Description
Die Erfindung betrifft die Herstellung von neuen mehrfach halogenierten Steroiden, und zwar von 9a,21-Dihalogen-llß, 17a-dihydroxy-6a-fluor-16ß-methyl-pregna-l,4-dien-3,20-dion-Verbindungen der Formel worin Xj Chlor oder Fluor, X2 Brom oder Chlor, R! Wasserstoff und R2 den Acylrest einer Carbonsäure darstellen, in der im Patentanspruch 1 definierten Weise.
Nachstehend mit «nieder» bezeichnete kohlenstoffhaltige 2c Verbindungen und Reste enthalten vorzugsweise bis und mit 7 Kohlenstoffatome.
Eine Acylgruppe Ac leitet sich vorzugsweise von den in der Steroidchemie gebräuchlichen Carbonsäuren z.B. mit höchstens 18 Kohlenstoffatomen, insbesondere von entsprechenden ali-25 phatischen, cycloaliphatischen, cycloaliphatisch-aliphatischen, aromatischen oder aliphatischen Carbonsäuren ab. Der Rest Ac ist insbesondere gegebenenfalls, z.B. durch gegebenenfalls veräthertes oder verestertes Hydroxy, wie Niederalkoxy, z.B. Methoxy oder Äthoxy, oder Phenoxy, oder Halogen, z.B. Chlor, 30 oder Carboxy substituiertes Niederalkanoyl, wie geradkettiges oder verzweigtes Niederalkanoyl, z.B. Acetyl, Propionyl, Buty-ryl, Isobutyryl, Valeryl, Isovaleryl, Pivaloyl, 2-Äthyl-butyryl, 2,2- oder 3,3-Dimethyl-butyryl, Hexanol oder Heptanoyl, Hydroxyniederalkanoyl, z.B. 3-Hydroxypropionyl, Phenoxynie-35 deralkynoyl, z.B. Phenoxyacetyl, Halogenniederalkanoyl, z.B. Chloracetyl, oder Carboxyniederalkanoyl, z.B. 3-Carboxy-pro-pionyl oder 4-Carboxy-butyryl, Alkenoyl, z.B. Undecylenoyl, Cycloalkylniederalkanoyl, z.B. Cyclopentylpropionyl oder Cy-clohexylacetyl, gegebenenfalls substituiertes Benzoyl, z.B. Ben-40 zoyl, oder gegebenenfalls substituiertes Phenylniederalkanoyl oder -niederalkenoyl, z.B. Phenylacetyl, sowie ein Carboxyniederalkanoyl in Form eines Salzes, in erster Linie eines Alkali-metall-, wie Natrium- oder Kaliumsalzes. Dabei ist das Natriumsalz einer Verbindung der Formel I besonders bevorzugt, 45 worin R2 für 3-Carboxypropionyl steht.
Die erfindungsgemässen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So zeichnen sie sich durch eine hervorragende antiinflammatorische Wirksamkeit bei einer verminderten systemischen Wirkung aus. So z.B. bei lokaler 50 Anwendung im Rohwattegranulomtest an der Ratte kann man die Hemmung der entzündlichen Vorgänge im Dosisbereich von etwa 0,001 bis etwa 0,10 mg/Pellet feststellen, wobei aber die ersten Anzeichen einer systemischen Wirkung erst an oder sogar oberhalb der oberen Grenze dieses Dosisbereichs bemerk-55 bar sind, und zwar die Gewichtsabnahme des Thymus oberhalb 0,03 mg/Pellet und des gesamten Körpers erst bei 1,0 mg/Pellet. Bei der topischen Verabreichung (Rattenohr-Test nach To-nelli) liegt ED50 sogar bei etwa 10 — 30 mcg/ml. - Wegen diesen biologischen Eigenschaften sind die neuen Verbindungen in al-60 len Indikationen, für die sich Glucocorticoid-Steroide mit entzündungshemmenden Eigenschaften eignen, insbesondere jedoch als topisch anzuwendende antiinflammatorische Gluco corticoïde, z.B. zur Behandlung von entzündlichen Dermatosen, wie Ekzemen, Dermatiden, oder partiell corticoidresistenten 65 Dermatosen, z.B. Psoriasis, verwendbar. Sie können zudem als wertvolle Zwischenprodukte zur Herstellung anderer nützlicher Stoffe, insbesondere anderer pharmakoligisch wirksamer Steroide, Anwendung finden.

