Es wurde gefunden, dass neue 2-Androsten-17-äther der allgemeinen Formel I

R1 und R2 gleich oder verschieden sein können und Alkyl mit 1 - 3 C-Atomen oder zusammen mit dem Stickstoffatom einen Pyrrolidin-, Piperidin-, Hexamethylenimin-, Piperazin- oder Morpholinrest bedeuten und n 2, 3 oder 4 ist und deren physiologisch unbedenkliche Säureadditionssalze bei guter Verträglichkeit hervorragende cholesterin spiegel senkende, östrogene, anti-östrogene und Antiferti litäts-Wirksamkeit sowie gute fungistatische bzw. fungizide (antimykotische) und bakteriostatische bzw. bakterizide Eigenschaften aufweisen.
So senkte z.B. eine orale Gabe von 10 mg/kg des Hydrochlorids des 17ss-(2-Diäthylaminoäthoxy)-2-androstens bei Ratten (Methodik vgl. Counsell et al., J. Med. Pharm.
Chem. 5, 720, 1224 (1962) den Cholesterinspiegel im Serum um 29%. Die gleiche Substanz wirkte z.B. fungistatisch bzw. fungizid gegen nachstehende Mikroorganis- men: Aspergillus fumigatus, Aspergillus niger, Candida albicans, Candida parapsilosis, Epidermophyton floccosum, Geotrichon, Histoplasma capsulatum, Microsporum audouini, Microsporum candidum, Mucor, Nocardia asteroides, Penicillium, Trichophyton mentagrophytes, Trichophyton rubrum.
Die neuen Verbindungen können daher als Arzneimittel, Fungistatica bzw. Fungizide, Bakteriostatica bzw.
Bakterizide und auch als Zwischenprodukte zur Herstellung anderer nützlicher Verbindungen verwendet werden.
Besonders bevorzugt sind Verbindungen der nachstehenden ausgewählten Formeln Ia, Ib und Ic sowie deren physiologisch unbedenkliche Säureadditionssalze:

Z eine gegebenenfalls funktionell abgewandelte OH Gruppe oder Hal bedeutet und n, R1 und R3 die oben angegebene Bedeutung haben, mit HZ abspaltenden Mitteln behandelt.
Eine erhaltene Base der Formel I kann durch Behandeln mit einer Säure in ein physiologisch unbedenkliches Säureadditionssalz überführt oder eine Base der Formel I kann aus einem erhaltenen Säureadditionssalz durch Behandeln mit einer Base in Freiheit gesetzt werden.
In den Verbindungen der Formeln I und II bedeutet die Gruppe -ICnH2a- bevorzugt -(CH2)n-, im einzelnen -CH2CH3-, -CH2CH2CH2- oder -CH2CH2CH2CH2-. Sie kann aber auch -CH(CH3)-, -CH(CH3)CH2-, -CH2CH(CH3)-, -CH(C2H5)-, -CH(CH3)CH2CH3-, -CH2CH(CH3)CH2-, -CH2CH2CH(CH3)-, -CH{CeH)CH2-, -CH2CH(C2Hs)-, -CH(n-C3H,)- oder -CH(iso-C3H7)- bedeuten.
In den Verbindungen der Formel II kann Z ausser Hal und freien OH-(iruppen auch funktionell abgewandelte OH;Gruppen bedeuten. Als solche sind in erster Linie veresterte, insbesondere reaktionsfähig veresterte, OH-Gruppen zu nennen, z:B. mit einer Sulfonsäure (z.B.
mit 1 - 10 C-Atomen, wie Methan-, Äthan-, Benzol-, p -Toluol- oder Naphthalinsulfonsäure) oder Carbonsäure (z.B. mit 1 - 18, vorzugsweise 1 - 7 C-Atomen, z.B. einer Fettsäure, wie Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Valeriansäure, Isovaleriansäure, Trimethylessigsäure, Capronsäure, Isocapronsäure, Önanthsäure, Caprylsäure, Pelargonsäure, Caprinsäure, Laurinsäure, Myristinsäure, Palmitinsäure, Stearinsäure, ferner z.B. Crotonsäure, Ölsäure, Cyclohexancarbonsäure, Cyclohexylessig- und -propionsäure, Benzoesäure, Phenylessigsäure- und -propionsäure, Picolinsäure, Nicotinsäure, Isonicotinsäure oder Furan-2-carbonsäure) veresterte OH-Gruppen.
