Procédé de préparation de cétostéroïdes non saturés Parmi les hormones stéroïdes à activité cortisoni- que utilisées en thérapeutique, la Al-déhydrocortisone (prednisone) et le Al-déhydrocortisol (prednisolone) ont acquis récemment une place très importante.
D'autres 3-cétostéroïdes diéniques 1,4 constituent des produits intermédiaires pour préparer soit l'un des composés précités, comme c'est le cas de la A1-déhy- dro substance S (17u,21-dihydroxy-Al,4-prégnadiène- 3,20-dione), soit, depuis plus longtemps, des estro- gènes comme c'est le cas de la Al@4-androstadiénolone ou des dérivés apparentés.
Pour accéder aux Ah4-3-cétostéroïdes de la série de l'androstadiène ou du prégnadiène, on peut soit utiliser des méthodes microbiologiques en mettant à profit les propriétés deshydrogénantes des ferments sécrétés par certains micro-organismes, comme l'ont montré parallèlement Vischer, Meystre et Wettstein (Helv. Chim. Acta, 195, 38, 835) et Hershberg et Collaborateurs (J.
Am. Chem. Soc., 1955,<I>77,</I> 4184), soit encore partir du dérivé 2,4-dihalogéné corres pondant au cétostéroïde diénique qu'on veut prépa rer, lequel, par double déhydrohalogénation, fournit le produit voulu, selon le schéma

D'après ce qui précède, cette double déhydro- halogénation constitue la dernière ou l'avant-dernière étape de la synthèse du composé diénique cherché et intervient donc à un moment où le rendement, et sur tout la .pureté du produit, exercent une influence pri mordiale sur le prix de revient et la production.
Lorsqu'on essaie de déhydrohalogéner ces dérivés 2,4-dihalogénés par les méthodes usuelles (traitement à la collidine, passage par les dinitro-phény1hydr- azones ou semi-carbazones, ou traitement aux halo génures de lithium dans le diméthylformamide selon la méthode de Holysz (J.
Am. Chem. Soc., 1953, <I>75</I> 4432)), on se heurte à différentes difficultés dont la principale est constituée par la migration concomi tante de la double liaison 1,2 en 6,7. Cette constata tion des inventeurs est corroborée par la publication de Fried et Collaborateurs (J. Am. Chem. Soc., 1955, <I>77,</I> 4432 note 14).
En effet, ces auteurs préconisent, comme méthode de préparation de choix de l'acétate de 6-déhydro-9a-fluoro-hydrocortisone, l'action du chlorure de lithium au sein du diméthylformamide sur le dérivé 2,4-dibromé correspondant. A cette dif ficulté s'ajoute, dans le cas du passage par la dinitro- phénylhydrazone ou la semi-carbazone, celle du cli vage de l'hydrazone diénique. La présence du diène. 4,
6 indésirable se révèle immédiatement à l'examen du spectre U.V. du produit obtenu qui montre la pré sence de deux maxima d'absorption avec très souvent prépondérance du diène 4,6.
Le produit réactionnel étant constitué par un mélange de diènes 1,4 et 4,6 ainsi que de produit de départ non transformé ou altéré, on est obligé de recourir à une purification laborieuse par cristallisa tion fractionnée très poussée ou par chromatographie, et les rendements en diène 1,4- cherché sont souvent assez bas. C'est ainsi que Herzog et Collaborateurs (J.
Am. Chem. Soc., 1955, 77, 4782) obtiennent, au départ de 20 g de 17a-hydroxy-21-acétoxy-prégnane- 3,11,20-trione, après dibromuration et débromhydra- tation, seulement 2,4 g d'acétate de Al-déhydro- cortisone.
Dans son brevet suisse No 324083, la titulaire a décrit un procédé de préparation de cétostéroïdes a-bromés convenant particulièrement bien à l'obten tion de 3-cétostéroïdes 2,4-dibromés, aussi bien en série allo qu'en série<I>normale</I> et permettant notam ment un accès facile à la 2,4-dibromo-androstane- 3,17-dione, aux esters en 17 de la 2,
4-dibromo an- drostanol-3-one et à la 2,4-dibromo-17a-hydroxy- prégnane-3,11,20-trione.
D'autres 3-cétostéroïdes 2,4-dibromés peuvent être préparés par des procédés qui font l'objet des brevets suisses Nos 337840, 339202, 340506 et 345003.
L'objet de la présente invention est un procédé de double déhydrohalogénation des 3-cétotstéroïdes dihalogénés en position 2,4- de la susdite formule A appartenant à la série allo ou à la série<I>normale,</I> les dits cétostéroïdes pouvant faire partie du groupe de l'androstane, de l'étiocholane, de l'allo-prégnane ou du prégnane et tous analogues à chaînes ramifiées en 17.
Ces cétostéroïdes peuvent porter dans les noyaux B, C et D d'autres substituants, notamment des grou pes hydroxyles libres ou estérifiés, des groupes car bonyles libres ou bloqués et, s'ils appartiennent à la série du prégnane ou de l'allo-prégnane, leur chaîne latérale comportera un carbonyle ou un ester d'énol en position 20, un groupe méthyle libre ou halogéné ou une fonction acyloxy en 21.
Le nouveau procédé selon l'invention est remar quable en ce qu'il fournit, de façon pratiquement exclusive, le diène 1,4 cherché, l'analyse spectrogra- phique révélant l'absence de l'isomère 4,6 facilement décelable par son maximum d'absorption caractéristi que différent du maximum d'absorption du diène 1,4.
On a en effet trouvé que si l'on traite à chaud un 3-cétostéroïde 2,4-dihalogéné par un excès (géné ralement un poids égal) de carbonate de lithium, en présence d'un amide disubstitué comme le diméthyl- formamide, il y a formation concomitante d'halogé nure de lithium, en même temps que s'opère la dou ble déhydrohalogénation en 1,4, sans formation simultanée de diène 4,6.
Le procédé selon l'invention est caractérisé en ce qu'on traite un 3-cétostéroïde dihalogéné dont le cycle A présente la structure

