  Verfahren zur Herstellung von Tetracyclinderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Tetracyclinderivate, und zwar neuer substituierter Aminotetracycline, in welchen die Aminogruppe in der 7- und/oder 9-Stellung des Tetracyclinkerns steht.



   Die erfindungsgemäss erhältlichen neuen Tetracyclinderivate entsprechen der Formel

 worin R Wasserstoff oder Methyl und   Rt    und R2 Wasserstoff,   Monomethylamino    oder Dimethylamino bedeuten, wobei   Rt    und R2 nicht beide Wasserstoff sein können.



   Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man ein entsprechendes Tetracyclin mit einer Aminogruppe in der 7- und/oder 9-Stellung mit Methyljodid oder einem Methylschwefelsäureester umsetzt.



   Diese neuen Tetracyclinderivate können hergestellt werden, indem ein Tetracyclin mit einer 7- und/oder 9-Aminogruppe mit einem oder zwei Molen Methyljodid oder eines Methylschwefelsäureesters je Aminogruppe umgesetzt wird. Wenn je Aminogruppe 2 Mol Methyljodid oder eines Methylschwefelsäureesters verwendet werden, kann das zweite Mol getrennt oder gleichzeitig mit der Bildung der Kohlenstoff-Stickstoff-Bindung eingeführt werden.



   Das Verfahren gemäss der Erfindung kann mit oder ohne blockierende Gruppe an der Dimethylaminogruppe in der 4-Stellung des der Alkylierung unterworfenen Tetracyclins durchgeführt werden. Falls eine blockierende Gruppe vorhanden ist, besteht sie vorzugsweise aus der Benzylgruppe. Die Benzylgruppe kann dann nach der Alkylierung der   7- und/oder    9-Aminogruppe des Tetracyclins durch übliche katalytische Entbenzylierungsverfahren abgespalten werden. So kann ein 7- und/oder 9 Nitrotetracyclin mit Benzylchlorid unter Bildung des entsprechenden 7- und/oder 9-Nitro-4-benzyldimethylaminotetracyclinquartärsalzes umgesetzt werden, die 7und/oder 9-Nitroverbindung dann in das 7- und/oder 9-Alkyl-, Aralkyl- oder (substituiert Aralkyl)-amino-4benzyl-dimethylaminotetracyclin übergeführt werden und das Endprodukt dann durch katalytische Entbenzylierung gebiIdet werden.



   Ein anderes Mittel zur Blockierung jeder Quaternisierung an der 4-Dimethylaminogruppe des Tetracyclinmoleküls im Verlauf der direkten Methylierung besteht in der Herstellung des Monosalzes. Dieses lässt sich leicht herstellen, da die 4-Dimethylaminogruppe basischer ist und infolgedessen die Salze leichter bildet, als dies die stärker aromatischen Aminogruppen in den 7- und/oder 9-Stellungen des der Alkylierung unterworfenen Tetra  cyclinmoleküls    tun. Die Reaktionsbedingungen für die Alkylierung sind ziemlich einfach und bestehen lediglich darin, dass das 7- und/oder 9-Aminotetracyclin mit dem gewünschten Alkylierungsmittel, üblicherweise in Gegenwart eines Lösungsmittels, gegebenenfalls unter Erhitzen, in Berührung gebracht wird.



   Spezifische Ausgangsmaterialien, die beim neuen Verfahren nach der Erfindung eingesetzt werden können, sind z. B.



  7-Amino-6-desoxy-6-desmethyltetracyclin, 9-Amino-6-desoxy-6-desmethyltetracyclin,   7, 9-Diamino-6-desoxy-6-desmethyltetracyclin, 7-Amino-6-desoxytetracyclin,    7,   9-Diamino-6-desoxytetracyclin,    9-Amino-7-nitro-6-desoxy-6-desmethyltetracyclin,   9-Amino-7-nitro-6-desoxytetracyclin, 9-Amino-5 -hydroxy-6-desoxytetracyclin,    11    -Brom-7-amino-6-desmethyl-6-desoxytetracyclin    und dergleichen.  



