Description

       

  Procédé de     préparation    de cétostéroïdes non saturés    Il a été trouvé que l'action du couple carbonate  de lithium-bromure de lithium, le premier étant géné  ralement employé en excès et le deuxième servant à  accélérer la réaction, permet, dans des conditions de  température et de durée bien définies et délimitées,  de créer, à partir d'un dérivé     polybromé    de la série  stéroïde, un composé à une ou plusieurs doubles  liaisons tout en     maintenant    au moins un atome de  brome présent     dans    la molécule.

   L'opération s'effec  tue avec enlèvement d'une seule molécule d'acide  bromhydrique lorsqu'il s'agit d'un dérivé     dibromé,     ou d'une ou plusieurs     molécules    d'acide bromhydri  que lorsqu'il s'agit d'un composé     polybromé    avec  création, selon le cas, d'une ou plusieurs doubles liai  sons. On évite la formation d'un diène 4,6 et on a  constaté que, lorsque le dérivé stéroïde qu'on veut       déhydrohalogéner    porte un brome en 17, on évite  la migration de la double liaison 16,17 formée vers  la position 14,15.  



  Le procédé selon l'invention se rapporte à la  préparation de     cétostéroïdes    non saturés, contenant  au moins un atome de brome dans la molécule, par       déhydrohalogénation    partielle de dérivés     polybromés     saturés     correspondants    et est     caractérisé    par le fait  qu'on traite le stéroïde     polybromé,    en     présence    d'un  couple     halogénure    de     lithium-carbonate    de lithium  au sein d'un amide     N,N-disubstitué    d'un acide ali  phatique inférieur,

   à une température     comprise    entre       70o    et 1000 C, le temps de réaction étant     compris     entre 20 minutes et une heure et en ce qu'on isole  le dérivé non saturé bromé.  



  Bien entendu, les stéroïdes     polybromés    que l'on  soumet au     procédé    selon l'invention peuvent     porter,     en dehors des atomes de brome, divers     autres    substi  tuants des noyaux A, B, C, D ou de la chaîne laté-         rale,    notamment des groupes carbonyle ou hydroxyle  ou des dérivés de ces fonctions facilement     reconver-          sibles    en cétones ou alcools     comme    des acétals,     ènes,          amines,

      éthers     d'énols    pour le     carbonyle    et des esters  ou éthers-oxydes facilement     clivables    pour     l'hy-          droxyle.    Le procédé     s'applique    aussi bien en série  normale (noyaux A et B     cis),    qu'en série     allo    (noyaux  A et B     trans)    et les stéroïdes peuvent     appartenir    à  la série des     génines    comme     l'hecogénine,    à la série  des     stérols    ou de leurs produits de dégradation  comme les     androstanes,

          allopregnanes    ou encore à  celle des acides     biliaires,    des     pregnanes    ou     des        étio-          cholanes,    tout en comportant les substitutions sus  mentionnées.  



  Le nouveau procédé fournit notamment comme  le montrent les exemples, une voie très pratique pour  passer avec     d'excellents    rendements d'un     3,20-dicéto-          pregnane    ou d'un     3-hydroxy-20-céto-pregnane        poly-          bromé    de     formule

    dans laquelle les deux R représentent de l'hydrogène,  ou l'un des deux représente un groupe     hydroxy    ou       oxo    ou encore.

   un dérivé simple de ces deux derniers  groupements d'où ceux-ci peuvent être régénérés  facilement, R' est un groupe     oxo    ou     hydroxy    ou un      dérivé facilement clivable de     ces    fonctions et chaque  X, X', X" et X"' représentent de l'hydrogène ou du  brome, à la     21-bromo-Als-pregnène-3,

