Die Verbindungen der Formel I werden erfin dungsgemäss erhalten, wenn man ein Diazepinderivat der Formel:

worin R1, R4 und R5 die oben genannte Bedeutung besitzen und X Halogen oder eine höchstens 3 C- Atome aufweisende Alkoxy- oder Alkylthiogruppe darstellt, mit Ammoniak bzw.
einem primären oder sekundären Amin der Formel HNR.R3, beispiels weise Diäthylamin, Piperidin, Morpholin, N-Methyl- piperazin, alkylierte Alkylendiamine und dergleichen, umsetzt. Zur Bildung quaternärer Ammoniumderivate kann das Reaktionsprodukt nachträglich mit einem Quaternisierungsmittel behandelt werden.
Die Umsetzung des Imid-halogenids, Imido-äthers oder Imido-thioäthers mit dem Amin erfolgt zweck mässig, indem man die Komponenten, je nach ihren physikalischen Eigenschaften, evtl. unter Verwendung eines inerten Verdünnungsmittels, wie Dioxan, Xylol, Mesitylen, Decalin und dergleichen, während 1 bis 3 Tagen auf eine Temperatur von vorzugsweise über 150 C erhitzt,
sei es durch Rückflusserhitzung oder unter Verwendung des Einschlussrohres. Das Amin wird dabei vorzugsweise in mindestens dreifachem molarem überschuss verwendet. Die Reaktion wird oftmals durch Säure katalysiert, wofür dem Reak tionsgemisch einige Tropfen Eisessig (z. B. 5 Tropfen auf 10 g der Diazepinverbindung) zugesetzt werden können.
Nach Einengen des Reaktionsgemisches ver teilt man den Rückstand zweckmässig zwischen Äther und Wasser und entzieht die gebildete Base z. B. durch Extraktion mit verdünnter Salzsäure oder Essig säure.
Aus der nötigenfalls mit Kohle geklärten Hydrochlorid- oder Acetatlösung kann man die Base mit Ammoniak ausfällen und, falls sie :gut kristalli siert und in Äther schwerlöslich ist, direkt durch Abfltrieren isolieren, andernfalls. in Äther aufnehmen und die ätherische Lösung in üblicher Weise durch Auswaschen mit Wasser und Trocknen mit Natrium sulfat aufarbeiten.
Die weitere Reinigung kann durch Umkristallisieren oder Hochvakuumdestillation erfol gen.
Es versteht sich, dass man die basische Seiten kette, soweit R2 und R3 nicht gleichzeitig Wasserstoff bedeuten, auch schrittweise einführen kann, indem man die Diazepinverbindung der Formel II zunächst mit Ammoniak oder einem primären Amin umsetzt und R2 und/oder R3 nachträglich einführt,
gegebenenfalls unter gleichzeitiger Quaternisierung. Auch weitere N-Atome der basischen Seitenkette können gegebe- nenfalls nachträglich alkyliert werden.
Soweit von einem Ausgangsstoff mit Rl=Wasser- stoff ausgegangen wurde, kann auch in 5-Stellung gegebenenfalls nachträglich eine Alkylgruppe einge führt werden.
Die allfällige Quaternisierung des Umsetzungs- produktes erfolgt beispielsweise durch Behandeln mit ein, bzw. zwei molaren Äquivalenten eines Dialkyl- sulfats, Alkylhalogenids oder Sulfonsäurealkylesters.
Die erhaltenen Basen sind ,gelb, in vielen Fällen kristallisierbar, sonst im Hochvakuum' unzersetzt destillierbar, und besitzen schon auf Grund der Amidingruppierung, abgesehen von allfälligen weite ren basischen Stickstoffatomen, genügende Basen stärke, um mit anorganischen oder organischen Säu ren, beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, Essig säure, Oxalsäure, Malonsäure,
Bernsteinsäure, Malein- säure, Äpfelsäure, Weinsäure, Toluolsulfonsäure und dergleichen, in Wasser beständige Salze zu bilden.
Als Ausgangsstoffe der Formel II werden die Imido-thioäther bevorzugt, da sie am leichtesten zugänglich sind. Man erhält sie beispielsweise durch thermische Cyclisierung entsprechend substituierter o-Nitro-.diphenylamino-o'-carbonsäuren zum Lactam, dessen tautomere Form der Formel 1I entspricht, worin X eine Hydroxylgruppe ist.
Durch Behandeln des Lactams mit Phosphorpentasulfid in siedendem Pyridin erhält man das in Alkalilauge lösliche, gelbe Thiolactam (Formel Il; X = SH; tautomere Form), und daraus durch Alkylierung mit Alkali und Dialkyl- sulfat den Imido-tihoäther (Formel I; X = S-Alkyl), beide Stufen in guter Ausbeute.
Die in erfindungsgemässer Weise erhaltenen Basen, ihre quaternären Ammoniumderivate und Säure-Addi- tionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimitteln Verwendung finden können, insbe sondere als Analgetika, Chemotherapeutika, Anti- histaminika, Antiallergika, Sedativa,
Adrenolytika und Neuroplegika. Einzelne davon eignen sich zur Be handlung psychotischer Zustände.
Beispielsweise zeigt das gemäss Beispiel 27 erhal tene 8-Chlor-1 1-(N-methyl)piperazino-SH- dibenzo:[b,e],[1,4]:diazepin im Tierversuch die Eigenschaften eines Neuroplegi- kums mit starker analgetischer, parasympathicoly- tischer und sympathicolytischer Wirkung.
Die analge- tische Wirkung wird durch Messung der Schmerz schwelle bei der elektrischen Reizung der Zahnpulpa beim Kaninchen bestimmt. Sie wird in der folgenden Tabelle I zur Wirkung bekannter Analgetika in Ver gleich gesetzt.

