Abstract

Przedmiotem zgłoszenia jest sposób wytwarzania polilaktydowych rusztowań gąbczastych do hodowli komórek nabłonka walcowatego, zgodnie z którym w reaktorze szklanym umieszcza się roztwór polilaktydu w dioksanie, dichlorometanie lub chloroformie o stężeniu 1,5 - 6% wag., roztwór miesza się przez 20 - 28 h z szybkością 550 - 700 obr/min., w temperaturze 20 - 30°C, do całkowitego rozpuszczenia polimeru, następnie do roztworu dodaje się nierozpuszczalnik polilaktydu, którym jest woda w takiej ilości, aby stężenie nierozpuszczalnika wynosiło od 0 - 5% wag. w stosunku do masy polimeru, po czym, po uzyskaniu jednorodnego roztworu polimeru, roztwór zamraża się przez 20 - 28 h w temperaturze -10 do -20°C i uzyskane zamrożone rusztowanie poddaje się żelowaniu w wodzie z metanolem i/albo etanolem w stosunku objętościowym wody do alkoholu 0,01 - 0,1, przy czym objętość kąpieli żelującej jest minimum 30 razy większa niż objętość roztworu polimeru w rozpuszczalniku, zaś etap żelowania prowadzi się w temperaturze od -10°C do -20°C przez 30 - 48 h, a następnie otrzymane rusztowanie oczyszcza się w wodzie w ciągu 0,5 - 1,5 h, po czym suszy się z pozostałości wody i rozpuszczalników przez 60 - 80 h pod zmniejszonym ciśnieniem, w temperaturze poniżej 50°C.The subject of the application is a method for producing spongy polylactide scaffolds for the cultivation of cylindrical epithelial cells, according to which a solution of polylactide in dioxane, dichloromethane or chloroform at a concentration of 1.5 - 6% by weight is placed in a glass reactor, the solution is mixed for 20-28 h at a speed of 550 - 700 rpm, at a temperature of 20-30 ° C, until the polymer is completely dissolved, then a polylactide non-solvent is added to the solution, which is water in such an amount that the concentration of the non-solvent is from 0-5 wt. based on the weight of the polymer, after which, after obtaining a homogeneous polymer solution, the solution is frozen for 20 - 28 h at -10 to -20 ° C and the obtained frozen scaffold is gelatinized in water with methanol and / or ethanol in a volume ratio of water to alcohol 0.01 - 0.1, the volume of the gelling bath is a minimum 30 times greater than the volume of the polymer solution in the solvent, while the gelling stage is carried out at a temperature from -10 ° C to -20 ° C for 30 - 48 h, and then the obtained scaffold is cleaned in water within 0.5 - 1.5 h, after which it is dried from residual water and solvents for 60 - 80 h under reduced pressure, at a temperature below 50 ° C.
