Description

Przedmiotem wynalazku jest nowy odczynnik do •oznaczania glikozy, cholesterolu, kwasu moczowego i hemoglobiny, zwlaszcza w próbkach biologicznych.Znanych jest kilka sposobów wykazywania i oznaczania ilosciowego glikozy, cholesterolu, kwa¬ su moczowego i hemoglobiny. Ze wzgledu na lat¬ wosc przeprowadzenia i taniosc oraz z przyczyn techniczno-pomiarowych najczesciej stosuje sie me¬ tody polegajace na chemicznym odczynie barwnym.Do pomiaru powyzszych substancji stosuje sie na ogól odczyn barwny z l-fenylo-2,3-dwumetylo- -3-amino-pirazolin-5-onu (zwanego dalej: 4-amino- -fenazon), fenolu, pochodnych fenolu i pochodnych kwasu salicylowego). Reakcja zachodzaca z fenolo- -4-aminofenazonem zostala zastosowana po raz pierwszy przez P. Trinder'a (Ann. Clin. Biochem. 3, 24, 1969) do oznaczania cukru we krwi. Lekarska waznosc tej metodyki jest tak duza, ze do tego celu wypracowano niezliczone. podobne metody.Ogólna wada tych metod polega na tym, ze cha¬ rakterystyka pomiaru nie jest liniowa i powstaja¬ ca barwa nie jest dostatecznie trwala lub jest swiatloczula (por. tablica 1 cytowanego artykulu i tablica III niniejszego opisu patentowego).Celem niniejszego wynalazku jest opracowanie nowego systemu, usuwajacego wady znanych me¬ tod.Wynalazek polega na nieoczekiwanym rozpozna¬ niu, ze gdy do tworzenia barwnego produktu sto¬ suje sie 4-amino-fenazon i tymol, w optymalnych warunkach zachodzi doskonala, bardzo dobrze na¬ dajaca sie do stosowania (trwala, nie swiatloczula, charakterystyka liniowa) i wykazujaca bardzo do¬ bra czulosc reakcja fotometryczna.Przedmiotem wynalazku jest odczynnik do ozna¬ czania glikozy, cholesterolu, kwasu moczowego, i hemoglobiny, korzystnie w próbkach biologicznych, który zawiera 4-amino-fenazon, znany roztwór bu¬ forowy, znany srodek stabilizujacy, znany detergent i ewentualnie katalizator enzymatyczny, charakte¬ ryzujacy sie tym, ze jako odczynnik zawiera ty¬ mol (para-izopropylo-meta-krezol).Wedlug korzystnej postaci wykonania odczyn¬ ników wedlug wynalazku rozpuszcza sie tymol w nizszym alkoholu, korzystnie w etanolu. Tym sa¬ mym przez specyficzne reakcje enzymatyczne two¬ rzy-sie nadtlenek wodoru. Z powstalego nadtlenku wodoit? uwalnia'* sie za pomoca peryksydazy tlen in statu nascendi, k^óry doprowadza do powstania 4r/2Mzopropylo-5'-mctylo-r,4'-benzochinono-mo- no-iniinp/-fenazonu (\o znaczy barwnego produktu).W przypadku substancji, które same sa zdolne do katalizowania rozkladu nadtlenku wodoru (np. he¬ moglobina), nie jest potrzebna uzupelniajaca reak¬ cja enzymatyczna, i doi mieszaniny reakcyjnej do¬ daje sie tylko nadtlenek wodoru, barwotwórczy 4- -amino-fenazón i tymol.Nowy odczynnik wedlug wynalazku ma kilka za¬ let, które mozna zreasumowac nastepujaco: a) Reakcja barwna zachodzi w dobrze okreslo¬ nym zakresie pH (pH = 7,0—8,0, korzystnie 7,5). b) Maksimum absorpcji swietlnej utworzonego pro¬ duktu barwnego przypada w zakresie dlugosci fal 465—480 nm; dlatego celowe jest przeprowadzenie pomiarów fotometrycznych w tym zakresie, w szcze¬ gólnosci 475 nm. c) Szczególnie wazne jest, ze wykrywalna foto- metrycznie barwa zostaje utworzona stosunkowo szybko. Stwierdzono, ze trwala barwa powstaje w temperaturze pokojowej w ciagu 25 minut i w temperaturze 37°C w ciagu 15 minut. d) Do nastawienia optymalnej wartosci pH na¬ daje sie rozmaite bufory (takie jak trój-fosforan, NafHP04—KH2P04, K2HP04—KH2P04 i boran so¬ du — kwas solny). e) Udalo sie okreslic korzystne stezenie tymolu i jego rozpuszczalnik. Celowe jest rozpuszczenie ty¬ molu w metanolu, etanolu lub izopropanolu, w szczególnosci w etanolu. Optymalizowanie stezenia tymolu jest podane w tablicy I. Wartosc optymal¬ na wynosi 2,5 mmoli/dm3. Pomiary przeprowadza sie za pomoca roztworu glikozy o podanym steze¬ niu (11,10 i 27,75 mmoli) dm3. 