Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Amine der Formel EMI1.1 worin Alk1 Alkyl mit bis zu 7 C-Atomen, alk2 Alkylen mit bis zu 3 C-Atomen, R Alkyl mit bis zu 7 C-Atomen oder Arylalkyl mit bis zu 12 C-Atomen und R1 Wasserstoff, Halogen, Trifluormethyl, Alkyl, Alkenyl, Alkoxy, Alkenyloxy oder Alkanoyl mit jeweils bis zu 7 C-Atomen bedeutet, sowie deren Salzen. Alkyl Alkl hat bevorzugt bis zu 4 C-Atome, wie iso- oder n Propyl, gerades oder verzweigtes, in beliebiger Stellung gebunde- nes Butyl, besonders Äthyl und vor allem Methyl. Alkylen alkz ist verzweigtes oder vor allem geradkettiges Alkylen mit vorzugsweise mindestens 2 C-Atomen in der Alkylenkette, insbesondere mit 2 oder 3 C-Atomen in der Alkylenkette, wie Propylen-1,3 oder vor allem Äthylen-1,2. Die Substituenten R1 werden wie folgt näher definiert: Halogen ist z.B. Fluor, Brom und insbesondere Chlor, Alkyl hat insbesondere die für Alkl gegebene Bedeutung. Alkenyl hat vor allem 2 bis 4 C-Atome, wie Vinyl, Methallyl und besonders Allyl. Alkoxy ist insbesondere solches, worin der Alkylteil die für Alk gegebene Bedeutung hat, wie Äthoxy, iso- oder n-Propoxy und besonders Methoxy. Alkenyloxy hat insbesondere 3 oder 4 C Atome im Alkenylteil, wie Methallyloxy oder vor allem Allyloxy. Alkanoyl hat insbesondere bis zu 4 C-Atome, wie Butyryl, Propionyl oder vor allem Acetyl. Vorzugsweise steht R1 in ortho Stellung zum 2-Hydroxypropoxyrest. Vor allem ist R1 aber Wasserstoff. Alkyl R hat bevorzugt bis zu 4 C-Atome, und ist unverzweigt oder bevorzugt verzweigt, insbesondere am x-C-Atom verzweigt, und ist z.B. sek.-Butyl oder insbesondere tert.-Butyl oder vor allem Isopropyl. Arylalkyl R hat bevorzugt bis zu 10 C-Atome, und ist im Alkylteil unverzweigt oder bevorzugt verzweigt, insbesondere am x-C-Atom des Alkylteils verzweigt. Der Arylteil stellt insbesondere einen Phenylrest dar, der gegebenenfalls mehrfach oder insbesondere einfach durch Alkyl oder Alkoxy mit bis zu 7 C-Atomen, Halogen, oder Trifluormethyl substituiert ist, bevorzugt aber unsubstituiert ist. Beispiele für Arylalkyl R sind l-Methyl-3-phenylpropyl und insbesondere l-Methyl-2-phenyl- äthyl. Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So blockieren sie cardiale FRezeptoren, wie sich bei der Bestimmung des Antagonismus der Tachykardie nach 0,5 ,ug/kg i.v. d/l-Isoproterenolsulfat an der narkotisierten Katze bei intravenöser Gabe von 0,02 bis 2 mg/kg zeigen lässt, so blockieren sie vasculäre p-Rezeptoren, wie sich bei der Bestimmung des Antagonismus der Vasodilatation nach 0,5 ug/kg i.v. d/l-Isoproterenolsulfat an der narkotisierten Katze bei intravenöser Gabe von 3 und mehr mg/kg zeigen lässt, und so blockieren sie cardiale FRezeptoren, wie sich bei der Bestimmung der Tachykardie nach 0,005 ug/ml d/l-Isoproterenolsulfat am isolierten Meerschweinchenherzen in vitro bei einer Konzentration von 0,02 bis 2 pg/ml zeigen lässt. Die neuen Verbindungen können daher als cardioselektive Antagonisten von adrenergischen ssRezeptorenstimulantien, z.B. zur Behandlung von Arrhythmien und Angina Pectoris, verwendet werden. Sie können aber auch als wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere pharmazeutisch wirksamer Verbindungen