  Die Tetracyclinausgangsmaterialien für das neue Verfahren nach der Erfindung können entsprechend den in der belgischen Patentschrift Nr. 577 854, der österreichischen Patentschrift Nr. 212 308, in den Veröffentlichungen von Beereboom et al., J.A.C.S. 82, 1003 (1960) und Boothe et al., J.A.C.S. 82, 1253 (1960) und der US Patentanmeldung Serial-No. 65 584 vom 28. Oktober 1960 beschriebenen Verfahren hergestellt werden. Die Tetracyclinausgangsmaterialien können bei dem erfindungsgemässen Verfahren sowohl in Form ihrer freien Basen als auch in Form ihrer Salze mit verschiedenen organischen und anorganischen Säuren eingesetzt werden, je nachdem, ob eine Löslichkeit in polaren oder nicht-polaren Lösungsmittelsystemen gewünscht wird.



   Typische Verbindungen, die nach dem erfindungsgemässen Verfahren hergestellt werden können, sind z. B.   



  7-Methylamino-6-desoxy-6-desmethyltetracyclin, 9-Methylamino-6-desoxy-6 -desmethyltetracyclin,    7-Dimethylamino-6-desoxy-6-desmethyltetracyclin, 9-Dimethylamino-6-desoxy-6-desmethyltetracyclin,   7-Methylamino-6-desoxytetracyclin    und   7,9-Di-(methylamino)-6-desoxytetracyclin   
Die nach dem erfindungsgemässen Verfahren erhältlichen Tetracycline sind biologisch aktiv und besitzen die Breitspektrum-Antibakterienaktivität der bisher bekannten Tetracycline. Insbesondere besitzt das 7-Methyl  amino -6- desoxy - 6- desmethyltetracyclin    eine aussergewöhnliche Aktivität sowohl bei oraler als auch bei parenteraler Verabreichung gegenüber Staphylococcus aureus, Stamm Smith, und Staphylococcus aureus, Stamm Rose, bei Infektionen von Mäusen.



   Staphylococcus aureus, Stamm Smith, wurde durch J. M. Smith und R. J. Dubos am Rockefeller-Institut untersucht und in Journ. Expt. Med. 103, 87 (1956) beschrieben. Staphylococcus aureus, Stamm Smith, ist Koagulase-positiv, Tellurit-negativ und ist gegenüber Tetracyclin, Penicillin, Streptomycin, Erythromycin, Carbomycin, Neomycin, Chloramphenicol und Novobiocin in vitro sensitiv. Versuche wurden unternommen, um diesen Stamm zu phagotypisieren, jedoch wurde festgestellt, dass er nicht typisierbar ist.



   Staphylococcus aureus, Stamm Rose, ATCC-Nr.



  14 154, wurde klinisch aus einem Abszess eines Patienten isoliert, welcher auf Behandlung mit den Tetracyclinen nicht ansprach. Dieser Organismus erwies sich als resistent gegenüber den klinisch verwendeten Tetracyclinen in vitro und in vivo. Staphylococcus aureus, Stamm Rose, ist Koagulase und Tellurit-positiv und resistent gegenüber Tetracyclin, Penicillin, Streptomycin und Erythromycin. Er ist gegenüber Carbomycin, Neomycin, Chloramphenicol und Novobiocin in vitro sensitiv. Staphylococcus aureus, Stamm Rose, wurde phagotypisiert mit den folgenden Ergebnissen:

Es wurde festgestellt, dass 7-Methylamino-6-desmethyl-6-desoxytetracyclin intravenös gegenüber Staphylococcus aureus, Stamm Smith, bei Infektionen von Mäusen etwa viermal so wirksam ist wie Tetracyclin.

