Description

아토피 피부염 치료용 조성물
본 발명은 아토피 피부염 치료용 조성물에 관한 것으로 더 상세하게는 톳( S. fusiforme) 부탄올 분획물을 유효성분으로 하는 아토피 피부염 치료용 조성물에 관한 것이다.
아토피 피부염(atopic dermatitis, AD)은 type2 사이토카인을 생산하는 보조 T세포(Th2)와 immunoglobulin E(IgE) 항체의 작용에 의해 나타나는 만성적 피부 염증 질환으로서 외부 환경 자극에 의한 과민성 염증반응에 대해 부적절한 면역 반응으로부터 야기 된다. 아토피 피부염의 초기 단계는 알러젠(allergen) 특이성을 보이는 활성화된 Th2-type 세포에 의해 유도되는 반면, 아토피 피부염이 진행됨에 따라, 인터루킨-2 및 인터페론-γ와 같은 Th1 사이토카인이 발현됨에 따라 아토피 피부염이 만성적 성향을 띠며 심각한 난치성 수준으로 변화한다. 아토피 피부염은 염증 반응에서 기인되는 증상으로, 만성적인 재발되는 비 전염성 피부염으로 다양한 피부 장애를 가져온다. 최근 10년 동안 아토피 피부염에 대한 많은 연구가 수행된 바 있으며, 기존의 연구들에 따르면 다양한 면역 이상을 수반하는 알레르기성 염증이 기본적인 원인이라고 알려져 있어 가장 효과적인 항아토피 피부염 약물은 면역 억제 기능 효과를 가져야 할 것이며, Th1/Th2 세포의 불균형을 조절하는 동시에 인체에 안전하고, 장기간 처리했을 때 재발을 방지해야 한다. 현재 아토피 피부염에 대한 대표적인 국소적 면역억제제로는 코르티코스테로이드(corticosteroids), 사이클로스포린 A(cyclosporine A), 타크로리무스(tacrolimus)및 피메크로리무스(pimecrolimus) 등이 알려져 있으나 제한적인 효과나 림프종 등부작용의 위험성으로 장기간 사용하는 것에 대한 우려가 증폭됨에 따라 천연물 유래의 효율적인 아토피 피부염의 예방 또는 치료제에 관한 연구가 진행되고 있다. 이와 관련하여 대한민국 등록특허 제1418916호는 톳 추출물을 유효성분으로 하는 아토피 피부염의 예방 또는 치료용 조성물을 개시하고 있다.
그러나 상기 선행기술의 경우, 추출과정에서 헥산(hexane) 분획물이 포함되어 아토피 피부염의 예방 또는 치료용 조성물로 사용하는데 규제상의 문제와 유효성분의 함유량이 낮은 문제점을 안고 있다.
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 보다 효과적이고 경제적으로 대량생산이 가능한 톳(S. fusiforme)을 이용한 아토피 피부염 치료용 조성물을 제공하는 것을 목적으로 한다. 그러나 이러한 과제는 예시적인 것으로, 이에 의해 본 발명의 범위가 한정되는 것은 아니다.
본 발명의 일 관점에 따르면, 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8;2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 질환 치료용 조성물이 제공된다.
본 발명의 다른 일 관점에 따르면, 치료적으로 유효한 양의 제1항 내지 제7항 중 어느 한 항의 조성물을 아토피 피부염에 걸린 개체에 투여하는 단계를 포함하는 아토피 피부염 치료방법이 제공된다.
본 발명의 다른 일 관점에 따르면, 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8;2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 개선용 건강기능식품이 제공된다.
본 발명의 다른 일 관점에 따르면, 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8:2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 피부 개선용 화장료 조성물이 제공된다.
본 발명의 다른 일 관점에 따르면, Ⅰ) 톳을 탄소수 1 내지 4의 저급알코올 또는 그의 수용액으로 추출하는 톳 저급알코올 추출물 제조단계; Ⅱ) 상기 톳 저급알코올 추출물을 물에 용해시킨 후 n-헥산을 가하여 분획하는 1차 분획단계; Ⅲ) 상기 1차 분획단계에서 분리된 물층에 에틸초산을 가하여 분획하는 2차 분획단계; 및 Ⅳ) 상기 2차 분획단계에서 분리된 에틸초산층으로부터 에틸초산을 제거하는 에틸초산 분획물의 제조단계를 포함하는 아토피 치료효과가 향상된 톳 추출물의 초산에틸 분획물의 제조방법이 제공된다.