Die Erfindung betrifft vorzugsweise diejenigen Verbindungen der Formel I, worin Xj Fluor oder Chlor, X2 Chlor, Rt Wasserstoff und R2 Niederalkanoyl, z.B. Propionyl, darstellen.
Die Erfindung betrifft vor allem insbesondere die in den Beispielen genannten Verbindungen der Formel I.
Erfindungsgemäss werden Verbindungen der Formel I in an sich bekannter Weise hergestellt, indem man eine 1,2-gesättigte Verbindung der Formel ch2x2

worin R2, X! und X2 die obgenannten Bedeutungen haben, dehydriert.
Die 1,2-Dehydrierung kann z.B. durch Behandeln mit einem geeigneten dehydrierenden Chinon, wie 2,3-Dichlor-5,6-dicyano-l,4-benzochinon, durchgeführt werden.
Die 1,2-Dehydrierung von Ausgangsstoffen der Formel VIII kann auch durch Behandeln mit Selendioxyd oder mikrobiologisch, z.B. mit geeigneten Mikroorganismen, wie Coryne-bacterium simplex oder Septomyxa affinis, erfolgen.
Die Ausgangsstoffe der Formel VIII können erhalten werden, indem man die 21-Hydroxygruppe einer 6a-Fluor-llß, 21-dihydroxy-16ß-methyl-9a-X1-17a-OR2-pregn-4-en-3,20-dion-Verbindung z.B. durch Behandeln mit einem geeigneten Sulfonsäurederivat, wie Methansulfonsäurechlorid, in eine 21-organische Sulfonyloxygruppe überführt und das Zwischenprodukt mit einem Lithiumhalogenid, z.B. Lithiumchlorid, behandelt.
Verfahrensgemäss erhältliche Verbindungen der Formel I mit salzbildenden Eigenschaften, d.h. Verbindungen, worin R2 einen Carboxy-niederalkanoylrest darstellt, können in an sich bekannter Weise durch Behandeln mit einer geeigneten Base, wie einem Alkalimetall-, wie Natrium- oder Kaliumhydroxid, -carbonat oder -hydrogencarbonat (wobei man vorzugsweise die äquivalente Menge der Base verwendet), üblicherweise in einem wässrigen oder wässrig-alkoholischen Medium, in ihre Salze überführt werden.
Umgekehrt kann man ein verfahrensgemäss erhältliches Salz einer Verbindung der Formel I mit salzbildenden Eigenschaften in die freie Verbindung überführen, z.B. ein entsprechendes Salz mit einer Säure, wie einer anorganischen Säure behandeln (z.B. durch Ansäuern eine Lösung des Salzes in einem wässrigen oder wässrig-organischen Medium auf einen pH-Wert von etwa 1-2 bringen.
Vorzugsweise setzt man in den obigen Verfahrensschritten diejenigen Ausgangsstoffe ein, die zu vorstehend als besonders bevorzugt beschriebenen Verbindungen führen.
Die gemäss der vorliegenden Erfindung hergestellten Endstoffe können als pharmazeutische Präparate enthaltend eine erfindungsgemässe Verbindung der Formel I als Wirkstoff, sowie zur Herstellung von solchen pharmazeutischen Präparaten, Verwendung finden.
Dabei kommen in erster Linie topisch anwendbare pharmazeutische Präparate, wie Cremen, Salben, Pasten, Schäume, Tinkturen und Lösungen, in Frage, die von etwa 0,005 % bis etwa 0,1 % des Wirkstoffs enthalten, ferner Präparate zur oralen Verabreichung, z.B. Tabletten, Dragées und Kapseln, und parenteralen Verabreichung. Die Herstellung solcher Präparate erfolgt in an sich bekannter Weise.
