PL 227 373 B1 2 Opis wynalazku Przedmiotem wynalazku jest nowy przewodzacy polimer, zawierajacy monomer funkcyjny bis(2,2'-bitienylo)-(4-aminofenylo)metan oraz monomer sieciujacy, 2,2'-bis(2,2'-bitiofeno-5-ylo)-3,3'- -bitionaften, molekularnie wdrukowany za pomoca 2,4,6-trinitrofenolu, i sposób wytwarzania tego po- limeru jak równiez jego zastosowanie jako warstwa rozpoznajaca fluorescencyjnego czujnika che- micznego do selektywnego wykrywania i/lub oznaczania nitroaromatycznych zwiazków wybuchowych, w tym 2,4,6-trinitrofenolu. 2,4,6-Trinitrofenol (ang. 2,4,6-trinitrophenol, TNP), znany równiez jako kwas pikrynowy, jest wy- buchowym zwiazkiem nitroaromatycznym. Zwiazek ten wykorzystywany jest do produkcji zapalek, baterii elektrycznych i kolorowego szkla, a takze do trawienia miedzi i barwienia materialów tekstyl- nych. TNP jest wysoce toksyczny. Jego spozycie prowadzi do zatrucia [Wyman, J. F., Serve, M. P., Hobson, D. W., Lee, L. H., Uddin, D. E., J. Toxicol. Environ. Health, 37 (1992) 13]. Objawy zatrucia TNP to, m.in. bóle glowy i brzucha, nudnosci, wymioty a takze zóltawy odcien skóry. Wysoka dawka TNP moze spowodowac uszkodzenie erytrocytów, niezyt zoladka i jelit, zapalanie nerek, zóltaczka lub krwiomocz. Jezeli TNP dostanie sie do oka, to moze spowodowac jego podraznienie lub wywolac stan chorobowy rogówki oka. Dawka smiertelna dla królika to 120 mg/kg TNP [Patnaik, P., A comprehen- sive Guide to the Hazardous Properties of Chemical Substances, John Wiley & Sons, New Jersey, 2007]. Dlatego wykrywanie i selektywne oznaczanie 2,4,6-trinitrofenolu jest tak wazne. Do oznaczania TNP w ilosciach sladowych stosuje sie kilka metod. Wykorzystuje sie w nich ta- kie techniki analityczne jak woltamperometria, wysokosprawna chromatografia cieczowa, grawimetria i techniki spektroskopowe [Singh, S., J. Hazard. Mater., 144 (2007) 15]. Wsród tych ostatnich, techniki optyczne, a w szczególnosci fluorescencyjne, znajduja coraz szersze zastosowanie [Salinas, Y. Marti- naez-Manez, R., Marcos M. D., Sancenon, F., Costero, A. M., Parra, M., Gil, S., Chem. Soc. Rev., 41 (2012) 1261; Kim, H. N. Guo, Z., Zhu, W., Yoon, J., Tian, H., Chem. Soc. Rev., 40 (2011) 79]. Popu- larnosc tych technik zwiazana jest z mozliwoscia szerokiego wyboru zwiazków z ugrupowaniami o charakterze fluorofora lub chromofora. Techniki te charakteryzuja sie wysoka wykrywalnoscia i czu- loscia, a takze latwoscia wykonywania pomiarów spektrofotometrycznych. Wiekszosc dotychczas opublikowanych prac na ten temat glównie skupia sie na wykorzystaniu jako fluorofory mediatorów, takich jak np. polimer przewodzacy, poli[2-metoksy-5-(2-etyloheksyloksy)-1,4-fenylenowinylen] [Chu, F., Tsiminis, G., Spooner, N. A. Monro, T., Sens. Actuators. B, 199 (2014) 22], fluorescencyjnych na- nokompozytów sferycznych [Ma, Y., Li, H., Peng, S., Wang, L., Anal. Chem. 84 (2012) 8415], badz porowatych sieci polimerów [Zhang, W., Qiu, L. -G., Yuan, Y. -P, Xie, A. -J., Shen, Y. -H., Zhu, J. -F., J. Hazard. Mater., 221–222 (2012) 147]. Jednakze w przewazajacej mierze metody te obejmuja ozna- czanie TNP w fazie gazowej. Wykrywaniu i oznaczaniu TNP w roztworach moga sprostac czujniki chemiczne z dedykowa- nymi warstwami polimerów wdrukowanych molekularnie (ang. molecularly imprinted polimers, MIPs) jako elementami rozpoznajacymi zdolnymi do selektywnego i odwracalnego przylaczenia TNP. Opra- cowano juz MIP wdrukowany za pomoca TNP [ Huynh, T.-P., Sosnowska, M. I., Sobczak, J. W., KC, C. B., Nestrov, V. N., D'Souza, F., Kutner, W., Anal. Chem. 85 (2013) 8361], ale do oznaczania TNP nie wykorzystano jeszcze detekcji spektrofluorymetrycznej. W niniejszym zgloszeniu przedstawiamy przygotowanie takiego MIPu i jego zastosowanie w postaci warstwy rozpoznajacej czujnika spektro- fluorymetrycznego do selektywnego oznaczania TNP. Celem niniejszego wynalazku jest opracowanie warstwy MIP i sposobu jej wytwarzania tak, aby mogla byc wykorzystana jako warstwa selektywnie rozpoznajaca TNP. Celem tym jest takze sposób jej osadzania na przetworniku sygnalu chemicznego za pomoca elektropolimeryzacji w warunkach potencjodynamicznych. Niniejszy wynalazek obejmuje rozpoznajacy przewodzacy polimer bisbitiofenowy, korzystnie w postaci warstwy polimeru molekularnie wdrukowanego, charakteryzujacy