  20-dione    ou  au     21-bromo-3-hydroxy-20-céto-Als-pregnène        corres-          pondants.        L'acétoxylation    en 21 suivie de l'hydrogé  nation de la double liaison permet     l'accès    aisé et       économique    aux     21-acétoxy-pregnanes        correspon-          dants    d'où l'on passe très     facilement,    selon les pro  cédés     connus    (formation du     3-cétostéroïde        4-halogéné     correspondant, et     déhydrohalogénation)

      aux     21-          acétoxy-A4-pregnène-diones        correspondantes.    On ac  cède     ainsi    grâce aux     excellents    rendements réalisés  lors de la première     débromhydratation    en 16,17 d'une  façon très économique à l'acétate de     désoxycorticos-          térone    ou à     l'acétate    de     11-déhydro-corticostérone     d'où l'on passe, par saponification, aux hormones  libres correspondantes.

   On peut également tirer pro  fit de la double liaison 16,17     ainsi    créée pour intro  duire selon     Plattner        (Helv.        Chim.        Acta,    1948, 31,  2210) et     Julian    (J.

   Am.     Chem.    Soc., 1949,<I>71,</I> 3575)  par     l'intermédiaire    de     l'époxyde,    l'hydroxyle en 17a,  voie qui a été peu     utilisée    jusqu'ici par suite des mau  vais rendements de la     débromhydratation    sélective,  effectuée généralement par l'acétate de     potassium    ou  des bases     tertiaires    aromatiques ou     hétérocycliques     et passer ainsi à des     composés    de la série de la cor  tisone, ou si l'on remplace au préalable le brome en  21 par un hydrogène,

   passer à la     17a-hydroxy-pro-          gestérone.       Le procédé permet également d'accéder au départ  d'un dérivé stéroïde     2,4-dibromé    au     Al-4-bromo-sté-          roïde    correspondant. Le remplacement de l'halogène  par un hydrogène     fournit    le     Al-stéroïde    correspon  dant, dont     certains    ne sont pas encore     décrits    ou  n'ont pas subi d'examen physiologique par suite des       difficultés    de préparation rencontrées jusqu'ici.

    Comme on     sait,    ces     Al-stéroïdes    sont une matière  première intéressante pour l'obtention des     1-hydroxy-          stéroïdes.       Enfin, si l'on part d'un     pregnane        2,4,17,21-tétra-          bromé,    on peut passer à volonté au     4,21=dibromo-          A1,16-pregnadiène    correspondant ou encore     préparer     la     21-bromo-Al,4:

  16_pregnatriène-3,20-dione    qui con  duit à la     àl,4,16_pregnatriène-3,20-dione.       En     utilisant    le procédé selon     l'invention    on peut  par exemple au     départ    de la     2,4-dibromo-17p-          acétoxy-étiocholane-3-one    passer à la     4-bromo-17p-          acétoxy-Al-étiocholène-3-one    et transformer ce dé  rivé en     17p-acétoxy-Al-étiocholène-3-one.       Par une voie     analogue,    au départ de la 2,4-di       bromo-17a-hydroxy-21-acétoxy-pregnane-3,11,

  20-          trione    on aboutit à la     17a-hydroxy-21-acétoxy-Al-          pregnène-3,11,20-trione    utilisable selon le brevet       américain    No 2737518 pour l'obtention de la     1-          hydroxy-cortisone.     



       Enfin,    la     A1,4,16-pregnatriène-3,20-dione    obtenue  par le procédé selon l'invention au départ de la 2,4,-         17,21-tétrabromopregnane-3,20-dione    fournit par       cracking,    saturation de la double liaison 16,17 et  réduction selon     Birch    la     19-norprogestérone.     



  Ces     déhydrohalogénations    sélectives     partielles     s'exercent de préférence en     position    16,17 et 1,2  ouvrant ainsi, comme il a été indiqué plus haut, un  accès, soit à des produits nouveaux, soit à des termes       intermédiaires    déjà connus mais peu     accessibles    jus  qu'ici.

