Abstract

Rekombinante proteinkomplekser som har human faktor VIII:C-aktivitet blir uttrykt i en eukaryot vertscelle ved å transformere vertscellen med første og andre ekspresjons- kassetter som henholdsvis koder for et første polypeptid som i det vesentlige er homologt til human faktor VIII:C A-område og et andre polypeptid som i det vesentlige er homologt til human faktor VIII:C C-området. I den foreliggende oppfinnelse kan det første polypeptidet bli forlenget slik at det har som sin C-terminale ende et humant faktor VIII:C B-område N-terminalt peptid, en polypeptidforlengelse på 3-40 aminosyrer og et humant faktor VIII:C B-område C-terminalt peptid. Produksjon av det andre polypeptidet blir forbedret ved å benytte en al- antitrypsin signalsekvens.
