Description
Verfahren zur Herstellung neuer Phenothiazin-Derivate
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Phenothiazinderivaten der Formel I (siehe Formelblatt), worin R, Wasserstoff, Halogen, die Trifluormethyl-, die Cyangruppe oder eine niedere Alkanoyl-, Alkoxy-oder Alkylthiogruppe, R2 Wasserstoff oder die Methylgruppe und R3 und R4 je eine niedere Alkylgruppe oder zusammen mit dem Stickstoffatom einen Pyrrolidin-, Piperidin-, Morpholin-oder N-Methyl-piperazinring bedeuten, sowie ihrer Salze mit anorganischen oder organischen Säuren.
Erfindungsgemäss werden die Verbindungen der Formel I erhalten, indem man Verbindungen der Formel II, worin Ri und R2 obige Bedeutung besitzen und X für Chlor, Brom, Jod oder den Rest einer organischen Sulfonsäure steht, in Gegenwart eines säurebindenden Mittels in einem unter den Reaktionsbedingungen inerten Lösungsmittel mit Piperidinderivaten der Formel III, worin Rs und R4 obige Bedeutung besitzen, umsetzt und die erhaltenen Verbindungen der Formel I gewünschten- falls in ihre Säureadditionssalze überführt.
Das Verfahren wird beispielsweise folgendermassen durchgeführt :
Man erhitzt eine Verbindung der Formel II in einem wasserfreien organischen Lösungsmittel, vorzugsweise abs. Toluol oder abs. Xylol, während längerer Zeit (10 bis 50 Stunden) mit einem Piperidinderivat der Formel III unter Rühren bei Normaldruck zum Sieden oder eventuell im Bombenrohr auf 130-180 . Als säurebin- dendes Mittel verwendet man z. B. eine anorganische oder tertiäre organische Base oder aber ein zweites Mol der Verbindung III. Das erhaltene Produkt wird in bekannter Weise aus dem Reaktionsgemisch isoliert und durch chromatographische Adsorption, Kristallisation oder durch tZberführung in ein geeignetes Salz gereinigt.
Die erfindungsgemäss hergestellten Verbindungen sind in der Literatur bisher nicht beschrieben worden.
Sie stellen basische Substanzen dar und bilden mit anorganischen oder organischen Säuren beständige, bei Raumtemperatur kristallisierte Salze, welche ebenfalls Gegenstand der vorliegenden Erfindung sind. Beispiele solcher Salze sind die Hydrochloride, Hydrobromide, Sulfate oder die Malonate, Succinate, Fumarate, Hydrogenfumarate, Maleinate, Tartrate, p-Toluolsulfonate, Methansulfonate usw.
Die Verbindungen der Formel I zeichnen sich durch sedative und neuroleptische Eigenschaften aus, wie z. B. narkosepotenzierende Wirkung, Hemmung bedingter und emotioneller Reaktionen sowie der motorischen Ak tivität usw. Daneben besitzen die Verbindungen ausge prägte adrenolytische, ferner auch hypotensive bzw. antihypertensive, bradykarde und analgetische Eigenschaften.
Die Tagesdosis soll 10-500 mg betragen. Die neuen Verbindungen können in der Inneren Medizin als Sedative, zur Behandlung von Kreislauferkrankungen, insbesondere des hypertonen Formenkreises, und zur Behebung von vegetativen Dystonie verwendet werden ; vorzugsweise in Form ihrer physiologisch verträglichen wasserlöslichen Salze.
Die Verbindungen können als Arzneimittel allein oder in entsprechenden Arzneiformen für enterale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arzneiformen werden die Wirkstoffe mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verarbeitet. Man verwendet beispielsweise für Tabletten und Dragées : Milchzucker, Stärke, Talk, Stearinsäure usw. ; für Injektionspräparate : Wasser, Alkohole, Glycerin, pflanzliche ) le und dergleichen. Zudem können die Zubereitungen geeignete Konservierungs-, Stabilisierungs-und Netzmittel, Lösungsvermittler, Süss-und Farbstoffe, Aromantien usw. enthalten.
Die als Ausgangsprodukte benötigten Piperidinderivate der Formel III sind neu. Zu ihrer Herstellung setzt man ein Acetamid der Formel IV, worin R3 und R4 die eingangs erwähnte Bedeutung besitzen, in Gegenwart eines Alkaliamids, wie Lithiumamid, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in flüssigem Ammoniak oder abs. Äther, abs. Dioxan usw. oder in Gemischen von flüssigem Ammoniak mit abs.
