Abstract

"ENGENHARIA METABóLICA PARA A PRODUçãO DE AMINOáCIDOS". A presente invenção é dirigida para a produção fermentativa de aminoácidos, provendo microorganismos, métodos e processos utilizáveis. Os microorganismos da invenção são capazes de converter glucose em aminoácidos diferentes de L-isoleucina, L-leucina e L-valina com maior eficiência do que a cepa parental. A invenção provê microorganismos que são tornados auxotróficos ou braditróficos para a síntese de um ou mais aminoácidos de cadeia ramificada por mutagênese e selecionados por sua capacidade de produzir maiores rendimentos percentuais do aminoácido desejado do que a cepa parental. Os microorganismos preferidos são Corynebacterium, Brevibacterium ou Escherichia coli produzindo L-lisina. A mutagênese é realizada por técnicas clássicas ou através de metodologia de rDNA. Os métodos da invenção são projetados para aumentar a produção de um aminoácido por mutagênese de uma cepa parental, selecionando células auxotróficas ou braditróficas para a síntese de um ou mais aminoácidos de cadeia ramificada e selecionando auxotrofos ou braditrofos de aminoácido de cadeia ramificada que produzem um maior rendimento percentual a partir de dextrose do desejado aminoácido do que a cepa parental. Os processos da invenção são projetados para a produção de um aminoácido compreendendo cultivar, em um meio, um microorganismo obtido por mutagênese de uma cepa parental e selecionando variantes que são capazes de converter glucose em aminoácidos diferentes de Lisoleucina, L-leucina e L-valina com maior eficiência do que a cepa parental.