상기한 바와 같이 이루어진 본 발명의 일 실시예에 따르면, 톳(S. fusiforme)으로부터 항아토피 효능을 나타내는 유효물질은 종래의 아토피 피부염의 국소적 면역억제제를 대체할 수 있어 아토피 피부염의 치료제로 유용하게 사용될 수 있다. 물론 이러한 효과에 의해 본 발명의 범위가 한정되는 것은 아니다.

용어의 정의:
본 문서에서 사용되는 용어 "톳(S. fusiforme)"은 대한민국, 일본, 중국 등지에 넓게 분포하며, 해안가 암초지대를 따라 흔히 관찰되는 갈조류 중 하나로 항암, 면역조절, 항염증 및 항산화 등 다양한 약리학적 효능이 있는 것으로 알려져 있다.
본 문서에서 사용되는 용어 "DPPH 라디칼"은 진한 보라색을 띠는 유기 질소 라디칼(organic nitrogen radicals)들 중 하나이며 일반적으로 다양한 항산화 물질(antioxidant substances)의 자유라디칼 소거활성을 평가하는 시약으로 주로 사용된다. 또한 DPPH는 안정된 자유라디칼이고, 안정한 반자성 분자(diamagnetic molecule)가 되기 위해서 전자나 수소라디칼(hydrogen redical)을 받아주는 작용으로 인해 본 발명의 톳 추출물의 항산화 효능을 초기에 평가하기 위하여 라디칼소거활성 분석에 사용되었다.
본 문서에서 사용되는 용어 "Raw 264.7 세포"는 마우스 대식세포주로 LPS에 의한 Raw 264.7 세포의 활성은 toll-like receptor-4의 신호전달로 시작되어, NF-κB 신호전달과정을 통해 염증을 유도하는데 이런 과정을 통해서 질소산화물(NO)과 프로스타글란딘E2(PGE2)의 생성을 유도하고, 염증유도 사이토카인 TNF-α, IL-1β 및 IL-6 등을 비롯한 다수의 사이토카인을 분비시킨다.
발명의 상세한 설명:
본 발명의 일 관점에 따르면, 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8;2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 질환 치료용 조성물이 제공된다.
상기 아토피 질환 치료용 조성물에 있어서 상기 톳(S. fusiforme) 추출물의 부탄올 분획물은 Ⅰ) 톳을 탄소수 1 내지 4의 저급알코올 또는 그의 수용액으로 추출하는 톳 저급알코올 추출물 제조단계; Ⅱ) 상기 톳 저급알코올 추출물을 물에 용해시킨 후 에틸초산을 가하여 분획하는 1차 분획단계; Ⅲ) 상기 1차 분획단계에서 분리된 물층에 부탄올을 가하여 분획하는 2차 분획단계; 및 Ⅳ) 상기 2차 분획단계에서 분리된 부탄올층으로부터 부탄올을 제거하는 부탄올 분획물의 제조단계를 포함하는 방법으로 제조될 수 있다.

상기 제조방법에 있어서, 상기 에틸초산 분획물을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8:2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 분획을 수집한 후 용매를 제거하여 크로마토그래피 활성분획을 제조하는 단계를 추가로 포함할 수 있다.
본 출원인은 톳 추출물을 유효성분으로 하는 아토피 피부염의 예방 또는 치료용 조성물에 관한 특허(대한민국 등록특허 제1418916호)를 출원하였으나 상기 특허의 경우 톳 추출물의 분획공정에서 헥산(hexane) 분획물이 포함되어 아토피 피부염 치료제 용도로 사용하기에는 규제상의 문제와 항아토피 효능을 나타내는 유효물질의 함량이 낮은 문제점을 가지고 있어 상기 문제점을 해결하고자 예의 노력한 결과, 톳 추출물의 분획공정에서 헥산 공정을 배제하여 보다 간단하면서도 톳 추출물의 항아토피 유효성분인 부탄산(butanoic acid)의 함량은 증가시키는 톳 추출물 2차 분획물을 제조하는데 성공하였다. 따라서 상기 부탄올 함량이 높은 항아토피 유효성분은 종래의 아토피 피부염의 국소적 면역억제제를 대체할 수 있다.
이하, 실시예를 통해 발명을 상세히 설명한다. 그러나 본 발명은 이하에서 개시되는 실시예에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 수 있는 것으로, 이하 실시예는 본 발명의 개시가 완전하도록 하며, 통상의 지식을 가진 자에게 발명의 범주를 완전히 알려주기 위해 제공되는 것이다.
