Abstract
Metode for påvisning av en forbindelse som etterligner, potensierer eller hemmer den fysiologiske effekt av leptin, hvilken metode omfatter: (a) for en forbindelse som etterligner den fysiologiske effekt av leptinet, bedømmelse av effekten av forbindelsen på en leptinaktivert signaloverfører og aktivator av transkripsjons- (STAT) DNA-responselement koblet til et rapportørgen; eller (b) for en forbindelse som potensierer eller hemmer den fysiologiske effekt av leptin, bedømmelse av effekten av forbindelsen på responsen på leptin av et leptinaktivert STAT DNA-responselement koblet til et rapportørgen;. hvor,. responselementet og rapportøren er uttrykt i den humane kaukasiske kolon-epitelcellelinje (CACO-2) eller responselementet og rapportøren er uttrykt i CACO-2-celler, hvilken cellelinje (den konstruerte cellelinje) også er transfektert med et polypeptid som kan mediere eller forsterke stimulering med leptin av et leptinaktivert STAT DNA-responselement og inneholder de passende STAT proteiner.A method of detecting a compound that mimics, potentiates or inhibits the physiological effect of leptin, which method comprises: (a) for a compound mimicking the physiological effect of leptin, assessing the effect of the compound on a leptin-activated signal transducer and activator of transcriptional (STAT) DNA response element linked to a reporter gene; or (b) for a compound that potentiates or inhibits the physiological effect of leptin, assessing the effect of the compound on the response to leptin by a leptin-activated STAT DNA response element linked to a reporter gene; where,. the response element and reporter are expressed in the human Caucasian colon epithelial cell line (CACO-2) or the response element and reporter are expressed in CACO-2 cells, which cell line (the constructed cell line) is also transfected with a polypeptide that can mediate or enhance stimulation with leptin of a leptin-activated STAT DNA response element and contains the appropriate STAT proteins.
