Semisynthetische bereiding van humane insuline
De uitvinding heeft betrekking op de semisynthetische bereiding van humane insuline uit dierlijke insuline waarvan het B30 aminozuur en eventueel daaraan carboxyeindstandig gekoppelde groepen zijn 5 verwijderd en dat overigens op alle plaatsen identiek is aan humane insuline, door des(B30)insuline te koppelen met een threonine-derivaat en uit dit koppelingsproduct humane insuline te bereiden, alsmede op de koppelingsproducten verkregen bij deze werkwijze.

Hypoglycemie als gevolg van een tekort aan vrij insuline in het bloed (diabetes mellitus) kan behandeld worden door kunstmatig, bijvoorbeeld met behulp van injecties of met behulp van een doseerpomp, insuline 15 toe te dienen. Overwegend worden hiervoor gebruikt insulines geïsoleerd uit alvleesklieren van dieren zoals varkens of runderen.
Dergelijke insulinepreparaten blijken in bepaalde gevallen antigene of allergene werking te bezitten.
20 Deels kan dit een gevolg zijn van de aanwezigheid van verontreinigingen (bijvoorbeeld proinsuline, gedeeltelijke afbraakproducten daarvan, arginine-insuline, insuline-ethylester, monodeamino-insuline, of insuline aggregaten). Daarnaast kan de antigene werking ook 25 samenhangen met de structurele verschillen tussen dierlijke en humane insulines, waardoor het uit dieren afkomstige eiwit bij de mens als lichaamsvreemd wordt herkend. Zo verschillen varkens- en humane insuline van elkaar in één aminozuur namelijk het carboxyeindstandige 30 aminozuur van de z.g. B-keten. Dit zogenoemde B30 aminozuur is alanine in varkens-insuline maar threonine bij de mens. Varkens-insuline zal dus in humane insuline 8201650 ê * - 2 - omgezet kunnen worden door dit B30 aminozuur door threonine te vervangen, en hiertoe zijn ook een aantal methoden bekend.
Uiteraard starten bijna alle bekende methoden 5 met het verwijderen van het B30 aminozuur; in het geval van varkens-insuline wordt dus het alanine verwijderd. Dit geeft des(B30)insuline, ook vaak des(ala)insuline, kortweg DAI, genoemd. Voor de daaropvolgende bewerkingen is een aantal alternatieve werk-10 wijzen bekend die tot vorming van humane insuline leiden. Deze berusten op de al dan niet enzymatische koppeling van threonine, waarvan in het algemeen de carboxylgroep, bijvoorbeeld door een ester, beschermd is, aan des(B30)insuline. Die beschermende groep dient 15 na de threoninekoppeling verwijderd te worden. Een veel gebruikte beschermende groep is een tertiaire butoxy-groep; voor het verwijderen daarvan zijn vrij rigoreuze condities noodzakelijk, met name een extreme pH, die ook de rest van het insulinemolecuul aantasten. Dien-20 tengevolge ontstaan partiële degradatieproducten van insuline die vrijwel niet van het gewenste product, humane insuline, te scheiden zijn. Hierdoor krijgt men dus een zeer onzuiver insuline preparaat. De zuivering vormt ook een belangrijk probleem in een eerder 25 stadium, bij de scheiding van het gereageerde en ongereageerde des(B30)insuline uit het reactiemengsel waarin de threoninekoppeling is uitgevoerd. Met name bij bereiding op grote schaal leveren deze scheidingen onvoldoende zuiver materiaal, tenzij ten koste van erg 30 grote verliezen.
De onderhavige uitvinding maakt het echter mogelijk humane insuline te bereiden met een grote zuiverheid, zonder het gebruik van talrijke rendementsverlagende scheidings- en zuiveringsstappen.

De werkwijze volgens de uitvinding wordt daardoor gekenmerkt dat als threonine derivaat toegepast wordt de verbinding 5 H,N-CH-C^° -R1 ά I * HC-CH, I 3