dans laquelle X et X' constituent des halogènes iden tiques ou non, au sein d'un amide disubstitué d'un acide gras inférieur, à chaud, avec un produit ren fermant du carbonate de lithium.
La réaction, assez lente au début, progresse ensuite plus rapidement, toujours en présence d'un excès de carbonate de lithium. Ce dernier n'étant pas soluble dans le milieu réactionnel, on peut le mainte nir en suspension par une bonne agitation. Après précipitation par l'eau acidulée pour dissoudre le carbonate de lithium, on obtient le cétostéroïde di- énique cherché, qui est généralement pur après une seule recristallisation. Les rendements sont élevés.
Un mode d'exécution du présent procédé con siste à employer un mélange halogénure de lithium- carbonate de lithium et peut ainsi accélérer la ré action.

On fait passer un courant d'azote et ajoute 5,52 g (0,01 mol.) de 17-benzoate de 2,4-di- bromo-androstane-17p-ol-3-one de formule A (préparé selon l'exemple 5 du brevet suisse NI, 324083) et chauffe le mélange sous agitation et sans interrompre le barbotage d'azote pendant quinze heures à 90-950 C. On refroidit à la tem pérature ambiante et verse dans 200 cm3 d'eau contenant 5 cm3 d'acide acétique. On essore, lave à l'eau, sèche et recristallise dans l'acétone.
On obtient ainsi 3,1 g de 17-benzoate de A14-andro- stadiène-170-ol-3-one de formule B, soit un ren dement de 80%, F=221 o C, [a]20 =+122, (c=1%, chloroforme), a max. = 235 m[. avec F,= 24450 et 1. = 283,
5 m#t avec F=1500. Le produit ne renferme donc pas de diène 4,6.

<I>b) Par le mélange chlorure de lithium-</I> <I>carbonate de lithium</I> On opère comme dans l'exemple a), mais on introduit au début 2 g de carbonate de lithium. Le produit obtenu est constitué exclusivement par la AM-androstadiène-3,17-dione, F = 139o C, [ ]20 = + 1140 (c =1 0/0, chloroforme), @max. = 244 ml, avec s = 16 200.