   Wenn 7  Methylamino-6-desmethyl-6-desoxytetracyclin    in einer einzigen oralen Röhrendosis gegen Staphylococcus aureus, Stamm Smith, verabreicht wird, ist es 17mal wirksamer als Tetracyclin. Zusätzlich liegen die Blutplasmaspiegel von   7- Methylamino -6- desmethyl -6- desoxytetracyclin    nach einer einzelnen oralen Röhrendosis bei Mäusen etwa 5mal höher als diejenigen, die mit Desmethyl-chlortetracyclin erzielt werden.



   Von besonderer Bedeutung ist die Tatsache, dass   7-Methylamino-6-desmethyl-6-desoxytetracyclin    unter den Tetracyclinen insofern einzigartig aktiv ist, als sie eine unerwartet hohe Aktivität gegen den Tetracyclinresistenten Stamm Staphylococcus aureus, Stamm Rose, besitzt. Es stellt das einzige Tetracyclin, das bis jetzt bekannt ist, dar, welches eine orale Aktivität gegenüber dieser Tetracyclin-resistenten Infektion bei Mäusen aufweist.



   So blieben bei so niedrigen oralen Dosen, wie 16mg/kg von 20 mit Staphylococcus aureus, Stamm Rose, infizierten Mäusen, welche mit 7-Methylamino-6desmethyl-6-desoxytetracyclin behandelt worden waren, 16 Stück 7 Tage nach der Infektion am Leben, während von 20 der infizierten nicht-behandelten Kontrollmäuse 19 Stück innerhalb eines Tages starben. Unter denselben Testbedingungen ist Tetracyclin unwirksam bei der maximal erträglichen Dosis von 2048 mg/kg.



   Beispiel I
Umsetzung von   7-Amino-6-desmethyl-6-desoxy-    tetracyclin mit Methyljodid
Eine Lösung von 265 mg 7-Amino-6-desmethyl-6desoxytetracyclin in 25 cm3  Methylcellosolve  wurde bei Raumtemperatur 24 Stunden mit 0,5 cm3 Methyljodid behandelt. Es wurde ein Gemisch von 7-Dimethyl  amino-6-desmethyl-6-desoxytetracyclin    und 7-Methylamino-6-desmethyl-6-desoxytetracyclin erhalten.



   Beispiel 2
Umsetzung von 7-Amino-6-desmethyl-6-desoxy tetracyclin mit Methylsulfat
Eine Lösung von 265 mg 7-Amino-6-desmethyl-6desoxytetracyclin in 25 cm3  Methylcellosolve  wurde bei Raumtemperatur 24 Stunden mit 1 cm3 Methylsulfat behandelt. Es wurde ein Gemisch von 7-Dimethylamino6-desmethyl-6-desoxytetracyclin und 7-Methylamino-6desmethyl-6-desoxytetracyclin erhalten.



   In jedem der obigen Beispiele 1 und 2 wurde auf Papierchromatogrammen   7-Dimethylamino-6-desmethyl-    6-desoxytetracyclin nachgewiesen und erwies sich durch Vergleich in drei verschiedenen Papierchromatogrammsystemen als mit dem bekannten   7-Dimethylamino-6-des-    methyl-6-desoxytetracyclin identisch: 1) Butanol/Ammoniumhydroxyd, pH   8,0;    Rf   0,70.   



  2)    Versene /Butanol/Puffer,    pH   8,3;    Rf   0,69.   



  3) Nitromethan/Pyridin/Benzol/Puffer, pH   3,4;    Rf 0,69.   



  
 



  Process for the preparation of tetracycline derivatives
The invention relates to a process for the production of new tetracycline derivatives, specifically new substituted aminotetracyclines in which the amino group is in the 7- and / or 9-position of the tetracycline nucleus.



   The new tetracycline derivatives obtainable according to the invention correspond to the formula

 where R is hydrogen or methyl and Rt and R2 are hydrogen, monomethylamino or dimethylamino, where Rt and R2 cannot both be hydrogen.