   Or, d'après les     résultats    exposés dans le bre  vet principal on aurait pu conclure qu'un excès de       réactif        débromhydraterait    totalement le stéroïde       polyhalogéné.    D'autre     part,        comme    le montreront       certains    exemples, l'emploi d'un halogénure de       lithium    seul ne conduit point au     résultat    précité,  mais aboutit à la formation de diènes     conjugués          A14,13,-AIG,

  17    de même que dans le cas     des        cétosté-          roïdes        2,4-dibromés    il     fournissait    du diène     A4.5,A6.7.     



  Pour la mise en pratique du procédé, on traite  le stéroïde     polybromé,    de préférence, par le couple  bromure de lithium-carbonate de lithium en     présence     de     diméthylformamide,    pendant une durée comprise  entre 20 minutes et une heure et à une température       comprise    entre     70()    et     100,1    C.

   Le carbonate de  lithium, généralement employé en     excès,    n'étant     pas     soluble dans le     milieu    réactionnel, doit de préférence  être     maintenu        constamment    en suspension par une  bonne agitation pour     neutraliser    toute     trace    d'hy  dracide libre dans le milieu. Après précipitation, de  préférence par l'eau acidulée qui détruit l'excès de  carbonate de lithium, on obtient le stéroïde partiel  lement     déhydrohalogéné    qui est généralement pur  après une seule     recristallisation.    Les rendements  avoisinent souvent 90 % de la théorie.  



  La température et la durée de réaction optima  peuvent être déterminées au préalable par un     essai     fait en petit, en suivant le spectre     d'absorption    U.V.  ou le titre en brome du produit précipité.  



  Les proportions du couple halogénure de     lithium-          carbonate    de lithium peuvent varier entre d'assez  larges     limites,    l'excès de ce dernier étant unique  ment limité par des     raisons        d'économies    ou d'agita  tion. Quant à la quantité d'halogénure de lithium  nécessaire     pour        assurer    une     vitesse    réactionnelle  satisfaisante, elle est généralement comprise entre  0,1 et 1 molécule par molécule de stéroïde     poly-          bromé.     



  La préparation des dérivés     polybromés        servant     de produit de départ et figurant dans les exemples  n'est indiquée que lorsque le produit n'est pas     décrit     dans la littérature et figure,     lorsqu'il    y a lieu, au  début des     exemples.    A la fin,     des        exemples        d7utilisa-          tion    des composés     partiellement        débromhydratés     figurent également pour montrer la valeur du pro  cédé dans l'obtention de     certains    dérivés hormonaux.  



       Les        points    de fusion sont des points de fusion  instantanée déterminés sur     bloc    de     Maquenne.

    On dissout 0,9 g de bromure de lithium anhydre  dans 25     cm3    de     diméthylformamide    et ajoute 2 g de  carbonate de lithium sec.

   A la suspension agitée et  chauffée à 950 C on ajoute sous courant d'azote  5 g de     17,21-dibromo-3a-acétoxy-pregnane-20-one,     F.     145p    C,     [a]D    = + 220 (c = 1 %,     éthanol)    dont la  préparation se fait selon les conditions décrites pour  l'isomère     (3    par Marker et     coll.    (J. Am.     Chem.    Soc.  1942, 64, 213).  



  On maintient à la même température pendant  30 minutes et verse dans l'eau renfermant un peu       d'acide    acétique pour solubiliser le carbonate de    lithium,     essore    le produit précipité, lave à l'eau et  sèche. Après     cristallisation    dans l'acétone, on obtient  3,8 g de     21-bromo-3a-acétoxy-Al5-pregnène-20-one     pure, soit un rendement de 90 %. F.     158-160p    C,       [a]D    = +     600    (c = 1 %, éthanol).

  <I>b) Par le bromure de lithium seul</I>  On opère selon a) mais en absence de carbonate  de     lithium.    On obtient un produit brut, avec un ren  dement de 84 % fondant en dessous de<B>1000</B> C.       [a]D    = +<B>2720</B> (c = 1 %,     éthanol).    Il s'agit d'un pro  duit qui, comme le montre le spectre U.V., a été  transposé en majeure partie en diène     015,1s_20-one     = y     max.    309     m[,    avec<B>El'/ </B>     '/m    = 186.  