5 Die gemäss der vorliegenden Erfindung hergestellten neuen Verbindungen verwendet man vorzugsweise zur Behandlung von Entzündungen, in erster Linie als lokal anzuwendende antiinflammatorische Glucocorticoide, üblicherweise in Form von pharmazeutischen Präparaten, besonders in Form von topisch ioanwendbaren pharmazeutischen Präparaten.
Die nachfolgenden Beispiele illustrieren die oben beschriebene Erfindung; sie sollen jedoch diese in ihrem Umfang in keiner Weise einschränken, Temperaturen werden in Celsiusgraden angegeben.

Beispiel 1
Eine Lösung von 228 mg 21-Chlor-6a,9a-difluor-llß,-17 a-dihydroxy-16 ß-methyl-pregn-4-en-3,20-dion-17-pro-2Q pionat in 11,4 ml Dioxan wird nach Zugabe von 230 mg 2,3-Dichlor-5,6-dicyano-l,4-benzochinon (DDQ) während 20 Stunden unter Stickstoffatmosphäre zum Rückfluss erhitzt. Das Reaktionsgemisch wird im Wasserstrahlvakuum eingedampft und der amorphe Rückstand über eine 100-fache Gewichtsmen-25 ge Kieselgel (Stufensäule) chromatographiert. Die mit einem Gemisch von Methylenchlorid-Methanol (99:1) eluierten Fraktionen ergeben das reine 21-Chlor-6a,9a-difluor-llß,17a-dihydroxy-16ß-methyl-pregna-l,4-dien-3,20-dion-17-propio-nat, das nach Umlösen aus Methylenchlorid/Äther bei 220-30 221° schmilzt.
Das als Ausgangsstoff zu verwendende 17-Monopropionat wird wie folgt hergestellt:
Eine Suspension von 1,0 g 21-Chlor-9ß,llß-epoxy-17a-hydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propionatin 35 5,4 ml Dioxan und 1,4 ml Orthoameisensäuretriäthylester wird mit 0,9 ml einer Lösung, bestehend aus 277 mg p-Toluolsulfon-säure, 2,7 ml Dioxan und 0,55 ml Äthylalkohol, versetzt und 90 Minuten bei Zimmertemperatur gerührt. Nach Zugabe von 3,8 ml Pyridin wird das Reaktionsgemisch mit Äthylacetat ver-40 dünnt, dreimal mit gesättigter Natriumchlorid-Lösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Der so erhaltene rohe 3-Äthylenoläther (3-Äthoxy-21-chlor-9ß,llß-epoxy-17a-hydroxy-16ß-methyI-pregna-3,5-dien-20-on-17-propionat) wird in einem Gemisch von 50 ml Tetra-45 hydrofuran und 10 ml Wasser gelöst und während 40 Minuten bei Zimmertemperatur mit gasförmigem Perchlorylfluorid behandelt. Nachdem man anschliessend während 10 Minuten Stickstoff durchgeleitet hat, wird die Reaktionslösung mit Äthylacetat verdünnt, nacheinander mit 10% Kaliumjodid-Lö-50 sung, mit 10% Natriumthiosulfat-Lösung und dreimal mit gesättigter Natriumchlorid-Lösung gewaschen, getrocknet und im Wasserstrahlvakuum eingedampft. Das anfallende Rohprodukt wird über eine 100-fache Gewichtsmenge Kieselgel (Stufensäule) chromatographiert. Die mit einem Gemisch von Toluol-55 Äthylacetat (90:10) eluierten Fraktionen ergeben, aus Methylenchlorid/Äther umkristallisiert, das 21-Chlor-6a-fluor-9ß,llß-epoxy-17a-hydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propionat, Smp. 174-177°.
