Abstract
A process for preparing tetracycline derivatives containing a 7- and/or 9-amino group, in which at least one of said amino groups is substituted by one or two radicals of the formula ** (See formula) ** wherein R3 and R4 are the same or different and are hydrogen atoms or lower alkyl, aryl, substituted aryl, aralkyl or substituted aralkyl groups, characterized in that (a) a tetracycline having a 7-amino group and/or 9-amino, in which the amino group can be formed in situ with one or two moles by amino groups of an alkylating, aralcohlylating or substituted aralcohlylating reagent capable of forming a carbon bond with the nitrogen atom of the amine, (b) and, if necessary, separately or simultaneously with operation (a), react the product of operation (a) with hydrogen to reduce any unsaturated carbon to nitrogen bond, or to replace any other radical with hydrogen of hydrogen in the alkyl portion of the substituted alkyl, aralkyl or aralkyl group; with the proviso that when there is a carbonyl oxygen atom in the carbon bonded to the nitrogen of the amine, the product of operation (a) which is reacted with hydrogen, is then only a 7-formylamino tetracycline and/or -formylamino tetracycline, (c) and if two moles of the alkylating, aralcohlylating and/or substituted aralcohlylating reagent are used, the second mole can be introduced separately or simultaneously with the formation of the carbon-to-nitrogen bond and/or reaction with hydrogen.
