157, Avenue du Roi, à Forest, Bruxelles, Procédé pour la fabrication d'une préparation de fer injectable. (Priorité d'une demande de brevet, déposée en Suisse, en date du
7 juillet 1944) 
On ne connaît jusqu'ici que très peu de préparations de fer qui soient'injectables par voie parentérale. Les sels de fer administrés par voie buccale ne sont résorbés que lentement ou produisent dans beaucoup de cas des actions secondaires désagréables.

   Les préparations de fer utilisées jusqu'ici pour injections présentent de gros inconvénients, dus, avant tout, au fait qu'elles exercent ou bien une influence néfaste sur

 l'organisme, lorsqu'elles ne sont pas utilisées à des très fortes dilutions, ou bien elles n'ont que très peu d'effet thérapeutique en raison de la liaison trop complexe du fer. 



   On a maintenant trouvé dans le complexe de l'acide

 di-(c,y-dioxy-3,-dimétYyl-butyrato)-ferrique, une prépara- tion de fer qui, injectée sous forme de solution neutralisée, possède une action correspondante à sa teneur en fer, cela même en concentration élevée et sans produire d'actions

   L'acide di-(oc,Y-dioxy-p,-diméthyl-butyrato)- ferrique est obtenu en préparant des solutions de sel ferrique de l'acide Ó,y-dioxy-ss, ss-diméthyl-butyrique selon les méthodes connues (par exemple en faisant réagir du sulfate de fer avec des sels alcalino-terreux de l'acide Ó,y-dioxy-ss,ss-diméthylbutyrique en solution aqueuse et en séparant cette solution du sulfate alcalino-terreux précipité), à évaporer jusqu'à siccité, à redissoudre le résidu de solvants organiques et   à   préparer des solutions aqueuses neutres. 



   Ce nouvel acide se présente sous forme d'une poudre jaune-brune, non hygroscopique qui se dissout facilement dans l'eau avec une réaction acide. Il se dissout aussi facilement dans les alcools méthylique et éthylique, il est insoluble par contre dans l'acétone et l'éther. Comparativement à la constitution admise pour des dérivés ferriques d'autres oxyacides (Gmelin, Handbuch der anorganischen Chemie, 8me édition, Fer, partie B, page 531), on peut lui attribuer la formule suivante:

Cet acide donne avec des alcalis des sels bruns-jaunes, facilement solubles dans l'eau en réaction neutre. Ils sont difficilement solubles dans les solvants organiques habituels. 



  Ces sels alcalins, contrairement   à   la plupart des sels de fer organiques, sont extraordinairement stables. On peut stériliser les solutions neutres par chauffage. Elles conviennent parfaitement pour les traitements ferrugineux par voie parentérale.

  L'acide di-(oc,r-dioxy-,-diméthyl-butyrato)-ferriue peut être utilisé comme médicament sous forme de solution neutralisée. 



   Exemple. 



   On dissout 135 parties de sel de barium de l'acide

  *Y-dioxy-pep-diméthyl-butyrique (CHOBa.H0) dans 1350 parties d'eau. On fait tomber, goutte à goutte, à la température ambiante en agitant une solution d'environ 45 parties de sulfate de fer   (Fe2(S04)3.3   h2O) dans 300 parties d'eau jusqu'à ce qu'on ne puisse plus mettre en évidence dans la solution soit des ions de barium ou de sulfate. 



  On sépare ensuite du sulfate de barium par filtration sur amiante et on évapore la solution jaune-brune dans le vide jusqu'à siccité. Le gâteau cristallin, légèrement coloré en brun, qui reste est dissous dans une quantité double d'alcool méthylique en réchauffant légèrement et en précipitant ensuite

   Par neutralisation avec la soude ou le bicarbonate de soude en solution aqueuse, ce qui nécessite une quantité équivalente, on obtient des solutions brunes, stables à la chaleur, du sel sodique. On peut alors obtenir le sel sodique jaunebrun sous forme solide par évaporation dans le vide. Pour l'emploi thérapeutique, on peut utiliser directement la solution neutralisée sans isoler le sel.

   This new acid comes in the form of a yellow-brown, non-hygroscopic powder that easily dissolves in water with an acid reaction. It also dissolves easily in methyl and ethyl alcohols, it is insoluble on the other hand in acetone and ether. Compared to the constitution admitted for ferric derivatives of other oxyacids (Gmelin, Handbuch der anorganischen Chemie, 8th edition, Fer, part B, page 531), we can attribute the following formula:

This acid gives with alkalis yellow-brown salts, easily soluble in water in neutral reaction. They are hardly soluble in the usual organic solvents.



  These alkali salts, unlike most organic iron salts, are extraordinarily stable. Neutral solutions can be sterilized by heating. They are ideally suited for parenteral ferruginous treatments.

   Dissolve 135 parts of barium salt from the acid

   By neutralization with sodium hydroxide or sodium bicarbonate in aqueous solution, which requires an equivalent amount, brown, heat-stable solutions of the sodium salt are obtained. The yellow-brown sodium salt can then be obtained in solid form by evaporation in a vacuum. For therapeutic use, the neutralized solution can be used directly without isolating the salt.

Procédé pour la fabrication d'une préparation de fer injectable, consistant à évaporer jusqu'à siccité des solutions de sel ferrique de l'acide Ó,r-dioxy-ss,ss-diméthyl-butyrique, préparées par les méthodes habituelles, à redissoudre le résidu de solvants organiques et à préparer des solutions aqueuses neutres.