실시예 1: 톳 추출물 제조
1-1: 톳 용매추출 및 분획물 제조
본 발명에 사용된 톳(S. fusiforme)은 2015년 10월에 동해에서 채취하였으며, 상기 채취한 톳은 염분과 모래를 수돗물로 닦아낸 후 80% 에탄올을 이용하여 살균처리하고 증류수로 세척한 후 동결건조하여 사용하였다. 건조된 톳을 곱게 파쇄한 톳 분말(dried S. fusiforme powder, 100 g)은 1 L의 80% 에탄올에 침지하여 실온에서 24시간 주기로 3일 동안 추출하였다. 상기 추출방법의 개선을 위해 80% 에탄올 추출 동결건조물 10 g을 증류수 1L에 용해시킨 후 상기 증류수에 용해된 에탄올 추출물:에틸초산(부피비=1:1)으로 희석하여 1차 분획하였고, 에틸초산으로부터 분리된 물층 및 에틸초산층 중 물층을 분리하여 동일 부피의 부탄올을 가한 후, 2차 분획하여, 각각, 에틸초산 분획(EA), 부탄올 분획(BuOH) 및 잔사(물분획)을 얻었으며 각각의 수율은 에틸초산 분획 4.2%, 1차 물분획 89%. 부탄올 분획 56% 및 2차 물분획 28%를 나타내었다(도 1).
1-2: 톳 에틸초산 분획물 제조과정
본 발명의 톳 추출물의 항아토피 효능 개선을 위해 80% 에탄올 추출물 10 g을 n-헥산(n-헥산:물=1:1)으로 3회 분획 후 잔존물(residue)을 다시 에틸초산(에틸초산:물=1:1)으로 용매 분획하였다. 그 후, 톳 추출물에 다량 포함된 클로로필(chlorophyll)을 제거하기 위해 100% 에틸초산 1L를 이용하여 실리카 겔60(230~400 메쉬)로 오픈컬럼을 수행하였다(도 18).
실시예 2: 가스 크로마토그래피-매스 스펙트로메트리 ( GC -MS) 분석
본 발명의 일 실시예에 따라 수득한 상기 모든 시료들은 메탄올에 녹인 뒤 5975C GC/MS 기구(Agilent Technologies, 미국)를 이용하여 분석하였고 크로마토그래피(chromatography) 분리는 헬륨 캐리어 가스와 HP-5 컬럼(250 μm × 0.25 μm × 30 m, Agilent Technologies, 미국)을 이용하여 수행하였다.
구체적으로, 각 시료들은 1 μL씩 인젝터(injector)에 주입하였으며, 주입방식은 분리(split) 방식으로 비율은 5:1로 설정하였고, 컬럼(column)의 흐름은 각각 5 mL/min 및 1 mL/min로 설정하였다. 상기 인젝터의 온도는 250℃, 트랜스퍼라인(transfer line)의 온도는 250℃로 설정하였으며, GC 오븐의 온도는 50℃에서 3분 동안 유지한 후, 50℃에서 280℃까지 10 ℃/min씩 증가시켰다. 또한 이온소스(ion source) 온도는 250℃로 설정하였으며, 이온화 에너지는 -70 V로 고정하였다. 이후 분석 결과는 1059 V하에서 4분 간격으로 35 - 250 m/z의 범위에서 수집되었고 분리된 성분들은 머무름 시간(retention time)과 질량범위(mass spectra)를 바탕으로 Wiley 7n library data와 비교하여 분석하였다.
GC-MS 분석결과, 각 분획물은 머무름 시간(retention times) 4.154분에서 21.706분 사이에서 다양한 성분들을 확인하였고 각 분획물의 상대적 부탄산(butanoic acid) 함유정도를 확인하기 위해 표준물 탐지시간인 4.154-7분 사이의 면적 수준을 비교한 결과 2차 물분획을 제외한 모든 분획물에서 부탄산이 관찰되었으며 에탄올 추출물(25%), 에틸초산 분획물(9%) 및 부탄올 분획물(10%)를 포함하는 것으로 확인되어 톳의 항아토피 효능의 유효물질로 예상되는 부탄산 포함 분획물 획득과정 방법을 개선하였다(도 2).
실시예 3: 라디칼 소거활성 및 단백질 보호분석법
3-1: 라디칼 소거활성(Radical scavenging)
본 발명의 일 실시예에 따른 톳 분획물의 항산화 효능을 초기에 평가하기 위하여 DPPH 라디칼을 이용하여 라디칼 소거활성을 분석을 수행하였다.