720 mg der letztgenannten Verbindung werden in einem 60 Kunststoffgefäss mit 14,5 ml eines Reagens, zubereitet durch Vermischen von 10 g Harnstoff mit 13,25 g wasserfreiem flüssigem Fluorwasserstoff, Übergossen und während 3 Stunden unter Eiskühlung gerührt. Das Reaktionsgemisch wird auf 55 ml eiskalte gesättigte Ammoniak-Lösung gegossen, mit Essigsäure 65 schwach angesäuert und zweimal mit Chloroform extrahiert. Die organischen Phasen werden vereinigt, mit eiskalter verdünnter Natronlauge gewaschen, getrocknet und am Wasserstrahlvakuum eingedampft. Das Rohprodukt wird über eine

100-fache Gewichtsmenge Kieselgel (Stufensäule) chromatographiert. Die mit einem Gemisch von Toluol-Essigester (90:10) eluierten Fraktionen ergeben, aus Methylenchlorid/ Äther umkristallisiert, das 21-Chlor-6a,9a-difluor-llß, 17 a-dihydroxy-16 ß-methyl-pregn-4-en-3,20-dion- 17-pro-pionat, Smp. 212-213°.
Beispiel 2
Eine Lösung von 350 mg 9a,21-Dichlor-6a-fluor-llß, 17-dihydroxy-16ß-methyl-pregn-4-en-3,20-dion-17-propio-nat und 525 mg 2,3-Dichlor-5,6-dicyan-l,4-benzochinon in 17,5 ml Dioxan wird 20 Stunden in einer Stickstoffatmosphäre unter Rückfluss gekocht. Das abgekühlte Gemisch wird ge-nutscht, das Filtrat eingedampft, in Methylenchlorid gelöst und durch die 15-fache Gewichtsmenge neutrales Aluminiumoxid (Aktivität II) filtriert. Das Filtrat wird eingeengt und durch preparative Dünnschichtchromatographie (Silicagel; ein 9:1-Ge-
misch von Toluol-Methanol als Laufmittel bei 3-maligem Lauf) gereinigt. Die gewünschte Zone wird durch UV-Licht von Wellenlänge 254 nm lokalisiert, mit Äthylacetat aus dem Adsorbens eluiert und aus Methylenchlorid-Äther umkristallisiert, womit s das gewünschte 9a,21-Dichlor-6a-fluor-llß,17-dihydroxy-16ß-methyl-pregna-1,4-dien-3,20-dion-17 -propionat, Smp. 212-213° (Zersetzung), resultiert.
Das Ausgangssteroid wird folgendermassen erhalten: In eine Lösung von 400 mg 21-Chlor-6a-fluor-9ß,ll-epoxy-17-iohydroxy-16ß-methyl-pregn-4-en-3,20-dion-propionatin 20 ml Chloroform wird während 30 Minuten bei 0° Chlorwasserstoffgas eingeleitet. Das Gemisch wird weitere 30 Minuten bei 0° stehen gelassen, mit Chloroform verdünnt, mit einer eiskalten gesättigten Natriumhydrogencarbonat-Lösung gewa-15 sehen, getrocknet und im Vakuum eingedampft. Das erhaltene rohe 9a,21-Dichlor-6a-fluor-llß,17-dihydroxy-16ß-me-thyl-pregn-4-en-3,20-dion-17-propionat kann ohne Reinigung in die Dehydrierungsreaktion eingesetzt werden.

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel VIII mit einem dehydrierenden Chinon behandelt.2PATENTANSPRÜCHE 1. - 3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass man einen Ausgangsstoff der Formel VIII mit 2,3-Dichlor-5,6-dicyan-l,4-benzochinon behandelt.
- 4. Verfahren gemäss einem der Ansprüche 1-3, dadurch gekennzeichnet, dass man Verbindungen der Formel I, worin Xj Fluor oder Chlor, X2 Chlor, Rj Wasserstoff und R2 Niederalk-anoyl mit 2-5 Kohlenstoffatomen bedeuten, herstellt.
- 5. Verfahren gemäss einem der Ansprüche 1-3, dadurch gekennzeichnet, dass man 9a,21-Dichlor-6a-fluor-llß,17 a-dihydroxy-16ß-methyl-pregna-1,4-dien-3,20-dion-17-propionat herstellt.
- 6. Verfahren gemäss einem der Ansprüche 1-3, dadurch gekennzeichnet, dass man 21-Chlor-6a,9ot-difluor-llß, 17a-dihydroxy-16ß-methyl-pregna-l,4-dien-3,20-dion-17-propionat herstellt.
