Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer mehrbasischer Verbindungen der For

oder von Salzen davon. In Formel I bedeutet A Phenylen, Rs-substituiertes Phenylen, Phenylenamino oder Rl-substituiertes Phenylenamino. Ri ist Halogen, Amino, Nitro, Hydroxy, Alkyl, Alkoxy, Alkylthio, Acyl- amino oder Aminocarbonyl.
Die Reste R sind direkt oder über das Zwischenglied-NH-mit dem Kern ver knüpfte basische Gruppen der Formel :

worin R2 und R3 gleich oder verschieden sind und Wasserstoffatome, Alkoxyalkylgruppen oder gerade oder verzweigte Alkylgruppen mit höchstens 7 C-Atomen bedeuten, oder zusammen eine Di-oder Trimethylengruppe bilden, worin einzelne Wasserstoffatome durch Alkylgruppen mit zusammen höchstens 6 C-Atomen ersetzt sein können, und R4 Wasserstoff oder eine Alkylgruppe mit höchstens 6 C-Atomen bedeutet.
Die gewünschten Verbindungen werden erhalten, wenn man ein Amin der Formel :

Die Umsetzung zwischen Amin und Isocyanat, bei welcher anstatt vom freien Amin auch von einem Salz oder einem reaktionsfähigen Derivat ausgegangen werden kann, wird zweckmässig in einem Lösungsmittel, zum Beispiel Dimethylformamid oder Pyridin, bei Tem peraturen zwischen etwa 15 und 250 C durchgeführt.
Zur Herstellung von Verbindungen der Formel I, worin A Phenylenamino oder Rl-substituiertes Phenylen amino bedeutet, wird die Reaktion zweckmässig so durchgeführt, dass man-das Amin der Formel It direkt mit einem Diisocyanat der Formel :

worin R1 die genannte Bedeutung hat, reagieren lässt.
Hierbei entsteht intermediär eine Verbindung der Formel III, in welcher A Phenylenamino oder Rl-substi- tuiertes Phenylenamino bedeutet.
Die Verbindungen entsprechend Formel I können als freie Basen oder in Form ihrer Salze mit anorganischen oder organischen Säuren gewonnen werden. Als Salze der Basen gemäss Formel I seien diejenigen der Schwefelsäure, Salzsäure, Bromwasserstoffsäure, Iodwasser stoffsäure, Phosphorsäure, Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Weinsäure, Maleinsäure, Oxalsäure, Citronensäure, Salicylsäure und dergleichen erwähnt. Durch besonders gute Löslichkeit zeichnen sich die Salze der Hydroxycarbonsäuren, Ketocarbonsäuren und Aminocarbonsäuren aus, insbesondere die Salze der Glykolsäure, Milchsäure, Zuckersäure, Schleimsäure, Ascorbinsäure, Heptagluconsäure, Galactosidogluconsäure, Gelactosido-heptagluconsäure, Lävulinsäure und der Glutaminsäure.
Die Herstellung löslicher Salze erfolgt zweckmässig, indem man die mehrbasische Verbindung in Wasser aufschlämmt und die zur Neutralisation erforderliche Menge der gewünschten Säure zusetzt, wobei die Base in Lösung geht. Gewünschtenfalls kann man das Salz durch Eindampfen oder Acetonzusatz in fester Form gewinnen.
Die erhaltenen löslichen Salze ergeben haltbare, sterilisierbare Lösungen, die sich für Injektionszwecke eignen.
Die Lösungen können auch weitere Substanzen enthalten, doch ist zu beachten, dass diese keine Fällungsmittel sein dürfen. So ist zur Herstellung isotonischer Lösungen Kochsalz nicht verwendbar, wenn das Chlorion die mehrbasische Verbindung ausfällen würde ; in solchen Fällen eignet sich für diesen Zweck z. B. Glucose.
Die in der beschriebenen Weise erhaltenen mehrbasischen Verbindungen und ihre Salze sind neue Verbindungen. Sie besitzen pharmakologische Wirkung und eignen sich vor allem als Chemotherapeutika, insbesondere Tuberkulostatika und zur Therapie von Trypanosomenerkrankungen, sowie zur Krebsbekämpfung, insbesondere zur Bekämpfung der Leukämie. Ausserdem können sie als Zwischenprodukte zur Herstellung weiterer, insbesondere pharmakologisch wirksamer Verbindungen benutzt werden.
Die antileukämische Wirkung wird bei der durch Übertragung von Krebszellen künstlich leukämisch gemachten Maus mit Wirkstoffmengen von etwa 1 bis 500 mg/kg/Tag erzielt und äussert sich in einer Verlänge- rung der Uberlebenszeit gegenüber unbehandelten Kontrolltieren. Die Oberlebenszeit der Kontrolltiere zu 100 /o gesetzt, beträgt die Uberlebenszeit bei täglicher Verabreichung von 1 bis 500mg/kg erfindungsgemäss erhaltener Produkte bis zu 400 ouzo und mehr. Der Wirkstoff wird in gegebenenfalls isotonisch gemachter wässriger Lösung beziehungsweise Suspension i. v. oder i. p. gespritzt.
Zur Bekämpfung anderer Krebsarten sowie für allgemein chemotherapeutische Zwecke, insbesondere zur Tuberkulosebekämpfung, eignen sich auch andere Arzneiformen und Applikationsweisen. Zum Beispiel können Carcinome, Sarcome oder Tuberkuloseherde lokal behandelt werden, wobei eine Depotwirkung auftreten kann. Neben Lösungen beziehungsweise Suspensionen kommen für solche Zwecke auch pulver-oder salbenförmige Präparate in Frage, die ausser dem Wirkstoff die üblichen Hilfsstoffe enthalten.
Beispiel 1
4, 86 g p-Phenylen-diisocyanat (0, 03 Mol) und 14, 04 g p-Imidazolinoanilin-Dihydrochlorid (0, 06 Mol) werden in einem Gemisch aus 70 ml Dimethylformamid und 20 ml absolutem Pyridin erhitzt. Während kurzer Zeit tritt völlige Auflösung ein, dann erscheint ein kristalliner Niederschlag. Dieser wird abgenutscht, mit Dimethylformamid und Alkohol gewaschen, in konzentrierter Ammoniaklösung suspendiert und während 4 Stunden stehen gelassen. Die freie Base wird abgenutscht und in heisser verdünnter Essigsäure gelöst. Auf Zusatz von Kochsalzlösung scheidet sich sofort das Dihydrochlorid der Formel