자유라디칼 소거활성을 측정하기 위해 먼저, 상기 분획물을 DPPH 용액으로 반응시켰고(Espin JC et al., J. Agric . Food Chem ., 48:648-656, 2000) 상기 각 동결건조한 분획물은 스톡 용액(100 mg/mL)으로서 DMSO(dimethyl sulfoxide, 시그마알드리치, 미국)로 용해하였으며 각 분획물(에탄올(EtOH), 에틸초산(EA), 에틸초산-물(EA-H2O), 부탄올(BuOH) 및 부탄올-물(BuOH-H2O))은 메탄올에 용해한 DPPH 용액 0.3 mM로 반응하였다. 지정된 농도(100 μg/mL)의 각 분획물을 실온에서 20분 동안 DPPH 라디칼 용액에서 반응시킨 후 517 nm 파장에서 흡광도를 측정하였다. DPPH 자유 라디칼소거활성(%)은 하기의 공식을 이용하여 계산하였으며 양성대조군으로는 0.1 mM 아스코르브산을 사용하였고 10% DMSO는 음성대조군(vehicle control)으로 사용하였다. 하기 공식에서 Ac는 대조군 DPPH 용액의 시료 흡광도를 나타내고, A는 DPPH 용액에서의 시료의 흡광도, As는 시료 자체의 흡광도를 나타낸다:
DPPH 라디칼소거활성(%) = [Ac - (A - As)]/Ac × 100.
그 결과, DPPH 분석에서 톳의 각 분획물이 100 μg/mL에서 라디칼 소거활성을 나타내었고 에틸초산 및 부탄올 분획물에서 각각 높은 활성에 도달하였는데, 이는 톳이 가지는 생물학적 활성 중 항산화효능을 나타내고 있어 분획 추출이 잘 되었음을 말해준다(도 3). 도 3에 나타난 바와 같이 에틸초산과 부탄올의 라디칼소거활성은 유사하게 나타났으나 에틸초산의 수율이 낮아 경제적 타산성이 떨어지므로 이후 실험에서는 배제하였다.
3-2: 단백질 보호분석(protein protection assays)
본 발명의 일 실시예에 따른 톳 분획물의 단백질 보호분석을 위해 종래에 보고된 방법에 약간의 변형을 가하여 금속-촉매 반응을 이용한 수산기 라디칼-매개 산화 실험을 수행하였다(Mayo JC et al., Biochim. Biophys. Acta., 1620:139-150, 2003).
구체적으로, 표적 단백질인 BSA(bovine serum albumin)는 최종 농도가 0.5 mg/mL이 되기까지 150 mL의 인산완충용액(phosphate buffer, pH 7.3)에 용해시킨 후 상기 BSA 용액에 각 6.25-100 μg/mL의 각기 다른 농도의 분획물 및 아스코르브산염(Asc)을 첨가하였고 100 μM의 Cu2+ 및 2.5 mM의 H2O2의 존재 또는 부존재하에 배양 하였다. 이때 PBS(phosphate buffered saline)에 용해된 0.1 mM의 아스코르브산염(Asc)을 대조군 항산화제(control antioxidant)로 사용하였고 상대적으로 0.1 mM의 아스크로브산은 양성 대조군으로 10% DMSO는 음성대조군(vehicle control)으로 사용하였다. 반응은 37 ℃가 유지되는 진탕항온수조(shaking water bath)에서 뚜껑이 없는 튜브에 넣어 수행하였고 BSA를 포함하여 모든 반응물은 2시간 동안 배양하였다. 반응이 끝난 후에, 상기 각각의 혼합물은 10% SDS-PAGE 전기영동으로 분리한 후, 0.1% 쿠마시 블루 용액(Coomassie Blue solution)으로 염색하여 단백질을 시각화하였다.
그 결과, 각 분획물의 전 농도에서 Cu2+ 및 H2O2에 의해 생성되는 수산기 자유 라디칼에 의한 공격으로부터 BSA 단백질의 분해를 완벽하게 보호하는 단백질 수준의 항산화 활성을 나타내었다(도 4). 따라서 상기 각 분획물이 효과적으로 DPPH 라디칼 및 수산기 자유 라디칼과 같은 자유 라디칼을 소거시킬 수 있음을 확인하였고 이는 항산화 효과를 통해 아토피 피부염등을 악화시키는 산화물질을 효과적으로 제거하는 효능을 가지고 있음을 증명하였다.
실시예 4: 항염증활성 분석
4-1: 산화질소 생산 측정
본 발명의 일 실시예에 따른 톳 분획물의 항산화 효과를 더 심도있게 관찰하기 위해 마우스 대식세포주인 Raw264.7로부터 산화질소 생성을 측정하였다.
구체적으로, 24-웰 조직배양 플레이트에 Raw264.7 세포(1 X 106 세포/mL)를 분주하고 세포가 안정하게 부착 되도록 37℃에서 12시간 동안 선 배양하였다. 이어서, 세포를 PBS로 씻어내고, 샘플들[LPS(lipopolysaccharide), 각 분획물(50 μg/mL)]을 포함하는 1% FBS DMEM을 다시 보충해주고 24시간 동안 배양하였다. 그 후 그리스시약(Promega)을 이용하여 배지 속의 질산염의 양을 측정함으로써 산화질소의 생산을 측정하였다. 각 분획물에 의한 산화질소 생성 억제효과는 Raw264.7 세포주에서 측정하였는데 산화질소 생성을 자극하기 위해, 1 μg/mL의 LPS를 첨가하고 각 분획물 50 μg/mL을 24시간 까지 세포에 처리하였다.