verwendet werden. Von den Aminen der Formel I sind insbesondere solche hervorzuheben, worin Alkl obige Bedeutungen hat, alk2 Äthylen1,2 oder Propylen-1,3 ist, R x-verzweigtes Alkyl mit bis zu 4 Catomen oder Phenylalkyl mit & verzweigtem Alkylteil mit bis zu g C-Atomen ist, und R1 obige Bedeutungen hat. Besonders sind Amine der Formel I hervorzuheben, worin 91kl Alkyl mit bis zu 4 C-Atomen, insbesondere Methyl, ist, alk2 Propylen-1,3 oder insbesondere Äthylen-1,2 ist, R Phenylalkyl mit bis zu 10 C-Atomen und einem x-verzweigten Alkylteil, insbe ;ondere 1-Methyl-2-phenyläthyl, oder vor allem x-verzweigtes alkyl mit bis zu 4 C-Atomen, wie tert.-Butyl oder vor allem [sopropyl ist, und R1 Alkyl mit bis zu 4 C-Atomen, insbesondere Methyl, Alkenyl mit bis zu 4 C-Atomen, insbesondere Methallyl oder vor allem Allyl, Alkoxy mit bis zu 4 C-Atomen, insbesondere Methoxy, Alkenyloxy mit bis zu 4 C-Atomen, insbesondere Methallyloxy oder vor allem Allyloxy, Alkanoyl mit bis zu 4 C-Atomen, insbesondere Acetyl, Halogen, insbesondere Chlor, oder vor allem Wasserstoff ist. Besonders zu nennen sind das l-[4-(2-Methylthioäthoxy) phenoxyj-2-hydroxy-3-t-butylaminopropan, das 1-[3-Chlor-4-(2- methylthioäthoxy)-phenoxy]-2-hydroxy-3-isopropylaminopropan, das 1 -[2-Methyl-4-(2-methylthioäthoxy)-phenoxy]-2-hydroxy- 3-isopropylaminopropan, das 1-[4-(3-Methylthiopropoxy)- phenoxyj-2-hydroxy-3-isopropylaminopropan, das 1-[2-Allyl s(2-methylthioäthoxy)-phenoxy]-2-hydroxy-3-isopropylamino- propan, das 1-[4-(2-t-Butylthioäthoxy)-phenoxy]-2-hydroxy-3- isopropylaminopropan, das l-[4-(2-Äthylthioäthoxy)-phenoxy]- 2-hydroxy-3-isopropylaminopropan, das 1-[4-(1-Methyl-2-methyl- thioäthoxy)-phenoxyj-2-hydroxy-3-isopropylaminopropan und vor allem das 1 -[4-(2-Methylthioäthoxy)-phenoxyj-2-hydroxy-3- isopropylaminopropan, das cardiale ,SRezeptoren blockiert, wie sich bei der Bestimmung des Antagonismus der Tachykardie nach 0,5 Fg/kg i.v. d/l-lsoproterenolsulfat an der narkotisierten Katze bei intravenöser Gabe von 0,03 bis 1 mg/kg zeigen lässt, das vasculäre FRezeptoren blockiert, wie sich bei der Bestimmung des Antagonismus der Vasodilatation nach 0,5 ,ug/kg i.v. d/l Isoproterenolsulfat an der narkotisierten Katze bei intravenöser Gabe von 3 und mehr mg/kg zeigen lässt, und das cardiale P- Rezeptoren blockiert, wie sich bei der Bestimmung der Tachykardie nach 0,005 ug/ml d/l-Isoproterenolsulfat am isolierten Meerschweinchenherzen in vitro bei einer Konzentration von 0,03 bis 1 ,ug/ml zeigen lässt. Nach dem erfindungsgemässen Verfahren werden die neuen Verbindungen in an sich bekannter Weise erhalten, indem man eine Verbindung der Formel EMI1.2 mit einer Verbindung der Formel H2N-R (III) oder einem Salz davon, worin Alkyl, alk2, R und R1 obige Bedeutung haben, X1 für Hydroxy steht und Z eine reaktionsfähige veresterbe Hydroxygruppe bedeutet, oder X1 und Z zusammen Epoxy bedeuten, um setzt. Eine reaktionsfähige, veresterte Hydroxylgruppe ist insbesondere eine durch eine starke anorganische oder organische Säure, vor allem eine Halogenwasserstoffsäure, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Jodwasserstoffsäure, ferner Schwefelsäure oder eine starke organische Sulfonsäure, wie eine starke