It is known that determining the number of platelets and leukocytes is a common method in clinical laboratories. Platelet counts can be determined by various methods, such as those described below. Electronic particle counting using automated devices designed for platelet counting. While this method is quick and simple, it requires very expensive equipment and cannot always replace microscopic counting. The reproducibility of this method is also inferior to that of microscopic counting. Platelet counting is performed using a phase-contrast microscope, using a solution containing cocaine or novocaine, or another solution that allows such counting. A phase-contrast microscope is necessary to use this method, and another significant disadvantage is that it is very tiring, exhausting, and harmful to the eyes of those performing the calculations, especially if there are many of them. Platelet counting by staining. This method is the simplest and can be performed using a microscope. In known methods of this type, crystal violet (gentian violet), which is hexamethyl-p-rosaniline hydrochloride, is used as a dye. These methods are justifiably criticized for giving distorted results compared to the phase contrast method, because not all thrombocytes are stained clearly.2 142 043 Leukocyte counting is the most widely used method in hematological diagnostics, and Turk's solution, described in the Hungarian Pharmacopoeia (Hi« Hg» VI#), is generally suitable for this purpose and gives good results, but this solution is not suitable for determining the number of thrombocytes, which is a serious disadvantage, because it is very often necessary to conduct both of these widely used laboratory tests together. To determine the number of thrombocytes and leukocytes in whole blood using a regular light microscope, a dye solution is necessary, which (a) causes complete hemolysis of the erythrocytes without reducing the number of thrombocytes and leukocytes, (b) fixes both thrombocytes and leukocytes, and (c) contains a dye which binds strongly to both these bodies and thus fixes them in ordinary light. A solution which meets the first two of these requirements is known. (Scand* J* Clin# Eivest. 33, 121 (1974)/, but when using it, the platelet count must be carried out using a phase contrast microscope. The invention aims to produce such a dye solution by means of which the number of platelets and leukocytes in whole blood can be determined using a conventional optical microscope, while avoiding the above-described disadvantages of the reagent. Surprisingly, it has been found that by combining a hemolyzing and fixing solution with a basic high-affinity dye, a solution of the agent is obtained which enables the joint determination of the number of platelets and leukocytes using a conventional optical microscope. Both conditions can be met if the hemolyzing and fixing solution is compatible with the dye, i.e. it does not form any precipitate. The formation of microscopic precipitates should be avoided. should be avoided, as they may have the character of thrombocytes and thus cause errors in the calculation. The solutions of cresyl violet dyes used so far only partially meet these conditions, and the calculation solutions containing cresyl violet dye components have a weakly alkaline reaction, so the thrombocytes are only faintly colored and are not clearly visible. The invention is based on the finding that the so-called thiazine dyes are very well suited for this purpose. A feature of thiazine dyes is the presence of two chromophore groups. Thiazine compounds are used in medical practice in various fields, e.g. for staining bacteria and in histochemistry, but neither the technical nor patent literature gives any indication that they can be useful for the production of the agent that is the subject of the invention. The most important Representatives of this group of compounds are: toluidine blue of formula I, thionine of formula II and methylene blue of formula III. The agent according to the invention contains 0.1-5 parts by weight of acetone, 0.05-2.0 parts by weight of formaldehyde and/or glutaraldehyde, 0.001-0.1 parts by weight of a thiazine dye, preferably toluidine blue, 0.1-2.0 parts by weight of a mineral salt, preferably sodium chloride, and the remainder up to 100 parts by weight is water. A particular advantage of the dyes used in accordance with the invention is that they have a very high affinity also for the acidic macromolecules of thrombocytes and leukocytes, thanks to which a very strong and clear visibility is achieved. The most important advantage of the agents according to the invention is that they are much more sensitive than known reagents. This fact is confirmed by the tests described below. From samples with an unknown number of thrombocytes and leukocytes, the number of thrombocytes and leukocytes is determined by 20 consecutive counts. The accuracy of the cell count is then satisfactory when the normal deviation is less than 10. The results of such tests carried out using the agent according to the invention and using known reagents are given in the drawing in the form of graphs, in which the numbers of thrombocytes and leukocytes are determined according to the 31 system as G values/liter, This is giga(cell count)/liter, which is equal to the number of cells multiplied by 109 in 1 liter. Figure 1 shows the results of determining the number of platelets using the agent according to the invention. For comparison purposes, thrombocytes were counted using the TKROKCFIK reagent manufactured by Godecke, and leukocytes were counted using a reagent known as Turk's solution. The tests were carried out with blood drawn from 41 patients, and thrombocytes and leukocytes were counted in parallel. The results in the Cartesian coordinate system are given in Figures 2 and 4, respectively. The sum of the counted platelets in 10 right angles of the Burker cell is multiplied by 2 in the case of ethylenediaminetetraacetic acid, or by 2.2 in the case of citrate as an anticoagulant, to obtain the platelet count in grams/liter. Leukocyte count. The number of leukocytes found in the 4 squares bounded by 3 lines is divided by 18 (or multiplied by 0.055) in the case of citrate, or divided by 20 (or multiplied by 0.05) in the case of ethylenediaminetetraacetic acid as an anticoagulant, and the leukocyte count in grams per liter is obtained. Example III. Determination of the thrombocyte count and the leukocyte count in capillary blood. The disinfected fingertip or heel is pricked, the first drops of blood are removed, and 25 µl of blood is sucked out with a pipette with a plastic syringe (e.g., Finn, Gilson, or Eppendorf pipette).