sie tym, ze stanowi go polimer zawierajacy bis(2,2'-bitienylo)-(4-aminofenylo)metan jako monomer funkcyjny, oraz 2,2'- -bis(2,2'-bitiofeno-5-ylo)-3,3'-bitionaften jako monomer sieciujacy, oraz luki molekularne wdrukowane za pomoca czasteczek 2,4,6-trinitrofenolu (TNP). Korzystnie, zastosowany monomer sieciujacy spel- nia równoczesnie role fluorofora. Ponadto obecny wynalazek obejmuje sposób wytwarzania rozpoznajacego przewodzacego po- limeru bisbitiofenowego, okreslonego powyzej, metoda wdrukowania molekularnego 2,4,6-trinitro- fenolu (TNP) za pomoca elektropolimeryzacji, korzystnie w warunkach potencjodynamicznych, charak- PL 227 373 B1 3 teryzujacy sie tym, ze prowadzi sie elektropolimeryzacje monomeru funkcyjnego stanowiacego bis(2,2'-bitienylo)-(4-aminofenylo)metan, w obecnosci monomeru sieciujacego stanowiacego 2,2'- -bis(2,2'-bitiofeno-5-ylo)-3,3'-bitionaften oraz w obecnosci 2,4,6-trinitrofenolu (TNP) jako szablonu, który to szablon jest wdrukowywany molekularnie w trakcie tej elektropolimeryzacji z wytworzeniem rozpoznajacego przewodzacego polimeru bisbitiofenowego z wdrukowanym molekularnie szablonem 2,4,6-trinitrofenolu (TNP), z którego wyekstrahowywuje sie szablon z uzyciem metanolu po osadzeniu tak otrzymanego polimeru w postaci warstwy na przetworniku sygnalu chemicznego/elektrodzie pracu- jacej i otrzymuje sie rozpoznajacy przewodzacy polimer bisbitiofenowy z lukami molekularnymi po wdrukowanym szablonie; przy czym monomer sieciujacy wystepuje w podwójnej roli, tj. z jednej strony sieciuje MIP-TNP a z drugiej jest fluoroforem, przy czym do elektropolimeryzacji stosuje sie roztwór mieszaniny rozpuszczalników, korzystnie acetonitrylu i toluenu, w stosunku objetosciowym, korzystnie, 95 : 5, zawierajacy 2,4,6-trinitrofenol (TNP), bis(2,2'-bitienylo)-(4-aminofenylo)metan i 2,2'-bis(2,2'-bitiofeno-5-ylo)-3,3'-bitionaften, korzyst- nie w stosunku molowym, 1:1:2, oraz elektrolit podstawowy, korzystnie chloran(VII) tetrabutyloamo- niowy, (TBA)CIO4, korzystnie 0,1 M, oraz przykladany potencjal jest liniowo zmieniany w sposób cykliczny w zakresie od 0 do 1,5 V, wzgledem Ag/AgCI, ze stala szybkoscia w zakresie od 5 mV/s do 5 V/s. Korzystnie, w sposobie wedlug wynalazku, przykladany potencjal jest liniowo zmieniany w spo- sób cykliczny w zakresie od 0,5 do 1,30 V wzgledem Ag/AgCI, ze stala szybkoscia 20 mV/s. Korzystnie, w sposobie wedlug wynalazku, do osadzania stosuje sie przetwornik sygnalu che- micznego/elektrode pracujaca wybrana sposród grupy stalych elektrod przewodzacych i pólprzewo- dzacych, korzystnie w postaci plytki szklanej z napylonym zlotem i plytki szklanej z warstwa tlenku cynowo-indowego. Wynalazek równiez obejmuje zastosowanie rozpoznajacego polimeru bisbitiofenowego, ko- rzystnie w postaci warstwy polimeru, molekularnie wdrukowanego za pomoca 2,4,6-trinitrofenol (TNP), okreslonego powyzej, jako elementu rozpoznajacego czujnika chemicznego do wykrywania i/lub se- lektywnego oznaczania 2,4,6-trinitrofenolu w próbkach biologicznych, jak i w czujnikach do wykrywa- nia i/lub oznaczania substancji wybuchowych, na uzyteczny sygnal analityczny za pomoca pomiarów spektrofluorymetrycznych. Korzystne przyklady wykonania wynalazku P r z y k l a d 1 Osadzanie warstwy polimeru wdrukowanego molekularnie za pomoca 2,4,6-trinitrofenolu na róznych elektrodach Przed osadzeniem warstwy MIP, zarówno plytki szklane pokryte warstwa zlota jak i plytki szkla- ne pokryte warstwa przewodzaca tlenku cynowo-indowego oczyszczono przez oplukanie toluenem, a nastepnie zanurzenie na 20 min w acetonie a nastepnie w wodzie, po czym na 10 min w izopro- panolu. Warstwy MIPów osadzono na ww. plytkach za pomoca elektropolimeryzacji potencjodynamicz- nej w kilku cyklach zmian potencjalu, z szybkoscia 20 mV/s, w zakresie od 0,50 do 1,30 V vs Ag/AgCI. Polimeryzacje przeprowadzono w ukladzie elektrochemicznym, w którym przeciwelektroda byla plytka szklana pokryta warstwa zlota umieszczona naprzeciw i równolegle do elektrody pracujacej. Na Fig. 1 przedstawiono zmiany pradu, towarzyszace zmianom potencjalu, prowadzace do osadzenia warstwy MIPów na plytkach szklanych pokrytych (a) zlotem i (b) ITO. Do elektropolimeryzacji zastosowano roztwór 0,3 mM 2,4,6-trinitrofenolu, 0,3 mM bis(2,2'- -bitienylo)-(4-aminofenylo)metanu