Pour accéder aux ¯ 1,4-3-cétostéroïdes de la série de l'androstadiène ou du pregnadiëne,on peut soit utiliser des métho- des microbiologiques en mettant à profit les propriétés déshydro- génantes des ferments secrétés par certains microorganismes, com- me l'ont montré parallèlement VISCHER, KEYSTRE et WETTSTEIN (Helv.
Chim.Acta, 1955, 38, 835) et HERSHBERG et Collaborateurs (J. Am.
Chem. Soc.,1955,77, 4184), soit encore partir du dérivé 2,4-diha- logéné correspondant au cétostéroide diénique qu'on veut préparer, lequel par double déhydrohalogénation fournit le produit voulu, selon le schéma :

D'après ce qui précède, cette double déhydrohalogéna- tion constitue la dernière ou l'avant-dernière étape de la synthè- se du composé diénique cherché et intervient donc à un moment où le rendement, et surtout la pureté du produit, exercent une in- fluence primordiale sur le prix de revient et la production.
Lorsqu'on essaie de déhydrohalogéner ces dérivés 2,4- dihalogénés par les méthodes usuelles (traitement à la collidine, passage par les dinitrophénylhydrazones ou semicarbazones, ou traitement aux halogénures de lithium dans le diméthylformamide selon la méthode de HOLYSZ (J. Am. Chem.Soc., 1953, 75, 4432)), on se heurte à différentes difficultés dont la principale est constituée par la migration concomitante de la double liaison 1,2 .en 6,7. Cette constatation des inventeurs est corroborée par

la publication de FRIED et Collaborateurs (J.Am.Chem.
Soc., 1955', 77, 4182 note 14). En effet, ces auteurs préconisent, comme métho- de de préparation de choix de l'acétate de 6-déhydro 9 Ó -fluoro hydrocortisone, l'action du chlorure de lithium au sein du dimé- thylformamide sur le dérivé 2,4-dibromé correspondant. A cette difficulté s'ajoute, dans le cas du passage par la dinitrophényl- hydrazone ou la semicarbazone, celle du clivage de l'hydrazone diénique. La présence du diène 4,6 indésirable se révèle immédia- tement à l'examen du spectre U. V. du produit obtenu qui montre la présence de deux maxima d'absorption avec très souvent prépon- dérance du diène 4,6.
Le produit réactionnel étant constitué par un mélange de diènes 1,4 et 4,6 ainsi que de produit de départ non transfor- mé ou altéré, on est obligé de recourir à une purification labo- rieuse par cristallisation fractionnée très poussée ou par chroma- tographie, et les rendements en diène 1,4-cherché sont souvent assez bas. C'est ainsi que HERZOG et Collaborateurs (J. Am. Chem.
Soc., 1955, 77, 4782) obtiennent, au départ de 20 g de 17 Ó -hy- droxy 21-acétoxy pregnane 3,11 ,20-trione, après dibromuration et débromhydratation seulement 2,4 g d'acétate de ¯ -déhydro- .cortisone.
Dans son brevet français n 1.085.842 du 20 mai 1953, la Société demanderesse a décrit un procédé de préparation de cétostéroïdes Ó-bromes convenant particulièrement bien à l'ob- tention de 3-cétostéroides 2,4-dibromés, aussi bien en série allô qu'en série normale et permettant notamment un accès facile à la 2,4-dibromo androstane 3,17-dione, aux esters en 17 de la 2,4-di- bromo androstanol 3-one et à la 2,4-dibromo 17 Ó-hyroxy pregnane 3,11 , 20-trione.
D'autres 3-cétostéroïdes 2,4-dibromés ont fait l'ob- jet des brevets belges intitulés - " Pregnadiènes substitués et leur procédé d'obten-