  Par     recristallisation    dans l'acétone, on obtient la       21-bromo-OlE-pregnène-3,20-dione    pure, identique à  celle obtenue selon a) avec un rendement de 7 %  seulement.

   Am.     Chem.    Soc.,  <I>77,</I> 1064 (l955), dans 300     cm3    de méthanol absolu  contenant 3     cm3    d'acide sulfurique pur à     66o        Bé    et  fait     bouillir    à reflux     41/.    h tout en continuant l'agi  tation. La solution obtenue est glacée à environ -10 .  La     3a-hydroxy-17,21-dibromopregnane-11,20-dione     cherchée se sépare. On essore, lave au méthanol       glacé    et empâte le produit avec de l'eau très chaude  pour     éliminer    le méthanol de solvatation.

   On essore  et sèche à     40 .    On obtient 86 g, soit un rendement  de 93 %, du produit cherché. F. 1900 C,     [a]2'     =     0o    + 2- (c = 1 %, chloroforme).

Une nouvelle     débromhydratation    par le même  couple, mais conduite cette fois-ci à     150,,    C pendant  1 heure, fournit la     17(3-acétoxy-Al,4-androstadiène-          3-one    avec un rendement de 95 %, F.     153o    C,       [a]D    = +     34,,    (c= 1 %,     éthanol).         Spectre U.

   V.     h        max.    244     mu,    avec     s    =16000 (éthanol).  Le produit de départ est obtenu selon le     procédé     décrit dans le brevet suisse No 324083, et dont les    détails figurent dans le brevet principal, et possède  les     constantes    suivantes : F.     256o    C,     [a]D    = +     9o     (c =1 %, chloroforme).

Chauffé à     95 C,    pendant 15 heures     dans    les con  ditions de l'exemple 1,     avec    le     couple        déhydrohalo-          génant,    il fournit, avec un rendement de 80 %,       l'acétate    de     41-déhydrocortisone    décrit dans la litté  rature.  



  <I>b) Par le bromure de lithium seul</I>       Dans    les conditions de l'exemple 1, mais en  opérant en absence de carbonate de lithium, on  obtient, avec un rendement de 80 %, un produit brut  coloré, F.     150o    C,     renfermant    12 % de brome dont  le pouvoir rotatoire     [a]D    = + 1530 (c =1 %,     tétra-          hydrofuran).     



  La cristallisation fractionnée ne permet d'isoler  aucun composé pur. Ce     produit    est un mélange  de     4[i-bromo-17a--hydroxy-21-acétoxy-Ol-pregnène-          3,11,20-trione,    de     17a-hydroxy-21-acétoxy-01,4-pre-          gnadiène-3,11,20-trione    et de     17a-hydroxy-21-          acétoxy-44.6-pregnadiène-3,11,20-trione.     



  Le spectre U.V. montre un     h        max.    238     mu,    avec  El      19    =164.

    Par     chauffage    dans     l'acétone    en     présence    d'iodure  de sodium et précipitation dans l'eau on obtient  la     4(3-bromo,-21-iodo-A',16-pregnadiène-3,20-dione     qui, par traitement au     bisulfite    de sodium, conduit  à la     4p-bromo-Al-16-pregnadiène-3,20-dione,    F.  230- C, [a]20 = + 2000 (c = 0,5 %,     dioxane).    Sou  mis à une nouvelle     débromhydratation    selon l'une  des méthodes connues et appliquées en série sté  roïde il fournit la     AI.4.1c-pregnatriène-3,20-dione,       identique au     produit    décrit par     Rubin,         ishinsky    et       Franca        Bompard,    J. Am.     Chem.        Soc.,    73, 2338  (l951), pouvant donner la     norprogestérone    par  cracking puis hydrogénation de la double     liaison     16,17 et réduction selon     Birch.     



