~5 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
본 발명의 조성물이 에멀젼으로 제형될 경우 음이온성, 비이온성, 양이온성 또는 양쪽성 계면활성제가 포함될 수 있다.
상기 계면활성제로는 PEG-40-스테아레이트 및 세틸알코올 중에서 선택된 1종 이상을 0.1~10 중량%(더욱 상세하게는, 1~5 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
상기 보존제는 본 발명의 조성물이 부패 내지 산화되는 것을 방지하는 역할을 한다.
상기 보존제로는 메틸파라벤, 프로필파라벤 및 EDTA-이나트륨 염 중에서 선택된 1종 이상을 0.1~10 중량%(더욱 상세하게는, 0.1~1 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
상기 피부컨디셔닝제는 피부를 부드럽고 유연하게 유지하는데 도움을 주는 역할을 한다.
상기 피부컨디셔닝제로는 다이에탄올아민 세틸포스페이트를 0.1~10 중량%(더욱 상세하게는, 0.1~5 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
상기 보습제는 물과 결합함으로써 피부 표면에 수분을 유지시켜 주는 습윤제 역할을 한다.
상기 보습제로는 1,2-프로판다이올, 글리세린, 폴리프로필렌글리콜, 폴리에틸렌글리콜, 락트산, 피롤리돈카복실산, 유레아, 인지질, 콜라겐, 엘라스틴, 세라마이드, 레시틴, 솔비톨 및 PEG-4 중에서 선택된 1종 이상을 0.5~8 중량%(더욱 상세하게는, 1~5 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
상기 중화제는 유화제 또는 발포증강제/안정제와 같은 성분을 중화시키는 역할을 한다.
상기 중화제로는 수산화칼륨을 0.01~8 중량%(더욱 상세하게는, 0.5~5 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
상기 물은 전술한 기능성 성분들을 균일하게 분포시키는 용매 역할을 한다.
상기 물(정제수)로는 탈이온수를 30~70 중량%(더욱 상세하게는, 40~60 중량%)의 함량으로 사용할 수 있으나, 반드시 이에 한정되는 것은 아니다.
또한, 본 발명의 조성물에는 지질 상(phase)으로서 광물성 오일 및 광물성 왁스, 실리콘 오일, 예컨대 다이메틸폴리실록산, 다이에틸폴리실록산, 다이페닐폴리실록산, 사이클로메치콘 및 이들의 혼합물이 포함될 수 있다.
본 발명의 피토스핑고신-1-포스페이트(PhS-1-P) 및 UV-필터링 물질을 포함하는 조성물은 화장품을 비롯해 피부 보호(케어), 피부 질환 예방 및 노화 방지를 위한 다양한 기능성 제품군에 적용될 수 있다.
예를 들어, 본 발명의 화장료 조성물은 일광 방지/일광 차단 조성물 또는 메이크업 용품(make-up product)과 같이 자외선(예컨대, UV-A) 조사로 인한 손상으로부터 인체 표피를 보호(케어)하기 위한 조성물로서 유용하다. 특히 본 발명의 조성물은 UV 차단 효과와 더불어 산화적 스트레스를 억제함에 따라 피부 노화를 방지하는 상승적 효과를 발휘하는바, 항노화에 특화된 UV 차단제로서 매우 적합하게 사용될 수 있다.
이하, 실시예를 통해 본 발명을 보다 구체적으로 설명한다. 그러나 하기 실시예는 본 발명의 이해를 돕기 위한 것일 뿐 어떠한 의미로든 본 발명의 범위가 이들 실시예로 한정되는 것은 아니다.
실시예: 피토스핑고신 유도체를 함유하는 광범위 스펙트럼 일광 차단 로션의 제조
하기 표 1에 기재된 함량에 따라 각 성분을 균일하게 배합하여 에멀젼 타입의 로션을 제조하고, 이를 인체 피부에 적용한 다음, 태양광에의 장기 노출에 따른 피부 손상 여부를 조사하였는바, 본 발명의 기능성 화장품은 자외선에 기인한 피부 노화 방지에 매우 효과적인 것으로 확인되었다.