tion " n 547.014 (où la Société demanderesse a décrit la prépa- prégnane ration de la 2,4-dibromo 17 Ó -hydroxy 21-acétoxy/3, 11, 20-trio- ne lévogyre).
-"Dérivés bromés du pregnane et leur procédé d'obten- tion " n 547.015, qui décrit la préparation de la 17C6 -hydroxy 2,4,21-tribromo pregnane 3,11,20-trione sous ses deux formes sté- réoisomères.
- " Dérivés bromés de la série du pregnane et leur procédé d'obtention " n 548.945, relative à l'une des formes stéréoisomères du composé précité.
- " Procédé d'obtention d'une pregnadiène-dione sub- stituée " n 549.904, où la Société demanderesse a décrit la pré- paration de la 2,4-dibromo 11ss, 17Ó- -dihydroxy 21-acétoxy pre- gnane 3,20-dione.
L'objet de la présente invention est un procédé de double déhydrohalogénation des 3-cétostéroïdes dihalogénés en po- sition 2,4- de formule A appartenant à la série allo ou à la sé- rie normale, lesdits cétostéroïdes pouvant faire partie du groupe de l'androstane, de l'étiocholane, de l'allo-pregnane ou du pre- gnane et tous analogues à chaînes ramifiées en 17.
Ces cétostéroïdes peuvent porter dans les noyaux B, C et D d'autres substituants, notamment des groupes hydroxyles li- bres ou estérifiés, des groupes carbonyles libres ou bloqués et, s'ils appartiennent à la série du pregnane ou de ltallo-pregnane, leur chaîne latérale-comportera un carbonyle ou un ester d'énol en position 20, un groupe méthyle libre ou halogéné ou une fonction acyloxy en 21.
Le nouveau procédé selon l'invention est remarquable en ce qu'il fournit, de façon pratiquement exclusive,le diène 1,4 cherché, l'analyse spectrographique révélant l'absence de l'isomère 4,6 facilement décelable par son maximum d'absorption caractéris-'

tique différent du maximum d'absorption du diène 1,4.
Les inventeurs onten effet trouvé que si l'on traite à chaud un 3-cétostéroide 2,4-dihalogéné par un excès (généralement un poids égal) de carbonate de lithium, en présence d'un amide disubstitué,comme le diméthylformamide, il y a formation concomi- tante d'halogénure de lithium, en même temps que s'opère la dou- ble déhydrohalogénation en 1,4,sans formation simultanée de diène @,6. La réaction, assez lente au début, progresse ensuite plus rapidement, toujours en présence d'un excès de carbonate de li- thium. Ce dernier agent, n'étant pas soluble dans le milieu réac- tionnel,doit être maintenu constamment en suspension par une bon- ne agitation pour neutraliser toute trace d'hydracide libre dans le milieu.
Après précipitation par l'eau acidulée pour dissoudre le carbonate de lithium, on obtient le cétostéroide diénique ' cherché qui est généralement pur après une seule recristallisa- tion. Les rendements sont élevés.
Un mode d'exécution du présent procédé consiste à préformer le couple halogénure de lithium-carbonate de lithium si l'on veut accélérer la réaction.
Le procédé de la société demanderesse se caractérise par l'emploi du carbonate de lithium et par le fait qu'il conduit à un résultat nouveau, la formation des diènes 1,4 sans conforma- tion appréciable des diènes 4,6-correspondants.
Les exemples suivants illustrent l'invention sans tou- tefois la limiter et montrent également, par des essais compara- tifs, les avantages du nouveau procédé. Bien entendu, sans s'éloi- gner pour cela du cadre de l'invention; on peut utilisera la place des dérivés 2,4-dibromés, des analogues dihalogénés ou partir d'un cétostéroide portant en 2 un halogène différent de celui qui substitue le carbone en 4. Comme le montrent les exemples, les proportions du couple halogénure de lithium-carbonate de lithium peuvent varier entre d'assez larges limites.