oraz 0,6 mM 2,2'-bis(2,2'-bitiofeno-5-ylo)-3,3'-bitionaftenu w mie- szaninie dwóch rozpuszczalników organicznych, acetonitylu i toluenu, zmieszanych w stosunku obje- tosciowym jak, odpowiednio, 95 :5. Jako elektrolit podstawowy zastosowano chloran(VII) tetrabutylo- amoniowy, (TBA)CIO4, korzystnie o stezeniu 0,1 M. Po osadzaniu na plytkach, warstwe MIP-TNP oplukano acetonitrylem w celu usuniecia z niej resztek roztworu elektrolitu podstawowego. Podczas elektropolimeryzacji, zarówno na plytce szklanej pokrytej warstwa zlota jak i pokrytej warstwa ITO, zaobserwowano pik anodowy opowiadajacy elektroutlenieniu ugrupowania bis(2,2'- -tienylo)metanu. Dla plytki szklanej pokrytej warstwa zlota, pik pojawil sie w pierwszym cyklu przy potencjale -1,0 V vs Ag/AgCI. Nastepnie, w kolejnych dwóch cyklach, pik ten obnizal sie i przesuwal w kierunku bardziej dodatnich potencjalów. Bylo to spowodowane obnizeniem przewodnosci warstwy MIPu wraz ze wzrostem jej grubosci. Osadzana warstwa stanowi bariere, o rosnacej opornosci, utrud- niajaca dalsze elektroutlenianie ugrupowania bis(2,2'-tienylo)metanu w zadanym zakresie potencja- lów. Podczas osadzania warstwy MIP-TNP na plytce szklanej pokrytej warstwa ITO pik anodowy po- jawil sie przy potencjale -0,95 V vs Ag/AgCI i wzrósl w kolejnych dwóch cyklach. Oznacza to, ze poli- mer jest przewodzacy, a wzrost pradu moze byc zwiazany ze wzrostem powierzchni polimeru spowo- dowanym wzrostem szorstkosci warstwy osadzonego MIPTNP wraz ze wzrostem jej grubosci. Dla porównania przygotowano równiez warstwy niewdrukowanych polimerów (ang. non- imprinted polimers, NIPs). Osadzono je wedlug procedury opisanej powyzej jednakze z roztworu, który nie zawieral szablonu TNP. PL 227 373 B1 5 P r z y k l a d 2 Ekstrakcja szablonu 2,4,6-trinitrofenolu z osadzonej warstwy MIP-TNP W celu sprawdzenia czy szablon TNP jest calkowicie usuwany z warstwy MIPu za pomoca eks- trakcji, warstwe MIPu osadzono na dyskowej elektrodzie platynowej za pomoca elektropolimeryzacji w warunkach potencjodynamicznych stosujac kilka cykli zmiany potencjalu z szybkoscia 50 mV/s w zakresie od 0,50 do 1,25 V vs Ag/AgCI, z roztworu 0,3 mM 2,4,6-trinitrofenolu, 0,3 mM bis(2,2'- -bitienyl)-(4-aminofenyl)metanu, 0,6 mM 2,2'-bis(2,2'-bitiofeno-5-ylo)-3,3'-bitionaftenu i 0,1 M (TBA)CIO4 dwóch rozpuszczalników organicznych, acetonitylu i toluenu, zmieszanych w stosunku objetosciowym jak, odpowiednio, 95:5. Nastepnie szablon TNP wyekstrahowano z warstwy MIP-TNP metanolem. Ekstrakcje prowadzono przez 4 godz. przy ciaglym mieszaniu na mieszadle magnetycznym. Postep ekstrakcji kontrolowano za pomoca róznicowej woltamperometrii pulsowej (ang. differential pulse vol- tammetry, DPV), w zakresie potencjalów od 0 do 0,60 V vs Ag/AgCI, dla acetonitrylowego roztworu 0,1 mM ferrocenu i 0,1 M (TBA)CIO4 (fig. 2). Zaobserwowano pik przy potencjale 0,30 V, który malal wraz z postepem ekstrakcji. To obnizenie wysokosci piku zwiazane jest z obnizeniem przewodnosci warstw MIP spowodowane usuwaniem TNP z wdrukowanych luk molekularnych MIP-TNP. P r z y k l a d 3 Morfologia warstwy MIP W celu okreslenia morfologii i porównania osadzonych warstw MIPów na plytce szklanej z war- stwa zlota i na plytce szklanej z warstwa ITO wykonano zdjecia obszaru (0,5 x 0,5) ?m 2 mikroskopii sil atomowych (ang. atomie froce microscope, AFM) za pomoca mikroskopu MultiMode® 8 firmy Bruker sterowanego kontrolerem Nanoscope V w trybie Tapping Mode. Na podstawie tych zdjec oszacowano grubosc i szorstkosc tych warstw. Warstwa MIP-TNP osadzonej na plytce szklanej z warstwa zlota byla homogeniczna o niewielkiej szorstkosci (2,7±0,4 nm) (Fig. 3a), natomiast warstwa MIP-TNP osa- dzona na plytce szklanej pokrytej ITO byla niehomogeniczna o rozpoznawalnych ziarnach srednicy 10 nm (Fig. 3b). Warstwa ta byla stosunkowo szorstka (14,4 ± 0,9 nm). P r z y k l a d 4 Pomiary spektroskopii odbiciowo-absorpcyjnej z modulacja polaryzacji w podczerwieni dla warstwy MIP-TNP Wykonano pomiary spektroskopii odbiciowo-absorpcyjnej z modulacja polaryzacji w podczer- wieni (ang. polarization-modulation infrared reflection-absorption spectroscopy, PM-IRRAS), aby po- twierdzic osadzenie kompleksu TNP z monomerem funkcyjnym (Fig. 4). Do pomiarów zastosowano plytke szklana pokryta warstwa zlota