Äther, abs. Dioxan usw., mit 1-Benzyl-4-piperidon um und spaltet im erhaltenen Kondensationsprodukt die Benzylgruppe hydrogenolytisch ab.
Praktisch verfährt man beispielsweise so, dass man eine Suspension von Lithiumamid in flüssigem Ammoniak langsam mit einem Acetamid der Formel IV versetzt und nach ca. 30minütigem Rühren 1-Benzyl-4- piperidon tropfenweise zugibt. Nach ca. zweistündigem Rühren zersetzt man das Reaktionsgemisch durch Zugabe von festem Ammoniumchlorid, lässt den Ammoniak verdampfen, versetzt den Rückstand unter Eisküh- lung mit Wasser und extrahiert mit einem mit Wasser nicht mischbaren Lösungsmittel, z. B. Benzol. Die organischen Phasen werden vereinigt, mit Wasser gewaschen, getrocknet und eingedampft.
Zur hydrogenolytischen Abspaltung der Benzylgruppe wird die als Rückstand erhaltene Verbindung z. B. in Eisessig über einem Palladiumkatalysator bei 50 und ca. 5 atü in einem Druckgefäss hydriert.
Die Verbindungen der Formel II sind bereits bekannt oder aus den entsprechenden Phenothiazinen der Formel V, worin Ri die eingangs erwähnte Bedeutung besitzt, leicht zugänglich, beispielsweise durch Umsetzung mit 1, 3-Dihalogenpropanen oder 3-Halogen-1-pro- panolen, welche in 2-Stellung eine Methylgruppe tragen können, in Gegenwart einer starken Base, wie Natriumamid in flüssigem Ammoniak, und anschliessende Behandlung der in letzterem Falle erhaltenen 3-Hydroxypropyl-phenothiazine mit Thionylchlorid, Phosphortribromid oder p-Toluol-, Methan-, Äthan-sulfonylchlorid usw.
In den nachfolgenden Beispielen, welche die Durchführung des Verfahrens erläutern, den Umfang der vorliegenden Erfindung jedoch in keiner Weise einschrän- ken sollen, erfolgen alle Temperaturangaben in Celsiusgraden ; die Schmelz-und Siedepunkte sind nicht korrigiert.

Beispiel 1 2-Chlor-10- [3- gy-4-dimethylcarbamoyl-methyl- 1-piperidyl) propyl] phenothiazin
Ein Gemisch von 9, 7 g 2-Chlor-10- (3-chlorpropyl) phenothiazin und 11, 6 g 4-Hydroxy-4-dimethylcarba- moylmethyl-piperidin wird in 40 ml Xylol im Bombenrohr während 48 Std. auf 170 erhitzt. Der Rohrinhalt wird mit 100 ml Benzol herausgespült und mit Wasser neutral gewaschen. Man trocknet über Natriumsulfat und dampft das Lösungsmittel ab.
Der erhaltene Rück- stand wird in 100 ml Aceton gelöst, über Aktivkohle filtriert und das Filtrat mit einer heissen Lösung der berechneten Menge Fumarsäure in Äthanol versetzt. Nach längerem Stehen im Eisschrank kristallisiert das reine 2-Chlor-10- [3- (4-hydroxy-4-dimethylcarbamoylmethyl- 1-piperidyl) propyl] phenothiazin-hydrogenfumarat aus ; Smp. 197-200 (Zers.).
Das als Ausgangsmaterial benötigte 4-Hydroxy-4dimethylcarbamoylmethyl-piperidin wird z. B. wie folgt hergestellt : a) l-Benzyl-4-hydroxy-4-dimethylcarbamoylmethyl- piperidin
1, 4 g Lithium werden in 300 ml flüssigem Ammoniak in Gegenwart von Eisen-III-nitrat zu Lithiumamid umgesetzt. Diese Lithiumamidsuspension wird darauf tropfenweise mit 18, 3 g N, N-Dimethylacetamid versetzt.
Man rührt das Gemisch während 30 Min. bei der Temperatur des flüssigen Ammoniaks und tropft darauf langsam 18, 9 g 1-Benzyl-4-piperidon zu. Nach zweistündigem Rühren versetzt man vorsichtig mit 10 g Am- moniumchlorid, verdampft den Ammoniak und versetzt den Rückstand unter Eiskühlung mit 100 ml Wasser.