   The process according to the invention is characterized in that a corresponding tetracycline with an amino group in the 7- and / or 9-position is reacted with methyl iodide or a methyl sulfuric acid ester.



   These new tetracycline derivatives can be prepared by reacting a tetracycline with a 7- and / or 9-amino group with one or two moles of methyl iodide or a methylsulfuric acid ester per amino group. If 2 moles of methyl iodide or a methyl sulfuric acid ester are used per amino group, the second mole can be introduced separately or simultaneously with the formation of the carbon-nitrogen bond.



   The process according to the invention can be carried out with or without a blocking group on the dimethylamino group in the 4-position of the tetracycline subjected to the alkylation. If a blocking group is present, it preferably consists of the benzyl group. After the alkylation of the 7- and / or 9-amino group of the tetracycline, the benzyl group can then be split off by customary catalytic debenzylation processes. Thus, a 7- and / or 9-nitrotetracycline can be reacted with benzyl chloride to form the corresponding 7- and / or 9-nitro-4-benzyldimethylaminotetracycline quaternary salt, and the 7- and / or 9-nitro compound can then be converted into the 7- and / or 9-alkyl , Aralkyl- or (substituted aralkyl) -amino-4benzyl-dimethylaminotetracycline can be converted and the end product can then be formed by catalytic debenzylation.



   Another means of blocking any quaternization on the 4-dimethylamino group of the tetracycline molecule in the course of direct methylation is to make the mono-salt. This is easy to prepare because the 4-dimethylamino group is more basic and consequently forms the salts more easily than do the more aromatic amino groups in the 7 and / or 9 positions of the tetracycline molecule subjected to the alkylation. The reaction conditions for the alkylation are quite simple and consist only in contacting the 7- and / or 9-aminotetracycline with the desired alkylating agent, usually in the presence of a solvent, optionally with heating.



   Specific starting materials that can be used in the new process according to the invention are e.g. B.



  7-amino-6-deoxy-6-desmethyltetracycline, 9-amino-6-deoxy-6-desmethyltetracycline, 7, 9-diamino-6-deoxy-6-desmethyltetracycline, 7-amino-6-deoxytetracycline, 7, 9- Diamino-6-deoxytetracycline, 9-amino-7-nitro-6-deoxy-6-desmethyltetracycline, 9-amino-7-nitro-6-deoxytetracycline, 9-amino-5-hydroxy-6-deoxytetracycline, 11 -Bromo- 7-amino-6-desmethyl-6-deoxytetracycline and the like.



  The tetracycline starting materials for the new process according to the invention can be prepared according to the methods described in Belgian patent specification No. 577 854, Austrian patent specification No. 212 308, in the publications by Beereboom et al., J.A.C.S. 82, 1003 (1960) and Boothe et al., J.A.C.S. 82, 1253 (1960) and US patent application Serial-No. 65 584 of October 28, 1960 described method are produced. The tetracycline starting materials can be used in the process according to the invention both in the form of their free bases and in the form of their salts with various organic and inorganic acids, depending on whether solubility in polar or non-polar solvent systems is desired.

The tetracyclines obtainable by the process according to the invention are biologically active and have the broad-spectrum antibacterial activity of the tetracyclines known to date. In particular, the 7-methyl amino -6-deoxy-6-desmethyltetracycline has exceptional activity, both when administered orally and when administered parenterally, against Staphylococcus aureus, Smith strain, and Staphylococcus aureus, Rose strain, in infections of mice.



   Staphylococcus aureus, Smith strain, has been studied by J. M. Smith and R. J. Dubos at the Rockefeller Institute and published in Journ. Expt. Med. 103, 87 (1956). Staphylococcus aureus, strain Smith, is coagulase-positive, tellurite-negative and is sensitive to tetracycline, penicillin, streptomycin, erythromycin, carbomycin, neomycin, chloramphenicol and novobiocin in vitro. Attempts have been made to phagotype this strain but have been found to be non-typable.