  Le composé de départ utilisé dans cet exemple  est obtenu comme suit  10g de     17,21-dibromo-pregnane-3,20-dione,    F.       162-165 C,    dont la     préparation    est décrite dans le  brevet suisse No 336796, sont dissous dans 70     cm3     de     dioxane    anhydre à     20 C.    A la solution agitée,  on ajoute très rapidement, après avoir     amorcé    la  réaction avec très peu de brome, 7 g de cet halogène.  L'absorption est instantanée. Au cours de l'introduc  tion, la température monte à 350 C.  



  La solution est versée dans l'eau. Il se forme une  huile incolore qui     cristallise    peu à peu. On     essore    et  empâte dans un mélange à     parties    égales de     méthanol     et d'éther     isopropylique.    On essore et lave le pro  duit à l'éther     isopropylique.    On obtient 4,65 g (soit  un rendement de 35 %) de     2a,4@,17a,(31-tétra-bro-          mopregnane-3,20-dione,    F.

       230     C, dont le     point    de  fusion ne change pas par     recristallisation,   <B>MD</B> =       --50     - 2 (c = 1 %,     dioxane).

The formation of a diene 4,6 is avoided and it has been observed that, when the steroid derivative which is to be dehydrohalogenated carries a bromine at 17, the migration of the 16,17 double bond formed towards the 14,15 position is avoided. .



  The process according to the invention relates to the preparation of unsaturated ketosteroids, containing at least one bromine atom in the molecule, by partial dehydrohalogenation of corresponding saturated polybrominated derivatives and is characterized in that the polybrominated steroid is treated, in presence of a lithium halide-lithium carbonate couple in an N, N-disubstituted amide of a lower aliphatic acid,

   at a temperature between 70 ° and 1000 C, the reaction time being between 20 minutes and one hour and in that the unsaturated brominated derivative is isolated.



  Of course, the polybrominated steroids which are subjected to the process according to the invention can carry, apart from the bromine atoms, various other substituents of the A, B, C, D rings or of the side chain, in particular carbonyl or hydroxyl groups or derivatives of these functions which are easily reconverted into ketones or alcohols, such as acetals, enes, amines,

       Finally, the A1,4,16-pregnatriene-3,20-dione obtained by the process according to the invention starting from the 2,4, - 17,21-tetrabromopregnane-3,20-dione provided by cracking, saturation of the 16,17 double bond and reduction according to Birch 19-norprogesterone.



  These partial selective dehydrohalogenations are preferably exerted in positions 16, 17 and 1,2 thus opening, as indicated above, access either to new products or to intermediate terms already known but not very accessible until 'here.

   However, from the results presented in the main patent, it could have been concluded that an excess of reagent would completely dehydrate the polyhalogenated steroid.   For the practice of the process, the polybrominated steroid is treated, preferably, with the lithium bromide-lithium carbonate couple in the presence of dimethylformamide, for a period of between 20 minutes and one hour and at a temperature of between 70 ( ) and 100.1 C.

   Lithium carbonate, generally used in excess, not being soluble in the reaction medium, should preferably be kept constantly in suspension by good stirring in order to neutralize any trace of free hy dracid in the medium. After precipitation, preferably with acidulated water which destroys the excess of lithium carbonate, the partially dehydrohalogenated partial steroid is obtained which is generally pure after a single recrystallization. Yields are often around 90% of theory.



  The optimum reaction temperature and time can be determined beforehand by a small test, following the U.V. absorption spectrum or the bromine titer of the precipitated product.



  The proportions of the lithium halide-lithium carbonate couple can vary between fairly wide limits, the excess of the latter being only limited by reasons of economy or agitation. As for the quantity of lithium halide necessary to ensure a satisfactory reaction rate, it is generally between 0.1 and 1 molecule per molecule of polybrominated steroid.



  The preparation of the polybrominated derivatives serving as starting material and appearing in the examples is only indicated when the product is not described in the literature and appears, where appropriate, at the start of the examples. At the end, examples of the use of partially dehydrated compounds are also given to show the value of the process in obtaining certain hormonal derivatives.