그 결과, 부탄산이 관찰되지 않은 부탄올-물 분획물을 제외한 전 분획물에서 산화질소의 생성을 효과적으로 억제하였고, 이는 25 μmol/L의 L-NMMA(질소산화물 합성효소 저해제)와 거의 유사한 결과이다. 상기 결과들은 부탄산을 포함하고 있는 톳 분획물이 산화질소를 제거하거나 산화질소의 생성을 억제함으로써 세포 수준에서 염증물질인 산화질소의 농도 수준을 조절할 수 있다는 것을 증명하였다(도 5).
4-2: 항염증활성(anti-inflammatory activity)
본 발명의 일 실시예에 따른 톳 분획물의 항염증활성을 Raw264.7를 이용하여 세포수준에서 관찰하였다.
구체적으로, 항염증활성 실험을 위해 Raw264.7 세포(1 X 106 세포/mL)를 6-웰 조직배양 플레이트에 분주하고 세포가 안정하게 부착 되도록 37℃에서 12시간 동안 선 배양하였다. 이어서, 상기 세포를 PBS로 씻어내고, 각 샘플들[LPS(lipopolysaccharide), 각 분획물(50 μg/mL)]을 포함하는 1% FBS DMEM을 다시 보충해주고 24시간 동안 배양하였다. 반응 종료 후 배양배지를 제거하고 총 RNA 추출을 위해 TRIZOL(Invitrogen, 미국) 용액 1 mL를 각 웰에 넣고 제조사의 매뉴얼에 따라 총 RNA를 추출하였다. cDNA 합성은 1 μg의 총 RNA와 Improm-II 역전사 시스템(Promega, 미국)과 올리고 dT 프라이머를 사용하여 총량이 20 μL 되도록 조성한 뒤 역전사 반응을 수행하였고 하기 표 1에 제시된 프라이머(Bioneer, 한국)를 이용하여 PCR을 수행하였다. 정량적 실시간 PCR(quantitative real-time PCR)은 SYBR Green PCR 마스터믹스(Qiagen, 미국)을 사용하여 총 20 μL 반응물이 되도록 섞은 뒤 Rotor-Gene 6000(Corbett Research, 호주)를 이용하여 수행되었다. 정량적 실시간 PCR 마스터믹스는 10 μL의 2×효소 마스터믹스, 7.0 μL의 RNase-free 증류수, 각 프라이머 1 μL(10 pM) 및 1 μL cDNA 주형을 혼합하였고 PCR 조건은 95℃ 전배양(pre-incubation), 10분을 유지한 후 95℃ 변성(denaturation) 15초, 52℃ 결합(annealing) 15초, 그리고 72℃ 중합(polymerization) 10초를 1주기로 하여 총 45주기 반복하였다. 예상된 정량적 실시간 PCR 산물의 구조를 확인하기 위하여 용융곡선 분석(melting curve analysis)을 사용하였으며, 정량적 실시간 PCR 산물의 크기는 1.2% 아가로스겔 상에서 전기영동을 통해 확인하였다. 각 유전자의 정량적 실시간-PCR 결과에 대한 표준 곡선은 내부-작동 칼리브레이터(inter-run calibrator)를 사용하였고, 각 샘플의 유전자발현 수준은 Rotor-Gene 6000 Series Software 1.7을 사용하여 측량하였으며, β-actin 발현과 상대적으로 비교하여 다른 유전자 증폭의 변동성을 검량하였다.
그 결과, LPS에 의한 Raw264.7 세포의 염증유도 사이토카인(IL-1β, IL-6 및 TNF-α) 유전자 발현이 톳 분획물에 의해 감소하는 것을 확인하였고, 세포독성은 나타내지 않았다(도 6 내지 10). 또한 염증반응 시 NO 생성에 관여하는 효소인 iNOS 및 COX-2 염증유전자 발현 억제활성이 나타나 부탄산을 포함하는 톳 추출물이 LPS를 통해 염증반응이 유도된 Raw 264.7 세포에서 염증유도 사이토카인 생성을 억제하는 항염증활성을 나타내어 아토피 피부염 완화효능이 있음을 증명하였다. 본 발명에서 사용된 모든 프라이머 정보를 하기 표 1에 표시하였다.