   Staphylococcus aureus, strain Rose, ATCC No.



  14 154, was clinically isolated from an abscess in a patient who did not respond to treatment with the tetracyclines. This organism was found to be resistant to the clinically used tetracyclines in vitro and in vivo. Staphylococcus aureus, strain Rose, is coagulase and tellurite positive and resistant to tetracycline, penicillin, streptomycin and erythromycin. It is sensitive to carbomycin, neomycin, chloramphenicol and novobiocin in vitro. Staphylococcus aureus, strain Rose, was phagotyped with the following results:

7-Methylamino-6-desmethyl-6-deoxytetracycline was found to be about four times as effective as tetracycline intravenously against Staphylococcus aureus, Smith strain, in infections of mice.

   When 7-methylamino-6-desmethyl-6-deoxytetracycline is administered in a single oral tube dose against Staphylococcus aureus, Smith strain, it is 17 times more effective than tetracycline. In addition, the blood plasma levels of 7-methylamino -6-desmethyl -6-deoxytetracycline after a single oral tube dose in mice are about 5 times higher than those obtained with desmethylchlorotetracycline.



   Of particular importance is the fact that 7-methylamino-6-desmethyl-6-deoxytetracycline is uniquely active among the tetracyclines in that it has an unexpectedly high activity against the tetracycline-resistant strain Staphylococcus aureus, strain Rose. It is the only tetracycline known to date that has oral activity against this tetracycline-resistant infection in mice.



   Thus, at oral doses as low as 16 mg / kg of 20 mice infected with Staphylococcus aureus, strain Rose, which had been treated with 7-methylamino-6desmethyl-6-deoxytetracycline, 16 remained alive 7 days after infection during out of 20 of the infected, non-treated control mice, 19 died within one day. Under the same test conditions, tetracycline is ineffective at the maximum tolerable dose of 2048 mg / kg.



   Example I.
Reaction of 7-amino-6-desmethyl-6-deoxy-tetracycline with methyl iodide
A solution of 265 mg of 7-amino-6-desmethyl-6-deoxytetracycline in 25 cm 3 of methyl cellosolve was treated with 0.5 cm 3 of methyl iodide at room temperature for 24 hours. A mixture of 7-dimethyl amino-6-desmethyl-6-deoxytetracycline and 7-methylamino-6-desmethyl-6-deoxytetracycline was obtained.



   Example 2
Reaction of 7-amino-6-desmethyl-6-deoxy tetracycline with methyl sulfate
A solution of 265 mg of 7-amino-6-desmethyl-6-deoxytetracycline in 25 cm 3 of methyl cellosolve was treated with 1 cm 3 of methyl sulfate at room temperature for 24 hours. A mixture of 7-dimethylamino-6-desmethyl-6-deoxytetracycline and 7-methylamino-6-desmethyl-6-deoxytetracycline was obtained.



   In each of the above Examples 1 and 2, 7-dimethylamino-6-desmethyl-6-deoxytetracycline was detected on paper chromatograms and, by comparison in three different paper chromatogram systems, was found to be identical to the known 7-dimethylamino-6-des-methyl-6-deoxytetracycline : 1) butanol / ammonium hydroxide, pH 8.0; Rf 0.70.



  2) Versene / butanol / buffer, pH 8.3; Rf 0.69.



  3) nitromethane / pyridine / benzene / buffer, pH 3.4; Rf 0.69.

 

Claims (1)

PATENTANSPRUCH Verfahren zur Herstellung von Tetracyclinderivaten der Formel EMI2.1 worin R Wasserstoff oder Methyl und Rt und R3 Wasserstoff, Monomethylamino oder Dimethylamino bedeuten, wobei Rj und R2 nicht beide Wasserstoff sein können, dadurch gekennzeichnet, dass man ein entsprechendes Tetracyclin mit einer Aminogruppe in der 7- und/oder 9stellung mit Methyljodid oder einem Methylschwefelsäureester umsetzt.