est uniquement limité par des raisons économiques et d'agitation, la quantité d'halogénure pour assurer une vitesse réactionnelle satisfaisante étant comprise entre 0,1- et 1 molécule par molécule de cétostéroide dihalogéné. Les températures optima peuvent s'é- chelonner entre 70 et le reflux du solvant, la durée de réaction étant généralement comprise entre dix minutes et vingt heures. Les points de fusion sont des points de fusion instantanée déterminés sur bloc de Maquenne. Les spectres U.V. ont été tous déterminés dans l'éthanol.
La préparation des dérivés dibromés non décrits encore dans la littérature est donnée à titre indicatif, le procédé em- ployé étant celui décrit dans les brevets ou demandes de brevets cités plus haut. Les formules des produits de départ et des diènes obtenus figurent sur le schéma joint.
EXEMPLE 1 : Débromhydratation du 17-benzoate de la 2,4-dibromo androstane 17 ss -ol 3-one (Schéma I). a) Par le bromure de lithium seul.
A 50 cm3de diméthylformamide, on ajoute : - Bromure de lithium anhydre : 2 g 6 (0,03 mol.)
On fait passer un courant d'azote dans la solution et on ajoute 5,52 g (0,01 mol. ) de 17-benzoate de 2,4-dibromo andro- stane 17 ss -ol 3-one et chauffe sous agitation et barbotage d'azo- te pendant deux heures à 90-95 . On refroidit et verse dans 200 cm3 d'eau. Le produit est traité comme en b) F = 180 C.
D'après le spectre U.V. # max. 231,5 m avec = 18.000 min 251 mu avec 6 = 9.250 # max. 283,5 m avec ± = 16.900 le produit de débromhydratation contient 60-61 % de 17-benzoate de ¯ -androstadiène 17 ss -ol 3-one et 36-37 % de 17-benzoate de ¯-1,4-androstadfiène 17 ss -ol 3-one. b) Par le couple bromure de lithium-carbonate de lithium.

A 35 cm3de diméthylformamide, on ajoute : - Bromure de lithium anhydre 2,6 g (0,03 mol.) et introduit, sous courant d'azote et agitation, 5,6 g (0,01 mol.) de 2 Ó, 4/3 -dibromo 17 Ó -hydroxy 21-acétoxy pregnane 3, 11, 20- belge trione, VA, décrite dans l'exemple 7 du brevet/intitule' " Pregna- diènes substituée et leur procédé ¯de préparation " n 547.014, [Ó]D20= 19 (c = 0, 5 acétone) . On chauffeà 90-95 sous at- mosphère d'azote pendant trois heures. On refroidit et verse le mélange dans 200 cm3 d'eau, essore, lave à l'eau et sèche. On ob- tinos 2,8 g de 21-acétate de ¯ -déhydrocortisone, VB, brut, souil- lé de résines, soit un rendement de 70 %.
Après purification dans l'acétone, on obtient 2 g (soit un rendement de 50 %) d'un produit dont l'examen du spectre U. V. révèle la présence de 80 % d'acétate de A 1-déhydrocortisone et 10% d'acétate de ±\ -déhydrocortisone.

Dans 35 m3 de diméthylformamide, on introduit, sous- courant d'azote et agitation, 4,5 g de carbonate de lithium anhydre (0,06 mol.), puis on ajoute 5,6 g de 2 Ó, 4/3 -dibromo 17 Ó, 21- acétoxy pregnane 3, 11, 20-trione (0,01 mol.). On chauffe pendant cinquante heures à 95-100 en maintenant le courant d'azote et l'a- gitation. On refroidit et !erse le mélange dans 200 cm3d'eau et 10 cm d'acide acétique; Après essorage, lavage à l'eau et séchage, on obtient 3,6 g (soit un rendement de 90 %) de 21-acétate de ¯ 1-déhydrocortisone brute exempte de résines.
Le spectre U.V. révèle : # max. 239,5 m avec = 11.000 # max. 285 m avec ¼ = 700