In den Verbindungen der Formel II kann der Substituent Z in a- oder iP-;Stellung stehen; Ausgangsverbindungen der Formel II, in denen Z in p-Stellung steht, sind bevorzugt, da sie präparativ leichter zugänglich sind.
Die als Ausgangsmaterial verwendbaren Steroide der Formel II können in 3-Stellung entweder ein Ghlor-, Brom- oder Jodatom oder eine freie oder funktionell abgewandelte Hydroxygruppe besitzen. Die Hydroxygruppe kann beispielsweise als Estergruppe oder als leicht spaltbare Äthergruppe (acetalartige Äthergruppe), z.B. als Tetrahydropyranyläthergruppe, vorliegen.
Unter den Estern sind die Sulfonsäureester der zugrundeliegenden 17-substituierten Androstan-3-ole (in denen der Sulfonsäurerest vorzugsweise 1 - 10 C-Atome besitzt), z.B. die Methansulfonate, Äthansulfonate, Benzolsulfonate und besonders die p-Toluolsulfonate, bevorzugt; aber auch Car- bonsäureester (in denen der Carbonsäurerest vorzugsweise 1 - 18, insbesondere 1 - 7, C-Atome besitzt), z.B. die Formiate, Acetate, Propionate oder Benzoate, ferner Xanthogenate und Carbonate sind für die iEliminierungs- reaktion geeignet.
Die Einführung einer 2(3}ständigen Doppelbindung unter Abspaltung von HZ gelingt bei Temperaturen zwischen 0 und 3000, vorzugsweise zwischen 20 und 2000.
Obwohl die Abspaltung schon durch einfaches Erhitzen (z.B. der 3-Xanthogenate, 3-Carbonate oder 3-Benzoate) bewirkt werden kann, ist es zweckmässig, Reagenzien zuzu setzen, die die Abspaltung begünstigen. Als solche eignen sich, wenn man die Alkohole II (Z = OH) als Ausgangsstoffe verwendet, Dehydratisierungsmittel, z.B. Säuren bzw. Lewis-Säuren, wie Schwefelsäure, Polyphosphorsäure, P205, Oxalsäure, Trifluoressigsäure, p-Toluolsulfonsäure, Benzolsulfonsäure, ferner Dimethylsulfoxid, Kaliumhydrogensulfat oder Jod.
Verwendet man Ester II (Z = veresterter OH) oder Halogenide II (Z = Hal) als Ausgangsstoffe, so ist es vorteilhaft, zur Bindung der abgespaltenen Säure HZ eine Base zuzusetzen, z.B. Pyridin, Kollidin, Lutidin, Chinolin, N,N-Dimethylanilin, Triäthylamin, NaOH oder KOH. Auch neutral reagierende Salze, wie LiCl, LiBr, eignen sich als Katalysatoren für die Abspaltung. Die Umsetzung kann mit oder ohne Gegenwart eines zusätzlichen Lösungsmittels erfolgen; als Lösungsmittel dienen z.B. Kohlenwasserstoffe, wie Benzol, Toluol, Xylol; halogenierte Kohlenwasserstoffe, wie Chloroform, Tetrachlorkohlenstoff, 1,2-Dichloräthan, Chlorbenzol; Sulfoxide, wie Dimethylsulfoxid; Amide, wie Dimethylformamid.
Auch ein Überschuss des Eliminierungsmittels, z.B. der zugesetzten Base, kann als Lösungsmittel dienen.
Eine besonders bevorzugte Ausführungsform ist das Erhitzen der 3p-p-Toluolsulfonate II (Z = -p-CTH7SO2- -0-) mit überschüssigem Kollidin; diese Reaktion benötigt etwa 1 bis 12 Stunden zum Abschluss.
Ferner ist es möglich, eine Base der Formel I mit einer Säure in das zugehörige Säureadditionssalz überzuführen. Für diese Umsetzung kommen solche Säuren in Frage, die physiologisch unbedenkliche Salze liefern. So eignen sich organische und anorganische Säuren, z.B. aliphatische, alicyclische, araliphatische, aromatische oder heterocyclische ein- oder mehrbasige Garbon- oder Sulfonsäuren, wie Ameisensäure, Essigsäure, Propionsäure, Pivalinsäure, Diäthylessigsäure, Oxalsäure, Malonsäure, Bernsteinsäure, Pimelinsäure, Fumarsäure, Maleinsäure, Milchsäure, Weinsäure, Äpfelsäure, Aminocarbonsäure, Sulfaminsäure, Benzoesäure, Salicylsäure, Phenylpropionsäure, Citronensäure, Gluconsäure, Ascorbinsäure, Nicotinsäure, Isonicotinsäure, Methansulfonsäure,
Äthansulfonsäure, +p-Hydroxyäthansulfonsäure p-Toluolsulfon- säure, Naphthalinmono- und -disulfonsäuren, Schwefelsäure, Salpetersäure, Halogenwasserstoffsäure oder Bromwasserstoffsäure, oder Phosphorsäuren, wie Orthophosphorsäure.