10 20 25 30 35 40 f) Optymalizowanie stezenia 4-amino-fenazonir podano w tablicy II. Pomiary przeprowadza sie rów¬ niez w tym przypadku za pomoca roztworu gliko¬ zy i roztworu tymolu o stezeniu 2,5 mmoli/dm3.Wedlug tej tablicy maksymalna, nie zmieniajaca sie juz ekstynkcje otrzymuje sie przy stezeniu 0,600 mmoli/dm3. Dla pewnosci jako optymalne traktuje sie stezenie 0,800 mmoli/dm8. a g) Optymalizuje sie równiez stezenie nadtlenku potrzebne do pomiaru glikozy, cholesterolu i kwa¬ su moczowego (por. tablica III). Optymalne oka¬ zalo sie stezenie 15E/cm8. (1E = 1 jednostka mie¬ dzynarodowa; ilosc substancji, która wymienia 1 mmol substratu w ciagu 1 minuty). Zastosowana peroksydaza nie moze zawierac innych zanieczysz¬ czen enzymatycznych w stezeniach przeszkadzaja¬ cych.Z powyzszej tablicy widac, ze maksymala in¬ tensywnosc (ekstynkcje) utworzonej za pomoca gli¬ kozy barwy osiaga sie juz przy aktywnosci enzy¬ mu wynoszacej 14E/cm3; dla pewnosci stosuje sie aktywnosc 15E/cms. h) Przy oznaczaniu cholesterolu stosuje sie de¬ tergent aby zwiekszyc rozpuszczalnosc. Do tego ce¬ lu mozna stosowac 9-laurylo-etero-polidekanol (Sig¬ ma, St. Zjedn. Am.), Triton X-100 (oktylofenolopo- lietylenoglikoloeter, LOBA, Austria), albo Brij 35 (poUoksyetylenolauryloeter, BDH, Anglia). Opty¬ malny zakres stezenia powyzszych srodków wyno¬ si 0,4^1,2%.Praktyczna mozliwosc stosowania odczynników wedlug wynalazku przedstawiaja nastepujace pro^ cesy. 1) Doswiadczenia otrzymane za pomoca wzorco- wan. Several methods are known for detecting and quantifying glucose, cholesterol, uric acid, and hemoglobin. Due to their ease of use, low cost, and for technical and measurement reasons, methods based on chemical color reactions are most commonly used. The color reaction of 1-phenyl-2,3-dimethyl-3-amino-pyrazolin-5-one (hereinafter referred to as 4-amino-phenazone), phenol, phenol derivatives, and salicylic acid derivatives is generally used to measure the above substances. The reaction with phenol-4-aminophenazone was first used by P. Trinder (Ann. Clin. Biochem. 3, 24, 1969) to determine blood sugar. The medical importance of this methodology is so great that countless methods have been developed for this purpose. The general disadvantage of these methods is that the measurement characteristic is not linear and the resulting color is not sufficiently stable or is photosensitive (see Table 1 of the cited article and Table III of the present patent description). The aim of the present invention is to develop a new system that eliminates the disadvantages of the known methods. The invention is based on the unexpected finding that when 4-aminophenazone and thymol are used to create a colored product, under optimal conditions an excellent, very well-suited for use (stable, non-photosensitive, linear characteristic) and very sensitive photometric reaction occurs. The subject of the invention is a reagent for determining glucose, cholesterol, uric acid and hemoglobin, preferably in biological samples, which contains 4-aminophenazone, known a buffer solution, a known stabilizing agent, a known detergent and optionally an enzymatic catalyst, characterized in that it contains thymol (para-isopropyl-meta-cresol) as a reagent. According to a preferred embodiment of the reagents according to the invention, thymol is dissolved in a lower alcohol, preferably ethanol. Other chemical substances are dissolved in distilled water and the alcoholic and aqueous solutions are mixed with in a ratio of 1:4. As prepared, the reagent contains 1 mol, in a concentration of 2.5 + 0.2 mmol/dm3. The concentration of other chemical substances in the reagent is as follows: phosphate buffer 0.08 + 0.02 mol/dm3 4-aminophenazone 0.5 + 0.2 mmol/dm3 sodium azide 9.85 + 0.1 mmol/dm3 ethanol 330.0 + 20 mmol/dm3. The reagent according to the invention is used as follows: thymol and 4-aminophenazone are brought into contact with the marked substances (glucose, cholesterol, uric acid) in appropriate concentrations in In a buffered environment, hydrogen peroxide is formed through specific enzymatic reactions. Nascent oxygen is released from the resulting hydrogen peroxide by peroxidase, which leads to the formation of 4-(2-M-isopropyl-5'-methyl-1,4'-benzoquinone-mono-aminophenazone) (i.e., the colored product). In the case of substances that are themselves capable of catalyzing the decomposition of hydrogen peroxide (e.g., hemoglobin), no additional enzymatic reaction is necessary, and only hydrogen peroxide, the color-forming 4-aminophenazone, and