waarin 10 R = H of een enzymatisch afsplitsbare suikerrest en R1 = een enzymatisch afsplitsbare suikerrest, aminozuur of peptide, of een amide of ester van beide laatste, of R^ = H indien R Φ H, gevolgd door enzymatische afsplitsing van suiker, 15 aminozuur of peptide, waarna humane insuline uit het medium wordt geïsoleerd.
Insuline waarvan het B30 aminozuur ontbreekt, kan afzonderlijk bereid worden door het dierlijke insuline te behandelen met een geschikt enzym, bijvoor-20 beeld met carboxypeptidase A. Des(B30)insuline kan echter ook in situ bereid worden waarbij het verwijderen van het B30 aminozuur en het koppelen van het threonine-derivaat in een z.g. transpeptidase-reactie verzorgd wordt door een enkel enzym, bijvoor-25 beeld trypsine.
Koppeling van het threonine-derivaat aan de B29 lysine van vooraf bereid desB30 insuline kan bijvoorbeeld met behulp van trypsine, of bijvoorbeeld met een ander enzym met trypsin-achtige werking zoals lysylendo-30 peptidase uit Achromobacter lyticus plaatsvinden. Er kan gebruik qemaakt worden van dragergebonden enzymen, waarbij na het beëindigen van de reactie enzym en oplossing met gemodificeerd insuline eenvoudig te scheiden zijn.
De covalente binding tussen de groep R en/of R1 35 en threonine dient enzymatisch verbreekbaar te zijn.

Deze voorwaarde bepaalt in belangrijke mate de keuze van het aminozuur c.q. de aminozuren of de suiker die de groep R en/of R1 vormen.
Indien R1 bestaat uit aminozuur of -zuren gaat 5 de voorkeur uit naar mono-, di- of tripeptiden. Deze kunnen bestaan uit bijvoorbeeld hydrofobe aromatische aminozuren (fenylalanine, tryptofaan) en/of basische aminozuren (arginine, lysine). Bij toepassing van hydrofobe aminozuren wordt de zuivering onder 10 hydrofobe condities sterk bevorderd, terwijl scheiding en zuivering onder polaire of ionische omstandigheden verbeterd worden bij koppeling van bijvoorbeeld basische aminozuren.
Suikers welke volgens de uitvinding als groep 15 R en/of R^ toegepast kunnen worden zijn mono-, di- of polysacchariden. Met name mono- en disacchariden zoals glucose, fructose, mannose, galactose, sucrose, maltose, lactose verdienen de voorkeur. Door hun sterk polaire karakter hebben aan insuline gekoppelde suiker een 20 zeer gunstige invloed op scheiding en zuivering van het eiwit.
Teneinde humane insuline te bereiden dient na koppeling van het threonine-derivaat aan des(B30)-insuline de restgroep R en/of R1 enzymatisch verwijderd 25 te worden. De keuze van de restgroep bepaalt welk enzym of welke combinatie van enzymen hiertoe geëigend is.
Indien de groep R1 opgebouwd is uit één of meer aminozuren zal verwijdering daarvan met voordeel met 30 behulp van carboxypeptidase geschieden. Een carboxy-peptidase is een exopeptidase welke beginnend vanaf het carboxyuiteinde van de peptideketen telkens een aminozuur kan verwijderen, met een voorkeur voor bepaalde aminozuren.

Carboxypeptidase A(CPA) splitst met name hydrofobe aminozuren af, met een voorkeur voor de aromatische aminozuren fenylalanine, tyrosine en tryptofaan. Daarentegen splitst carboxypeptidase B(CPB) bij 5 voorkeur naast basische aminozuren. In combinatie met geschikt gekozen aminozuren in de groep R1 zijn CPA en/of CPB zeer geschikte proteolytische enzymen voor gebruik bij de werkwijze volgens de uitvinding. Doch ook andere carboxypeptidasen of 10 dipeptylcarboxypeptidasen zijn voor dit zelfde doel geschikt.
Wanneer de groep bestaat uit een via esterbinding gekoppelde suiker kan voor de afsplitsing daarvan gebruik gemaakt worden van een 15 esterase, en wanneer de groep R bestaat uit een via etherbinding gekoppeld suiker kan voor de afsplitsing een voor een dergelijke O-glycosidische binding specifiek enzyme zoals galactosidase worden gebruikt.
De uitvinding kan verder toegelicht worden aan 20 de hand van de volgende voorbeelden.