aromatische Sulfonsäure, beispielsweise Benzolsulfonsäure, 4-Brombenzolsulfonsäure oder 4-Toluolsulfonsäure, veresterte Hydroxylgruppe. So steht Z insbesondere für Chlor, Brom oder Jod. Diese Umsetzung wird in der üblichen Weise durchgeführt. Bei Verwendung eines reaktionsfähigen Esters als Ausgangsmaterial wird vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder mit einem Überschuss an Amin gearbeitet. Geeignete basische Kondensationsmittel sind z.B. Alkalihydroxyde, wie Natrium- oder Kaliumhydroxyd, Alkalicarbonate, wie Kaliumcarbonat, und Alkalialkoholate, wie Natriummethylat, Kaliumäthylat und Kaliumtertiärbutylat. Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. So können beispielsweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon, erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z.B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern. Andererseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind. Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure-; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin. Diese oder andere Salze der neuen Verbindungen, wie z.B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen frei macht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen. Die neuen Verbindungen können je nach der Wahl der Ausgangsstoffe und Arbeitsweisen, als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asymmetrische Kohlenstoffatome enthalten auch als Isomerengemische (Racematgemische) vorliegen. Erhaltene Isomerengemische (Racematgemische) können auf Grund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation. Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder durch Umsetzen mit einer, mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z.B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden Können, zerlegen. Besonders gebräuchliche optisch aktive Säuren sind z.B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Äpfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden. Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders erwähnten Gruppen von Endstoffen und besonders zu den speziell beschriebenen oder hervorgehobenen Endstoffen führen. Die Ausgangsstoffe sind bekannt oder können, falls sie neu sind, nach an sich bekannten Methoden erhalten werden. Die neuen Verbindungen können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem z.B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z.B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talg, planzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.B. als Tabletten, Dragees, Kapseln, Suppositorien, Salben, Crem oder in flüssiger Form als Lösungen (z.B. als Elixier oder Sirup), Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Präparate, welche auch in der Veterinärmedizin Verwendung finden können, werden nach üblichen Methoden gewonnen. Die folgenden Beispiele erläutern die Erfindung ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben. Beispiel 1: Eine Lösung von 20 g 1-[4-(2-Methylthioäthoxy)-phenoxy]- 2-hydroxy-3-chlorpropan und 30 g Isopropylamin in 50 ml Isopropanol werden 12 bis 14 Stunden unter Rückfluss erhitzt. Die flüchtigen Anteile werden hierauf bei reduzicrtem Druck abgedampft, der Eindampfrückstand in 