Description
Opis wynalazkuDescription of the invention
The subject of the invention is a method for producing poly(naringenin), intended for use as a stabilizer of polymer materials, materials based on biopolymers and environmentally friendly polymers, as well as as an antimicrobial additive to polymer packaging materials.
Naringenin is a natural polyphenol from the flavonoid group, found in citrus fruits. This bioflavonoid has been shown to have, among other things, antioxidant properties. Due to the ability to delay oxidation processes, scientific literature proposes the use of naringenin as a stabilizing agent for polymeric materials, and also describes the biological properties of this compound, including its antibacterial effect.
The valuable properties of polyphenols from the flavonoid group depend strictly on their chemical structure. Literature data show that polymeric forms of flavonoids may be characterized by stronger antioxidant properties, better antimicrobial activity and higher thermal stability.
Znane są następujące reakcje polimeryzacji flawonoidów: polimeryzacja enzymatyczna, fotopolimeryzacja, polimeryzacja katalizowana kwasem HCl, autokondensacja, polimeryzacja ze związkiem sieciującym. The following polymerization reactions of flavonoids are known: enzymatic polymerization, photopolymerization, polymerization catalyzed by HCl acid, autocondensation, polymerization with a cross-linking compound.
Gotowy produkt przemywa się cykloheksanem, a następnie mieszaniną wody z etanolem (czasopisma: Colloidsand Surface A: Physicochemical and Engineering Aspects 452 (2014) 173-180 oraz Materials Science and Engineering C 44 (2014) 9-16).There is also a known method of obtaining polymeric forms of quercetin and its glycoside rutin, which involves polymerization with a cross-linking compound in the form of glycerol diglycidyl ether (GDE), using L-α lecithin as a surfactant, in a cyclohexane environment in the case of obtaining poly(quercetin) or in a gasoline environment when obtaining poly(rutin). The finished product is washed with cyclohexane and then with a mixture of water and ethanol (journals: Colloidsand Surface A: Physicochemical and Engineering Aspects 452 (2014) 173-180 and Materials Science and Engineering C 44 (2014) 9-16).
The molecule of quercetin and rutin, flavonoids from the flavonol group, contains, like most flavonoids, a carbon skeleton with a ketone group in position 4, while naringenin is a flavonoid from the flavonone group and, unlike quercetin, it does not have a double bond in the C ring in its structure. Moreover, unlike from quercetin, naringenin does not have an ortho-dihydroxyl (catechol) group in the ring of the B structure.
Celem wynalazku jest opracowanie sposobu wytworzenia polimerycznej formy naringeniny. The aim of the invention is to develop a method for producing a polymeric form of naringenin.
The method of obtaining poly(naringenin), using the polymerization method with a cross-linking compound in the form of glyceryl diglycidyl ether (GDE), in the presence of L-α lecithin as a surfactant, in a cyclohexane environment, according to the invention, consists in first preparing a solution of (+)-naringenin in a 1 M aqueous sodium hydroxide solution using 1 g of (+)-naringenin per 10 ml of hydroxide solution. Then, the solution prepared in this way is added to a 0.1 M solution of lecithin in cyclohexane, using 2.67 ml of (+) naringenin solution per 100 ml of lecithin solution, and then the whole thing is stirred for 2 hours at a speed of 1000 rpm at room temperature. After this time, glycerol diglycidyl ether (GDE) is added in an equimolar amount in relation to the amount of (+) naringenin used and the mixture is mixed at a speed of 1000 rpm until the GDE is completely consumed, preferably 2 hours, after which the obtained poly(naringenin) is washed twice with cyclohexane with centrifugation at 6000 rpm for 30 minutes, at room temperature and dried at 35°C for 72 hours.
The obtained poly(naringenin) powder is characterized by increased resistance to oxidation and better thermal stability compared to monomeric naringenin and also has antimicrobial activity. Poly(naringenin) can be used as a stabilizer of polymeric materials, materials based on biopolymers and environmentally friendly polymers, as well as as an antimicrobial additive to polymeric packaging materials.