z osadzona warstwa MIP-TNP jako próbke badana i taka plytke z osadzona przez odparowanie z kropli roztworu acetonitrylu warstwa TNP jako próbke porównawcza. Do rejestracji widm PM-IRRAS zastosowano spektrofotometr Vertex 8v z transformacja Fouriera, z modulem PMA50 i detektorem MCT(Hg-Cd-Te) kontrolowanym oprogramowaniem OPUS 6.5 firmy Bruker. Widma byly zbierane z powierzchni 0,3 cm 2 . Dla próbki porównawczej TNP analizowano pasma zarejestrowane przy 875, 1157, 1248, 1365 i 1428 cm -1 . Porównanie widm doswiadczalnych i wygenerowanych teoretycznie (ponizej) wykazuja, ze ww. pasma odpowiadaja drganiom rozciagajacym wiazan w grupie –NO2 (Fig. 4a). Dla próbki ba- danej MIP-TNP zaobserwowano podobne pasma przy 876, 1158, 1178, 1202, 1227, 1377 i 1431 cm -1 (Fig. 4b). Wskazuje to na obecnosc grupy –NO2 w próbce badanej i potwierdza wdrukowanie TNP w warstwe MIP-TNP. Widma IR zostaly równiez wygenerowane teoretycznie za pomoca teorii funkcjonalu gestosci (ang. density functional theory, DFT) z zastosowaniem metody B3LYP/6-31G w przyblizeniu harmo- nicznym za pomoca oprogramowania Gaussian 09 [Frisch, M.J., et al, Gaussian, Inc., Wallingford, CT, 2009]. W celu przypisania odpowiednich drgan wiazan do rejestrowanych pasm zastosowano proce- dure dwuetapowa. W jej pierwszym etapie zastosowano analize rozkladu energii wibracyjnej (ang. vibrational energy distribution analysis, VEDA). Obliczone za pomoca VEDA [Jamróz, M.H., Spectro- chim. Acta Part A, 114 (2013) 220] drgania w widmie IR zostaly opisane we wspólrzednych wewnetrz- nych. W drugim etapie porównano zmierzone widma IR z widmami teoretycznymi. Do porównania tego zastosowano oprogramowanie SPESCA [Spectroscopy and Molecular Modeling Group, Institute of Nuclear Chemistry and Technology, Warsaw, pp. accessed: 2014, http://smmg.pl/software/sowtware-spesca.htmn. Oprogramowanie to wykorzystuje metode regresji liniowej wazonej do przypisania czestotliwo- sci teoretycznie wygenerowanych drgan wiazan do eksperymentalnie wyznaczonych polozen pasm. PL 227 373 B1 6 P r z y k l a d 5 Pomiary rezystencji warstwy MIP-TNP za pomoca elektrochemicznej spektroskopii impe- dancyjnej Wykonano pomiary opornosci elektrycznej warstwy podwójnej za pomoca elektrochemicznej spektroskopii impedancyjnej (ang. electrochemical impedance spectroscopy, EIS). Na dyskowej elek- trodzie platynowej o srednicy 1 mm osadzono warstwe MIP-TNP wedlug procedury opisanej w Przy- kladzie 2. Pomiary EIS przeprowadzono dla acetonitrylowego roztworu 0,1 mM ferrocenu i 0,1 M (TBA)CIO4. Pomiar prze- prowadzono przy potencjale otwartego obwodu dla czestotliwosci zmienianych w zakresie od 200 kHz do 100 mHz, co 10 mHz, przy amplitudzie 0,1 mV. Otrzymano trzy widma EIS (Fig. 5). Zarówno plytki szklane pokryte ITO jak i plytki szklane pokryte zlotem z osadzonymi warstwami MIP-TNP, z wyekstrahowanym szablonem, moco- wano w uchwycie do próbek stalych model J1933 pod katem 30° wzgledem promieniowania zródla wzbudzajacego. Pomiary byly prowadzone przy dlugosci fali wzbudzajacej 350 nm. Dzialanie chemosensora fluorescencyjnego polega na zmianie fluorescencji warstwy MIP-TNP z wyekstrahowanym szablonem po selektywnym zwiazaniu analitu TNP w lukach molekularnych polimeru. Dla plytki szklanej pokrytej zlotem przed pierwszym nakropleniem roztworu analitu zaobserwo- wano dwa piki fluorescencji, jeden przy 420 a drugi przy 600 nm (Fig. 6). Piki te stopniowo zanikaly wraz ze wzrostem stezenia nakraplanego, a nastepnie odparowywanego, acetonitrylowego roztworu analitu TNP. Pik przy 420 nm malal szybciej niz pik przy 600 nm. Natomiast w przypadku plytki pokry- tej ITO oba piki rosly wraz ze wzrostem stezenia nakraplanego roztworu analitu TNP. Wzrost ten byl najprawdopodobniej zwiazany z pecznieniem warstwy na skutek wnikania do niej czasteczek analitu i towarzyszacych im czasteczek rozpuszczalnika (Fig. 8). Aby skonstruowac krzywe kalibracyjne, wykreslono zaleznosc wzglednej fluorescencji od steze- nia analitu. W pomiarach tych oprócz analitu TNP nakraplane byly roztwory acetonitrylowe, o róznych stezeniach, substancji przeszkadzajacych w oznaczeniu TNP. Struktury tych substancji byly podobne do struktury analitu, tj. 2,4,6-trinitrotoluen, znany jako trotyl (ang. 2,4,6-trinitrotoluene, TNT) i 2,4-dinitrotoluenu (ang. 2,4-dinitrotoluene, DNT). Z nachylenia krzywych kalibracyjnych wyznaczono czulosci chemo- sensora z warstwa MIP-TNP wzgledem tych substancji. Selektywnosc chemosensora, której miara jest stosunek jego czulosci wzgledem TNP do czulosci wzgledem substancji przeszkadzajacej, jest dosc wysoka. Its consumption leads to poisoning [Wyman, J. F., Serve, M. P., Hobson, D. W., Lee, L. H., Uddin, D. E., J. Toxicol. Environ. Health, 37 (1992) 13]. Symptoms of PAH poisoning include: headache and abdominal pain, nausea, vomiting and a yellowish skin tone. A high dose of PAH can cause red blood cell damage, gastroenteritis, inflammation of the kidneys, jaundice, or haematuria. If PAH enters the eye, it may irritate the eye or cause a disease of the cornea of the eye. The lethal dose for rabbits is 120 mg / kg PAH [Patnaik, P., A comprehen- sive Guide to the Hazardous Properties of Chemical Substances, John Wiley & Sons, New Jersey, 2007]. This is why detection and selective determination of 2,4,6-trinitrophenol is so important. Several methods are used for the determination of TNP in trace amounts. Most of the work published on this subject so far focuses mainly on the use of mediators as fluorophores, such as, for example, a conductive polymer, poly [2-methoxy-5- (2-ethylhexyloxy) -1,4-phenylene vinyl] [Chu, F., Tsiminis , G., Spooner, NA Monro, T., Sens. A MIP has already been developed, printed with TNP [Huynh, T.-P., Sosnowska, M. I., Sobczak, J. W., KC, C. B., Nestrov, V. N., D'Souza, F., Kutner, W., Anal. Chem. 85 (2013) 8361], but spectrofluorimetric detection has not yet been used to determine the TNP. In this application we present the preparation of such a MIP and its use in the form of a recognition layer of a spectrofluorimetric sensor for the selective determination of TNP. The object of the present invention is to develop a MIP layer and a method of its production so that it can be used as a TNP selective recognition layer. This goal is also a method of its deposition on a chemical signal converter by means of electropolymerization under potentiodynamic conditions. The present invention comprises a conductive bisbitiophene polymer, preferably in the form of a molecularly imprinted polymer layer, characterized in that it is a polymer containing bis (2,2'-bitienyl) - (4-aminophenyl) methane as a functional monomer, and 2,2 ' - -bis (2,2'-bitiophene-5-yl) -3,3'-bitionaphthene as a cross-linking monomer, and molecular gaps imprinted with 2,4,6-trinitrophenol (TNP) molecules. Preferably, the cross-linking monomer used simultaneously acts as a fluorophore. Furthermore, the present invention includes a method for the preparation of a conductive bisbitiophene recognition polymer as defined above, a method for molecular imprinting of 2,4,6-trinitrophenol (TNP) by electropolymerization, preferably under potentiodynamic conditions, characterized by that the electropolymerization of the functional monomer bis (2,2'-bitienyl) - (4-aminophenyl) methane is carried out in the presence of the 2.2'-bis (2,2'-bitiophene-5-yl) cross-linking monomer - 3,3'-bitionaphthene and in the presence of 2,4,6-trinitrophenol (TNP) as a template, which template is molecularly imprinted during this electropolymerization to form a conductive recognizing bisbitiophene polymer with a molecularly imprinted 2,4,6-trinitrophenol template ( TNP), from which the template is extracted with methanol after depositing the polymer thus obtained in the form of a layer on the chemical signal converter / working electrode and a diagnosis is obtained conductive bisbitiophene polymer with molecular cavities after the printed pattern; the cross-linking monomer has a dual role, i.e. on the one hand it cross-links MIP-TNP and on the other hand it is a fluorophore, the electropolymerization using a solution of a mixture of solvents, preferably acetonitrile and toluene, in a volumetric ratio, preferably 95: 5, containing 2 , 4,6-trinitrophenol (TNP), bis (2,2'-bitienyl) - (4-aminophenyl) methane and 2,2'-bis (2,2'-bitiophene-5-yl) -3,3 ' -bitionaphthene, preferably in a molar ratio of 1: 1: 2, and a base electrolyte, preferably (VII) tetrabutylammonium chlorate, (TBA) ClO4, preferably 0.1 M, and the applied potential is linearly changed in a cyclic