Bij de volgende voorbeelden wordt steeds uitgegaan van des(B 30)insuline bereid uit varkensinsuline volgens de methode beschreven door E.W. Schmitt et al. (Hoppe Seyler's Z. Physiol. Chemie 359, 799 (1978), het z.g.
5 des(ala)insuline.(DAI).
Voorbeeld I - VI
Koppeling van di- en tripeptiden aan DAI
Uitgevoerd werd de koppeling met verschillende 10 substraten (di- en tripeptiden en HCl-zout van tri-peptide) met verschillende enzymsystemen (trypsine, en dragergebonden trypsine en lysylendopeptidase).
Deze koppelingen werden als volgt uitgevoerd: a. Een buffermedium wordt samengesteld uit 15 - een 1:1 mengsel van dimethylformamide met ethanol (= de z.g. organische fractie), en - een zekere hoeveelheid Tris HC1 buffer met -1 concentratie 0,5 mol.1 b. In dit buffermedium wordt een afgewogen hoeveel-20 heid substraat opgelost, alsmede een hoeveelheid DAI.
c. Dit mengsel wordt op de gewenste pH gebracht m.b.v. geconcentreerd NaOH (1 mol.1”1).
d. Een hoeveelheid van het koppelingsenzym wordt 25 toegevoegd, waarna de koppelingsreactie start, en welke verder plaatsvindt bij 37 °C.
e. Na verloop van de reactietijd wordt het reactie-product chromatografisch gescheiden; de opbrengst van het koppelingsproduct wordt bepaald uit het 30 chromatogram en uitgedrukt in % ten opzichte van de theoretische waarde.
De reactiecondities en opbrengsten voor de voorbeelden I-VI zijn vermeld in Tabel 1.

Voorbeeld VII
Chromatoqrafische scheiding van koppelingsproducten Vergeleken is het chromatografisch gedrag van peptide-gekoppeld DAI met dat van ester-gekoppeld 5 DAI.
Daartoe zijn bereid de koppelingsproducten van DAI met thr-O-methyl, thr-O-tBu, thr-phe-O-methyl en thr-phe-phe, op de onder voorbeeld ï-IV beschreven wijze.
10 De koppelingscondities waren: DAI concentratie 14 g.l”1, 0,06 mol.l"1 substraat, 2,5 g.l”1 trypsine, 67¾ organische fractie in medium van pH 6.4 en een reactietijd van een half uur.
De koppelingsproducten werden gescheiden op een 15 analytische HPLC kolom en de retentiewaarden (Rf) bepaald. in tabel 2 zijn de resultaten van deze scheiding weergegeven als het verschil in retentiewaarde van de koppelingsproducten met die van DAI (AR^).

Voorbeeld VIII
Verwijderen van gekoppelde groep phe-phe van DAI-thr-phe-phe
Na scheiding van onomgezet DAI en koppelings-5 product DAI-thr-pjhe-phe op een hydrofobe kolom werd een fractie van 23 ml verkregen die 0,85 mg. ml**1 koppelingsproduct bevatte. Dit koppelingsproduct werd gevriesdroogd en vervolgens opgelost in een 0,2 mol.l"1 ammoniumbicarbonaatbuffer pH 8,5 tot 10 een eindconcentratie van ongeveer 2,8 mg.ml" .
Aan deze oplossing werd 2 μΐ carboxypeptidase A toegevoegd (2,3 E). De reactie werd na 30 minuten gestopt door toevoeging van een gelijk volume 0,5 11101.1-1 citroenzuur. Hierna is het materiaal 15 gekristalliseerd.
Controle leerde dat het koppelingsproduct volledig is omgezet in -humane insuline zoals aangetoond werd met behulp van aminozuuranalyse.
In het supernatant na kristallisatie konden met 20 behulp van aminozuuranalyse naast fenylalanine geen andere aminozuren aangetoond worden.

Claims (6)
1. Werkwijze voor de semisynthetische bereiding van humane insuline uit dierlijke insuline waarvan het B30 aminozuur en eventueel daaraan carboxy-eindstandig gekoppelde groepen zijn verwijderd en dat overigens op alle plaatsen identiek is aan humane insuline, door des(B30)insuline te koppelen met een threonine-derivaat en hieruit humane insuline te bereiden, met het kenmerk, dat als threonine-derivaat wordt toegepast de verbinding H?N-CH-C^°— R1 ά j HC-CH. i J Q-R waarin R * H of een enzymatisch afsplitsbare suikerrest, en R^ = een enzymatisch af splitsbare suikerrest, aminozuur of peptide, of een amide of ester van beide laatste, of R^ = H indien R Φ H, gevolgd door enzymatische afsplitsing van suiker, aminozuur of peptide, waarna humane insuline uit het medium wordt geïsoleerd.
2. Werkwijze volgens conclusie 1, met het kenmerk, dat als threonine-derivaat een verbinding met R = H en R1 = een mono-, di- of tripeptide wordt toegepast.
3. Werkwijze volgens conclusie 2, met het kenmerk, dat een threonine-derivaat met R^ = een mono—, di- of tripeptide van fenylalanine wordt toegepast.