300 ml Äthylacetat gelöst und mit je 50 ml 2-n. Salzsäure extrahiert. Die wässrige Phase wird abgetrennt und mit konzentrierter Natronlauge stark alkalisch gestellt. Das ausgefallene Öl wird mit etwa 200 ml Äthylacetat extrahiert und im Kugelrohr bei 150 bis 160"/0,04 Torr destilliert. Man erhält so das 1-[4-(2-Methylthioäthoxy)- phenoxy]-2-hydroxy-3-isopropylaminopropan der Formel EMI2.1 als fast farbloses Öl, das kristallin erstarrt. Sein hydrochlorid schmilzt bei 102 bis 103 (kristallisiert aus Butanon). Beispiel 2: Auf analoge Weise wie in Beispiel 1 erhält man unter Verwendung von 35 g t-Butylamin das 1 -[4-(2-Methylthioäthoxy)- phenoxyj-2-hydroxy-3-t-butylaminopropan der Formel EMI3.1 vom F. 62-63 (aus Äther-Petroläther). Sein hydrochlorid schmilzt bei 131-133 (aus Aceton-Äther). Das als Ausgangsmaterial benötigte l-[4(2-Methylthio- äthoxy)-phenoxy]-2-hydroxy-3-chlorpropan kann man auf folgende Weise erhalten: 2-(p-Hydroxyphenoxy)-tetrahydropyran wird analog Beispiel 9 mit 2-Methylthioäthylchlorid umgesetzt und in Äthanol mit verdünnter Salzsäure 2 bis 3 Stunden hydrolysiert. Das so erhaltene p-(2-Methylthioäthoxy)-phenol wird roh weiter verwendet. 20 g rohes p-(2Methylthioäthoxy)-phenol, 30 ml Epichlorhydrin und 0,2 ml Piperidin werden 1 Stunden unter Rückfluss erhitzt. Das überschüssige Epichlorhydrin wird hierauf bei 100 Badtemperatur und 10 Torr abdestilliert. Das so erhaltene rohe 1 -4(2-Methylthioäthoxy)-phenoxyj-2-hydroxy- 3-chlorpropan wird direkt weiter verwendet. Beispiel 3: 21 g rohes 1-[3-Chlor-4-(2-methylthioäthoxy)-phenoxy]- 2,3-epoxypropan, 21 g Isopropylamin und 100 ml Isopropanol werden 3 Stunden unter Rückfluss erhitzt. Das durch Aufarbeiten wie in Beispiel 1 erhaltene rohe 1-[3-Chlor-4-(2-methylthio äthoxy)-phenoxy]-2-hydroxy-3-isopropalaminopropan der Formel EMI3.2 ergibt durch Umsetzen mit der Halben äquivalenten Menge Fumarsäure ein Kristallines, neutrales Fumarat vom F. 149-150' (aus Isopropanol-Äther). Das Ausgangsmaterial kann auf folgende Weise hergestellt werden: 43,3 g Chlorhydrochinon, 33 g 2-Methylthioäthylchlorid und 42 g Pottasche werden in 400 ml Aceton 20 Stunden unter Rückfluss und Rühren erhitzt. Das Reaktionsgemisch wird abgeköhlt und filtriert. Nach dem Abdampfen des Lösungsmittels im Vakuum wird der Eindampfrückstand in 500 ml Äthylacetat aufgenommen und mit insgesamt 500 ml 2-n. Natronlauge portionenweise extrahiert. Die wässrige Phase wird unter Eiskühlung mit 6-n. Salzsäure angesäuert und das abgeschiedene Öl mit Äthylacetat extrahiert. Das Äthylacetat wird im Vakuum abgedampft und der Rückstand im Kugelrohr destilliert. Die bei 140C Badtemperatur und 0,01 Torr siedende Fraktion enthält das 3-Chlor-4-(2-methylthioäthoxy)-phenol. 21,5 g 3-Chlor-4-(2-methylthioäthoxy)-phenol, 15 g Epichlorhydrin und 11 g Kaliumcarbonat werden in 250 ml Aceton 16 bis 20 Stunden unter Rückfluss erhitzt. Die ungelösten Salze werden hierauf abfiltriert und das Filtrat im Vakuum eingedampft. Der Eindampfrückstand wird in 300 ml Äthylacetat aufgenommen und mit 100 ml kalter 2-n. Natronlauge gewaschen. Nach dem Trocknen und Eindampfen erhält man das rohe 1-[3 Chlor-4-(2-methylthioäthoxy)-phenoxy]-2,3-epoxypropan, welches ohne weitere