Abstract
Zgłoszenie dotyczy sposobu wytwarzania środka przeciwzakrzepowego, z rzepiku pospolitego (Agrimonia eupatoria), obejmującego ekstrakcję substancji aktywnych alkoholem lub ketonem, a następnie wodą oraz wyodrębnianie ich poprzez ekstrakcję rozpuszczalnikami w układach dwufazowych, a po zatężeniu, oddzielenie składników aktywnych, poprzez wytrawianie alkoholem niższym z fazy stałej. Wyekstrahowane składniki aktywne, stanowiące mieszaninę koniugatów polifenolowo-polisacharydowych, o masie cząsteczek od 60 000 Da do 220 000 Da i współczynniku rozproszenia masy składników polimerowych wynoszącym od 3,4 do 3,6 oraz wartość lepkości względnej (?WZ) wynosi 1,11500 - 1,11750, poddaje się częściowej degradacji i homogenizacji na cząsteczki o masie cząsteczkowej od 10 000 do 45 000 Da, przy użyciu ultradźwięków o częstotliwości 20 kHz oraz mocy w zakresie 30 - 70 W. Otrzymaną mieszaninę poreakcyjną rozdziela się na membranie filtracyjnej, w celu usunięcia mniejszych niż 5 000 Da produktów degradacji, uzyskane składniki aktywne, przekształca się w sole sodowe lub sole wapniowe, a po uzyskaniu pH roztworu na poziomie 7,2 i po wysuszeniu otrzymuje środek przeciwzakrzepowy. Przedmiotem zgłoszenia jest również środek przeciwzakrzepowy, otrzymany z części naziemnych rzepiku pospolitego (Agrimonia eupatoria) stanowiący mieszaninę, koniugatów polifenoli z polisacharydami, o makrocząsteczkach częściowo zdegradowanych, a częściowo skondensowanych przy użyciu ultradźwięków, których masa cząsteczkowa wynosi od 10 000 Da do 45 000 Da, współczynnik rozproszenia masy składników polimerowych ma wartość od 1,2 do 1,5, oraz lepkość względną (?WZ) wynosi od 1,01617 - 1,02119. Zgłoszenie dotyczy też zastosowania środka przeciwzakrzepowego w kompozycjach farmaceutycznych, kompozycjach środków spożywczych specjalnego przeznaczenia medycznego, stosowanych w zakrzepicy żył głębokich, w chorobie niedokrwiennej serca, chorobie aorty i naczyń obwodowych, w powikłaniach zakrzepowych spowodowanych urazem lub zabiegiem operacyjnym oraz w kompozycjach kosmetycznych.The application relates to a method for producing an anticoagulant from agrimony (Agrimonia eupatoria), involving the extraction of active substances with alcohol or ketone, followed by water, and their isolation by extraction with solvents in two-phase systems, and after concentration, separation of active ingredients, by digesting with lower alcohol from the phase constant. Extracted active ingredients, being a mixture of polyphenol-polysaccharide conjugates, with a molecular weight from 60,000 Da to 220,000 Da and a mass dispersion factor of polymer components from 3.4 to 3.6 and a relative viscosity (?WZ) value of 1.111500 - 1.11750, is partially degraded and homogenized into particles with a molecular weight from 10,000 to 45,000 Da, using ultrasound at a frequency of 20 kHz and power in the range of 30 - 70 W. The resulting reaction mixture is separated on the filtration membrane in order to removing less than 5,000 Da of degradation products, the resulting active ingredients are converted into sodium salts or calcium salts, and after obtaining a solution pH of 7.2 and after drying receives an anticoagulant. The subject of the application is also an anticoagulant, obtained from the ground parts of agrimony (Agrimonia eupatoria) constituting a mixture of polyphenols conjugates with polysaccharides, with macromolecules partially degraded and partly condensed using ultrasounds, whose molecular weight is from 10,000 Da to 45,000 Da, the mass dispersion factor of polymer components has a value from 1.2 to 1.5, and the relative viscosity (lepWZ) is from 1.01617 - 1.02119. The application also relates to the use of an anticoagulant in pharmaceutical compositions, foodstuff compositions for special medical purposes, used in deep vein thrombosis, in ischemic heart disease, aortic and peripheral vascular disease, in thrombotic complications due to trauma or surgery, and in cosmetic compositions.