Schliesslich kann man eine Base der Formel I aus einem ihrer Säureadditionssalze in Freiheit setzen, indem man dasselbe, vorteilhaft in wässeriger Lösung, mit einer starken Base, wie NaOH oder KOH, behandelt.
Die neuen Verbindungen können im Gemisch mit festen, flüssigen und/oder halbflüssigen Arzneimittelträgern in der Human- oder Veterinärmedizin eingesetzt werden.
Als Trägersubstanzen kommen solche organischen oder anorganischen Stoffe in Frage, die für die parenterale, enterale oder topische Applikation geeignet sind und die mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Polyäthylenglykole, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, Vaseline, Cholesterin. Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise ölige oder wässerige Lösungen, sowie Suspensionen, Emulsionen oder Implantate. Für die enterale Applikation eignen sich Tabletten, Dragees, Lösungen, Sirupe und Säfte, für die topische Anwendung Salben, Cremes oder Puder.
Die angegebenen Zubereitungen können gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Konservierungs-, Stabilisierungs- oder Netzmitteln, Salzen zur Beeinflussung des osmotischen Druckes, Puffersubstanzen, Farb-, Geschmacks- und/oder Aromastoffen versetzt sein.
Zweckmässig wendet man die neuen Verbindungen oral in einer Einzeldosierung von 2 bis 300 mg an.
Beispiel I
15,2 g 17ss-(2-Diäthylaminoäthoxy)-androstan-3ss-ol-3 -p-toluolsulfonat (erhältlich durch Umsetzung von An drostan-3lp,17p-diol-3-tetrahydropyranyläther mit NaH und 2-Diäthylaminoäthylchlorid zu 17p-(2-Diäthylamino- äthoxy)-androstan-3l-ol-3-tetrahydropyranyläther, Spaltung mit Schwefelsäure zu 1 7- (2-Diäthylaminoäthoxy) - -androstan-3lp-ol und Veresterung mit p-Toluolsulfonsäurechlorid in Pyridin) werden in 300 ml Kollidin 5 Stunden gekocht. Anschliessend wird das Kollidin unter vermindertem Druck abdestilliert, der Rückstand in Äther aufgenommen und mit 2N Natronlauge ausgeschüttelt.
Danach wird die Ätherphase mit 100 ml mit Chlorwasserstoff gesättigtem Äther versetzt. Man erhält 17p-(2-Di- äthylamginoäthoxy)-2-androsten-hydrochlorid, F 23 5-23 60 (aus Aceton).
Analog erhält man aus den 3-p-Toluolsulfonsäureestern von 17ss-(2-Dimethylaminoäthoxy)-androstan-3ss-ol 17ss-(2-Di-n-propylaminoäthoxy)-androstan-3ss-ol 17ss-(2-Diisopropylaminoäthoxy)-androstan-3ss-ol 17ss-(2-Pyrrolidinoäthoxy)-androstan-3ss-ol 17ss-(2-Piperidinoäthoxy)-androstan-3ss-ol 17ss-(2-Hexamethyleniminoäthoxy)-androstan-3ss-ol 17ss-(2-Morpholinoäthoxy)-androstan-3ss-ol 17ss-(2-Piperazinoäthoxy)-androstan-3ss-ol 17ss-(3-Dimethylaminopropoxy)-androstan-3ss-ol 17ss-(3-Diäthylaminopropoxy)-androstan-3ss-ol 17ss-(3-Di-n-propylaminopropoxy)-androstan-3ss-ol 17ss-(3-Diisopropylaminopropoxy)-androstan-3ss-ol 17ss-(3-Pyrrolidinopropoxy)-androstan-3ss-ol 17ss-(3-Piperidinopropoxy)-androstan-3ss-ol 17ss-(3-Hexamethyleniminopropoxy)-androstan-3ss-ol 17ss-(3-Piperazinopropoxy)-androstan-3ss-ol 17ss-(1-Methyl-2-dimethylaminoäthoxy)-androstan-3ss-ol 17ss-(1-Methyl-2-diäthylaminoäthoxy)-androstan-3ss-ol

An Stelle der p-Toluolsulfonate können auch die entsprechenden 2-Chlor-, 2-Brom- oder 2-Jodverbindun- gen oder die entsprechenden Methan- oder Benzolsulfonate verwendet werden.