Reinigung verwendet werden kann. Beispiel 4: 18,9 g 1,2-Epoxy-3-[2-methyl-4-(2-methylthioäthoxy)-phen- oxy]-propan, 20 ml Isopropylamin und 100 ml Isopropanol werden 3 Stunden unter Rückfluss erhitzt. Eine Aufarbeitung analog Beispiel 1 ergibt das 1-[2-Methyl-4-(2-methylthioäthoxy) phenoxy]-2-hydroxy-3-isopropylaminopropan der Formel EMI3.3 als Öl, welches mit der halben theoretischen Menge Fumarsäure das neutrale Fumarat vom F. 134135 (aus Methanol Äther) bildet. Das Ausgangsmaterial kann wie folgt hergestellt werden: 50 g Toluhydrochinon, 50 g 2-Methylthioäthylchlorid und 45 g Kaliumcarbonat werden in 600 ml Aceton unter Rückfluss erhitzt. Nach 18 nach 33 und nach 55 Stunden werden nochmals je 9 g 2-Methylthioäthylchlorid und 11 g Kaliumcarbonat zugegeben. Danach wird noch 24 Stunden erhitzt. Das Reaktionsgemisch wird analog Beispiel 7 aufgearbeitet und ergibt das 2 Methyl-4-(2-methylthioäthoxy)-phenol als Öl vom Sdp. 130J/0,01 Torr 24 g 2-Methyl-4-(2-methylthioäthoxy)-phenol, 250 ml Diäthylketon, 18 g Epichlorhydrin und 18 g Kaliumcarbonat werden analog Beispiel 7 umgesetzt und zu öligem 1 -[2-Methyl- 4-(2-methylthioäthoxy)-phenoxy]-2,3-epoxypropan aufgearbeitet. Dieses wird roh weiterverwendet. Beispiel 5: 12 g 1 -[4(3-Methylthiopropoxy)-phenoxy]-2,3-epoxypropan, 120 ml Isopropanol und 15 ml Isopropylamin werden 3 bis 4 Stunden unter Rückfluss erhitzt. Nach Abdampfen der flüchtigen Anteile im Vakuum erhält man das rohe 1-[4-(3 Methylthiopropoxy)-phenoxy]-2-hydroxy-3-isopropylaminopropan der Formel EMI3.4 welches analog Beispiel 1 isoliert und hierauf in sein Hydrochlorid übergeführt wird. Das Hydrochlorid schmilzt nach Umkristallisation aus Aceton-Äther bei 102-103 . Das als Ausgangsmaterial benötigte Epoxypropan kann man wie folgt erhalten: 18 g Kaliumcarbonat, 26,2 g 1,3-Dibrompropan und 25 g 2-(p Hydroxyphenoxy)-tetrahydropyran werden in 500 ml Aceton 20 Stunden unter Rückfluss erhitzt. Das so erhaltene 2-[4-(3 Brompropoxy)-phenoxy]-tetrahydropyran wird in 100 ml Äthanol mit einer Lösung von 42 g Natrium-methyl-mercaptid in 400 ml Äthanol 3 Stunden bei 0-5" stehen gelassen. Nach Zugabe von konzentrierter Salzsäure bis pH 1 wird das Reaktionsgemisch 2 bis 3 Stunden bei Raumtemperatur stehen gelassen, hierauf iin Vakuum eingedampft und der Rückstand zwischen 100 ml Wasser und 300 ml Äther verteilt. Die Ätherphase wird mit 2-n. Natronlauge erschöpfend extrahiert, die wässrige Phase wird wiederum sauer gestellt und das 4-(3 Methylthiopropoxy)-phenol mit Äther extrahiert. Es siedet bei 125-128"/0,01 Torr. 10 g 4-(3-Methylthiopropoxy)-phenol, 100 ml Aceton, 8 g Epichlorhydrin und 8 g Kaliumcarbonat werden 48 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen werden die unlöslichen Bestandteile abfiltriert und das Filtrat wird eingedampft. Man erhält so das rohe l-[4-(3-Methylthiopropoxy)-phenoxyj-2,3- epoxypropan, welches ohne weitere Reinigung weiter verwendet wird. Beispiel 6: Eine Lösung von 11,0 g 1-[4-(2-Methylthioäthoxy)-2-allyl- phenoxy]-2,3-epoxypropan und 28 g Isopropylamin in 200 ml Isopropanol wird 14 Stunden unter Rückfluss erhitzt. Die flüchtigen Anteile werden hierauf bei reduziertem Druck abgedampft, der Eindampfrückstand in Äthylacetat