Abstract

Tidligere fremgangsmåter for dyrkning av humane embryonale stamceller har krevd enten fibroblast næringsceller eller et medium som er blitt eksponert for fibroblast næringsceller for å opprettholde stamcellene i en udifferensiert tilstand. Det er nå fiurmet at dersom høye nivåer av fibroblastvekstfaktor anvendes i et medium med gammaaminosmørsyre, pipecholinsyre, litium og lipider, vil stamcellene opprettholdes uendelig i udifferensiert tilstand gjermom mange passasjer, selv uten næringsceller eller kondisjonert medium. En humanisert matriks av humane proteiner kan anvendes som en grunnmatriks for å dyrke cellene. Nye linjer av humane embryonale stamceller er laget ved hjelp av disse kulturbetingelsene, der mediet og matriksen aldri er blitt eksponert for dyreceller, dyreproteiner, næringsceller eller kondisjonert medium.