A 100 cm3 de diméthylformamide, on ajoute 5,2 g (0,06 mol.) de bromure de lithium anhydre et 4,5 g (0,06 mol.) de carbonate de lithium anhydre.
Sous agitation et sous courant d'azote, on ajoute 4,9 g de 17-acétate de 2,4-dibromo-étio- cholane-170-ol-3-ane de formule E (0,01 mol.) et porte à l'ébullition sous reflux pendant une heure. Après purification par empâtage avec l'éther isopropylique, essorage et séchage, on obtient 3,02 g (soit un rendement de 92 0/0) de produit pur, F=152,50 C, [a]20 =+30 , (c = 1 0/0, éthanol), @max. = 245 mou avec s =15 100, ce qui dénote l'absence complète du diène 4,6.
En opérant dans des conditions identi ques avec seulement 0,087 g (0,001 mol.) de bro mure de lithium pour 4,5 g (0,06 mol.) de carbo nate de lithium et 4,9 g (0,01 mol.) de 17-acétate de 2,4-dibromo-étiocholane-17p-ol-3-one, on obtient 3,2 g (98 %) de 17-acétate de Al>4-andro- stadiène-17(3-ol-3-one, F = 150 C, [a]20 = + 32,5- (c = 1 0/0, éthanol).
Après purification par l'éther isopropylique, on recueille 2,95 g (90'0%), F. (instantanée) = 152,5 C, [a] D = 30 (c = 10/0, éthanol).
<I>b) Par le carbonate de lithium seul</I> Dans 40 ce de diméthy1formamide chauffé à 1000 C, on introduit, en agitant, sous barbotage d'azote, 2 g de carbonate de lithium anhydre, puis 2 g de 2,4-dibromo-17(3-acétoxy-étiocholane-3- one. On chauffe pendant une heure au reflux, defroidit, filtre et précipite par l'eau. Après lavage, on recristallise dans l'acétate d'éthyle.
On obtient ainsi 42 % de 17p-acétoxy-Al.4-andro- stadiène-3-one, F = 1520 C, [a]20 = + 32- 2 (c = 2%, chloroforme) ; @max. = 244,5 mil, E = 15 700, ce qui indique l'absence de diène 4,6.

<I>b) Par le mélange bromure de lithium-</I> <I>carbonate de lithium</I> A 35 ce de diméthylformamide, on ajoute 2,6 g (0,03 mol.) de bromure de lithium anhydre et 4,5 g (0,06 mol.) de carbonate de lithium anhydre.
Sous forte agitation et courant d'azote, on introduit 5,6 g (0,01 mol.) de 2u,4r-dibromo- 17a - hydroxy-21-acétoxy-prégnane-3,11,20-trione et, sans interrompre le courant d'azote et l'agita tion, on chauffe quinze heures à 95o C. On re froidit, verse le mélange dans 200 cm; d'eau et 10 ce d'acide acétique. On essore, lave à l'eau, sèche. On obtient 3,92 g (soit un rendement de 98 0/0) d'acétate de Al-déhydrocortisone pratique ment pur.
F = 222-232 C, [a]D =<B>4183a</B> (c = 1,%, dioxane) avec imax. = 239,5 mu. avec e = 14 300 et @ = 285 m#t avec s = 600. Le diène 4,6 correspondant est donc absent. Le produit est purifié pour analyse par recristallisation dans l'acétone.
Il fournit 3,6 g (soit un rendement de 90'0/0) d'acétate de Al-déhydrocortisone puris- sime, F = 225-2350 C, [ ]20 = + 1850 (c = 1 0/0, dioxane), @max. = 239,5 mit avec 3 = 14 950 et # = 285 m#t avec s = 600.