manner in range from 0 to 1.5 V, with respect to Ag / AgCI, with a constant rate in the range from 5 mV / s to 5 V / s. Preferably, in the method according to the invention, the applied potential is varied linearly cyclically in the range from 0.5 to 1.30 V with respect to Ag / AgCl, with a rate constant of 20 mV / s. Preferably, in the method according to the invention, a chemical signal converter / working electrode is used for deposition, selected from the group of solid conducting and semi-conducting electrodes, preferably in the form of a gold sputtered glass plate and a glass plate with an indium tin oxide layer. The invention also includes the use of a bisbitiophene recognition polymer, preferably in the form of a polymer layer, molecularly imprinted with 2,4,6-trinitrophenol (TNP), as defined above, as a chemical sensor recognition element for detection and / or selective determination 2, 4,6-trinitrophenol in biological samples as well as in sensors for the detection and / or determination of explosive substances into a useful analytical signal by means of spectrofluorimetric measurements. PREFERRED EMBODIMENTS OF THE INVENTION Example 1 Deposition of a molecularly imprinted polymer layer with 2,4,6-trinitrophenol on different electrodes Before depositing the MIP layer, both the gold coated glass plates and the indium tin oxide conductive glass plates were cleaned by rinsing with toluene followed by immersion for 20 min in acetone, then in water, then 10 min in isopropanol. The MIP layers were deposited on the above-mentioned plates by potentiodynamic electropolymerization in several cycles of potential changes, at a rate of 20 mV / s, in the range from 0.50 to 1.30 V vs Ag / AgCl. The polymerization was carried out in an electrochemical system in which the counter electrode was a glass plate covered with a gold layer placed opposite and parallel to the working electrode. Fig. 1 shows the changes in the current, accompanying the changes in potential, leading to the deposition of the MIP layer on glass plates coated with (a) gold and (b) ITO. For electropolymerization, a solution of 0.3 mM 2,4,6-trinitrophenol, 0.3 mM bis (2,2'-bitienyl) - (4-aminophenyl) methane and 0.6 mM 2,2'-bis (2 , 2'-bitiophene-5-yl) -3,3'-bithionaphthene in a mixture of two organic solvents, acetonitrile and toluene, mixed in a volume ratio of 95: 5, respectively. Tetrabutylammonium chlorate (VII), (TBA) ClO4, preferably at 0.1 M concentration, was used as the base electrolyte. After depositing on the plates, the MIP-TNP layer was rinsed with acetonitrile to remove any residual stock electrolyte solution. During the electropolymerization, both on the glass plate coated with a gold layer and an ITO layer, an anode peak was observed corresponding to the electro oxidation of the bis (2,2'-thienyl) methane moiety. For a gold coated glass plate, a peak appeared on the first cycle at a potential of -1.0 V vs Ag / AgCl. Then, in the next two cycles, this peak dropped and moved towards more positive potentials. This was due to the reduction in conductivity of the MIPu layer as its thickness increased. The deposited layer constitutes a barrier with increasing resistance, which hinders further electrooxidation of the bis (2,2'-thienyl) methane group in the given potential range. During the deposition of the MIP-TNP layer on the glass plate covered with the ITO layer, the anode peak appeared at a potential of -0.95 V vs Ag / AgCl and increased in the next two cycles. This means that the polymer is conductive and the increase in current may be related to an increase in the surface of the polymer due to an increase in the roughness of the layer of the deposited MIPTNP with its thickness. For comparison, layers of non-imprinted polymers (NIPs) were also prepared. They were deposited according to the procedure described above, however, from a solution that did not contain the TNP template. Example 2 Extraction of a 2,4,6-trinitrophenol template from a deposited MIP-TNP layer In order to check whether the TNP template is completely removed from the MIPu layer by extraction, the MIPu layer was deposited on a platinum disc electrode with electropolymerization under potentiodynamic conditions using several cycles of changing the