本例示的実施形態に係る高分子多孔質体の粉砕方法は、高分子多孔質体を粉砕する粉砕工程と、上記粉砕工程において粉砕中の上記高分子多孔質体を除電する除電工程と、を含む。以下、粉砕工程により粉砕された後の高分子多孔質体を「高分子ブロック」という。換言すれば、本例示的実施形態に係る方法は、高分子多孔質体を粉砕する粉砕工程と、前記粉砕工程において粉砕中の高分子多孔質体を除電する除電工程と、を実行することで、高分子ブロックを製造する方法である。以下、本例示的実施形態に係る粉砕方法と、高分子ブロック製造方法と、を総称して「本開示の方法」という。なお、本開示の方法は、粉砕工程と除電工程を行ったのち、再度粉砕工程と除電工程を繰り返すものであってもよい。粉砕工程と除電工程を行う回数としては、1~4回が好ましく、2~3回がより好ましい。
上述したように、本開示の方法は、高分子多孔質体を粉砕する粉砕工程を含む。本明細書において、「粉砕」とは、機械的エネルギーを与えることで固体及び固体の集合体を細分化することである。本開示の方法における粉砕手段の一例としては、乾式粉砕と湿式粉砕が挙げられるが、液体を分離除去するための乾燥等にかかる手間及びコストを省く観点から、乾式粉砕を用いることが好ましい。粉砕は、粉砕装置に備えられた粉砕部(詳細は後述)により行うことができる。
上述したように、本開示の方法は、上記粉砕工程において粉砕中の高分子多孔質体を除電する除電工程を含む。本明細書において、「除電」とは、帯電した物質から静電気を除去することを意味する。本開示の方法における除電方法の一例としては、放射線照射方法、コロナ放電方法、帯電防止剤添加方法及び加湿方法等が挙げられる。また、これらの除電方法のうち二つ以上を適宜組み合わせてもよい。なかでも、帯電防止剤等の添加、及び被粉砕物の摩擦係数に影響を与える可能性のある高湿度環境の形成等が不要という観点から、放射線照射方法及びコロナ放電方法が好ましい。特に、放射線照射方法によれば、除電能力を高めることができる。放射線の照射は、粉砕装置に備えられた照射部(詳細は後述)により行うことができる。
本開示の方法は、さらに高分子ブロック(粉砕後の高分子多孔質体)を分級する分級工程を含んでいてもよい。「分級」とは、性状の異なる高分子ブロックをそれらの性状に応じて分別する操作のことである。高分子ブロックの性状としては、大きさ、形状、密度、磁性、色及び化学成分等が挙げられる。高分子ブロックの大きさによって分級する方法としては、流体を介在させて行う流体分級、及びふるいによってふるいの網より大きな高分子ブロックとそれ以下の大きさの高分子ブロックに分けるふるい分け等が挙げられる。流体分級としては、サイクロンを適用してもよい。ふるい分けには、ふるい、メッシュ及びフィルタ等を適用してもよい。分級工程は、粉砕工程及び/又は除電工程と同時に行われてもよく、粉砕工程及び/又は除電工程の後に行われてもよい。ふるい分けによる分級工程は、粉砕装置に備えられた分級のためのふるい(詳細は後述)により行うことができる。
次に、本開示の別の例示的実施形態としての粉砕装置について説明する。図1を参照して、本例示的実施形態に係る粉砕装置10の構成の一例を説明する。図1は、粉砕装置10の構成を示す概略図である。粉砕装置10は、上記の例示的実施形態に係る方法における粉砕工程、除電工程及び分級工程を実行する機能を有する。

次に、本例示的実施形態において用いられる高分子多孔質体について説明する。「高分子」とは、分子量が大きい分子で、分子量が小さい分子から実質的又は概念的に得られる単位の多数回の繰り返しで構成した構造を有する分子を言う。高分子の例としては、ポリアミン、ポリサッカライド、ポリペプチド、タンパク質、ポリアミド、ポリエステル、ポリオレフィン、ポリエーテル及びポリヌクレオチド等が挙げられる。「多孔質体」とは、内部に孔(空隙)をもつ固体を意味する。高分子多孔質体は、例えば、高分子水溶液凍結体から水分を除去することにより得られる。