실시예 5: 정량적 실시간 PCR을 통한 Th1 / Th2 사이토카인 mRNA의 발현
본 발명의 톳 분획물의 처리에 따른 Th1/Th2 사이토카인(cytokine)의 mRNA 발현 비교분석을 위해 정량적 실시간-PCR을 수행하였다.
구체적으로, BALB/c 마우스에서 적출한 비장을 pH 7.2의 차가운 인산완충식염수(phosphate buffered saline)하에서 기계적인 방법을 통해 분쇄하여 단일 비장세포를 확보하였다. 또한, 염화암모늄(ammonium chloride)이 함유된 적혈구 분해 완충액(eBioscience, 미국)을 사용하여 상기 비장조직으로부터 적혈구를 제거하여 비장세포의 순도를 최대화한 후 10% 우태아혈청(fetal bovine serum)이 포함된 RPMI1640 완전배지(Hyclone, 미국)로 배양하였으며, 12-웰 플레이트에 1×107 cells/ml로 분주한 뒤 배양기 내 5% CO2 농도, 37 oC 배양 조건을 유지하였다. 2시간 안정화 시간이 경과한 후 24시간 동안 각 분획물(에탄올, 에틸초산, 에틸초산-물, 부탄올 및 부탄올-물)을 전처리하고 T 세포 의존성 자극을 위해 CD3 단일클론항체(anti-CD3)를 추가하고 반응을 유도하였다. anti-CD3 항체(in vitro)로 자극된 비장세포는 트리졸 방법(Invitrogen, 미국)을 이용하여 총 RNA를 추출하였다. cDNA 합성은 1 μg의 총 RNA와 Improm-II 역전사 시스템(Promega, 미국)과 올리고 dT 프라이머를 사용하여 총량이 20 μL 되도록 조성한 뒤 역전사 반응을 수행하였고 상기 표 1에 제시된 프라이머(Bioneer, 한국)를 이용하여 PCR을 수행하였다. 정량적 실시간 PCR(quantitative real-time PCR)은 SYBR Green PCR 마스터믹스(Qiagen, 미국)을 사용하여 총 20 μL 반응물이 되도록 섞은 뒤 Rotor-Gene 6000(Corbett Research, 호주)를 이용하여 수행하였다. 정량적 실시간 PCR 마스터믹스는 10 μL의 2×효소 마스터믹스, 7.0 μL의 RNase-free 증류수, 각 프라이머 1 μL(10 pM), 1 μL cDNA 주형을 혼합하였고 PCR 조건은 95℃ 전배양(pre-incubation), 10분을 유지한 후 95℃ 변성(denaturation) 15초, 52℃ 결합(annealing) 15초, 및 72℃ 중합(polymerization) 10초를 1주기로 하여 총 45주기 반복하였다. 예상된 정량적 실시간 PCR 산물의 구조를 확인하기 위하여 용융곡선 분석(melting curve analysis)을 사용하였으며, 정량적 실시간 PCR 산물의 크기는 1.2% 아가로스겔 상에서 전기영동을 통해 확인하였다. 각 유전자의 정량적 실시간-PCR 결과에 대한 표준 곡선은 내부-작동 칼리브레이터(inter-run calibrator)를 사용하였고, 각 샘플의 유전자발현 수준은 Rotor-Gene 6000 Series Software 1.7을 사용하여 측량하였으며, β-actin 발현과 상대적으로 비교하여 다른 유전자 증폭의 변동성을 검량하였다. 시료들의 발현정도는 대조 유전자(control gene)와 비교한 백분율로 표시하였고anti-CD3 항체에 의해 활성화된 비장세포에서 각 분획물에 의해 지속적으로 T 세포 수용체 매개 Th1/Th2 사이토카인 발현이 유전자 수준에서 억제 여부를 확인하였다. IL-2, IFN-γ, 그리고 IL-4 mRNA 발현 정도의 확인은 각 분획물 50 μg/mL 농도에서 수행되었고 사이클로스포린 A(cyclosporine A, CsA)는 양성 대조군으로 사용되었으며 실험은 3회 반복시행 후 평균 ± 표준 편차로 나타내었다.
그 결과, T 세포는 anti-CD3 항체를 처리할 때 높게 활성화되는데, 분획물 중 부탄산이 관찰되지 않은 부탄올-물 분획물을 제외한 모든 분획물에서 50 μg/mL 처리 시 IL-2, IFN-γ 및 IL-4 mRNA 발현양이 현저하게 감소되었고 군별 유전자 억제수준은 부탄산 면적값과 상응하게 나타났다. 상기 결과는 톳이 함유한 부탄산의 명백하고 전반적인 항아토피 효과를 보여주고 있다(도 11 내지 13).