La 2a,4(1-dibromo-17a-hydroxy-21-acétoxy-pré- gnane-3,20-dione de formule L, de départ, est pré parée comme suit, en appliquant le procédé de bro- muration dans le dioxane décrit dans les brevets cités à la page 4 de ce texte.
On dissout 39 g (0,1 mol.) de 17a-hydroxy-21- acétoxy-prégnane-3,20-dione, F = 197o C, [a]2 = + 68o 2 (c = 1 0/0, éthanol) dans 600 cm3 de di- oxane anhydre. On ajoute 1 cm3 d'acide acétique à 50 % d'acide bromhydrique sec,
puis introduit très rapidement 36 g de brome dans 100 cm3 d'acide acétique. La température s'élève de 10o C environ. Après avoir vérifié que tout le brome a été absorbé, on ajoute 300 cm3 d'eau distillée. La solution ainsi obtenue est versée lentement sous agitation dans 6 litres d'eau. Le produit dibromé précipité est essoré et lavé à l'eau, puis séché sous vide à la température ordinaire.
On obtient 54 g de produit brut titrant 29 à 29,5 % de brome (théorie 29,2'%). Après recris- tallisation dans le mélange acétate d'éthyle-hexane, on obtient la 2a,4(3-dibromo-17a-hydroxy-21-acét- oxy-prégnane-3,20-dione de formule L, pure, [ ]D = -16 4 (c = 0,5 0/0, acétone).
Calculé<B>:</B> Br 29,2 0/0 Trouvé : Br 29,1'% Le même produit peut également être obtenu, avec les mêmes rendements, à partir de la 4-bromo- 17a-hydroxy-21-acétoxy-prégnane-3,20-dione par bromuration dans les mêmes conditions.
A 35 cm3 de diméthylformamide, on ajoute 2,6 g (0,03 mol.) de bromure de lithium anhydre et 3 g (0,04 mol.) de carbonate de lithium anhydre.
On agite la suspension sous azote et introduit 5,4 g de composé de formule L préparé comme on vient de l'indiquer. Le mélange réactionnel est chauffé sans interrompre l'agitation ni le courant d'azote pen dant vingt heures à 95-100c, C. On refroidit et verse dans 100 cm3 d'eau et 10 cm3 d'acide acétique. On essore, lave à l'eau et sèche. On obtient 3,7 g de 21- acétate de Al-déhydro substance S de formule M.
Le produit est purifié par recristallisation dans le mélange acétone-hexane et fournit 2,7 g de 21-acét- oxy-17a-hydroxy-Ai>4-prégnadiène-3,20-dione, F = 2270 C, [a] D = + 780 (c = 0,5 0/0, acétone) ; spectre U.V. @max. = 245,5 mec avec E = 14 650.
Par saponification de l'acétate, on obtient la Al-déhydro substance S, F = 258o C.
<I>Analyse:</I> Cs1H.,804 = 344,44 Calculé: C 73,22 % H 8,19:()/o Trouvé : C 73,0 % H 8,2 0/0 Spectre U.V. ?.max. = 245 mu, avec 8 = 15 280. Le produit ne renferme pas de diène 4,6.

Après purification par recristallisation en acétone, on obtient l'acétate de d1-déhydrocortisol pur, F = 2440 C, [a]D = + 1150 (c = 1 /o, dioxane), ?,max. = 243 mu, avec e = 14 400.

On obtient 3,35 g de produit brut (soit un rendement de 80 /o), [a]20 = + 171 (c = 1 %, di- méthylformamide). Le spectre U.V. montre que Imax. = 239 mu avec e = 14 800.
Après empâtage dans l'acétone, on obtient la 21-bromo-17a-hydroxy- Ol,4-prégnadiène-3,11,20-trione pure de formule S, F (décomposition) =<B>2510</B> C, [a] D = + 1771, (c = 1 /o, diméthylformamide), #max. = 239 mu avec = 15 000.

From the foregoing, this double dehydrohalogenation constitutes the last or penultimate step in the synthesis of the sought diene compound and therefore occurs at a time when the yield, and over all the purity of the product, exerts a primary influence on cost price and production.

