Description
Verfahren zur Herstellung von äthinylsubstituierten 1-Phenoxy-2-hydroxy-3-alkylaminopropanen Die Erfindung betrifft ein Verfahren zur Herstel- lung neuer 1-Phenoxy-2-hydroxy-3-alkylaminopropane und ihrer Säureadditionssalze. Die neuen Verbindungen eignen sich zur Verwendung Aals Pharmazeutika.
Die neuen 1-Phenoxy-2-hydroxy-3-alkylaminopro- pane besitzen :die Formel I

worin R einen Alkylrest mit 1 bis 4 C-Atomen, vor zugsweise den Isopropyl- oder den tert.- Butylrest, R1 eine Äthinyl (-C=CH)gruppe, vorzugsweise in 2-Stel- lung des Benzolkerns, und R2 und R3, die gleich oder verschieden sein können, Wasserstoff oder eine Alkyl- oder Alkoxygruppe mit 1 Ibis 4 C-Atomen bedeuten, wobei Verbindungen, bei denen R2 und/oder R3 Was serstoff sind, bevorzugt werden.

worin Kt Wasserstoff oder ein Kation, beispielsweise ein Alkalimetallion, bedeutet, mit Epichlorhydrin in alkalischer Lösung hergestellt werden. Die hierzu be nötigten Ausgangsphenole der Formel VI (Kt = H) sind nach der in Bull. Chem. Soc. (Japan) 29 (l956), Seite 471 (referiert in Chem. Abstr. 51/Seite 8705b) beschriebenen Methode herstellbar.
Sie können durch Behandeln mit wässrigen Alkalien in :die entsprechenden Phenolate (Kt = Alkalimetallion) überführt werden.
Die erfindungsgemäss erhältlichen Substanzen der Formel I besitzen an der -CHOH-Gruppierung ein asymmetrisches Kohlenstoffatom und kommen daher in Form von Racematen wie auch von optisch aktiven Antipoden vor. Die optisch aktiven Verbindungen kön nen erhalten werden, indem man entweder von optisch aktiven Ausgangsverbindungen ausgeht - wo :
diese exi stieren - oder indem man die Racemate auf übliche Weise, beispielsweise mittels Ditoluylweinsäure, Di- benzoylweinsäure oder 3-Bromcampher-8-sulfonsäure in die diastereomeren Salze überführt und letztere durch fraktionierte Kristallisation trennt.
Die erhaltenen Verbindungen der Formel I können gewünschtenfalls in ihre physiologisch verträglichem Säureadditionssalze überführt werden. Für die Salzbil dung geeignete Säuren sind beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Methansulfon- säure, Maleinsäure, Essigsäure, Oxalsäure, Milchsäure, Weinsäure oder 8-Chlor-Theophyllin.
Die Verbindungen der Formel I, die üblicherweise in Form ihrer physiologisch verträglichen Säureaddi tionssalze zur Anwendung kommen, besitzen wertvolle therapeutische, insbesondere ss-adrenolytische Eigen schaften und können daher beispielsweise zur Behand lung oder Prophylaxe von Krankheiten der Herzkranz gefässe und zur Behandlung von Herzarrhythmien, ins besondere von Tachycardien, an Menschen eingesetzt werden.
Auch die blutdrucksenkenden Eigenschaften der erfindungsgemäss erhältlichen Substanzen sind thera- peutisch interessant. Als wertvoll haben sich dabei ins- besondere die Verbindungen herausgestellt, bei denen R den Isopropylrest oder den tert. Butylrest bedeutet.
Besonders wirksam sind davon wiederum Substan- zen, bei denen R1 in der 2-Stellung des Benzolkerns fixiert ist und R2 sowie R3 = Wasserstoff sind;
insbe sondere das 1-(2-Äthinylphenoxy)-2-hydroxy-3-isopro- pylaminopropan und das 1-(2-Äthinylphenoxy)-2-hy- droxy-3-tert.-butylaminopropan. Dabei ist das 1-(2-Äthinylphenoxy)-2 hydroxy-3-tert.- butylaminopropan dem 1-(2-Äthinylphenoxy)-2 hydroxy-3- isopropylaminopropan sowohl bezüglich seiner bradycardischen Eigenwirkung -als -auch bezüglich seiner isoproterenolantagonistischen Wirkung (beide getestet an Meerschweinchen)
und seiner geringen Toxizität (getestet an Mäusen) über legen und stellt bezüglich seiner Wirksamkeit die weit aus beste der erfindungsgemäss erhältlichen Verbindun gen dar.
Die übrigen erfindungsgemäss erhältlichen Ver- bindungen, beispielsweise das 1-(2-Äthinylphenoxy)-2-hydroxy-3- isopropylaminopropan, sind jedoch ihrerseits wiederum bekannten ss-Adreno- lytica wie dem 1-(1-Naphthoxy)-2 hydroxy-3-isopropyl- aminopropan überlegen.
Die Einzeldosis der erfindungsgemäss erhältlichen Verbindungen liegt bei 1 bis 300 mg .(0,016 bis 5 mg/kg), vorzugsweise bei 15 bis 100 mg (0,25 bis 1,66 mg/kg) für die orale Anwendung bzw. bei 0,1 bis 25 mg (0,002 bis 0,40 mg/kg) für die parenterale Anwendung am Menschen.
Die galenische Verarbeitung der Verbindungen der Formel I zu den üblichen Anwendungsformen wie Lö- sungen, Emulsionen, Tabletten, Dragees oder Depot formen kann -in bekannter Weise unter Heranziehung der dafür gebräuchlichen Hilfs-, Träger-, Spreng-, Binde-, Überzugs- oder Schmiermittel, Geschmackstoffe, Süssungsmittel,
Mittel zur Erzielung eines Depoteffektes oder Löslichkeitsvermittler erfolgen. Die erfindungsge- mäss erhältlichen Verbindungen können auch in Kom bination mit anderen pharmazeutischen Wirkstoffen wie beispielsweise herz- oder kreislaufwirksamen Sympa- thicomimetica oder Coronardilatatoren,
angewendet werden.. Das folgende Beispiel dient zur Erläuterung der Erfindung, ohne sie zu beschränken: <I>Beispiel</I> 1-(2-Äthinylphenoxy)-2-hydmoxy-3-tert. butylaminopropan - HCl 1,35 g (0,005 Mol) 3-tert. Butyl-5-(2-äthinyl- phenoxymethyl)-oxazolidinon-(2) werden in 8 ml Ätha- nol gelöst und nach Zugabe von 1,4g KOH (0,025 Mol) in. 3 ml H20 unter Rühren 3 Stunden am Rückfuss erhitzt.
Nach Abkühlung wird mit Wasser verdünnt, wobei geringe Mengen einer kristallinen Substanz aus fallen. Dieses nicht hydrolysierte Oxazolidinon wird ab gesaugt und die verbleibende Mutterlauge im Vakuum eingeengt. Der Rückstand wird mit verdünnter HCl digeriert und die saure, wässrige Lösung ausgeäthert. Die wässrige Phase wird dann mit NaOH alkalisch gestellt und ausfallende basische Anteile in Äther aufge nommen. Nach Waschen mit H20 wird die ätherische Lösung über MgSO4 :getrocknet, der Äther abdestilliert.
Der Rückstand wird in wenig Äthanol gedöst und mit ätherischer HCl versetzt. Das ausfallende Kristall isst wird abgetrennt und. getrocknet.
Fp: 160 bis 163 C.

