 <Desc/Clms Page number 4>

  "Process for preparing tetrahydroisoquinoline compounds".

   In Belgian patent application No. 340,622 filed August 30, 1940 with claim of priority for a filing made in Germany on August 30, 1939, tetrahydroisoquinoline compounds substituted on nitrogen by radicals or residues are described. aralkyl or aralkylene and containing in one of the two benzene rings at least two acylated hydroxyl groups, in particular acetylated, preferably located in position ortho to one another, said compounds exerting analgesic effects.

   However, it has been discovered that analogous compounds which are not substituted on the nitrogen atom, but on one of the carbon atoms of the tetrahydrogenated pyridic ring and contain in one of the benzene rings at least 2 acylated hydroxyl groups , in particular aoetylated, which are preferably in the ortho position to each other also have an analgesic effect. The compounds can also carry other

 <Desc / Clms Page number 2>

 very substituents, for example alkyl or alkylene groups at the various locations of the molecule and be substituted in the second benzene ring by a free hydroxyl group and / or by its functional derivatives.

   According to the invention, the aforementioned compounds are prepared by hydrogenating in a manner known per se non-hydrogenated or dihydrogenated isoquinolein compounds substituted on one of the carbon atoms of the pyridic ring by an aralkyl or aralkylene group, or quaternary ammonium salts of these isoquinoline compounds or else tetrahydrogenated isoquinoline compounds, substituted by an aralkylene group on one of the carbon atoms of the hydrogenated pyridic ring, which contain in one of the benzene rings at least two acylated, in particular acetylated, hydroxyl groups, preferably located in the ortho position to each other.

   The isoquinoline compounds employed as raw materials and their hydrogenated derivatives may be substituted, either by an aralkyl or aralkylene group and at least two acyloxy groups, by other substituents, for example by alkyl or alkylene groups at the the most diverse places of the molecule and / or, respectively, additionally in the second benzene ring by a free hydroxyl group and / or, respectively, its functional derivatives, such as alooxy groups or

 alkylenediozy. x, x> For the preparation of tetrsoquinoline substituted compounds with an aralaoyl, different raw materials can be used.

   Non-hydrogenated isoquinoline compounds or dihydroisoquinoline compounds substituted on one of the carbon atoms of the pyridic ring with an araleoyl and containing at least two acyloxy groups in a benzene ring can be hydrogenated in a manner known per se. , or where appropriate more strongly substituted or the quaternary ammonium salts of these compounds, so that the double bonds of the pyridic ring are transformed into single bonds.

   This hydro-

 <Desc / Clms Page number 3>

 generation can be carried out either with metals, eg zinc dust, optionally in the presence of the aid by which the hydroxyl groups are acylated, or catalytically, eg with platinum catalysts or palladium, such as platinum black, platinum oxide, palladium, palladium black, animal palladium-black.

  However, it is also possible to start from the corresponding non-hydrogenated or dihydrogenated isoquinoline compounds substituted with an aralkylene or their quaternary ammonium salts and catalytically hydrogenated, for example in the presence of platinum or palladium catalysts, so that the double bonds of the pyridic ring as well as the double bonds of the aralooylene radical are transformed into single bonds. Finally, it is possible to start from the corresponding tetrahydroisoquinoline compounds substituted with an aralkylene and also hydrogenate them oatalytically, the aralkylene substituents thus being converted into aralkyl substituents.

   On the other hand, if it is desired to prepare tetrahydroisoquinoline compounds of the type described above which are substituted by an aralooylene, one starts with non-hydrogenated or dihydrogenated isoquinoline compounds substituted on one of the carbon atoms of the. pyridic ring by an aralooylene and containing in a benzene ring at least 2 acyloxy groups, and optionally even more strongly substituted, or of their quaternary ammonium salts and they are hydrogenated with metals, if appropriate in the presence of the acid by which the hydroxyl groups are acylated, so that only the double bonds of the pyridic ring are transformed into single bonds, the double bonds of the aralooylene substituent however remaining maintained.

   The raw materials for the process according to the invention can be obtained, for example, by condensation of 1-methylisoquinolines or 1-methyl-3,4-dihydroisoquinolines, suitably substituted if appropriate, with aldehydes.

 <Desc / Clms Page number 4>

 suitably substituted, where appropriate, or alternatively by Bisehler-Napieralski synthesis from suitably substituted / 3 -phenylethylamines.

   The hydroxyl groups of the raw materials can be esterified with formic, acetic, propionic, butyric or other organic acids; preferably, acetylated compounds are employed.