thymol are added to the reaction mixture. The new reagent according to the invention has several advantages, which can be summarized as follows: a) The color reaction occurs within a well-defined pH range. (pH = 7.0-8.0, preferably 7.5). b) The maximum light absorption of the formed colored product occurs in the wavelength range of 465-480 nm; therefore, it is advisable to carry out photometric measurements in this range, especially at 475 nm. c) It is particularly important that the photometrically detectable color is formed relatively quickly. It has been found that a stable color develops at room temperature within 25 minutes and at 37°C within 15 minutes. d) Various buffers are suitable for adjusting the optimal pH value (such as triphosphate, NaHPO4-KH2PO4, K2HPO4-KH2PO4 and sodium borate-hydrochloric acid). e) The preferred thymol concentration and its solvent have been determined. It is advisable to dissolve thymol in methanol, ethanol or isopropanol, in particular in ethanol. Optimization of the thymol concentration is given in Table I. The optimum value is 2.5 mmol/dm3. Measurements are carried out using a glucose solution of the given concentration (11, 10 and 27.75 mmol) dm3. 10 20 25 30 35 40 f) Optimization of the 4-amino-phenazonir concentration is given in Table II. Measurements are also carried out in this case using a glucose solution and a thymol solution of 2.5 mmol/dm3. According to this table, the maximum, no longer changing extinction is obtained at a concentration of 0.600 mmol/dm3. For safety reasons, the concentration of 0.800 mmol/dm8 is considered as optimal. g) The concentration is also optimized peroxide needed to measure glucose, cholesterol and uric acid (see Table III). The optimal concentration was found to be 15E/cm3. (1E = 1 international unit; the amount of substance that exchanges 1 mmol of substrate in 1 minute). The peroxidase used must not contain other enzymatic contaminants in interfering concentrations. The table above shows that the maximum intensity (extinction) of the color formed with glucose is achieved already at an enzyme activity of 14E/cm3; for safety reasons, an activity of 15E/cm3 is used. h) In the determination of cholesterol, a detergent is used to increase solubility. 9-lauryl ether polydecanol (Sig¬ ma, United States), Triton X-100 (octylphenolpolyethyleneglycol ether, LOBA, Austria), or Brij 35 (polyoxyethylenelauryl ether, BDH, England). The optimum concentration range of the above agents is 0.4-1.2%. The practical possibility of using the reagents according to the invention is demonstrated by the following processes. 1) Experiments obtained by means of calibrations. The detectability and quantitative miscibility of glucose are determined in the concentration range of 2.775-33.300 mmol/dm³. The data are summarized partly in Table IV and partly in Figure 1 (after calculation of the matrix). The internationally recognized Bochringer ultraviolet hexokinase assay serves as the reference method. In this case, 45 human blood samples (without selection) are used as the sugar-containing solution. Figure 2 shows that there is no difference between the results obtained by these two methods, because the correlation coefficient (r = +0.9971) is quite close to the theoretical value of 1.000 and the steepness of the regression line differs from the theoretical value of 1.000 by only 0.4%. 3) Sensitivity, reproducibility, and process distortion are determined on the basis of the obtained measurement results. These statistical calculations give the following results: Sensitivity: ±0.17 mmol/dm* (relative to glucose), accuracy: ±0.09 mmol/dm8 (relative to glucose), distortion: ±0.35% (relative to glucose). The reagent according to the invention has, in addition to the above-mentioned advantages, further significant advantages: the reagent is suitable, on the one hand, for the quantitative determination of glucose, cholesterol, uric acid or hemoglobin (red blood pigment) present in biological samples of human and animal origin (e.g. blood, urine, cerebrospinal fluid) and, on the other hand, for the determination of the glucose content in beverages for human