Abstract

Metode for mutagenese ved hvilken en forutbestemt aminosyre blir innført i hver stilling av et valgt sett av stillinger i en forhåndsvalgt region (eller mange forskjellige regioner) av et polypeptid for å produsere et bibliotek av polypeptidanaloger. Metoden er basert på antagelsen at visse aminosyrer spiller en viktig rolle for strukturen og funksjonen av proteiner. Biblioteker kan dannes som inneholder bare ønskede polypeptidanaloger og har en rimelig størrelse for screening. Bibliotekene kan anvendes for å undersøke rollen til spesifikke aminosyrer for polypeptid-struktur og -funksjon og utvikle nye eller forbedrede polypeptider så som antistoffer, antistoffragmenter, enkelkjede antistoffer, enzymer og ligander.