Am. Chem. Soc., 1953, <I> 75 </I> 4432)), we come up against various difficulties, the main one of which is the concomitant migration of the double bond 1,2 in 6,7. This finding of the inventors is corroborated by the publication of Fried et al. (J. Am. Chem. Soc., 1955, <I> 77, </I> 4432 note 14).
In fact, these authors recommend, as the method of preparation of choice for 6-dehydro-9a-fluoro-hydrocortisone acetate, the action of lithium chloride within dimethylformamide on the corresponding 2,4-dibromine derivative. To this difficulty is added, in the case of passage through dinitro-phenylhydrazone or semi-carbazone, that of cleavage of the diene hydrazone. The presence of diene. 4,
6 undesirable is immediately revealed on examination of the U.V. spectrum of the product obtained which shows the presence of two absorption maxima with very often preponderance of diene 4,6.
The reaction product being constituted by a mixture of 1,4 and 4,6 dienes as well as of unprocessed or altered starting material, it is necessary to resort to a laborious purification by very thorough fractional crystallization or by chromatography, and the yields in 1,4-diene sought are often quite low. This is how Herzog et Collaborateurs (J.
Am. Chem. Soc., 1955, 77, 4782) obtain, starting with 20 g of 17α-hydroxy-21-acetoxy-pregnan-3,11,20-trione, after dibromuration and debromhydration, only 2.4 g of acetate of Al-dehydro-cortisone.
In its Swiss Patent No. 324083, the patentee described a process for the preparation of α-brominated ketosteroids particularly suitable for obtaining 2,4-dibrominated 3-ketosteroids, both in the allo series and in the <I> series. normal </I> and in particular allowing easy access to 2,4-dibromo-androstane-3,17-dione, to esters in 17 of 2,
4-dibromo an-drostanol-3-one and 2,4-dibromo-17a-hydroxy-pregnan-3,11,20-trione.
Other 2,4-dibrominated 3-ketosteroids can be prepared by methods which are the subject of Swiss patents Nos. 337840, 339202, 340506 and 345003.
The object of the present invention is a process for the double dehydrohalogenation of dihalogenated 3-ketotsteroids in position 2,4- of the aforesaid formula A belonging to the allo series or to the <I> normal series, </I> the so-called ketosteroids which may be part of the group of androstane, etiocholane, allo-pregnan or pregnan and all analogues with branched chains in 17.
These ketosteroids can carry in the rings B, C and D other substituents, in particular free or esterified hydroxyl groups, free or blocked carbonyl groups and, if they belong to the pregnan or allo- series. Pregnane, their side chain will contain a carbonyl or an enol ester in position 20, a free or halogenated methyl group or an acyloxy function in 21.
The new process according to the invention is remarkable in that it provides, practically exclusively, the sought-after 1,4 diene, the spectrographic analysis revealing the absence of the 4,6 isomer easily detectable by its. absorption maximum characteristic different from the absorption maximum of 1,4-diene.
It has in fact been found that if a 2,4-dihalogenated 3-ketosteroid is heat treated with an excess (generally an equal weight) of lithium carbonate, in the presence of a disubstituted amide such as dimethylformamide, there is concomitant formation of lithium halide, at the same time as the double dehydrohalogenation at 1,4 takes place, without simultaneous formation of diene 4,6.
The process according to the invention is characterized in that a dihalogenated 3-ketosteroid is treated, the ring A of which has the structure

in which X and X 'constitute halogens, which may or may not be identical, in an amide disubstituted from a lower fatty acid, when hot, with a product containing lithium carbonate.
The reaction, quite slow at the start, then progresses more rapidly, always in the presence of an excess of lithium carbonate. The latter not being soluble in the reaction medium, it can be kept in suspension by good stirring. After precipitation with acidulated water to dissolve lithium carbonate, the desired diene ketosteroid is obtained, which is generally pure after a single recrystallization. The returns are high.
One embodiment of the present process is to employ a lithium halide-lithium carbonate mixture and thus can accelerate the reaction.

A stream of nitrogen is passed and 5.52 g (0.01 mol.) Of 2,4-di-bromo-androstan-17p-ol-3-one 17-benzoate of formula A (prepared according to 'Example 5 of Swiss Patent NI, 324083) and heat the mixture with stirring and without interrupting the nitrogen bubbling for fifteen hours at 90-950 C. The mixture is cooled to room temperature and poured into 200 cm3 of water containing 5 cm3 of acetic acid. It is filtered off, washed with water, dried and recrystallized from acetone.