ab, welches abfiltriert, fünfmal mit Wasser gewaschen und am Vakuum getrocknet wird. Die Ausbeute beträgt 12, 0 g, entsprechend 75 der Theorie.
Das Produkt schmilzt bei 325 C unter Zersetzung.
Beispiel 2
2, 0 g p-Phenylen-diisocyanat und 8, 0 g m- (N, N' Dimethyl-amidino)-anilin-Dihydrochlorid werden in einem Gemisch aus 40 ml Dimethylformamid und 10 ml absolutem rynctin unter Schütteln gelost, dann wanrena einer Stunde auf dem Dampfbad erhitzt und über Nacht stehen gelassen. Hierauf filtriert man und versetzt das Filtrat mit 15 /0iger Salzsäure. Es bildet sich ein Niederschlag, welcher abgenutscht, dreimal mit verdünnter Salzsäure und zweimal mit Aether gewaschen und am Vakuum getrocknet wird. Man erhält 6, 8 g des Dihydrochlorids der Formel

Chematograpeutische Wirkung der Produkte
In der folgenden Tabelle II sind Angaben über die tuberkulostatische bzw. cancerostatische Wirkung erfindungsgemäss erhältlicher Produkte zusammengestellt.
Die tuberkulostatische Wirkung wurde in vitro bestimmt durch Messung der niedrigsten molaren Konzentration (Molekulargewicht in mg/ml) des Wirkstoffes, welche eben noch das Wachstum von Mycobacterium tuberculosis zu hemmen vermag. Die in der zweiten Kolonne angegebenen Werte entsprechen dem negativen Logarithmus dieser geringsten molaren Hemmungskonzentration.
Die cancerostatische Wirkung wurde an Mäusen bestimmt, in welchen künstlich verschiedene Krebsarten hervorgerufen worden waren.
Bei leukämisch gemachten Mäusen (Leukämie 1210*) wurde die Überlebenszeit der mit einem erfindungsgemäss erhaltenen Produkt behandelten Tiere gegenüber unbehandelten Kontrolltieren bestimmt, wobei die durchschnittliche Uberlebenszeit der Kontrollen zu 100 ouzo gesetzt wurde. Zum Beispiel bedeutet eine mit der angegebenen Dosis erreichte Uberlebenszeit von 200 /o, dass die behandelten Leukämie-Tiere doppelt so lang überlebten wie die unbehandelten.
An soliden Tumoren, deren Typus in der dritten Kolonne von rechts angegeben ist, wurden Adenocarcinom 755 (= Ca), Ehrlich Ascites* (= EA) und Sarcom 180* (= Sa) untersucht. Hier wurde das Gewicht der Tumoren von mit erfindungsgemäss erhaltenen Produkten behandelten Tieren mit dem Tumorgewicht von unbehandelten Tieren verglichen, wobei letzteres zu 100 /o gesetzt wurde. Eine in der Kolonne rechts angegebene Gewichtsreduktion auf z. B. 27 oxo bedeutet also, dass das Gewicht der Tumoren behandelter Mäuse lediglich 27 /o desjenigen bei den Kontrollen ausmachte.

Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung mehrbasischer Verbindungen der Formel : EMI16.1 oder von Säure-Additionssalzen davon, worin A Phenylen, Ri-substituiertes Phenylen, Phenylenamino oder Ri-substituiertes Phenylenamino bedeutet ; Ri Halogen, Amino, Nitro, Hydroxy, Alkyl, Alkoxy, Alkylthio, Acylamino oder Aminocarbonyl darstellt ;die Reste R direkt oder über das Zwischenglied-NH-mit dem Kern verknüpfte basische Gruppen der Formel : EMI16.2 darstellen, in welchen R2 und Ra gleich oder verschieden sind und Wasserstoffatome, Alkoxyalkylgruppen oder gerade oder verzweigte Alkylgruppen mit höchstens 7 C-Atomen bedeuten, oder zusammen eine Di-oder Trimethylengruppe bilden, worin einzelne Wasserstoffatome durch Alkylgruppen mit zusammen höchstens 6 C-Atomen ersetzt sein können, und R4 Wasserstoff oder eine Alkylgruppe mit höchstens 6 C-Atomen bedeutet ; dadurch gekennzeichnet, dass man ein Amin der Formel : EMI16.3 worin R die genannte Bedeutung hat, mit einem Isocyanat der Formel : EMI16.4 worin R und A die genannte Bedeutung haben, umsetzt und das Reaktionsprodukt als freie Base oder in Form eines Säure-Additionssalzes gewinnt.