potential with a rate of 50 mV / s in the range from 0.50 to 1.25 V vs Ag / AgCl, from a solution of 0.3 mM 2,4,6-trinitrophenol, 0.3 mM bis ( 2,2'-bitienyl) - (4-aminophenyl) methane, 0.6mM 2,2'-bis (2,2'-bitiophene-5-yl) -3,3'-bisthionaphthene and 0.1 M (TBA) ClO4 of two organic solvents, acetonitrile and toluene, mixed in a ratio by volume of 95: 5, respectively. The TNP template was then extracted from the MIP-TNP layer with methanol. Extractions were carried out for 4 hours. with constant stirring with a magnetic stirrer. Extraction progress was controlled by differential pulse voltammetry (DPV), in the potential range from 0 to 0.60 V vs Ag / AgCI, for acetonitrile 0.1 mM ferrocene and 0.1 M (TBA ) CIO4 (Fig. 2). A peak was observed at a potential of 0.30 V which decreased as the extraction progressed. This reduction in peak height is associated with a reduction in the conductivity of the MIP layers due to the removal of TNP from the imprinted MIP-TNP molecular voids. Example 3 Morphology of the MIP layer In order to determine the morphology and compare the deposited MIP layers on a glass plate with a gold layer and on a glass plate with an ITO layer, pictures of the area (0.5 x 0.5) μm2 of atomic force microscopy were taken. atomic froce microscope (AFM) using a Bruker MultiMode® 8 microscope controlled by a Nanoscope V controller in Tapping Mode. Based on these photos, the thickness and roughness of these layers were estimated. The MIP-TNP layer deposited on the glass plate with the gold layer was homogeneous with a slight roughness (2.7 ± 0.4 nm) (Fig. 3a), while the MIP-TNP layer deposited on the ITO-coated glass plate was non-homogeneous with recognizable grains 10 nm in diameter (Fig. 3b). This layer was relatively rough (14.4 ± 0.9 nm). Example 4 Measurements of infrared reflection-absorption spectroscopy with MIP-TNP layer Measurements of polarization-modulation infrared reflection-absorption spectroscopy (PM-IRRAS) were made to - affirm the deposition of the TNP complex with the functional monomer (Fig. 4). For the measurements, a glass plate coated with a gold layer with an deposited layer of MIP-TNP was used as the test sample and such a plate with a layer of TNP deposited by evaporation from a drop of acetonitrile solution as a reference sample. For the recording of PM-IRRAS spectra, a Vertex 8v Fourier transform spectrophotometer with a PMA50 module and an MCT (Hg-Cd-Te) detector controlled by the OPUS 6.5 software from Bruker was used. The spectra were collected from an area of 0.3 cm 2. For the comparative TNP sample, the bands recorded at 875, 1157, 1248, 1365 and 1428 cm -1 were analyzed. The comparison of the experimental and theoretically generated spectra (below) shows that the above-mentioned the bands correspond to the bond stretching vibrations in the –NO2 group (Fig. 4a). For the MIP-TNP test sample, similar bands were observed at 876, 1158, 1178, 1202, 1227, 1377 and 1431 cm-1 (Fig. 4b). This indicates the presence of the –NO2 group in the test sample and confirms the imprinting of TNP in the MIP-TNP layer. IR spectra were also theoretically generated with the use of the density functional theory (DFT) using the B3LYP / 6-31G method in a harmonic approximation using Gaussian 09 software [Frisch, MJ, et al, Gaussian, Inc. , Wallingford, CT, 2009]. A two-step procedure was used to assign appropriate vibrations to the recorded bands. In its first stage, vibrational energy distribution analysis (VEDA) was used. Calculated by VEDA [Jamróz, M.H., Spectrochim. Acta Part A, 114 (2013) 220] the vibrations in the IR spectrum have been described in internal coordinates. In the second step, the measured IR spectra were compared with the theoretical ones. The SPESCA software [Spectroscopy and Molecular Modeling Group, Institute of Nuclear Chemistry and Technology, Warsaw, pp. accessed: 2014, http://smmg.pl/software/sowtware-spesca.htmn. This software uses a weighted linear regression method to assign the frequency of the theoretically generated bond vibrations to the experimentally determined positions of the bands. MIP-TNP layer resistance measurements by electrochemical impedance spectroscopy The double layer electrical resistance was measured by electrochemical impedance