실시예 6: DNCB 처치 아토피 피부염 유발 생쥐의 조직학적 분석
본 발명의 톳 분획물을 아토피 피부염 유발 생쥐에 처리하고 조직학적 분석을 수행하였다.
구체적으로, 7주령의 수컷 BALB/c 생쥐를 구입하여(중앙실험동물, 한국) 1주 동안 실험실 환경(실내온도: 20 ± 2 ℃, 상대습도: 50%, 명암주기: 12 시간)에 적응시켰다. 상기 BALB/c 생쥐의 아토피 피부염 유사 피부질환을 유발시키기 위해 아세톤에 녹인 1% DNCB(dinitrochlorobenzene, Sigma-Aldrich, 미국) 20 μL를 양쪽 귀에 9일 동안 도포하여 1차적인 과민성 증상을 유도하였으며, 이 후 3주간 2일 간격으로 0.5% DNCB 20 μL를 추가적으로 도포하여 2차적인 증상으로 유발시켰다. 또한, 2차적인 증상을 유도하는 동시에 각각 음성 및 양성대조군으로 0.5 mg/kg 메디론(Methylprednisolone), 생리식염수에 현탁시킨 부탄올 분획물(10, 25 mg/kg)을 실험군으로 설정하여 매일 정해진 시간에 경구 투여하였다(도 14). 마지막 날에는 시료를 처리하지 않았으며, 피부를 적출하여 추가적인 분석을 수행하였다. 상기 아토피 유발 동물 실험을 통해 적출된 BALB/c 생쥐의 귀는 10% 중성 완충포르말린(neutral buffered formalin)으로 고정 후 파라핀 포매를 한 뒤, 5 μm 절편으로 표본을 제작하였고, 헤마톡실린-에오신(hematoxylin-eosin, H&E)으로 이중 염색하였으며 피부 두께, 세포 침투 정도 등을 측정하였다. 본 발명의 실시예 및 이미지 촬영은 암실에서 수행하였으며, Eclips Ti-S 도립현미경(inverted microscope, Nikon, 일본)을 사용하여 200배로 촬영하였다.
그 결과, DNCB 처리군은 정상 대조군과 비교했을 때 피부두께가 두꺼워진 상태인데 반해, 양성대조군(메디론)과 부탄올 분획물에서는 피부두께가 줄어든 것을 확인하였다(도 15). 특히 고농도의 부탄산을 다량포함하고 있는 부탄올 분획물 처리군에서는 양성대조군인 PC 치료군보다 피부조직의 상태가 더 개선된 것으로 나타나, 아토피 피부염과 유사한 피부 질환에 대해 보다 효과적인 회복작용을 할 수 있을 것으로 기대된다. 또한 아토피성 피부염의 가려움증과 연관된 IgE의 혈중 농도 측정결과 부탄올 분획물 고농도(25 mg/kg)에서 양성대조군인 메디론에 필적하는 수준의 효능을 나타냈으며(도 16) 경구 투여기간 동안 귀를 긁는 행동에서도 부탄올 분획물에서 양성대조군과 동등이상의 우수한 효능이 관찰되었다(도 17).
실시예 7: 톳 추출물의 활성 측정
본 발명의 최종 샘플의 활성을 측정하기 위해 n-헥산:에틸초산=8:2의 조건으로 오픈컬럼을 수행하여 TLC(thin-layer chromatography) 스폿을 기준으로 분획 1, 2, 및 3(F1~F3)을 나누었고 상기 분획물을 이용하여 아토피 유발인자에 대한 활성을 관찰하였다.
그 결과, F2 분획물에서 인터류킨-2(IL-2), 인터페론감마(IFNγ) 및 인터류킨-4(IL-4)) 유전자 발현이 현저히 감소되어 F2에 존재하는 성분이 톳의 주요 활성물질을 포함하는 것으로 확인하였으며 면역세포(T 세포)의 조기 활성인자인 CD25 유전자도 유의하게 감소하여 항아토피 효능이 입증되었다(도 19 내지 22). 또한 F2의 TLC(thin-layer chromatography) 결과 식별된 스폿은 8개였으며 Rf값은 각각 0.12, 0.33, 0.43, 0.49, 0.60, 0.65, 0.79, 0.88로 확인되었고 F1은 0.98, F2는 0.12로 나타났다(도 23).
본 발명은 상술한 실시예를 참고로 설명되었으나 이는 예시적인 것에 불과하며, 당해 기술분야에서 통상의 지식을 가진 자라면 이로부터 다양한 변형 및 균등한 다른 실시예가 가능하다는 점을 이해할 것이다. 따라서 본 발명의 진정한 기술적 보호 범위는 첨부된 특허청구범위의 기술적 사상에 의하여 정해져야 할 것이다.

