Abstract

Det er beskrevet en fremgangsmåte for fremstilling av 2-(4-hydroksy-3-morfolinyl)-2-sykloesenon (BTG-1675A) som omfatter trinnene: i) å omsette N-hydroksymorfolin med sykloesanon i nærvær av et oksidasjonsmiddel for således å oppnå et isoksazolidin med formel IV; og ii) konvertere isoksazolidinet med formel IV til 2-(4-hydroksy-3-morfolinyl)-2-sykloesenon. Fordelaktig blir oksidasjonsmidlet i trinn i) valgt fra gruppen som består av metalloksider, estere og amider av azodikarboksylsyre og trinnet ii) for konversjon blir utført ved basisk katalyse etterfulgt av triturering i et aromatisk hydrokarbon, fortrinnsvis toluen. Den beskrevne fremgangsmåte tillater at det oppnås BTG-1675A i henhold til oppfinnelsen i en mengde på hundreder av gram og på en industriell skala. Oppfinnelsen angår videre en ny fremgangsmåte til å fremstille hydroksylaminer, særlig N-hydroksymorfolin, som ble brukt i fremgangsmåten til fremstilling av BTG-1675A.