spectroscopy (EIS). On a platinum disc electrode with a diameter of 1 mm, the MIP-TNP layer was deposited according to the procedure described in Example 2. EIS measurements were carried out for an acetonitrile solution of 0.1 mM ferrocene and 0.1 M (TBA) ClO4. The SP-300 potentiostat from BioLogic SAS (Claix, France), computer-controlled with EC-LAB® software from the same manufacturer, was used for the measurements. The measurement was carried out at the open circuit potential for frequencies varying from 200 kHz to 100 mHz, every 10 mHz, at an amplitude of 0.1 mV. Three EIS spectra were obtained (Fig. 5). These spectra show the change of resistance (curve 1 in Fig. 5) before and (curve 2 in Fig. 5) after TNP extraction according to the procedure shown in Example 2, and also (curve 3 in Fig. 5) after immersion of the electrode with the deposited MIP-TNP layer with extracted TNP template up to 50 nM TNP for 20 min with constant stirring by means of a magnetic stirrer. Example 6 Determination of 2,4,6-trinitrophenol by spectrofluorimetry under stationary conditions For the determination of TNP by fluorescence spectroscopy, gold-coated glass plates and MIP-TNP-coated ITO-coated glass plates were used. These layers were deposited according to the procedure outlined in Example 1. A TNP pattern was extracted from these layers according to the procedure of Example 2. A Fluorolog-3 model FL3-22 spectrofluorimeter by Horiba Jobin Yvon was used for fluorescence measurements. Both the ITO-coated glass plates and the MIP-TNP-deposited gold-plated glass plates with the extracted template were mounted in the Model J1933 solid specimen holder at a 30 ° angle to the radiation of the exciter source. Measurements were carried out at an excitation wavelength of 350 nm. The effect of the fluorescent chemosensor is to change the fluorescence of the template-extracted MIP-TNP layer after selectively binding the TNP analyte in the molecular cavities of the polymer. For the gold coated glass plate, two fluorescence peaks were observed before the first spotting of the analyte solution, one at 420 and the other at 600 nm (Fig. 6). These peaks gradually disappeared as the concentration of the spotted and then evaporated acetonitrile TNP analyte solution increased. The peak at 420 nm was malal faster than the peak at 600 nm. In contrast, in the case of the plate coated with ITO, both peaks increased with increasing concentration of the spotted TNP analyte solution. This increase was most likely related to the swelling of the layer due to penetration of analyte particles and accompanying solvent particles (Fig. 8). Fluorescence measurements were performed on the ITO glass plate coated with a NIPu layer to confirm the imprinting of the MIP-TNP layer. The NIPu layer was deposited on the ITO plate according to the procedure outlined in Example 1. Measurement of the fluorescence of the NIPu layer showed that the TNP binding by NIP was much weaker than the TNP binding by the MIP-TNP layer with the TNP pattern extracted. Therefore, in the case of the NIPu layer, with the increase of the TNP concentration in the solution, the relative fluorescence increased only slightly. Example 7 Determination of the selectivity of the MIP-TNP chemosensor by spectrofluorimetry under steady-state conditions In order to determine the selectivity of the chemosensor, a series of measurements of the change in fluorescence intensity of the MIP-TNP layer with the extracted template deposited on a glass plate with ITO were carried out. The results of these measurements are presented in the form of calibration curves in Fig. 9. In these measurements, in addition to the TNP analyte, acetonitrile solutions of various concentrations of the interfering substances were spotted in the TNP determination. The structures of these substances were similar to that of the analyte, i.e. 2,4,6-trinitrotoluene, known as TNT, and 2,4-dinitrotoluene (2,4-dinitrotoluene). DNT). The sensitivity of the chemosensor with the MIP-TNP layer towards these substances was determined from the slope of the calibration curves. The selectivity of the chemosensor, as measured by the ratio of its sensitivity towards PAH to its sensitivity towards the interfering substance, is quite high.