- PATENTANSPRUCH Verfahren zur Herstellung von äthinylsubstituierten 1-Phenoxy-2-hydroxy-3-alkylaminopropanen der For mel I EMI0002.0120 und ihren Säureadditionssalzen, worin R einen Alkyl rest mit 1 bis 4 C-Atomen; R1 :die Äthinylgruppe; und R2 und R3, die gleich oder verschieden sein können, Wasserstoff, eine A:lkyl oder Alkoxygruppe :mit 1 bis 4 C-Atomen bedeuten, dadurch gekennzeichnet, dass man ein Oxazoüdinon der Formel II EMI0002.0136 hydrolysiert. UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekenn zeichnet, dass ,man Verbindungen :der Formel I her stellt, bei denen sowohl R. wie R3 ungleich Wasser stoff sind.2. Verfahren nach Patentanspruch, dadurch gekenn zeichnet, dass man 1-(2-Äthinylphenoxy)-2-hydroxy-3- tert. butylaminopropan herstellt. 3. Verfahren nach Patentanspruch, dadurch gekenn- zeichnet,dass man die erhaltenen Verbindungen der Formel I in ihre physiologisch verträglichen Säure- additionssalze überführt. 4. Verfahren nach den Unteransprüchen 2 und 3, dadurch gekennzeichnet, dass man das 1-(2-Äthinylphenoxy)-2,hydroxy-3-tert. butylaminopropan-hydrochlarird herstellt.
