Verfahren zur Herstellung neuer 1, 2, 6, 7, 8, 8a-Hexahydrobenz(cd) indole
Die Erfindung betrifft ein Verfahren zur Herstellung neuer 1,2,6,7,8,8a-Hexahydrobenz[cd]indole der Formel I, worin entweder X für Wasserstoff, Halogen, die Hydroxy-, Methoxyoder die Methylgruppe, Y und Z für Wasserstoff stehen, oder X, Y und Z sämtlich die Methoxygruppe bedeuten, oder X, Y und Z sämtlich die Hydroxygruppe bedeuten, oder X und Z füi die Methylgruppe und Y für Wasserstoff stehen und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin X, Y und Z obige Bedeutung besitzen, mit Äthylendiamin umsetzt und gewünschtenfalls die erhaltenen Verbindungen der Formel I in ihre Säureadditions salze überführt.
Beispielsweise geht man so vor, dass man eine Verbindung der Formel II mit Äthylendiamin vermischt, wobei man vorzugsweise eine geringe Menge Schwefelkohlenstoff, beispielsweise 0,5 bis 1%, bezogen auf die eingesetzte Menge der Verbindungen der Formel II, zugibt. Das Gemisch wird nun, vorzugsweise unter Sauerstoffausschluss, z. B. unter Stickstoffatmosphäre, während ca. 1 1/2 bis 4 Stunden erhitzt, z. B. auf ca 100 bis 1500.
Die Verwendung eines Lösungsmittels ist im allgemeinen nicht nötig.
Zur Aufarbeitung verreibt man z. B. das Reaktionsgemisch, nach Abdestillieren eines allfälligen Lösungsmittels, mit Wasser, trennt das ungelöste Produkt ab, löst es in Äthanol, setzt äthanolische Chlorwasserstoffsäure und Aktivkohle hinzu und erwärmt. Die Kohle wird abfiltriert, das Filtrat eingeengt, gegebenendalls mit einem anderen, unter den Reaktionsbedingungen inerten organischen Lösungsmittel verdünnt und zur Kristallisation stehen gelassen.
Die so erhaltenen Verbindungen der Formel I können als freie Basen oder in Form ihrer Salze nach bekannten Methoden gereinigt werden, z. B. durch Kristallisation aus einem unter den vorliegenden Bedingungen inerten Lösungsmittel oder Lösungsmittelgemisch wie Äthanol/Aceton, Äthanol/ Äther usw.
Die Verbindungen der Formel I sind basische Substanzen, welche mit anorganischen Säuren wie Chlorwasserstoff, Bromwasserstoff usw. oder mit organischen Säuren wie Methansulfonsäure, p-Toluolsulfonsäure, Maleinsäure, Weinsäure usw.
stabile, meist wasserlösliche Salze bilden.
Die Verbindungen der Formel I und ihre Säureadditionssalze, im folgenden kurz als neue Substanzen bezeichnet, sind in der Literatur bisher nicht beschrieben worden. Sie zeichnen sich in der pharmakologischen Prüfung an isolierten Organen und am Ganztier durch vielfältige und stark ausgeprägte Effekte aus und sollen daher als Heilmittel verwendet werden.
An der isoliert perfundierten Hinterextremität des Kaninchens führen die neuen Substanzen zu einer anhaltenden Vasokonstriktion bei einer Zugabe von etwa 10 bis 100 mg/min.
Aufgrund dieser Wirkung können sie als Vasokonstriktor verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 0,01 bis 0,15 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 0,1 bis 10 mg. Für orale Applikationen enthalten die Teildosen etwa 0,03 bis 5 mg der neuen Substanzen neben festen oder flüssigen Trägersubstanzen der Verdünnungsmittel.
Zudem bewirken sie am narkotisierten Hund beim Kreislaufversuch eine sympathomimetische sowie eine bradycarde Wirkung bei gleichzeitiger Carotisocclusionsreflexhemmung und können aufgrund dieser Wirkung zur Stabilisierung hypotoner Kreislaufstörungen verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 0,1 bis 10 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden.
Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 500 mg. Für orale Applikationen enthalten die Teildosen etwa 1,5 bis 250 mg der neuen Substanzen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Ausserdem zeigen sie psychotrope Eigenschaften, wie aus Beobachtungstesten an der Maus hervorgeht. Die zu verwen denden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 1 bis 10 mg/kg Körpergewicht erhalten: diese Dosis kann nötigenfalls in 2 bis 3 Anteiler oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 500 mg. Für orale Applikationen enthalten die Teildosen etwa 1,5 bis 250 mg der neuen Substanzen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Schliesslich zeigen sie beim Diurese-Versuch an der wachen Ratte eine salidiuretische Wirkung. Die zu verwendenden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 10 bis 30 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 10 bis 500 mg. Für orale Applikationen enthalten die Teildosen etwa 3 bis 250 mg der neuen Substanzen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Als Heilmittel können die neuen Verbindungen der Formel I bnv. ihre wasserlöslichen, physiologisch veträglichen Säureadditionssalze allein oder in geeigneten Arzneiformen wie Tabletten, Dragees, Injektionslösungen, Suppositorien usw.
enteral oder parenteral verabreicht werden. Ausser den üblichen anorganischen oder organischen pharmakologisch indifferenten Hilfsstoffen wie Milchzucker, Stärke, Talk, Stearinsäure, Wasser, Alkoholen, Glycerin, natürlichen oder gehärteten Ölen oder Wachsen usw. können die Zubereitungen auch geeignete Konservierungs-, Stabilisierungs- oder Netzmittel, Lösungsvermittler, Süss- oder Farbstoffe, Aromantien usw.

Zur Herstellung der als Ausgangsprodukte benötigten, bisher unbekannten Verbindungen der Formel II, welche ebenfalls von der vorliegenden Erfindung umfasst werden, kann man die Verbindungen der Formel III, worin X, Y und Z obige Bedeutung besitzen, mit Verbindungen der Formel IV, worin A Halogen, vorzugsweise Chlor oder Brom bedeutet, in Gegenwart eines säurebindenden Mittels, beispielsweise einer tertiären organischen Base wie Triäthylamin oder einer anorganischen Base, z. B. Alkalimetallkarbonate wie Kaliumkarbonat, Natriumkarbonat, usw., in einem unter den herrschenden Bedingungen inerten organischen Lösungsmittel, z. B. einem aromatischen Kohlenwasserstoff wie Benzol oder einem chlorierten Alkankohlenwasserstoff wie Chloroform oder einem niederen Alkanol umsetzen.
Die Umsetzung erfolgt vorzugsweise bei erhöhter Temperatur, insbesondere bei Siedetemperatur des Reaktionsgemisches am Rückfluss und dauert ca. 3 bis 20 Stunden.
Die als Ausgangsprodukt benötigten Verbindungen der Formel IIIa, worin entweder X' für Halogen, die Hydroxy-, Methoxy- oder Methylgruppe, Y und Z für Wasserstoff stehen, oder X', Y und Z' sämtlich die Methoxygruppe bedeuten oder X', Y' und Z' sämtlich die Hydroxygruppe bedeuten, oder X' und Z für die Methylgruppe und Y für Wasserstoff stehen, sind neu und können z. B. folgendermassen hergestellt werden:
1. Die Verbindungen der Formel IIIa', worin Hal für Halogen steht, erhält man beispielsweise, indem man 1,2,6,7,8,8a Hexahydrobenz[cd]indol nitriert, z. B. in schwefelsaurer Lösung durch Zusatz, unter Kühlung, von rauchender Salpetersäure und nachträgliche Erwärmung des Reaktionsgemisches auf ca. 90 während etwa 10 Minuten, die gebildete Nitro-Verbindung nach N-Acylierung, z.
B. mittels Acetanhydrid, zum 1-Acetyl-5-amino-1,2,6,7,8, 8a-hexahydrobenz[cd]- indol reduziert, die erhaltene Amino-Verbindung diazotiert, z. B. mittels Natriumnitrit in schwefelsaurer Lösung bei etwa 0 bis 5 , das entstandene Diazoniumsalz nach Sandmeyer umsetzt und die so erhaltene Halogenverbindung zu den Verbindungen der Formel lila' deacetyliert.
Die Reduktion des 1-Acetyl-1,2,6,7,8,8a-hexahydro-5- nitrobenz[cd]indols erfolgt z. B. katalytisch, beispielsweise in Gegenwart eines Palladium- oder Nickelkatalysators in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z. B. einem niederen Alkanol wie Methanol. Nach einer Variante wird diese Reduktion z. B. mit Hilfe von Hydrazinhydrat in Gegenwart von Raney-Nickel durchgeführt.