Claims (13)

  1. 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8;2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 질환 치료용 조성물.
  2. 제1항에 있어서,
    상기 톳(S. fusiforme) 추출물의 부탄올 분획물은
    Ⅰ) 톳을 탄소수 1 내지 4의 저급알코올 또는 그의 수용액으로 추출하는 톳 저급알코올 추출물 제조단계;
    Ⅱ) 상기 톳 저급알코올 추출물을 물에 용해시킨 후 에틸초산을 가하여 분획하는 1차 분획단계;
    Ⅲ) 상기 1차 분획단계에서 분리된 물층에 부탄올을 가하여 분획하는 2차 분획단계; 및
    Ⅳ) 상기 2차 분획단계에서 분리된 부탄올층으로부터 부탄올을 제거하는 부탄올 분획물의 제조단계를 포함하는 방법으로 제조되는, 아토피 질환 치료용 조성물.
  3. 제2항에 있어서,
    상기 톳 저급알코올 추출물 수용액 및 에틸초산의 부피비는 1 대 0.5 내지 2 인, 아토피 질환 치료용 조성물.
  4. 제2항에 있어서,
    상기 물층 및 부탄올의 부피비는 1 대 0.5 내지 2인, 아토피 질환 치료용 조성물.
  5. 제1항에 있어서,
    상기 에틸초산 분획물은
    Ⅰ) 톳을 탄소수 1 내지 4의 저급알코올 또는 그의 수용액으로 추출하는 톳 저급알코올 추출물 제조단계;
    Ⅱ) 상기 톳 저급알코올 추출물을 물에 용해시킨 후 n-헥산을 가하여 분획하는 1차 분획단계;
    Ⅲ) 상기 1차 분획단계에서 분리된 물층에 에틸초산을 가하여 분획하는 2차 분획단계; 및
    Ⅳ) 상기 2차 분획단계에서 분리된 에틸초산층으로부터 에틸초산을 제거하는 에틸초산 분획물의 제조단계를 포함하는 방법으로 제조되는, 아토피 질환 치료용 조성물.
  6. 제5항에 있어서,
    상기 톳 저급알코올 추출물 수용액 및 n-헥산의 부피비는 1 대 0.5 내지 2 인, 아토피 질환 치료용 조성물.
  7. 제5항 있어서,
    상기 물층 및 에틸초산의 부피비는 1 대 0.5 내지 2인, 아토피 질환 치료용 조성물.
  8. 치료적으로 유효한 양의 제1항 내지 제7항 중 어느 한 항의 조성물을 아토피 피부염에 걸린 개체에 투여하는 단계를 포함하는 아토피 피부염 치료방법.
  9. 제8항에 있어서,
    상기 투여는 경구, 정맥 내, 근육 내 또는 경피 투여인, 아토피 피부염 치료방법.
  10. 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8;2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 개선용 건강기능식품.
  11. 톳(S. fusiforme) 추출물의 부탄올 분획물, 상기 톳 추출물을 n-헥산과 물로 분획하여 층분리된 물층을 에틸초산으로 추가 분획한 에틸초산층으로부터 수득된 에틸초산 분획물 또는 상기 에틸초산 분획을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8:2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 크로마토그래피 분획을 유효성분으로 함유하는, 아토피 피부 개선용 화장료 조성물.
  12. Ⅰ) 톳을 탄소수 1 내지 4의 저급알코올 또는 그의 수용액으로 추출하는 톳 저급알코올 추출물 제조단계;
    Ⅱ) 상기 톳 저급알코올 추출물을 물에 용해시킨 후 n-헥산을 가하여 분획하는 1차 분획단계;
    Ⅲ) 상기 1차 분획단계에서 분리된 물층에 에틸초산을 가하여 분획하는 2차 분획단계; 및
    Ⅳ) 상기 2차 분획단계에서 분리된 에틸초산층으로부터 에틸초산을 제거하는 에틸초산 분획물의 제조단계를 포함하는 아토피 치료효과가 향상된 톳 추출물의 초산에틸 분획물의 제조방법.
  13. 제12항에 있어서,
    상기 에틸초산 분획물을 크로마토그래피로 전개해서 생성된 분획 중 n-헥산과 에틸 초산(8:2)을 전개용매로 하여 수행된 TLC 상에 Rf값 0.3 내지 0.9에 상응하는 분획을 수집한 후 용매를 제거하여 크로마토그래피 활성분획을 제조하는 단계를 추가로 포함하는, 아토피 치료효과가 향상된 톳 추출물의 초산에틸 분획물의 제조방법.
