Abstract
Przedmiotem zgłoszenia jest sposób wytwarzania koniugatów 1-amino-1,1-bisfosfonianów z betuliną o wzorze ogólnym 3, gdzie R1=Me, CH2CH2CONHCH(PO(OEt)2)2, R2=H,Me, który polega na tym, że pochodną betuliny o wzorze ogólnym 1, gdzie R1=Me, CH2CH2COOH, R2=H,Me, kontaktuje się w środowisku gazu inertnego z N,N'-dicykloheksylokarbodiimidem (DCC) w stosunku molowym w zakresie od 1:1 do 1:4,0 i 1-amino-1,1-bisfosfonianem o wzorze ogólnym 2 w stosunku molowym w zakresie od 1:2,0 do 1:4,0, w rozpuszczalnikach organicznych, korzystnie w dichlorometanie (DCM), w ilości od 6,0 cm3 do 7,5 cm3 na 0,1 mmola pochodnej betuliny o wzorze ogólnym 1, reakcję prowadzi się w temperaturze 40°C do 50°C z wykorzystaniem ultradźwięków w czasie od 2h do 4h, następnie odparowuje się rozpuszczalnik organiczny pod ciśnieniem od 10 mmHg do 20 mmHg, a surowy produkt ekstrahuje rozpuszczalnikami organicznymi, korzystnie octanem etylu, następnie odparowuje się części lotne pod ciśnieniem od 10 mmHg do 20 mmHg, po czym produkt oczyszcza się metodą chromatografii kolumnowej octan etylu/metanol, gradient w zakresie od 20:1 do 5:1; v/v.