Nach beendeter Reaktion filtriert man vom Katalysator ab und verdampft schonend zur Trockne. Die gebildete Amino Verbindung kann nach bekannten Methoden, z. B. durch Kristallisation aus einem unter den herrschenden Bedingungen inerten organischen Lösungsmittel, gereinigt werden.
Die Deacetylierung wird durch Hydrolyse, z. B. durch Erhitzen in einem Gemisch von Eisessig/konzentrierte Salzsäure/ Wasser, durch Sieden mit wässerig-methanolischer Natriumhydroxid-Lösung usw., durchgeführt.
Die Verbindungen der Formel lila' können auch durch Umsetzung des 1-Acetyl-5-amino- 1,2,6,7,8 ,8a-hexahydro- benz[cd]indols mit einer Lösung eines komplexen Kupferhalogenids, hergestellt durch Einleiten von Stickstoffmonoxid in eine Lösung von Kupfer(ll)halogenid in einem unter den herrschenden Bedingungen inerten organischen Lösungsmittel wie Acetonitril, und darauffolgende Deacetylierung des so entstandenen Reaktionsproduktes erhalten werden.
2. Zur Herstellung der Verbindung der Formel IIIal', worin entweder X" für die Methoxygruppe, Y' und 1' für Wasserstoff stehen, oder X", Y" und Z" sämtlich die Methoxygruppe bedeuten, oder X" und Z" für die Methylgruppe und Y" für Wasserstoff stehen, bromiert man z. B. eine Verbindung der Formel V, worin xi', y" und Z" obige Bedeutung besitzen, z. B. mit N-Bromsuccinimid in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel wie Acetonitril, setzt die erhaltene 8-Bromtetralon-Verbindung zum entsprechenden 8-Cyanotetralon um, beispielsweise durch Sieden mit Kupfercyanid in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.
B. einem di(nieder)Alkylamid einer aliphatischen Carbonsäure wie Dimethylformamid, cyclisiert diese Tetralon-Verbindung, z. B. nach den unten beschriebenen Verfahren, zu den 1,2,6,7-Tetrahydro benz[cdl)indol-2-on-Verbindungen der Formel VI, worin X", y" und Z" obige Bedeutung besitzen, reduziert diese Verbindungen katalytisch, z. B. in Gegenwart eines Platinkatalysators in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.
B. in einem niederen Alkanol wie Äthanol, bei Raumtemperatur und Normaldruck, und reduziert das so erhaltene Lactam der Formel VII, worin X", y und Z" obige Bedeutung besitzen, zur entsprechenden Verbindung der Formel IIIa", beispielsweise mittels Diboran in einem unter den herrschenden Bedingungen inerten organischen Lösungsmittel, z. B. einem offenkettigen oder cyclischen Äther wie Tetrahydrofuran.
Die Cyclisierung der 8-Cyanotetralone zu den Verbindungen der Formel VI kann entweder direkt, z. B. durch kurzes Erhitzen in einer alkalischen Lösung oder via die Säure, hergestellt durch Verseifen der 8-Cyan-Verbindung, durch Erhitzen mit flüssigem Ammoniak im Autoklaven, durchgeführt werden.
Die Verbindungen der Formel IIIa' können auch direkt aus den entsprechenden 8-Cyanotetralonen durch katalytische Hydrierung unter alkalischen Bedingungen, z. B. durch Reduktion in Gegenwart von Raney-Nickel in einer konzentrierten Ammoniaklösung, hergestellt werden. Bei dieser Reaktion entstehen auch Verbindungen der Formel VII, wobei das Verhältnis der erhaltenen Reaktionsprodukte von den Reaktionsbedingungen abhängig ist. Die Verbindungen der Formel VII können nach bekannten Verfahren, z. B. durch fraktionierte Kristallisation, von den Verbindungen der Formel IIIa" getrennt werden.
3. 1 ,2,6,7,8,8a-Hexahydro-5-hydroxybenz[cd]indol und 1,2,6,7,8,8a-Hexahydro-3 ,4,5-trihydroxybenz[cd]indol kann man nach bekannten Methoden aus den entsprechenden Methyläthern, beispielsweise durch Erhitzen mit Jodwasserstoffsäure, erhalten.