以下、本例示的実施形態における高分子多孔質体が含み得る、生体親和性及び生分解性を有し、骨等の組織再生能を有するリコンビナントゼラチンについて説明する。本開示において、リコンビナントゼラチンとは、遺伝子組み換え技術により作られたゼラチン類似のアミノ酸配列を有するポリペプチドもしくは蛋白様物質を意味する。下記のリコンビナントゼラチンは、生体内に埋植されることにより当該部位における組織の形成に寄与する組織修復材料として用いることができる。なお、「組織修復材料」とは、埋植部位に通常存在する正常組織の形成に寄与するものに限定されず、瘢痕組織等を含む非正常組織の形成を促進する材料も包含するものである。また、組織修復材料により修復可能な「組織」は、例えば歯及び骨等の硬組織であってもよく、下記のリコンビナントゼラチンは特に骨再生用基材として好適である。

高分子ブロックの大きさ(すなわち、粉砕された後の高分子多孔質体の大きさ)は、0.01mm以上、10mm以下であることが好ましい。高分子ブロックを上記の範囲内にすることで、組織修復材に適した大きさとすることができる。また、0.1mm以上、5mm以下であることがより好ましく、0.3mm以上、1.4mm以下であることが更に好ましい。なお、高分子ブロックの大きさとは、一つ一つの高分子ブロックの大きさを意味するものであり、複数の高分子ブロックの大きさの代表値(例えば平均値及び中央値等)を意味するものではない。

本実施例においては、高分子多孔質体が含むリコンビナントゼラチンとして、リコンビナントペプチドCBE3を用いた。CBE3としては、以下に記載のものを用いた(WO2008/103041A1に記載)。

アミノ酸数:571個
RGD配列:12個
イミノ酸含量:33%
ほぼ100%のアミノ酸がGXYの繰り返し構造である。
CBE3はERGD配列を有している。
CBE3のアミノ酸配列には、セリン残基、スレオニン残基、アスパラギン残基、チロシン残基及びシステイン残基は含まれていない。
等電点:9.34
高分子中の親水性繰返し単位比率:26.1%

