Przedmiotem zgłoszenia jest sposób funkcjonalizacji eksfoliowanego heksagonalnego azotku boru, który charakteryzuje się tym, że eksfoliowany heksagonalny azotek boru zanurza się w nadtlenku wodoru w stosunku 1 : 2, gotuje się pod chłodnicą zwrotną w temperaturze 110°C przez 48 godzin, suszy się i miesza z wodą w stosunku 2 : 50 w stosunku do wody. Następnie pod chłodnicą zwrotną doprowadza się do wrzenia i dodaje kwas chlorozłotowy w stosunku 4 : 3 w odniesieniu do heksagonalnego azotku boru i podgrzewa przez 5 minut w temperaturze 100°C. Dodaje się kroplami cytrynian sodu w stosunku 5 : 1 do ilości kwasu chlorozłotowego i prowadzi się reakcję przez 1 godzinę w stanie wrzenia, po czym otrzymany roztwór studzi się, odwirowuje się otrzymany związek i przemywa wodą, w celu wyrównania pH do 7. Otrzymano eksfoliowany heksagonalny azotek boru w następujący sposób. Sproszkowany heksagonalny azotek boru w ilości 0,75 g zmieszano z manganianem (VII) potasu w stosunku 1:4 i mieszano mieszadłem magnetycznym pod chłodnicą zwrotną. Stopniowo dolewano kwas siarkowy (VI) o stężeniu 96% w ilości 60 ml, podgrzewano do temperatury 40°C przez 6 godzin i studzono. Następnie dodano nadtlenek wodoru w ilości 200 ml i całość utrzymywano w niskich temperaturach z wykorzystaniem łaźni wodnej z lodem. Otrzymany eksfoliowany chemicznie heksagonalny azotek boru poddano oczyszczaniu w wirówce, wyrównując pH do 7 i przemyto wodą. Następnie poddano go eksfoliacji mechanicznej. Eksfoliacja mechaniczna polegała na tym, że heksagonalny azotek boru w ilości 0,3 g zmieszano z N-metylopirolidonem w ilości 15 ml a następnie poddano sonikacji tipem przez 30 minut. Po odparowaniu rozpuszczalnika otrzymano heksagonalny azotek boru cienkich warstwach. Stosowano sonikator o mocy 600 W (25%), w którym moduł pulsowania ustawia się na 5 sekund, (5 sekund włączony, 5 sekund wyłączony).Exfoliated hexagonal boron nitride was obtained as follows. After evaporation of the solvent, hexagonal boron nitride was obtained in thin layers. A 600 W (25%) sonicator was used, in which the pulsing module was set to 5 seconds (5 seconds on, 5 seconds off).

Określenie biokompatybilności zsyntetyzowanego nanozwiązku stanowiło istotny etap charakterystyki kompozytu h-BN_AuNP i obejmowało - określenie potencjalnej zmiany morfologii komórek oraz aktywności metabolicznej.Determination of the biocompatibility of the synthesized nanocompound was an important step in the characterization of the h-BN_AuNP composite and included the determination of a potential change in cell morphology and metabolic activity.
W analizie biologicznej wykorzystano komórki dwóch adhernetnych linii komórkowych - L929 (lina komórkowa mysich fibroblastów, ATCC® CCL-1 ™) oraz MCF-7 (linia komórkowa raka piersi, adenocarcinoma, ATCC® HTB-22 ™). Każda z linii została poddana działaniu kompozytu zawieszonego w pełnym medium hodowlanym DMEM uzupełnionym o bydlęcą surowicę płodową - FBS (10%), r-r antybiotyków (penicylinę-streptomycynę 50 IUml-1), r-r L- glutaminy (2 mM) oraz r-r amfoterycyny B (2.5 pgml-1). Komórki oby linii inkubowano z nanomateriałem w następujących stężeniach - 0,0, 3,125, 6,25, 10,0, 12,5, 25,0, 50,0, 100,0, 200,0 pgml-1 przez okres 24 godzin (w standardowych warunkach dla hodowli komórkowych - 37°C, 95% wilgotności, 5% CO2).Two adhernetic cell lines were used in biological analysis - L929 (murine fibroblast cell line, ATCC® CCL-1 ™) and MCF-7 (breast cancer, adenocarcinoma cell line, ATCC® HTB-22 ™). Cells of both lines were incubated with the nanomaterial at the following concentrations - 0.0, 3.125, 6.25, 10.0, 12.5, 25.0, 50.0, 100.0, 200.0 pgml -1 for a period of 24 hours (under standard conditions for cell culture - 37 ° C, 95% humidity, 5% CO2).
Analizę morfologiczną komórek prawidłowych i nowotworowych oparto o obserwację mikroskopową (kontrast fazowy, mikroskop odwrócony, Nikon Eclipse TS-100) oraz holograficzną (HoloMonitor™ M4, Phase Holographic).Morphological analysis of normal and neoplastic cells was based on microscopic observation (phase contrast, inverted microscope, Nikon Eclipse TS-100) and holographic (HoloMonitor ™ M4, Phase Holographic) observation.
W przypadku obu obserwacji mikroskopowych komórki hodowano w butelkach typu T-flask 25 (o powierzchni wzrostu 25 cm2), po uprzednim ich wysianiu w ilości 7,5x105/naczynie hodowlane. Mikrografię z użyciem mikroskopu odwróconego wykonano pod koniec 12-godzinnej inkubacji z testowanym czynnikiem. Natomiast analiza holograficzna polegała na poklatkowej dokumentacji holograficznej obu linii komórkowych w czasie rzeczywistym, podczas 12-godzinnej inkubacji w odstępie 1 min.