Man extrahiert mehrmals mit Benzol, wäscht die vereinigten Extrakte zweimal mit Wasser, trocknet über Natriumsulfat und dampft das Benzol ab. Der Rückstand wird unter stark vermindertem Druck destilliert, wobei das reine 1-Benzyl-4-hydroxy-4-dimethylcarbamoylmethyl-piperidin bei 164¯167 /10-4 mm Hg über- geht. b) 4-Hydroxy-4-dimethylcarbamoyltnetlayl-piperidin
23, 0 g 1-Benzyl-4-hydroxy-4-dimethylcarbamoylmethyl-piperidin werden in 170 ml Eisessig gelöst und in Gegenwart von 4, 5 g eines Palladiumkatalysators (10 /o auf Kohle) bei einem Anfangsdruck von 5 atü und einer Temperatur von 50 hydriert. Die für die Debenzylierung berechnete Menge Wasserstoff ist nach 2 Std. absorbiert.
Man filtriert vom Katalysator ab, dampft das Filtrat zur Trockne ein und nimmt den Rückstand in 40"/piger wässriger Kaliumcarbonatlösung und Chloroform auf. Man sättigt die wässrige Phase annähernd mit festem Kaliumcarbonat, extrahiert mehrmals mit Chloroform und dampft die über Natriumsulfat getrockneten Extrakte ein. Der Rückstand wird unter stark vermindertem Druck destilliert, wobei das reine 4-Hydroxy-4dimethylcarbamoylmethyl-piperidin bei 140-160 /0, 01 mm Hg übergeht (Temperatur im Luftbad gemessen).
Beispiel 2 10-3- [4-Hydroxy-4- (dimetlzylcarbcfnoydrnethyl)- piperidino]propyl}phenothiazin
Ein Gemisch von 23,8 g 10-(3-Chlorpropyl)phenothiazin, 16, 1 g 4-Hydroxy-4- (dimethylcarbamoylmethyl) piperidin und 35, 8 g Kaliumkarbonat wird in 400 ccm Xylol während 15 Std. unter Rühren zum Sieden erhitzt. Man filtriert von den anorganischen Bestandteilen ab und wäscht das Filtrat mit Wasser neutral. Man tocknet das Filtrat anschliessend über Magnesiumsulfat, dampft bei 60-70 zur Trockne ein und löst den Rück- stand in Toluol.
Die Lösung wird dreimal mit 10 /oiger Weinsäurelösung extrahiert, die Weinsäureextrakte mit festem Kaliumkarbonat alkalisch gestellt und mit Benzol extrahiert. Die vereinigten Benzolauszüge werden über Magnesiumsulfat getrocknet und bei 50 unter vermindertem Druck eingedampft. Das erhaltene Rohprodukt wird einmal aus Benzol und darauf noch einmal aus Aceton umkristallisiert. Smp. 112-113, 5 .
Beispiel 3 2-Brom-10-f3- [4-hydroxy-4- (diinett'zylcarbanioyl- methyl) piperidinolpropyllpheizothiazin
Aus 20, 25 g 2-Brom-10- (3-chlorpropyl) phenothiazin, 10, 65 g 4-Hydroxy-4- (dimethylcarbamoylmethyl) piperidin und 23, 7 g Kaliumcarbonat in 300 ccm Xylol erhält man nach dem im Beispiel 1 beschriebenen Ver f ahren 2-Brom-10-f 3- [4-hydroxy-4- (dimethylca-rbamoyl- methyl) piperidino] propyl} phenothiazin, das bei 100 bis 101 (aus Isopropanol) schmilzt.
Beispiel 4 2-Methylmercapto-10- 3- [4- (dimethylcarbamoyl- methyl)-4-hydroxypiperidino] propyl} phenothiazin
Ein Gemisch von 17, 1 g 2-Methylmercapto-10- (3- chlorpropyl)-phenothiazin, 9, 9 g 4- (Dimethylcarbamoyl- methyl)-4-hydroxy-piperidin und 22, 0 g Kaliumcarbonat wird in 250 ccm abs. Xylol während 16 Std. unter Rüh- ren zum Sieden erhitzt. Man filtriert von den anorganischen Bestandteilen ab, wäscht das Filtrat mit Wasser neutral, trocknet über Magnesiumsulfat und dampft das Lösungsmittel unter vermindertem Druck bei 60-70 ab. Zur Überführung der erhaltenen Rohbase in das Hydrogenfumarat wird in Aceton gelöst, mit der berechneten Menge Fumarsäure versetzt, kurz aufgekocht und kristallisieren gelassen.