4. 1,2,6,7 ,8,8a-Hexahydro-5-methylbenz[cd]indol kann man z. B. herstellen, indem man 1-Acetyl-5-amino-1,2,6,7,8,8ahexahydrobenz[cd]indol diazotiert, z. B. mittels Natriumnitrit in schwefelsaurer Lösung bei etwa 0 bis 5o, das entstandene Diazoniumsalz z. B. nach Sandmeyer zum 1-Acetyl-5-cyan 1,2,6,7,8,8a-hexahydrobenz[cd]indol umsetzt, die gebildete Cyanverbindung in das 1-Acetyl-5-formyl-1,2,6,7,8,8ahexahydrobenz[cd]indol überführt und diese Formyl-Verbindung z. B. nach Wolff-Kishner bzw. nach Modifikation und Verbesserungen dieses Verfahrens (z. B. Huang-Minlon-Verfahren) reduziert, wobei gleichzeitig die.Acetylgruppe hydrolytisch abgespalten wird.
Das 1-Acetyl-5-formyl-1,2,6,7,8,8a-hexahydrobenz[cd]- indol kann man z. B. erhalten, indem man die entsprechende Cyan-Verbindung mit Natriumhypophosphit und Raney-Nickel in einem Gemisch von Eisessig/Pyridin/Wasser versetzt und während längerer Zeit bei einer Temperatur von etwa 10 bis 500 rührt. Zur Aufarbeitung filtriert man vom Katalysator ab und dampft das Filtrat ein. Der Rückstand kann weiter aufgearbeitet werden, indem man ihn zwischen Wasser und einem damit nicht mischbaren, unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z. B. einem chlorierten aliphatischen Kohlenwasserstoff wie Methylenchlorid ausschüttelt und die vereinigten organischen Phasen eindampft.
Die so erhaltenen Verbindungen der Formel IIIa können auf übliche Weise isoliert und nach bekannten Methoden gereinigt werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Im nachfolgenden Beispiel, welches die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.

Beispiel 1,2,6,7,8,8a-Hexahydro-1- (2-imidazolin-2-ylmethyl)benz[cd]indol
Eine Mischung von 6,95 g t-Cyanomethyl-1,2,6,7,8,8a- hexahydrobenz[cd]indol, 2,58 g Äthylendiamin und 0,08 ml Schwefelkohlenstoff wird unter Stickstoffatmosphäre auf 1200 erwärmt. Nach 1 1/2-stündigem Erhitzen auf 1200 wird die schon bei ca. 90O geschmolzene Mischung wieder fest. Nach Abkühlen und Zugabe von 50 ml Eiswasser wird die Masse zerdrückt, abfiltriert und mit Wasser gewaschen. Man nimmt den Filterrückstand in wenig Äthanol auf und versetzt bis zur sauren Reaktion mit ätherischer Chlorwasserstofflösung. Die ausgefallenen Kristalle werden abfiltriert, mit Äther gewaschen, in heissem Äthanol gelöst und mit Aktivkohle behandelt. Nach Entfernen der Kohle durch Filtration setzt man Äther zu und lässt zur Kristallisation stehen.
Die Kristalle werden abfiltriert, mit Äther gewaschen und am Hochvakuum 20 Std. bei 1000 getrocknet; das erhaltene Dihydrochlorid der im Titel genannten Verbindung schmilzt bei 235o unter Zersetzung.
Das als Ausgangsmaterial verwendete 1-Cyanomethyl 1,2,6,7,8,8a-hexahydrobenz[cd]indol kann wie folgt hergestellt werden: In eine Lösung von 38,7 g 1,2,6,7,8,8a-hexahydro- benz[cd]indol in 200 ml Äthanol werden bei Raumtemperatur, unter Rühren und unter Stickstoffatmosphäre, 49,4 g Natriumcarbonat eingetragen. Hierauf lässt man eine Lösung von 26,6 g Chloracetonitril in 126 ml Äthanol zutropfen. Die Mischung wird 5 1/2 Stunden am Rückfluss zum Sieden erhitzt und dann heiss filtriert. Das Filtrat lässt man zur Kristallisation stehen. Die ausgefallenen Kristalle werden abfiltriert, mit Äthanol und Wasser gewaschen und 20 Std. bei 70O und 12 mm Hg getrocknet; das 1-Cyanomethyl-1,2,6,7,8,8a-hexahydrobenz[cd]indol zeigt nach Kristallisation aus Äthanol einen Smp. von 1321350.