le produit renferme donc 70% d'acétate de -déhydrocortisone avec absence complète du dièrie 4,6. Il reste environ 20 % de pro- duit non transformé. c) Par le couple bromure de lithium-carbonate de li- thium.
A 35 cm3 de diméthylformamide, on ajoute : - Bromure de lithium anhydre : 2,6 g (0,03 mol.) - Garbonate de lithium anhydre: 4,5 g (0,06 mol.)
Sous forte agitation et courant d'azote, on introduit 5,6 g (0,01 mol.) de 2Ó-4ss -dibromo 17 ss -hydroxy 21-acétoxy pre- gnane 3, 11, 20-trione et, sans interrompre le courant d'azote et l'agitation, on chauffe quinze heures à 95 . On refroidit, verse le mélange dans 200 cm3d'eau et 10 cm3d'acide acétique. On esso- re, lave à l'eau, sèche. On obtient 3,92 g (soit un rendement de 98 %) d'acétate de ¯ -déhydrocortisone pratiquement pur. F =222- 232 , [Ó]D20= + 183 (c= 1 %, dioxane) avec #max. 239,5m avec ±= 14.300 et # min. 285 m avec 6.= 600.
Le diène 4,6 cor- respondant est donc absent. Le produit est purifié pour analyse par recristallisation dans l'acétone. On dissout 39 g (0,1 mol. ) de 17 Ó-hydroxy 21-acétoxy 20 pregnane 3,20-dione, F= 197 , /"<7 D20 = + 68 ¯ 2 (c= 1 % ,éthanol) dans 600 cm de dioxane anhydre. On ajoute 1 cm3 d'acide acétique à 50 % d'acide bromhydrique sec,puis introduit très rapidement 36 g de brome dans 100 em3 d'acide acétique. La température s'élève

de 10 environ. Après avoir vérifié que tout le brome a été absor- bé, on ajoute 300 cm3 d'eau distillée. La solution ainsi obtenue est versée lentement sous agitation dans 6 litres d'eau. The object of the present invention is a process for the double dehydrohalogenation of dihalogenated 3-ketosteroids in position 2,4- of formula A belonging to the allo series or to the normal series, said ketosteroids possibly being part of the group of androstane, etiocholane, allo-pregnane or pregnane and all 17-branched chain analogues.
These ketosteroids can carry in the rings B, C and D other substituents, in particular free or esterified hydroxyl groups, free or blocked carbonyl groups and, if they belong to the pregnane or ltallo-pregnane series, their side chain will contain a carbonyl or an enol ester in position 20, a free or halogenated methyl group or an acyloxy function in 21.
The new process according to the invention is remarkable in that it provides, practically exclusively, the sought-after 1,4-diene, the spectrographic analysis revealing the absence of the 4,6 isomer easily detectable by its maximum of. absorption characteristic

The inventors have in fact found that if a 2,4-dihalogenated 3-ketosteroid is heat treated with an excess (generally an equal weight) of lithium carbonate, in the presence of a disubstituted amide, such as dimethylformamide, there is a concomitant formation of lithium halide, at the same time as the double dehydrohalogenation takes place at 1,4, without simultaneous formation of diene @, 6. The reaction, quite slow at the start, then progresses more rapidly, always in the presence of an excess of lithium carbonate. The latter agent, not being soluble in the reaction medium, must be kept constantly in suspension by good stirring to neutralize any trace of free hydracid in the medium.
After precipitation with acidulated water to dissolve lithium carbonate, the desired diene ketosteroid is obtained which is generally pure after a single recrystallization. The returns are high.
One embodiment of the present process consists in preforming the lithium halide-lithium carbonate pair if the reaction is to be accelerated.
The process of the applicant company is characterized by the use of lithium carbonate and by the fact that it leads to a new result, the formation of 1,4 dienes without appreciable conformity of the corresponding 4,6-dienes.
The following examples illustrate the invention without, however, limiting it and also show, by comparative tests, the advantages of the new process. Of course, without departing for this from the scope of the invention; it is possible to use the 2,4-dibromo derivatives, dihalogenated analogs or from a ketosteroid bearing at 2 a halogen different from that which substitutes the carbon at 4. Optimum temperatures may range from 70 to reflux of the solvent, the reaction time generally being between ten minutes and twenty hours. The melting points are instantaneous melting points determined on a Maquenne block. The U.V. spectra were all determined in ethanol.
The preparation of dibromo derivatives not yet described in the literature is given by way of indication, the process employed being that described in the patents or patent applications cited above. The formulas of the starting products and of the dienes obtained appear on the attached diagram.
EXAMPLE 1 Dehydration of 17-benzoate of 2,4-dibromo androstane 17 ss -ol 3-one (Scheme I). a) By lithium bromide alone.