Nach Kristallisieren des Rohhydrogenfumarates aus Athanol erhält man das reine 2-Methylmercapto-10- {3- [4- (dimethylcarbamoylme thyl)-4-hydroxypiperidino] propyl} phenothiazin-hydro- genfumarat vom Smp. 162-164 .
Nach dem in Beispiel 4 beschriebenen Verfahren werden die folgenden Verbindungen erhalten :
Beispiel 5 2-Methoxy-10- {3- [4- (dimethylcarbamoylmethyl)-4- hydroxypiperidino] propyl} phenotlaiazin aus 15, 3 g 2-Methoxy-10- (3-chlorpropyl) phenothiazin, 9, 3 g 4- (Dimethylcarbamoylmethyl)-4-hydroxypiperidin und 20, 7 g Kaliumcarbonat in 250 ccm abs. Xylol.
Hydrogenmaleinat Smp. 120-122 (Aceton).
Beispiel 6 2-Chlor-10-f 3- [4-hydroxy-4- (piperidinocarbonyl- methyl) piperidino] propylphenothiazin aus 9, 2 g 2-Chlor-10- (3-mesyloxypropyl) phenothiazin, 5, 6 g 4-Hydroxy-4- (piperidinocarbonylmethyl) piperidin und 10, 4 g Kaliumcarbonat in 200 ccm abs. Toluol.
Hydrogenmaleinat Smp. 162-165 (Aceton/Petroläther).
Das als Ausgangsmaterial benötigte 4-Hydroxy-4 (piperidinocarbonylmethyl) piperidin wird wie folgt hergestellt : a) 1-Benzyl-4-hydroxy-4- (piperidinocarbonyl- methyl) piperidin
1, 4 g Lithium werden in 300 ml flüssigem Ammoniak in Gegenwart von Eisen-III-nitrat zu Lithiumamid umgesetzt. Diese Lithiumamidsuspension wird darauf tropfenweise mit 25, 7 g 1-Acetylpiperidin versetzt, das Gemisch während 30 Min. bei der Temperatur des flüs- sigen Ammoniaks gerührt und darauf langsam 18, 9 g 1-Benzyl-4-piperidon zugetropft. Nach zweistündigem Rühren wird vorsichtig mit 10 g Ammoniumchlorid versetzt, der Ammoniak verdampft und der Rückstand unter Eiskühlung mit 100 ccm Wasser versetzt.
Man extrahiert mehrmals mit Benzol, wäscht die vereinigten Extrakte zweimal mit Wasser, trocknet über Natriumsulfat und dampft das Benzol ab. Das als Rückstand erhaltene rohe 1-Benzyl-4-hydroxy-4-(piperidinocarbo- nylmethyl) piperidin wird in der folgenden Stufe ohne weitere Reinigung eingesetzt. b) 4-Hydroxy-4- (piperidinocarbonylmethyl) piperidin
23, 0 g 1-Benzyl-4-hydroxy-4-(piperidinocarbonyl- methyl) piperidin werden in 170 ml Eisessig gelöst und in Gegenwart von 4, 5 g eines Palladiumkatalysators (101/o auf Kohle) bei einem Anfangsdruck von 5 atü und einer Temperatur von 50 hydriert. Die für Debenzylierung berechnete Menge Wasserstoff ist nach 2 Std. absorbiert.
Man filtriert vom Katalysator ab, dampft das Filtrat zur Trockne ein und nimmt den Rückstand in 40"/piger wässriger Kaliumcarbonatlösung und Chloroform auf. Die wässrige Phase wird annähernd mit festem Kaliumcarbonat gesättigt, mehrmals mit Chloro- form extrahiert und der über Natriumsulfat getrocknete Extrakt eingedampft. Der Rückstand wird unter stark vermindertem Druck destilliert, wobei das reine 4-Hy droxy-4- (piperidinocarbonylmethyl) piperidin bei 180 bis 200 /0, 1 mm Hg übergeht (Temperatur im Luftbad gemessen).
Beispiel 7 2-Chlor-10-3- [4- (diäthylcarbamoylmethyl)-4-hydroxy- piperidino] propyl} phenothiazin aus 18, 5 g 2-Chlor-10- (3-mesyloxypropyl) phenothiazin, 10, 7 g 4-(Diäthylcarbamoylmethyl)-4-hydroxypiperidin und 20, 7 g Kaliumcarbonat in 300 ccm abs. Toluol.
