Claims (3)
- 5.5 ± 0.5의 pH에서
- 인간 IgG1 서브클래스의 불변 중쇄 영역을 포함하는 120 mg/mL ± 18 mg/mL의 이중특이적 항-VEGF/ANG2 항체,
- 15 내지 35 mM의 염화나트륨, 및
- 15 내지 25 mM의 히스티딘 아세테이트 완충액
을 포함하고,
상기 이중특이적 항-VEGF/ANG2 항체는 2가이고, 인간 VEGF에 특이적으로 결합하는 제1 항원 결합 부위를 포함하고, 인간 ANG-2에 특이적으로 결합하는 제2 항원 결합 부위를 포함하되,
i) VEGF에 특이적으로 결합하는 상기 제1 항원 결합 부위는 중쇄 가변 도메인에서 서열번호 1의 CDR3H 영역, 서열번호 2의 CDR2H 영역, 및 서열번호 3의 CDR1H 영역을 포함하고, 경쇄 가변 도메인에서 서열번호 4의 CDR3L 영역, 서열번호 5의 CDR2L 영역, 및 서열번호 6의 CDR1L 영역을 포함하고,
ii) ANG-2에 특이적으로 결합하는 상기 제2 항원 결합 부위는 중쇄 가변 도메인에서 서열번호 9의 CDR3H 영역, 서열번호 10의 CDR2H 영역, 및 서열번호 11의 CDR1H 영역을 포함하고, 경쇄 가변 도메인에서 서열번호 12의 CDR3L 영역, 서열번호 13의 CDR2L 영역, 및 서열번호 14의 CDR1L 영역을 포함하고,
iii) 상기 이중특이적 항체는 넘버링이 카밧(Kabat)의 EU 인덱스를 따르는 돌연변이 I253A, H310A, 및 H435A 및 돌연변이 L234A, L235A 및 P329G를 포함하는 인간 IgG1 서브클래스의 불변 중쇄 영역을 포함하는, 액체 약제학적 제형으로서,

i) VEGF에 특이적으로 결합하는 상기 제1 항원 결합 부위는 중쇄 가변 도메인 VH로서 서열번호 7의 아미노산 서열을 포함하고, 경쇄 가변 도메인 VL로서 서열번호 8의 아미노산 서열을 포함하고,
ii) ANG-2에 특이적으로 결합하는 상기 제2 항원 결합 부위는 중쇄 가변 도메인 VH로서 서열번호 15의 아미노산 서열을 포함하고, 경쇄 가변 도메인 VL로서 서열번호 16의 아미노산 서열을 포함하는, 액체 약제학적 제형. - 제2항에 있어서, 상기 이중특이적 항-VEGF/ANG2 항체는 2가이고, 인간 VEGF에 특이적으로 결합하는 제1 항원 결합 부위를 포함하고, 인간 ANG-2에 특이적으로 결합하는 제2 항원 결합 부위를 포함하며,
iv) 카밧(Kabat)의 EU 인덱스를 따르는 넘버링에 따라, 상기 불변 중쇄 영역에서 S354C 및 T366W 돌연변이는 하나의 CH3 도메인에 포함되고, Y349C, T366S, L368A 및 Y407V 돌연변이는 다른 CH3 도메인에 포함되는, 액체 약제학적 제형.