Beispiel 2
0,8 ml einer einmolaren Lösung von Kalium-tert.-butylat werden zu einer Lösung von 410 mg 3α-Chlor-17ss- -(2-diäthylaminoäthoxy)-androstan (erhältlich aus 17ss-(2 -Diäthylaminoäthoxy)-androstan-3ss-ol-tosylat und KCl in Tetrahydrofuran) in 20 ml tert.-Butanol hinzugegeben.
Man kocht 16 Stunden, arbeitet auf und chromatographiert das Rohprodukt an basischem Al2O3. Mit Benzol wird 17ss-(2-Diäthylaminoäthoxy)-2-androsten eluiert.

Beispiel 3
Eine Lösung von 1 g des Methansulfonats vÏn 17ss- -(2-Diäthylaminoäthoxy)-androstan-3ss-ol, 1 g Kaliumacetat und 20 ml Essigsäure wird 9 Stunden gekocht und dann zur Trockne eingedampft. Man extrahiert mit Äther, trocknet die ätherische Lösung über Natriumsulfat, dampft ein und kocht den Rückstand mit 20 ml 5%iger äthanolischer KOH 2 Stunden.
Das nach der Aufarbeitung erhaltene Rohprodukt wird an basischem A1208 chromatographiert, wobei man als Hauptprodukt 17ss-(2-Diäthylaminoäthoxy)-2-androsten mit Benzol eluiert: Hydrochlorid, F. 235 - 236 . Daneben erhält man eine geringe Menge 17ss-(2-Diäthylaminoäthoxy)-androstan-3α
Beispiel 4
1 g des Benzolsulfonats von 17p-(2-Diäthylamino- äthoxy)androstan-33-ol werden in 30 ml Benzol gelöst und an 100 g basischem A1203 adsorbiert. Man gibt noch 1 ml Benzol auf die Säule und lässt sie verschTossen 18 Stunden lang bei Rauntemperatur stehen.
Elution mit 300 ml Benzol gibt als Hauptprodukt 17p-(2-Diäthylami- noäthoxy)-2-androsten. Deneben entsteht etwas 17ss-(2 -Diäthylaminoäthoxy)-androstan-3α-ol und 17ss-(2-Di äthylaminoäthoxy)-androstan-3ss-ol
Beispiel S
100 mg 17ss-(2-Diäthylaminoäthoxy)-androstan-3ss-ol werden mit 1 ml Pyridin und 0,11 ml POCL3 über Nacht bei Raumtemperatur stehengelassen. Man zersetzt unter Kühlung mit Wasser, extrahiert mit Äther, trennt ab, wäscht die Ätherschicht mit Wasser, trocknet und dampft ein. Das erhaltene rohe 17ss-(2-Diäthylamino-äthoxy)-2 -androsten wird durch Chromatographie an basischem Al203 gereinigt. Hydrochlorid, F. 235 - 236 .
Analog erhält man aus den in Beispiel 16 angegebenen 17l#-substituierten Androstan-3p-olen die entsprechenden 17p-substituierten 2-Androstene.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von basischen 2-Andro- sten-17-äthern der allgemeinen Formel I EMI3.1 worin RÚ und R2 gleich oder verschieden sein können und Alkyl mit 1 - 3 C-Atomen oder zusammen mit dem Stick stoffatom einen Pyrrolidin-, Piperidin-, Hexamethylen- imin-, Piperazin- oder Morpholinrest bedeuten und n 2, 3 oder 4 ist und deren physiologisch unbedenklichen Säureadditionssalzen, dadurch gekennzeichnet, dass man eine Verbin dung der Formel II, EMI4.1 worin Z eine gegebenenfalls funktionell abgewandelte OH Gruppe oder Hal bedeutet und n, Rl und R2 die oben angegebene Bedeutung haben,mit HZ abspaltenden Mitteln behandelt UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekenn- zeichnet, dass man eine erhaltene Base der Formel I durch Behandeln mit einer Säure in ein physiologisch unbedenkliches Säureadditionssalz der Formel I überführt.2. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man ein erhaltenes Säureadditionssalz der Formel I durch Behandeln mit einer Base in die freie Base der Formel I überführt.