Hydrogenfumarat Smp. 137-139 (Aceton).
Das als Ausgangsmaterial benötigte 4-(Diäthylcar- bamoylmethyl)-4-hydroxypiperidin wird wie in Beispiel la und lb hergestellt : a) l-Benzyl-4-(diäthylcarbamoylmethyl)-4-hydroxy- piperidin aus 4, 2 g Lithium in 700 ccm flüssigem Ammoniak, 56, 7 g 1-Benzyl-4-piperidon und 69 g N, N-Diäthylacet- amid. Sdp. 167-168 /0, 005 mm Hg. b) 4-(Diäthylcarbamoylmethyl)-4-hydroxypiperidin aus 51, 4 g 1-Benzyl-4-(diäthylcarbamoylmethyl)-4-hy- droxypiperidin in 450 ccm Eisessig, 5, 2 g Palladium Kohle 10 ouzo bei 20 atü und 60 während 12 Stunden.
Sdp. 121-123 /0, 1 mm Hg.
Beispiel 8 2-Trifluor-10- 3- [4- (diäthylcarbamoylmethyl)-4- hydroxypiperidino] propyl} phenothiazin aus 13, 2 g 2-Trifluormethyl-10- (3-mesyloxypropyl) phenothiazin, 7, 0 g 4-(Diäthylcarbamoylmethyl)-4-hy- droxypiperidin und 13, 6 g Kaliumcarbonat in 250 ml abs. Toluol. Hydrogenfumarat Smp. 153-155 (Aceton).
Beispiel 9 2-Chlor-10- (3- [4-hydroxy-4- (4-methyl-l-piperazinyl- carbonylmethyl) piperidino] propyl} phenothiazin aus 8, 7 g 2-Chlor-10- (3-mesyloxypropyl) phenothiazin, 5, 7 g 4-Hydroxy-4- (4-methyl-1-piperazinylcarbonylme- thyl) piperidin und 9, 7 g Kaliumcarbonat in 200 ccm abs.
Toluol. Dihydrobromid Smp. 247-249 (Zers.) (Ätha- nol).
Das als Ausgangsmaterial verwendete 4-Hydroxy-4- (4-methyl-1-piperazinylcarbonylmethyl) piperidin wird, wie in Beispiel 6 unter a) und b) beschrieben, durch Reaktion von 4-Methyl-1-acetylpiperazin mit 1-Benzyl-4piperidon und anschliessende Debenzylierung erhalten.
Sdp. 175-178 /0, 08 mm Hg (Heissluftbad).
Beispiel 10 2-Chlor-10-3- [4-hydroxy-4- (1-pyrrolidinyl-carbonyl- methyl piperidino] propyl} phenothiazin aus 18, 5 g 2-Chlor-10- (3-mesyloxypropyl) phenothiazin, 10, 6 g 4-Hydroxy-4- (1-pyrrolidinylcarbonylmethyl) piperidin und 20, 6 g Kaliumcarbonat in 200 ccm abs. Toluol. Hydrogenmaleinat Smp. 163-165 (Aceton).
Das als Ausgangsmaterial verwendete 4-Hydroxy-4 (1-pyrrolidinylcarbonylmethyl) piperidin wird, wie inBei- spiel 6 unter a) und b) beschrieben, durch Reaktion von 1-Acetylpyrrolidin mit 1-Benzyl-4-piperidon und anschliessende Debenzylierung erhalten. Sdp. 160-180 / 0, 09 mm Hg (Heissluftbad).
Beispiel 11 2-Chlor-10-T, 3-[4-hydroxy-4-(morpholinocarbonyl- methyl) piperidino] propyl phenothiazin aus 43, 8 g 2-Chlor-10- (3-mesyloxypropyl) phenothiazin, 27, 1 g 4-Hydroxy-4- (morpholinocarbonylmethyl) piperidin und 49, 2 g Kaliumcarbonat in 1000 ccm abs. Toluol.
Hydrogenfumarat Smp. 159-162 (Aceton).
Das als Ausgangsmaterial verwendete 4-Hydroxy-4 (morpholinocarbonylmethyl) piperidin wird, wie in Beispiel 6 unter a) und b) beschrieben, durch Reaktion von 1-Acetylmorpholin und 1-Benzyl-4-piperidon und anschliessend Debenzylierung erhalten. Die Base wird roh umgesetzt. Hydrogenmaleinat Smp. 151-152 (aus Ätha- nol/Äther).