Wyniki przedstawiono na wykresach na fig. 1 rysunku. Widma w podczerwieni przedstawiono na wykresach na fig. 1 rysunku. Zgodnie z danymi literaturowymi dotyczącymi widm w poczerwieni polimerycznych form flawonoidów (kwercetyny i rutyny), obecność następujących pasm w widmie w podczerwieni była charakterystyczna dla polimerycznej formy flawonoidu: 370-1250 cnr1 - drgania rozciągające arylu, 3700-3000 cnr1 - szerokie pasma odpowiadające tworzeniu wolnego OH pochodzącego z GDE, 1061 cm1 - C-CO-C w ketonach, 750-790 cnr1 i 800-900 cm1 - epoksydy (od GDE).The results are plotted in Figure 1 of the figure. The infrared spectra are plotted in Figure 1 of the figure. As can be seen from Fig. 1, the infrared spectrum of the obtained powder differed from the reference spectrum of (+)-naringenin, which confirmed the fact that a compound with a different structure was obtained from the - (+)-naringenin monomer. According to the literature data on the infrared spectra of polymeric forms of flavonoids (quercetin and rutin), the presence of the following bands in the infrared spectrum was characteristic of the polymeric form of the flavonoid: 370-1250 cnr 1 - aryl stretching vibrations, 3700-3000 cnr 1 - broad bands corresponding to formation of free OH derived from GDE, 1061 cm 1 - C-CO-C in ketones, 750-790 cnr 1 and 800-900 cm 1 - epoxides (from GDE).
Ponadto na widmie obecne były piki charakterystyczne dla grup funkcyjnych obecnych we flawonoidach:In addition, the spectrum included peaks characteristic of functional groups present in flavonoids:
2922 cm1 - Ar-CH3,2922 cm 1 - Ar-CH 3 ,
1560-1570 cm1 i 1450-1500 cm1 - drgania wibracyjnie od pierścieni aromatycznych flawonoidu, 1165 cm1 -C-OH.1560-1570 cm 1 and 1450-1500 cm 1 - vibration vibrations from the aromatic rings of the flavonoid, 1165 cm 1 -C-OH.
The appearance of bands characteristic of polymeric forms of flavonoids in the poly(naringenin) spectrum indicated the cross-linking reaction of (+)-naringenin and the formation of a high-molecular/polymeric compound.
Otrzymany proszek poli(naringeniny) poddano analizie termograwimetrycznej TGA. 2 rysunku. The obtained poly(naringenin) powder was subjected to TGA thermogravimetric analysis. Powder samples were heated at temperatures from 25°C to 800°C under dynamic argon flow (50 ml/min). For comparison, TGA analysis of the reference (+)-naringenin was also performed. The results are presented in Table 1 below and in the thermograms shown in Fig. 2 of the drawing.

T10, T50, T60 oznaczają odpowiednio temperatury, w których nastąpił ubytek masy badanej próbki wynoszący 10% i 50% i 60%.T10, T50, T60 mean the temperatures at which the mass loss of the tested sample occurred by 10%, 50% and 60%.
Monomeryczna naringenina rozkłada się jednoetapowo. W zakresie temperatur 290-380°C następuje 67% ubytek masy próbki.Monomeric naringenin is degraded in one step. In the temperature range of 290-380°C, there is a 67% loss in sample mass.