gelöst und mit je 25 ml 2-n. Salzsäure extrahiert. Die wässrige Phase wird abgetrennt und mit konzentrierter Natronlauge stark alkalisch gestellt. Das ausgefallene Öl wird mit Äthylacetat extrahiert, der Extrakt mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und eingedampft. Man erhält das 1-[2-Allyl-4-(2-methyl thioäthoxy)-phenoxy]-2-hydroxy-3-isopropylaminopropan der Formel EMI4.1 als fast farbloses Öl. Sein Hydrogenoxalat schmilzt bei 116-117 (kristallisiert aus Aceton). Das als Ausgangsstoff benötigte Espoxypropanderivat kann auf folgende Weise erhalten werden: 42,0 g (0,276 Mol) 2-Allylhydrochinon, gelöst in 300 ml Aceton, 39 g Kaliumcarbonat und 28 ml 2-Methylthioäthylchlorid werden 20 Stunden unter Rühren und Rückfluss erhitzt. Filtrieren und Eindampfen des Filtrats ergibt 67 g Eindampfgut als braunes Öl. Dieses wird in Äther gelöst und die Lösung viermal mit insgesamt 300 ml 2-n. Natronlauge extrahiert. Die alkalischen Extrakte werden durch Einleiten von Kohlendioxyd auf pH 9 gebracht und hierauf dreimal mit Äther extrahiert. Die ätherischen Extrakte werden mit Natriumsulfat getrocknet, filtriert, eingedampft und das Eindampfgut im Kugelrohr destilliert. Die Fraktion mit dem Siedeintervall 110-150'/0,05 Torr liefert nach Kristallisation aus Tetrachlorkohlenstoff das 4 (2-Methylthioäthoxy)-2-allylphenol vom F. 11 7- 119 . 13,0 g 4-(2-Methylthioäthoxy)-2-allylphenol, gelöst in 100 ml Diäthylketon, werden mit 8,4 g Kaliumcarbonat und 6 ml Epichlorhydrin 36 Stunden unter Rühren und Rückfluss erhitzt. Dann wird im Vakuum zur Trockne eingedampft und der Rückstand zwischen Wasser und Essigester verteilt. Die organische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und eingedampft. Man erhält das 1-[4-(2 Methylthioäthoxy)-2-allylphenoxy]-2,3-epoxypropan als gelbliches ö1. Beispiel 7: 12 g 1 -[4-(2-Methylthioäthoxy)-phenoxy]-2,3-epoxypropan, 7,5 g l-Methyl-3-phenylpropylamin und 100 ml Isopropanol werden 4 Stunden unter Rückfluss erhitzt. Nach Abdampfen der flüchtigen Anteile, zuletzt im Hochvakuum bei 130' i0,01 Torr, wird das Reaktionsgemisch in einem Gemisch von je 50 ml Äthylacetat und Äther aufgenommen, durch Hyflo klar filtriert und die Lösung mit 100 ml 2-n. Salzsäure geschüttelt. Dabei fällt das 1 -[4-(2-Methylthioäthoxy)-phenoxy]-2-hydroxy-3-k 1 -methyl- 3'-phenylpropyl)-amino]-propan als Hydrochlorid kristallin aus. Nach Umkristallisation aus Wasser unter Zusatz von Aktivkohle schmilzt es bei 154-156- (sintert ab 1482). Das als Ausgangsstoff verwendete Epoxyd kann auf folgende Weise hergestellt werden: Aus Hydrochinonmonobenzyläther und 2,3-Dihydropyran in Benzol wird unter Katalyse von p-Toluolsulfonsäure das 2-(4 Benzyloxyphenoxy)-tetrahydropyran hergestellt (Schmelzpunkt 67-69t nach Kristallisation aus Petroläther). Dieses wird unter Zusatz von Palladiumkohle-Katalysator debenzyliert und ergibt so das 2-(4-Hydroxyphenoxy)-tetrahydro- pyran vom Schmelzpunkt 90-93 nach Umkristallisation aus Äther/Petroläther. Umsetzung des letzteren Phenols mit der doppelten äquivalenten Menge an Methylthioäthylchlorid und Pottasche in Aceton während 3 Tagen bei Rückfluss-Temperatur und anschliessender saurer Hydrolyse führt zum 4-(2-Methylthioäthoxy)-phenol, einem