Beispiel 12 2-Acetyl-10-f3- [4- (dimethylcarbamoylmethyl)-4- hydroxypiperidino] propyl} phenothiazin
Ein Gemisch von 17, 2 g 2-Acetyl-10- (3-chlorpro- pyl) phenothiazin, 10, 0 g 4- (Dimethylcarbamoylmethyl)- 4-hydroxypiperidin und 22, 4 g Kaliumcarbonat wird in 350 ccm abs. Xylol während 18 Std. unter Rühren zum Sieden erhitzt. Man filtriert von den anorganischen Bestandteilen ab und wäscht das Filtrat mit Wasser neutral. Anschliessend wird dreimal mit total 1000 ccm 10 /oigerWeinsäurelösung extrahiert, die sauren Extrakte einmal mit Äther ausgezogen und darauf durch Zugabe von Kaliumcarbonat alkalisch gestellt. Man extrahiert die freigesetzte Base mit Benzol, trocknet über Magnesiumsulfat und dampft zur Trockne ein.
Nach Umkristallisation des verbleibenden Rückstandes aus Essigester/Pentan erhält man das reine 2-Acetyl-10- {3- [4- (dimethylcarbamoylmethyl)-4-hydroxypiperidino] pro pyl} phenothiazin vom Smp. 98-101 (sintert bei 85 ).
Nach dem in Beispiel 12 beschriebenen Verfahren werden die folgenden Verbindungen erhalten :
Beispiel 13 2-Cyano-10-{3-[4-(dimethylcarbamoylmethyl)-4hydroxypiperidino]propyl}phenothiazin aus 15, 6 g 2-Cyano-10- (3-chlorpropyl) phenothiazin, 9, 7 g 4- (Dimethylcarbamoylmethyl)-4-hydroxypiperidin und 21, 6 g Kaliumcarbonat in 300 ccm abs. Xylol. Smp.
125, 5-126, 5 (aus Isopropanol).
Beispiel 14 2-Trifluormethyl-10- {3- [4- (dimethylcarbamoylmethyl)- 4-hydroxypiperidino] propyl phenothiazin aus 17, 2 g 2-Trifluormethyl-10- (3-chlorpropyl) phenothiazin, 9, 3 g 4- (Dimethylcarbamoylmethyl)-4-hydroxy- piperidin und 20, 7 g Kaliumcarbonat in 300 ccm abs.
Xylol. Semp. 120, 5-121, 5 (aus Isopropanol).
Beispiel 15 2-Chlor-10- {3- [4- (dimethylcarbamoylmethyl)-4- hydroxypiperidino]-2-methylpropyl} phenothiazin
Ein Gemisch von 33, 4 g 2-Chlor-10- (2-methyl-3- chlorpropyl)-phenothiazin und 38, 4 g 4- (Dimethylcar- bamoylmethyl)-4-hydroxypiperidin in 500 ccm abs.
Xylol wird während 48 Std. im Autoklaven auf 170 erhitzt. Der Autoklaveninhalt wird mit Wasser neutral gewaschen und anschliessend, wie in Beispiel 12 be schrieben, aufgearbeitet. Hydrogenmaleinat Smp. 196 bis 198 (Äthanol).

Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung neuer Phenothiazinderivate der Formel I, worin Rj Wasserstoff, Halogen, die Trifluormethyl-, die Cyangruppe oder eine niedere Alkanoyl-, Alkoxy-oder Alkylthiogruppe, R2 Wasserstoff oder die Methylgruppe und Ra und R4 je eine niedere Alkylgruppe oder zusammen mit dem Stickstoffatom einen Pyrrolidin-, Piperidin-, Morpholin-oder N-Me thyl-piperazinring bedeuten, und ihrer Säureadditions- salze, dadurch gekennzeichnet, dass man Verbindungen der Formel II, worin Ri und R2 obige Bedeutung besitzen und X für Chlor, Brom, Jod oder den Rest einer organischen Sulfonsäure steht, in Gegenwart eines säurebindenden Mittels in einem unter den Reaktionsbedin gungen inerten Lösungsmittel mit Piperidinderivaten der Formel III,worin R3 und R4 obige Bedeutung besitzen, umsetzt und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