To prepare the previously unknown compounds of the formula II required as starting materials, which are also encompassed by the present invention, the compounds of the formula III, in which X, Y and Z are as defined above, with compounds of the formula IV in which A is halogen , preferably chlorine or bromine, in the presence of an acid-binding agent, for example a tertiary organic base such as triethylamine or an inorganic base, e.g. B. alkali metal carbonates such as potassium carbonate, sodium carbonate, etc., in an inert organic solvent under the prevailing conditions, e.g. B. an aromatic hydrocarbon such as benzene or a chlorinated alkane hydrocarbon such as chloroform or a lower alkanol.
The reaction is preferably carried out at an elevated temperature, in particular at the boiling point of the reaction mixture under reflux, and takes about 3 to 20 hours.
The compounds of formula IIIa required as starting material, in which either X 'is halogen, the hydroxy, methoxy or methyl group, Y and Z are hydrogen, or X', Y and Z 'are all methoxy group or X', Y ' and Z 'all represent the hydroxyl group, or X' and Z represent the methyl group and Y represent hydrogen, are new and can e.g. B. be manufactured as follows:
1. The compounds of the formula IIIa ', in which Hal is halogen, are obtained, for example, by nitrating 1,2,6,7,8,8a hexahydrobenz [cd] indole, e.g. B. in sulfuric acid solution by adding, with cooling, of fuming nitric acid and subsequent heating of the reaction mixture to about 90 for about 10 minutes, the nitro compound formed after N-acylation, z.
B. by means of acetic anhydride, reduced to 1-acetyl-5-amino-1,2,6,7,8,8a-hexahydrobenz [cd] indole, the amino compound obtained is diazotized, e.g. B. by means of sodium nitrite in sulfuric acid solution at about 0 to 5, the resulting diazonium salt converts according to Sandmeyer and the halogen compound thus obtained to the compounds of the formula purple 'deacetylated.
The reduction of 1-acetyl-1,2,6,7,8,8a-hexahydro-5-nitrobenz [cd] indole is carried out, for. B. catalytically, for example in the presence of a palladium or nickel catalyst in an inert organic solvent under the reaction conditions, e.g. B. a lower alkanol such as methanol. According to a variant, this reduction is z. B. carried out with the aid of hydrazine hydrate in the presence of Raney nickel.
After the reaction has ended, the catalyst is filtered off and gently evaporated to dryness. The amino compound formed can by known methods, for. B. by crystallization from an inert organic solvent under the prevailing conditions.
The deacetylation is carried out by hydrolysis, e.g. B. by heating in a mixture of glacial acetic acid / concentrated hydrochloric acid / water, by boiling with aqueous-methanolic sodium hydroxide solution, etc. carried out.
The compounds of the formula "Lila" can also be prepared by reacting 1-acetyl-5-amino-1,2,6,7,8,8a-hexahydrobenz [cd] indole with a solution of a complex copper halide, prepared by introducing nitrogen monoxide in a solution of copper (II) halide in an inert organic solvent under the prevailing conditions, such as acetonitrile, and subsequent deacetylation of the reaction product thus formed.
2. For the preparation of the compound of the formula IIIal 'in which either X "represents the methoxy group, Y' and 1 'represent hydrogen, or X", Y "and Z" all represent the methoxy group, or X "and Z" represent the Methyl group and Y "are hydrogen, brominated, for example, a compound of the formula V in which xi ', y" and Z "have the same meaning, for example with N-bromosuccinimide in an organic solvent which is inert under the reaction conditions, such as Acetonitrile, converts the 8-bromotetralone compound obtained to the corresponding 8-cyanotetralone, for example by boiling with copper cyanide in an organic solvent which is inert under the reaction conditions, e.g.

B. by the method described below, to the 1,2,6,7-tetrahydrobenz [cdl) indol-2-one compounds of the formula VI, in which X ", y" and Z "have the above meaning, these compounds are reduced catalytically, for example in the presence of a platinum catalyst in an organic solvent which is inert under the reaction conditions, e.g.
B. in a lower alkanol such as ethanol, at room temperature and normal pressure, and reduces the resulting lactam of formula VII, wherein X ", y and Z" have the above meaning, to the corresponding compound of formula IIIa ", for example by means of diborane in a below Organic solvents which are inert to the prevailing conditions, e.g. an open-chain or cyclic ether such as tetrahydrofuran.
The cyclization of the 8-cyanotetralones to the compounds of formula VI can either be carried out directly, e.g. B. by brief heating in an alkaline solution or via the acid, prepared by saponifying the 8-cyano compound, by heating with liquid ammonia in an autoclave.