アルミニウム合金(JIS A5056合金)製の円筒カップ状容器を用意した。容器は、曲面を側面としたとき、側面及び底面が閉鎖され、上面は開放された形をしている。底面は厚さ5mmであり、底面内周がR2mmで面取りされている。側面及び底面の内部が、FEP(テトラフルオロエチレン・ヘキサフルオロプロピレン共重合体)でコートされ、結果として容器の内径は104mmとなっている。以下、この容器を「円筒形容器」という。
上記リコンビナントゼラチンを含む溶液を精製後、30℃にて4.0質量%まで限外ろ過により濃縮した。得られたゼラチン水溶液を凍結乾燥した後、凍結乾燥体に注射用水を加えて30分かけて37℃まで昇温して再溶解し、7.5質量%のゼラチン水溶液を改めて得た。このゼラチン水溶液を、円筒形容器に約20g流し込んだ後、約-35℃に予冷した350×634×20mmアルミ板上に1mm厚のガラス板を介して14個設置し、蓋をして1時間静置することによって凍結したゼラチン凍結体を得た。このゼラチン凍結体を、凍結乾燥機((株)アルバック製DFR-5N-B)を用いて凍結乾燥して水分を除去することにより、凍結乾燥体を作製した。この凍結乾燥体が、本開示の高分子多孔質体の一例である。また、この凍結乾燥体について、空隙率は93.9%、孔径は50μm、密度は0.0748g/cm3であった。
上記のようにして手で割った高分子多孔質体約40gを、図1に示す構成の粉砕装置10により粉砕した。粉砕部20としてはスクリーンミル(クアドロ社製コーミルU10)の粉砕部を用いた。照射部30としては軟エックス線照射装置(浜松ホトニクス(株)製L14471)を用いた。換気部40としては集塵機(チコーエアテック(株)製CKU-450AT-HC)を用いた。粉砕は、それぞれ孔径が異なるスクリーン22を用いて、2回行った。
ガラスバイアルに充填された高分子ブロックを、ガラスバイアルの底からルーペで観察し、ガラスバイアルを揺すりながら焦げ(すなわち着色)の有無を目視で検査した。高分子ブロックを充填した30本のガラスバイアルのうち、焦げを認めたガラスバイアルの割合を、表1に示す。
図1の構成において、3台の照射部30A~30Cにより高分子多孔質体に照射される照射線量を評価するため、上記の高分子多孔質体の粉砕とは別途、照射線量の測定を行った。まず、照射部30による照射線量が既知の位置(900mmにおいて70μm線量当量率が702mSv/h)に、24×30mmの歯科用D感度エックス線インスタントフィルム((株)阪神技術研究所製DIC-100)(以下、「フィルム」という)を設置し、軟エックス線を照射した。専用のキット((株)阪神技術研究所製プッシャーシステム)を用いて、フィルムの現像、定着及び硬膜化処理等を実施し、透過濃度計(X-Rite社製310)を用いてフィルムの透過濃度を測定した。同様に、別のフィルムを用いて、照射時間を変えながら軟エックス線の照射及び透過濃度の測定を繰り返し行った。
図1の構成において、3台の照射部30A~30Cが粉砕部20を除電する能力を評価するため、上記の高分子多孔質体の粉砕とは別途、除電時間の測定を行った。粉砕部20におけるスクリーン22の内部に、チャージプレートモニタ(トレック社製MODEL156A)の1inch角チャージプレート(トレック社製156P-C25X25-S3M)を配置し、3台の照射部30A~30Cにより軟エックス線を照射した状態で、プレート電圧が+1000Vから+100Vへ変化するまでの除電時間を測定した。スクリーン22内部の除電時間の測定結果を表1に示す。
図3に、本実施例2に係る粉砕装置10の構成を示す。図3に示すように、本実施例2においては、上記実施例1(図1参照)とは照射部30Bの粉砕部20に対する照射角度が変わっている。具体的には、図3に示すように、粉砕装置10の鉛直方向(一点鎖線)に対して片側にそれぞれ60度及び5度の角度に設けられた2台の照射部30A及び30Bと、排出部14側に設けられた照射部30Cと、の合計3台の照射部によって、粉砕部20に対して軟エックス線を照射した。
照射部30による除電及び焦げ抑制の効果を検証するため、上記実施例1において、照射部30による軟エックス線の照射を行わずに、高分子多孔質体の粉砕を行った。その他の装置構成、リコンビナントゼラチン、高分子多孔質の作製方法、高分子多孔質体の粉砕方法、及び高分子ブロックの焦げの評価方法については、上記実施例1と同様であるので、説明を省略する。本比較例1において、焦げを認めたガラスバイアルの割合を表1に示す。
Claims (17)
- 高分子多孔質体を粉砕する粉砕工程と、
前記粉砕工程において粉砕中の前記高分子多孔質体を除電する除電工程と、
を含む粉砕方法。 - 前記除電工程において、前記高分子多孔質体に放射線を照射することで、前記高分子多孔質体を除電する
請求項1に記載の粉砕方法。 - 前記放射線は、エックス線及び紫外線のうち少なくとも一方である
請求項2に記載の粉砕方法。 - 前記放射線は、線源の管電圧が4kV以上、50kV以下のエックス線である
請求項3に記載の粉砕方法。 - 前記除電工程において、前記高分子多孔質体に照射される前記放射線の70μm線量当量率は、1mSv/h以上、200Sv/h以下である
請求項2から請求項4の何れか1項に記載の粉砕方法。 - 前記除電工程において、前記高分子多孔質体を粉砕する粉砕部の少なくとも一部における電位が+1000Vから+100Vに変化するまでの除電時間は、0.01秒以上、10秒以下である
請求項1から請求項5の何れか1項に記載の粉砕方法。 - 前記粉砕工程において、乾式粉砕を行う粉砕部を用いて、前記高分子多孔質体を粉砕する
請求項1から請求項6の何れか1項に記載の粉砕方法。 - 前記粉砕部は、スクリーン及び回転インペラを備える
請求項7に記載の粉砕方法。 - 前記粉砕部は、1×104回/h以上、1×107回/h以下の換気回数で換気される

(A)配列番号1のアミノ酸配列からなるペプチド。
(B)配列番号1のアミノ酸配列中、1個又は数個のアミノ酸残基が改変されたアミノ酸配列からなり、かつ生体親和性を有するペプチド。
(C)配列番号1のアミノ酸配列中、第4番目~第192番目のアミノ酸残基からなる部分アミノ酸配列と80%以上の配列同一性を有する部分配列を有するアミノ酸配列からなり、かつ生体親和性を有するペプチド。 - 粉砕前の前記高分子多孔質体の大きさは、0.1mm以上、50mm以下である
請求項1から請求項12の何れか1項に記載の粉砕方法。 - 粉砕後の前記高分子多孔質体の大きさは、0.01mm以上、10mm以下である