consumption (e.g. alcoholic beverages, juices, soft drinks) or for measuring other solutions containing glucose, cholesterol or uric acid. It should be noted that the reagent according to the invention clearly outperforms known tests. The quantitative comparison shows that according to the test method according to the invention, the color remains completely unchanged for at least 240 minutes, whereas according to the known methods, after a short maximum phase (after 45 minutes), a continuously decreasing color intensity is observed (Fig. 3). The reagent according to the invention and its use are illustrated by the following examples, without limiting the scope of protection to these examples. Example I. Determination of glucose in blood serum. Solution A: In 0.1 mol/dm3 of phosphate buffer solution (pH = 7.5; NatHPO4-2HtO and KH2PO4), the following substances are dissolved: glycosidases: peroxidases: 4-aminophenazone: sodium azide: 12.5 E/cm3 18 E/cm* 1.0 mmol/dm3 12.3 mmol/dm3. Solution B: 12.5 mmol/dm3 thymol in 96% concentrated ethanol (with distilled water). The reagent is prepared by mixing solutions A and B in a 4:1 ratio. Standardization is carried out using a glucose solution of 11.1 mmol/dm3 (dissolved in cold saturated benzoic acid). Marking: 3 cm3 of reagent, 0.02 cm3 of standard or 0.02 cm3 of blood serum in further test tubes are weighed. The substances are mixed and left at room temperature for 30 minutes or at 37°C for 15 minutes. In the latter case, the color formed after 10 minutes of cooling is compared photometrically with a blank sample (cuvette thickness 1 cm, wavelength 475 nm). The blood serum concentration [in glucose mmol/dm1]7 is calculated according to the following equation: sample extinction XII,1. standard extinction The process is linear in the range 0—30 mmol/dm3. Example II. Determination of total cholesterol content in blood serum. Solution A: in phosphate buffer (0.1 mol/l; pH = 7.5) the following substances are dissolved: peroxidase 18 E/cm3 cholesterosterase 12.5 E/cm3 cholesteroxidase 6 E/cm3 4-aminophenazone 1.0 mmol/l sodium azide 12.3 mmol/l 9-lauryl ether 5.0 g/l or Triton X-100 5 g/l or Brij-35 5 g/l. Solution B: 12.5 mmol/l thymol (in 96% freshly distilled ethanol). The reagent is prepared by mixing solutions A and B in a 4:1 ratio before use. Calibration is carried out using a cholesterol standard dissolved in ethanol [5.2 mmol/dm3]. Determination: To each 2.0 cm3 of the reagent, 0.02 cm3 of the standard solution or serum is added. The mixture is mixed and left at room temperature for 30 minutes or at 37°C for 15 minutes. The color developed after 10 minutes of cooling is compared photometrically with a blank sample (cuvette thickness 1 cm, wavelength: 475 nm). Calculation: cholesterol concentration in blood serum (mmol/dm3) = sample extinction X 5.2 standard extinction. The process is linear in the range 0—13 mmol. Example III. Determination of uric acid in blood serum. Solution A: in phosphate buffer (0.1 mol/l; pH 7.5) the following substances are dissolved: peroxidase 18 E/cm3 uricase 6 E/cm3 4-aminophenazone 1.0 mmol/l sodium azide 12.3 mmol/l 984 8 Solution B: 12.5 mmol/l thymol (in 96% ethanol). The reagent is prepared by mixing solutions A and B in a 4:1 ratio. Standardization is performed using a uric acid standard (595 µmol/dm3). Determination: Each time, 0.2 cm3 of the standard or serum sample is added to 0.2 cm3 of the reagent. The mixture is mixed and left for 60 minutes at room temperature or for 10 minutes at 30 minutes at 37°C. The color developed is compared with a blank sample photometrically (cuvette thickness 1 cm, wavelength 475 nm). Calculation: uric acid concentration in blood serum (µmol/dm3), sample extinction X595, standard extinction. The process is linear in the range 0—900 µmol/dm3. Claims 1. A reagent for determining glucose, cholesterol, uric acid and hemoglobin, preferably in biological samples, which comprises 4-aminophenazone, a known buffer solution, a known stabilizing agent, a known detergent and optionally an enzymatic catalyst, characterized in that it comprises thymol (para-isopropyl-meta-cresol) as a reagent. 2.