Poli(naringenina) rozkłada się dwuetapowo. Pierwszy etap rozkładu następuje około 250°C, a ubytek masy wynosi 10%. Poly(naringenin) is broken down in two stages. The first stage of decomposition occurs around 250°C and the weight loss is 10%. The second stage of decomposition occurs in the temperature range of 270-370°C. The second stage is accompanied by a loss of sample mass of 36.2%. The decomposition of poly(naringenin) begins at a lower temperature than the decomposition of naringenin (T10 poly(naringenin) = 144°C, T10 naringenin = 307°C). The half-decomposition temperature of poly(naringenin) T50 is 20°C lower than the T50 of monomeric naringenin. This may be due to the addition of the cross-linking compound GDE, which may reduce the T10 and T50 values. The final decomposition temperature of polymeric naringenin T60 is 94°C higher than T60 of monomeric naringenin, which means higher thermal stability of the polymeric form of naringenin.
Otrzymany proszek poddano także różnicowej kalorymetrii skaningowej DSC.The obtained powder was also subjected to differential scanning calorimetry (DSC).
3 rysunku. The samples were heated from -80 to 400°C at a rate of 10°C/minute in an air atmosphere. For comparison, DSC of monomeric naringenin was also performed. The results are presented in Table 2 below and in the thermograms in Fig. 3 of the drawing.

Tablica 2Table 2
Tg oznacza temperaturę zeszklenia, AHm - entalpię topnienia, Tm - temperaturę topnienia, ΔΗ0 entalpię utleniania i degradacji, To - temperaturę utleniania i degradacji.T g is the glass transition temperature, AH m - melting enthalpy, T m - melting point, ΔΗ 0 oxidation and degradation enthalpy, T o - oxidation and degradation temperature.
Badania prowadzono metodą dynamiczną „fiask shake methods”.Poly(naringenin) has a lower melting point than monomeric naringenin. This is due to the addition of the cross-linking compound GDE, which can lower the Tm. The melting enthalpy of poly(naringenin) is approximately 2.6 times higher than the melting enthalpy of monomeric naringenin. Poly(naringenin) has a higher final oxidation temperature T o (by 13.79°C) and a higher oxidation enthalpy ΔΗ 0 (about 6.8 times). The polymeric form of naringenin is therefore more resistant to oxidation. The obtained poly(naringenin) powder was also tested for antifungal activity. The research was conducted using the dynamic fiasco shake method.
Określono współczynniki zamierania mikroorganizmów D:The tests used fungal test strains: Candida albicans ATCC 10231 and Aspergillus niger ATCC 16404. The number of microorganisms in the test tubes after 24 hours of incubation was determined by the culture method on MEA medium (fungi). Additionally, control samples (microorganisms only) were counted at the beginning of the experiment (t = 0). Results were reported as the number of colony-forming units/ml of medium (CFU/ml). The death rates of microorganisms D were determined:
D = (log liczby mikroorganizmów t = o - log liczby mikroorganizmów t = 24).D = (log number of microorganisms t = o - log number of microorganisms t = 24).
Wyniki badań zestawiono w poniższej tablicy 3.The test results are summarized in table 3 below.
Tablica 3Table 3
Po 24 godzinach inkubacji obserwowano przyrost liczby wszystkich organizmów w pożywce kontrolnej bez związków polifenolowych. After 24 hours of incubation, an increase in the number of all organisms was observed in the control medium without polyphenolic compounds.
Przeanalizowano wzrost liczby komórek drożdży Candida albicans w hodowli monomerycznej naringeniny i poli(naringeniny). The increase in the number of Candida albicans yeast cells in the culture of monomeric naringenin and poly(naringenin) was analyzed. The death rate of microorganisms D in the poly(naringenin) culture was 0.64 logarithm values, which meant that the poly(naringenin) sample had bacteriostatic activity. The reference monomeric naringenin did not show such activity.
Monomeryczna naringenina i poli(naringenina) wykazują porównywalną aktywność antygrzybową.The number of Aspergillus niger mold cells after 24 hours increased by more than one order only in the control sample without polyphenols. In the remaining samples containing monomeric naringenin and poly(naringenin), a reduction in the number of cells in cultures with both compounds was noted, which means that they have antifungal activity. Monomeric naringenin and poly(naringenin) show comparable antifungal activity.