The compounds of formula IIIa 'can also be obtained directly from the corresponding 8-cyanotetralones by catalytic hydrogenation under alkaline conditions, e.g. B. by reduction in the presence of Raney nickel in a concentrated ammonia solution. This reaction also gives compounds of the formula VII, the ratio of the reaction products obtained being dependent on the reaction conditions. The compounds of formula VII can be prepared by known methods, e.g. B. by fractional crystallization, are separated from the compounds of formula IIIa ".
3. 1, 2,6,7,8,8a-hexahydro-5-hydroxybenz [cd] indole and 1,2,6,7,8,8a-hexahydro-3, 4,5-trihydroxybenz [cd] indole can be obtained from the corresponding methyl ethers by known methods, for example by heating with hydriodic acid.
4. 1,2,6,7, 8,8a-hexahydro-5-methylbenz [cd] indole can e.g. B. prepared by diazotizing 1-acetyl-5-amino-1,2,6,7,8,8ahexahydrobenz [cd] indole, e.g. B. by means of sodium nitrite in sulfuric acid solution at about 0 to 5o, the resulting diazonium salt z. B. according to Sandmeyer to 1-acetyl-5-cyano 1,2,6,7,8,8a-hexahydrobenz [cd] indole, the cyano compound formed in the 1-acetyl-5-formyl-1,2,6, 7,8,8ahexahydrobenz [cd] indole and this formyl compound z. B. after Wolff-Kishner or after modification and improvements of this process (z. B. Huang-Minlon process) reduced, at the same time die.Acetylgruppe is hydrolytically split off.
The 1-acetyl-5-formyl-1,2,6,7,8,8a-hexahydrobenz [cd] - indole can be z. B. obtained by adding the corresponding cyano compound with sodium hypophosphite and Raney nickel in a mixture of glacial acetic acid / pyridine / water and stirring at a temperature of about 10 to 500 for a long time. For work-up, the catalyst is filtered off and the filtrate is evaporated. The residue can be worked up further by dividing it between water and an immiscible organic solvent which is inert under the present conditions, e.g. B. shakes out a chlorinated aliphatic hydrocarbon such as methylene chloride and evaporates the combined organic phases.
The compounds of the formula IIIa thus obtained can be isolated in a customary manner and purified by known methods.
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
In the following example, which explains the invention in more detail but is not intended to limit its scope in any way, all temperatures are given in degrees Celsius and are uncorrected.

Example 1,2,6,7,8,8a-Hexahydro-1- (2-imidazolin-2-ylmethyl) benz [cd] indole
A mixture of 6.95 g of t-cyanomethyl-1,2,6,7,8,8a-hexahydrobenz [cd] indole, 2.58 g of ethylenediamine and 0.08 ml of carbon disulfide is heated to 1200 under a nitrogen atmosphere. After 1 1/2 hours of heating to 1200, the mixture, which has already melted at about 90O, solidifies again. After cooling and adding 50 ml of ice water, the mass is crushed, filtered off and washed with water. The filter residue is taken up in a little ethanol and an ethereal hydrogen chloride solution is added until the reaction is acidic. The precipitated crystals are filtered off, washed with ether, dissolved in hot ethanol and treated with activated charcoal. After removing the charcoal by filtration, ether is added and the mixture is left to crystallize.
The crystals are filtered off, washed with ether and dried in a high vacuum for 20 hours at 1000; the resulting dihydrochloride of the compound mentioned in the title melts at 235 ° with decomposition.
The 1-cyanomethyl 1,2,6,7,8,8a-hexahydrobenz [cd] indole used as starting material can be prepared as follows: In a solution of 38.7 g 1,2,6,7,8,8a- hexahydrobenz [cd] indole in 200 ml of ethanol are added 49.4 g of sodium carbonate at room temperature, with stirring and under a nitrogen atmosphere. A solution of 26.6 g of chloroacetonitrile in 126 ml of ethanol is then added dropwise. The mixture is refluxed for 5 1/2 hours and then filtered hot. The filtrate is left to crystallize. The precipitated crystals are filtered off, washed with ethanol and water and dried for 20 hours at 70O and 12 mm Hg; 1-cyanomethyl-1,2,6,7,8,8a-hexahydrobenz [cd] indole has a melting point of 1321350 after crystallization from ethanol.