Beispiel <I>1</I> 3,4 g 11-Methylthio-5H-dibenzo[b,e][l,4]diazepin werden in 40 ml absolutem Xylol gelöst, mit 4,3 g N-Methylpiperazin versetzt und nach Zusatz von 2 bis 3 Tropfen Eisessig 5 Stunden auf Rückfluss er- hitzt. Das Reaktionsgemisch wird mit Wasser versetzt und mit konzentrierter Natronlauge alkalisch .gemacht. Die Xylolschicht wird abgetrennt, mit Wasser neutral gewaschen und mit Salzsäure extrahiert.
Die sauren Auszüge werden mit konzentrierter Ammoniaklösung alkalisch gemacht, und das ausgefallene Öl wird aus- geäthert. Die ätherische Lösung wird mit Wasser gewaschen und eingedampft. Der Rückstand wird aus Aceton/Petroläther kristallisiert. Man erhält 3,6 g 11-(N Methyl)piperazino-5H dibenzo- [b,e][1,4]diazepin vom Schmelzpunkt 184-185 C in einer Ausbeute von 87 % der Theorie.
<I>Beispiel 2</I> Eine Lösung von 2,1 .g 5-Methyl-11-(N-Methyl)- piperazino-5H-dibenzo[b,e][1,4]diazepin, hergestellt gemäss dem späteren Beispiel 15, in 10 ml Benzol, wird mit einer Lösung von 0,87 g (äquimolare Menge) neutralem Dimethylsulfat in 30 ml Benzol versetzt, wobei sofort unter Erwärmung ein quaternäres Am moniumsalz kristallin ausfällt. Dieses wird nach Stehenlassen über Nacht abgenutscht und mit Äther nachgewaschen.
Die Ausbeute ist fast quatitativ, es handelt sich somit um das monoquaternäre Salz. Es ist hygroskopisch und lässt sich nicht umkristallisie- ren. Der Umstand, dass das UV-A .bsorptionsspektrum. mit demjenigen des Ausgangsmaterials identisch ist, spricht .dafür, dass das bereits eine Methylgruppe tra gende basische Stickstoffatom des N-Methyl- piperazin-Rests quaternisiert worden ist.
<I>Beispiel 3</I> 1,5 g 11-(N - Methyl)piperazino - 5H - dibenzo- [b,e][1,4]-diazepin, hergestellt gemäss Beispiel 1, wer den mit 10 ml Methanol und 4,0 ml (6,9 ,g) Methyl- bromid im Einschlussrohr 15 Stunden auf 100 C erhitzt.
Das in weiterem Methanol aufgenommene Reaktionsprodukt wird nach Abfiltrieren von einer geringen Menge unlöslichen Niederschlags -zur Trockne eingedampft. Durch Umkristallisieren aus Isopropanol/Essigester erhält man 1,93 g eines sehr stark hygroskopischen gelben Salzes mit dem Zer setzungspunkt 198-200 C.
Es handelt sich dabei um ein bisquaternäres Salz der Formel:

Claims (1)
- PATENTANSPRUCH I Verfahren zur Herstellung von Amidinen der Dibenzo[b,el[1,4]-diazepin-Reihe der Formel: EMI0006.0024 oder von Säure-Additionssalzen davon, worin R1 Wasserstoff oder eine Alkylgruppe mit höchstens 5 C-Atomen bedeutet, R.,und R3 gleich oder ver schieden sind und Wasserstoff oder gegebenenfalls durch Hetero@atome unterbrochene Alkylreste oder zusammen mit dem Stickstoff einen heterocyclischen Rest bedeuten, und worin R4 und R5 gleich oder verschieden sind und Wasserstoff, Halogenatome, Hydroxylgruppen oder 1 bis 3 C-Atome enthaltende Alkyl-,Alkoxy- oder Alkylmercaptogruppen bedeu ten, dadurch gekennzeichnet, dass man ein Diazepin- derivat der Formel: EMI0006.0049 worin X Halogen oder eine höchstens 3 C-Atome aufweisende Alkoxy- oder Alkylthiogruppe darstellt, mit Ammoniak bzw. einem primären oder sekundä ren Amin der Formel: HNR2R3 umsetzt. UNTERANSPRÜCHE 1.Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man als Ausgangsstoff der For mel 1I einen Imidothioäther verwendet. 2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man Amidine der Formel I, in denen R1 Wasserstoff bedeutet, nachträglich in 5-Stellung alkyliert. 3.Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man in erhaltenen Amidinen der Formel I, in denen R2 und/oder R3 Wasserstoff bedeuten, nicht Wasserstoff bedeutende Reste R2 und/oder R3 nachträglich einführt. 4.Verfahren nach Patentanspruch I oder Unter anspruch 3, dadurch gekennzeichnet, dass man in der Seitenkette gegebenenfalls vorhandene weitere N-Atome nachträglich alkyliert. PATENTANSPRUCH 1I Verwendung der nach Patentanspruch I erhal tenen Amidine der Formel I zur Herstellung ihrer quaternären Ammoniumderivate, dadurch gekenn zeichnet, dass man die Amidine der Formel I mit einem Quatemisierungsmittel behandelt.