Öl, das bei 120-130 /0,03 Torr siedet. Ein Gemisch von 142 g 4-(2-Methylthioäthoxy)-phenol 171 g Kaliumcarbonat, 115 g Epichlorhydrin wird während 40 Stunden unter Rühren zum Rückfluss erhitzt. Die dunkle Suspension wird abgekühlt, filtriert und das Filtrat im Vakuum eingedampft. Das verbleibende dunkelbraune Öl wird in 1500 ml Äther gelöst und zweimal mit je 500 ml 2-n. Natronlauge extrahiert. Die Ätherlösung wird hierauf mit je 500 ml Wasser dreimal gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Man erhält rohes, öliges 1-[4-(2-Methylthioäthoxy)-phenoxy]-2,3- epoxypropan, welches nach Umkristallisation aus Äther/Petrol äther bei 53-58 schmilzt.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung neuer Amine der Formel EMI5.1 worin Alkl Alkyl mit bis zu 7 C-Atomen, alk2 Alkylen mit bis zu 3 C-Atomen, R Alkyl mit bis zu 7 C-Atomen oder Arylalkyl mit bis zu 12 C-Atomen und R1 Wasserstoff, Halogen, Trifluormethyl, Alkyl, Alkenyl, Alkoxy, Alkenyloxy oder Alkanoyl mit jeweils bis zu 7 C-Atomen bedeutet, sowie deren Salzen dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI5.2 mit einer Verbindung der Formel H2N-R (III) oder einem Salz davon, worin Alkyl, alk2, R und R1 obige Bedeutung haben, X1 für Hydroxy steht, und Z eine reaktionsfähige veresterte Hydroxygruppe bedeutet, oder X1 und Z zusammen Epoxy bedeuten, umsetzt, und gegebenenfalls erhaltene freie Basen in ihre Salze oder erhaltene Salze in die freien Basen umwandelt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man Amine der Formel I herstellt, worin Alkl obige Bedeutung hat, alk2 Äthylen-1,2 oder Propylen-1,3 ist, R x-verzweigtes Alkyl mit bis zu 4 C-Atomen oder Phenylalkyl mit a-verzweigtem Alkyl mit bis zu 4 C-Atomen ist, und R1 obige Bedeutung hat.2. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man Amine der Formel I herstellt, worin Alkl Methyl, alk2 Propylen-1,3 oder Äthylen-1,2, R l-Methyl-2-phenyläthyl, tert. Butyl oder Isopropyl, und R1 Methyl, Methallyl, Allyl, Methoxy, Methallyloxy, Allyloxy, Acetyl, Chlor oder Wasserstoff bedeutet.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man das 1-[4-(2-Methylthioäthoxy)-phenoxy]-2-hydroxy-3-t- butylaminopropan, das 1-[3-Chlor-4-(2-methylthioäthoxy) phenoxy].2-hydroxy-3-isopropylaminopropan, das 1 -[2-Methyl- 4-(2-methylthioäffioxy)-phenoxy]-2-hydwxy-3-isopropylamino propan, das 1-[4-(3-Methylthiopropoxy)-phenoxy]-2-hydroxy- 3-isopropylaminopropan, das 1-[2-Allyl-4-(2-methylthioäthoxy)phenoxy]-2-hydroxy-3-isopropylaminopropan,das 1-[4-(2-t Butylthioäthoxy)-phenoxyj-2-hydroxy-3-isopropylaminopropan, das 1 -[(2-Äthyfthioäthoxy)phenoxy]-2-hydrnxy-3-isopropyl- aminopropan oder das 1-[4-(1-Methyl-2-methylthioäthoxy)-phen- oxy]-2-hydroxy-3-isopropylaminopropan herstellt.4. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man das 1-[4-(2-Methylthioäthoxy)-phenoxy]-2-hydroxy- 3-isopropylaminopropan herstellt.5. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die neuen Verbindungen in Form ihrer therapeutisch verwendbaren Salze herstellt.6. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man erhaltene Isomerengemische in die Isomeren und/oder erhaltene Racemate in die optischen Antipoden aufspaltet.
