Abstract

Foreliggende oppfinnelse er en ny fremgangsmåte for påvisning og lokalisering av spesifikke nukleinsyresekvenser i en prøve med høy grad av sensitivitet og spesifisitet. Fremgangsmåten og de nye preparater som benyttes i fremgangsmåten, omfatter anvendelse av probenukleinsyrer, fremstilling av bindingsområder i nukleinsyren og anvendelse av målbindingsansamlinger i nukleinsyren for påvisning og lokalisering av spesifikke målnukleinsyrer. Påvisning og lokalisering av målnukleinsyren oppnås selv i nærvær av nukleinsyrer med lignende sekvenser. Fremgangsmåten tilveiebringer en høy grad av amplifisering av signalet som dannes ved hver spesifikke bindingsbegivenhet. Nærmere bestemt presenteres fremgangsmåter og preparater for påvisning av HIV- og HPV-nukleinsyre i prøver. Disse fremgangsmåter og preparater kan anvendes ved diagnose av sykdom, genetiske undersøkelser, retts~ medisinske formål og analyse av nukleinsyreblandinger. Noen av de nye preparater som benyttes i påvisningsfremgangsmåten, er anvendbare for forebyggelse eller behandling av patogene tilstander.
